Measurement of Subcellular Metabolite Concentrations in Relation to Phloem Loading.

The key step of carbon export from green leaves is the loading of sugars into the phloem. To fully understand and quantify this process, it is essential to know the concentration of sugars in the different compartments of the cells along the phloem loading pathway. However, determining subcellular metabolite concentrations has been technically challenging. This paper describes a technique to measure metabolite levels in the chloroplast, the cytosol, and the vacuole of mesophyll cells with high accuracy. The nonaqueous fractionation (NAF) technique is arguably the method of choice to analyze th

Local and Systemic Effects of Brassinosteroid Perception in Developing Phloem.

The plant vasculature is an essential adaptation to terrestrial growth. Its phloem component permits efficient transfer of photosynthates between source and sink organs but also transports signals that systemically coordinate physiology and development. Here, we provide evidence that developing phloem orchestrates cellular behavior of adjacent tissues in the growth apices of plants, the meristems. Arabidopsis thaliana plants that lack the three receptor kinases BRASSINOSTEROID INSENSITIVE 1 (BRI1), BRI1-LIKE 1 (BRL1), and BRL3 ("bri3" mutants) can no longer sense brassinosteroid phytohormones

EDTA-Facilitated Collection of Phloem Exudates for Chemical Analyses.

Phloem exudates can be collected using several methods, including the ethylenediaminetetraacetic acid (EDTA)-facilitated method. Plant parts, such as leaves or inflorescences, are cut, and the cut edges are immediately placed into EDTA solution, which prevents plugging of sieve tubes. After this first inoculation, plant parts are transferred to water, into which phloem exudates are collected for a few hours. Inoculation and collection take place in the dark at high relative humidity. The samples can subsequently be used for different chemical analyses of RNA, proteins and various primary and s

Non-Invasive Determination of Phloem Transport Speed with 14C.

Studying the phloem, the tissue responsible for distributing organic substances between plant organs, presents significant challenges due to its location deep within the plant body and its sensitivity to manipulation. One effective approach to investigate phloem transport is the use of tracers to measure transport speed with a noninvasive approach. This chapter provides a detailed protocol for using 14C-labeled photoassimilates to study phloem transport velocity. A key advantage of this method is its noninvasive nature, as the isotope is supplied in the form of 14CO2, which is assimilated by s

Relative Quantification of Phloem Loading with 13CO2 in Grass Leaves.

A central challenge in plant physiology research is tracking the movement of photoassimilates, especially following their allocation via the phloem. To obtain better insight into movement of photoassimilates, we present a stable isotope-based approach to measure photoassimilate export from leaves. In this simple procedure, 13CO2 pulse label is supplied to whole leaves or defined sections of the leaves, sugars are extracted at two timepoints and the reduction of 13C -signal quantified. The method uses the stable 13C isotope of carbon, is safe to use, and has been successfully applied in both mo

Investigation of phloem transport of foliar-absorbed Cd in wheat.

The transport pathway of foliar absorbed Cd in wheat has not been clarified. Hydroponically raised wheat seedling stems were steam girdled to disrupt phloem transport, then the leaves were exposed to Cd solutions (40 or 60 mg L-1) containing 20 mg L-1 Rb (a phloem transport indicator) for 16 h. Plants were harvested 7 days later and divided into roots and leaves, while the stem was divided into two parts above and below the girdling location, with phloem sap collected from the upper part. Samples were analyzed for Cd, Rb, Cu, Zn and Mn concentrations; root glutathione concentration was also an

Role of the phloem in the biochemistry and ecophysiology of benzylisoquinoline alkaloid metabolism.

Benzylisoquinoline alkaloids (BIAs) are a diverse group of biologically active specialized metabolites produced mainly in four plant families. BIA metabolism is likely of monophyletic origin and involves multiple enzymes yielding structurally diverse compounds. Several BIAs possess defensive properties against pathogenic microorganisms and herbivores. Opium poppy (Papaver somniferum: Papaveraceae) has emerged as a model system to investigate the cellular localization of BIA biosynthesis. Although alkaloids accumulate in the laticifer cytoplasm (latex) of opium poppy, corresponding biosynthetic

Synthesis and phloem mobility of acidic derivatives of the fungicide fenpiclonil.

A series of derivatives of the phenylpyrrole fungicide fenpiclonil was synthesized in which a carboxyl group was present at various sites of this non-phloem-mobile molecule. Using the Kleier model, all these acidic analogues were predicted to be moderately phloem-mobile, especially the N-substituted derivatives. One of these latter molecules, N-carboxymethyl-3-cyano-4-(2,3-dichlorophenyl)pyrrole, exhibited some fungicidal activity on the pathogenic fungus Eutypa lata, and was then tested as a phloem-mobile pesticide in the Ricinus system. The compound was indeed mobile in the sieve tubes and w

Identification of Phloem Cell Transcriptome via Single-Cell RNA-Sequencing.

Single-cell RNA sequencing (scRNA-seq) has emerged as a powerful tool for dissecting cellular heterogeneity in plants at a high resolution. In particular, scRNA-seq has been applied to unravel the complex cell identities and regulatory networks of distinct phloem cell types. Because phloem cells are small and under high osmotic pressure, isolating and maintaining viable protoplasts is a challenge for successful phloem scRNA-seq. Here, we present an optimized protocol for isolating vasculature-enriched protoplasts from mature Arabidopsis thaliana leaves. Following tissue harvesting, protoplasti

Exogenous Melatonin Modulates Rice Responses to Phloem-Feeding Insect Stress.

The white-backed planthopper (WBPH; Sogatella furcifera) is a destructive phloem-feeding hemipteran that significantly limits rice productivity by depleting assimilates and transmitting viral pathogens. Although melatonin is recognised as a multifunctional plant-signalling molecule, its defensive role against WBPH remains inadequately characterised. In this study, we demonstrate that exogenous melatonin application markedly enhances rice resilience to WBPH through coordinated regulation of growth, defence signalling, antioxidant activity and ionic homoeostasis. Melatonin increased seedling sur

Phloem-specific localization of benzylisoquinoline alkaloid metabolism in opium poppy.

Opium poppy is the only commercial source of the narcotic analgesics morphine and codeine, and semi-synthetic derivatives of the natural opiate precursor thebaine, including oxycodone and the opioid antagonist naloxone. The plant also accumulates the vasodilator and antitussive agents papaverine and noscapine, respectively, which together with morphine, codeine and thebaine comprise the major benzylisoquinoline alkaloids (BIAs) in opium poppy. A majority of enzymes involved in the highly branched BIA metabolism in opium poppy have now been discovered, with many specifically localized to sieve

Regulatory DNA Sequences Controlling Gene Expression in Leaf Phloem Cells.

This chapter discusses the regulatory DNA sequences that enable cell-type-specific gene expression in leaf phloem tissues, focusing on phloem parenchyma, companion cells, and sieve elements. Molecular biology techniques, including reporter gene analysis, single-cell RNA sequencing, and live imaging, especially when combined with bioinformatic analysis, are powerful tools for identifying key promoter regions and/or cis-regulatory elements responsible for targeted expression. However, identifying conserved, cell type-specific promoter sequences across diverse plant species for cell-specific regu

Characterizing the circular RNA landscape in phloem sap of Brassica napus.

In plants, long-distance transport and phloem-mediated signal distribution play crucial roles in regulating stress adaptation and development. Phloem sap contains various types of RNAs, including small RNAs (sRNAs), messenger RNAs (mRNAs), and long noncoding RNAs (lncRNAs). Recently, endogenous circular RNAs have been identified in phloem sap of apple trees. Some phloem RNAs have been shown to have long-distance signaling functions, but for most, no functions have yet been determined. Due to their stability, circRNAs are interesting candidates with potential functions in long-distance signalin

Preparation of Tissue Sections for Light-Microscopic Analysis of Phloem Anatomy.

Accurate analysis and description of plant tissues often rely on the preparation of high-quality anatomical slides, a task that becomes particularly challenging when dealing with heterogeneous tissues such as phloem, which contains both soft and rigid components. This chapter provides a comprehensive protocol outlining key techniques for the optimal preparation of phloem tissue samples for light microscopy. The protocol encompasses essential steps such as fixation, softening, embedding, sectioning, staining, and mounting, and is adaptable for examining phloem and adjacent tissues in both woody

Live-Cell Imaging of Fluorescently-Tagged Phloem Proteins with Confocal Microscopy.

Confocal laser scanning microscopy can enable observation of phloem cells in living tissues. Here, we describe live imaging of phloem cells in the leaves and roots of Arabidopsis thaliana using fluorescently-tagged proteins, either expressed in the vasculature using phloem-specific promoters or constitutively expressed in reference marker lines. We provide instructions for identifying the majority of phloem cell types and a step-by-step protocol for the precise cellular and subcellular localization of phloem proteins.

Methods for Assessing the Role of Phloem Transport in Plant Stress Responses.

Delivery of carbohydrates to tissues under stress is important for plant defenses, recovery, and survival. Yet, little is known on how phloem function is altered under stress, and how that influences plant responses to stress. This chapter summarizes current and emerging methods for quantifying phloem transport and assessing how environmental stress affects phloem function at whole plant scale. The techniques discussed range from isotopic and fluorescent tracer studies to advanced non-invasive imaging (MRI, NMR, and PET), carbon balance approaches, anatomical analysis, and stem diameter monito

Using 13C to Quantify Phloem Transport on Tall Plants in the Field.

Phloem transport dynamics play a crucial role in understanding the movement of photoassimilates within plants, particularly in tall species, where tracing carbon allocation and monitoring transport velocities present unique challenges. The difference in time lags between a labeling pulse of 13CO2 of the foliage and the appearance of labeled C in the respiration at different locations along the stem of a tall plant is used to estimate at which velocities the isotope tracer, i.e., the labeled carbohydrates, are transported in the phloem sap. Here, we describe a method for pulse labeling tall pla

Evolution of the JULGI-SMXL4/5 module for phloem development in angiosperms.

Bifacial cambium, which produces xylem and phloem, and monopodial architecture, characterized by apical dominance and lateral branching from axillary buds, are key developmental features of seed plants, consisting of angiosperms and gymnosperms. These allow seed plants to adapt to diverse environments by optimizing resource allocation and structural integrity. In seed plants, SUPPRESSOR OF MAX2-LIKE (SMXL) family members function in phloem development and strigolactone-induced inhibition of axillary bud outgrowth. Although strigolactone signaling regulates most SMXL family members, the only kn

Plant-Derived BtHCYP Promotes Phloem Feeding and Fecundity in Bemisia tabaci.

Bemisia tabaci is a destructive agricultural pest with a remarkable capacity to exploit diverse host plants. Horizontally transferred genes (HTGs) have recently been implicated in this adaptive success, yet the functions of most HTGs in the whitefly remain unclear. Feeding behavior is crucial for nutrient acquisition and reproduction in piercing-sucking insects, but whether HTGs contribute to host adaptation by regulating feeding remains largely unknown. Here, we identified BtHCYP, a plant-derived HGT gene in B. tabaci encoding a cysteine protease. Biochemical and in vivo assays confirmed that

Defence-mediated phloem restriction of a plant virus facilitates insect transmission.

Plant viruses are causal agents of devastating diseases in crops and pose a threat to food security. Viruses transmitted by phloem-feeding insects are frequently restricted to the phloem; the mechanism determining this tissue tropism and its consequences for the viral cycle have remained elusive. Here we show that phloem restriction of tobacco curly shoot virus (TbCSV) depends on the allele of the C4 gene it carries. The Y35 allele produces a plasma membrane-associated C4 protein; TbCSV(Y35) is phloem-limited. The Y41 allele, however, produces two C4 variants, targeted to the plasma membrane a
