Evidence for the presence of both P1 and P2 purinoceptors in the rat myometrium.

1. Adenosine, adenosine triphosphate (ATP) and some stable analogues of adenosine inhibited field stimulation-induced contractions of the uterus from rats treated with oestradiol cypionate (20 micrograms/kg, s.c.) 1 day previously. Adenosine was twice as potent as ATP; both were potentiated by dipyridamole (10 mumol/L). 2. The order of agonist potency of adenosine and its analogues was: 5'-N-ethylcarboxamidoadenosine (NECA) > N6-cyclohexyladenosine > or = R-phenylisopropyladenosine = S-phenylisopropyladenosine = 2-chloroadenosine > or = adenosine > or = ATP > > 2-p-(2-carboxyethyl) phenethylam

Na+ amplifies adenosine receptor-mediated inhibition of adipocyte adenylate cyclase.

Membrane preparations of hamster adipocytes were used to study the inhibition of adenylate cyclase by the adenosine analogue, N6-phenylisopropyladenosine (PIA). Inhibition by PIA was half-maximal and maximal (about 40%) at about 0.03 and 1 micro M, respectively, and was antagonized by 3-isobutyl-1-methylxanthine in a competitive manner. Like other hormone receptor-mediated inhibitions of the adipocyte adenylate cyclase, the adenosine receptor-mediated action on the enzyme was absolutely dependent on GTP and was amplified by sodium ions.

[125I]Aminobenzyladenosine, a new radioligand with improved specific binding to adenosine receptors in heart.

The density of adenosine receptors in membranes derived from rat hearts in 25 times lower than the density of receptors in rat brain membranes. Consequently, adenosine radioligands which are useful in brain such as l-[3H]phenylisopropyladenosine, [3H]cyclohexyladenosine, [3H]-2-chloroadenosine and l-[125I]hydroxyphenylisopropyladenosine are of limited usefulness in heart, due to a high ratio of nonspecific to specific binding. We have synthesized a new radioligand, [125I]-N6-4-aminobenzyladenosine, which binds to rat heart membranes with one-sixth the nonspecific binding of the other radioliga

Effects of some adenosine analogs on morphine-induced analgesia and tolerance.

1. The analogs of adenosine D- and L-phenylisopropyladenosine (D- and L-PIA) and chloroadenosine (CADO) induced analgesia in mice (hot-plate test). 2. The antinociceptive effects of the three adenosine agonists were antagonized by caffeine but were unaffected by naloxone. 3. Morphine-induced antinociception was increased by pretreatment with adenosine agonists. 4. Whereas CADO significantly attenuated the induction of morphine tolerance, D- and L-PIA did not affect the process.

Characterization of the human peripheral lung adenosine receptor.

Because adenosine narrows asthmatic airways, is released during hypoxia and by mast cells, and is antagonized by theophylline, it may play a role in asthma. I characterized the first step in pulmonary responses to adenosine: its adenosine receptor. Plasma membranes, prepared from macroscopically normal human peripheral lung, were incubated with 10 nM 5'-N-ethylcarboxamido[3H]adenosine ([3H]NECA) and various concentrations of competing ligand under experimentally determined optimal conditions: 4 degrees C, pH 7.4, 5 mM MgCl2, 1.8 mg protein/ml, 30-min incubation time. Bound and free ligand were

Behavioral and cardiovascular effects of analogs of adenosine in cynomolgus monkeys.

The behavioral and cardiovascular effects of six metabolically stable analogs of adenosine were studied in cynomolgus monkeys. Behavioral effects were determined in monkeys trained to respond under a 30-response fixed-ratio (FR) schedule of food presentation; cardiovascular effects were evaluated in a separate group of unanaesthetized monkeys seated at rest. Dose-effect curves for each drug were determined by administering cumulative doses i.v. during sequential components of the experimental session. All six analogs of adenosine produced dose-related decreases in FR responding with the follow

Central effects of adenosine analogs on stress-induced gastric ulcer formation.

Rats subjected to restraint stress developed gastric lesions that could be reduced by R-phenylisopropyladenosine (R-PIA) administered intracerebroventricularly. This protective effect was reversed by 8-sulfophenyltheophylline given centrally, and by peripherally administered 8-phenyltheophylline. These results suggest that central adenosine receptors mediate the effect. In subsequent studies it was found that if the absolute level of ulcer formation in control rats was low, R-PIA had no ulcer protective effect. Thus, although it appears that adenosine receptors are important in attenuating pat

Effects of caffeine and theophylline on adenosine and benzodiazepine receptors in human brain.

The binding of various adenosine receptor ligands and of [3H]diazepam, as well as their inhibition of methylxanthines, have been studied in human brain cerebral cortex membranes. Caffeine and theophylline competitively inhibit binding of [3H]cyclohexyladenosine, [3H]diethylphenylxanthine, [3H]phenylisopropyladenosine and [3H]diazepam. Both caffeine and theophylline are more potent as inhibitors of adenosine receptor ligand binding compared to [3H]diazepam binding. Theophylline was more potent than caffeine in its ability to compete with adenosine receptor ligand binding while the reverse was t

Intrathecal and systemic R-phenylisopropyl-adenosine reduces scratching behaviour in a rat mononeuropathy model.

The aim of the present study was to investigate the effect of intravenous or intrathecal (i.t.) administration of R-phenylisopropyladenosine (R-PIA), a selective A1 adenosine receptor agonist, on spontaneous scratching behaviour, a phenomenon presumably related to pain in a mononeuropathy model (sciatic nerve ligation) in rats. The acute effect of daily i.t. R-PIA injections was studied up to 21 days following nerve ligation. The results demonstrate that both i.v. (30 nmol) and i.t. (3 nmol) R-PIA, in doses not producing any motor impairment, significantly reduces scratching behaviour in this

The influence of naloxone on the effects of adenosine receptor agonists in analgesic tests and binding studies.

Naloxone (1 mg/kg ip) reduced analgesic effect of R-phenylisopropyladenosine (R-PIA-0.2 mg/kg sc) in hot plate and tail-immersion tests in mice and in tail-immersion test in rats. Also the effect of 2-chloroadenosine (2-CADO-2 mg/kg sc) was significantly reduced by naloxone in mice in both nociceptive tests. Naloxone induced partial reduction of analgesic effects of 5'-N-ethylcarboxamideadenosine (NECA-0.02-0.05 mg/kg sc) in mice and rats. Binding studies revealed that the affinity of adenosine agonists (R-PIA and NECA) to opioid receptors was about 5000 times weaker than the corresponding aff

Adenosine analogs inhibit fighting in isolated male mice.

The potent adenosine analogs N-ethylcarboxamide adenosine (NECA) and phenylisopropyladenosine (PIA) inhibit fighting and associated agonistic behaviors in isolated male mice. These effects are reversed by methylxanthines; moderate doses of NECA which inhibit fighting have minimal effects on spontaneous locomotor activity. At very low doses, both NECA and PIA increase fighting in parallel with previously reported increases of motor activity. Brain levels of [3H]-NECA and [3H]-PIA achieved at behaviorally effective doses suggest an involvement of adenosine receptors. The biochemical mechanism of

Binding of adenosine and receptor-specific analogues to lymphocytes from control subjects and patients with common variable immunodeficiency.

Studies were performed on the binding of tritiated adenosine and its analogues, 5'-N-ethylcarboxamide adenosine (NECA) and N6-phenylisopropyladenosine (PIA), to human peripheral blood lymphocytes. These revealed binding only of adenosine (Kd, 1-10 microM, 14,000 binding sites/cell), which was abolished by dipyridamole, a specific adenosine transport inhibitor, suggesting that the binding is to the nucleoside transporter. The absence of high affinity (Kd less than or equal to 1 microM) binding of adenosine or of the two analogues. NECA and PIA suggests that the previously reported effects of ad

A subtype of adenosine receptors mediating pigment dispersion in leucophores of the medaka: evidence for an A2-receptor.

Beta adrenergic agonists and forskolin synergistically augmented the purine action, while Li+ blocked it competitively. 5. The results suggest that medaka leucophores p

The anticonvulsive action of adenosine: a postsynaptic, dendritic action by a possible endogenous anticonvulsant.

Neural afterdischarges generated in the presence of penicillin or low extracellular calcium concentrations were found to be inhibited by adenosine in the rat hippocampus in vitro. This anticonvulsant effect of adenosine is observed in the absence, as well as in the presence, of chemical synaptic transmission and apparently occurs at a postsynaptic site which is most sensitive in the apical dendritic region of the CA1 pyramidal cells. The methylxanthine theophylline antagonizes the effect of adenosine; and, the anticonvulsant action of the L-isomer of the adenosine analogue phenylisopropyladeno

Up-regulation of brain [3H]diazepam binding sites in chronic caffeine treated rats.

Brain [3H]diazepam and [3H]L-phenylisopropyladenosine binding sites in caffeine treated (75 mg/kg/day, i.p. 12 days) and caffeine withdrawn (30 days) rats were examined. Treatment with caffeine (75 mg/kg/day) for 12 days increases the Bmax (maximum binding capacity) for [3H]diazepam binding by 30.9% whereas the same treatment increases the Bmax for [3H]L-PIA binding by 165%. The Bmax for [3H]diazepam binding sites returns to slightly below control levels but [3H]L-PIA binding sites remain elevated after 30 days of caffeine withdrawal. These findings suggest that the up-regulation of [3H]diazep

Evidence for the action of endogenous adenosine in the rabbit retina: modulation of the light-evoked release of acetylcholine.

Much evidence has accumulated supporting the hypothesis that the purine nucleoside adenosine may indeed function as a neuromodulator in the mammalian retina, but to date no reports have directly illustrated a physiological role for this nucleoside. In other regions of the CNS, adenosine agonists decrease transmitter release, whereas antagonists increase release. A similar role for adenosine in the retina is now apparent. The cholinergic amacrine cells of the rabbit retina were labeled with [3H]choline, and the effects of enzymatic adenosine degradation or adenosine antagonists on the light-evo

Binding of [3H]KF17837S, a selective adenosine A2 receptor antagonist, to rat brain membranes.

The potential of 8-(3,4-dimethoxystyryl)-1,3-dipropyl-7-[3H] methylxanthine ([3H]Kf17837S) as a highly selective antagonist radioligand for the adenosine A2A receptor was examined and compared with the properties of the adenosine A2A receptor agonist radioligand 2-[p-(2-[3H]carboxyethyl)phenethylamino]-5'-N-ethyl- carboxamidoadenosine ([3H]CGS21680). Affinities of barbiturates for the GABA-receptor complex and A1 adenosine receptors: a possible explanation of their excitatory effects.

The effects of barbiturates on the GABA-receptor complex and the A1 adenosine receptor were studied. At the GABA-receptor complex the barbiturates inhibited the binding of [35S]t-butylbicyclophosphorothionate ([35S]TBPT) and enhanced the binding of [3H]diazepam. Kinetic and saturation experiments showed that both effects were allosteric. Whereas all barbiturates caused complete inhibition of [35S]TBPT binding, they showed varying degrees of maximal enhancement of [3H]diazepam binding; (+/-)methohexital was identified as the most efficacious compound for this enhancement. At the A1 adenosine re
