Lewis Acid-Mediated Deoxygenative Synthesis of Trivalent Sterically Hindered Phosphines from Aldehydes.

The efficient synthesis of trisubstituted trivalent phosphines remains challenging. Herein, we report a novel approach for rapid construction of these compounds from aldehydes under Lewis acid-mediated conditions. By using sterically hindered disubstituted phosphines, aldehydes can undergo a deoxygenative reaction to afford trivalent phosphorus compounds. This modular and practical method is characterized by operational simplicity, a broad substrate scope, and excellent functional group tolerance, which holds great potential for applications in organic synthesis.

Evaluation of the immunomodulatory effects of novel phosphinic acid derivatives in vitro and in a murine colitis model.

T cells and cytokines are key contributors to the pathogenesis of inflammatory bowel disease (IBD), where dysregulated immune activation drives chronic intestinal inflammation. Phosphinic acid compounds are biologically active molecules that function as enzyme inhibitors and have been investigated for diverse pharmacological applications, including their potential therapeutics in inflammatory diseases. In this study, we investigated the immunomodulatory effects of seven novel phosphinic acid derivatives, including PAC-tryptophan (PAC-Trp), PAC-leucine (PAC-Leu), PAC-proline (PAC-Pro), PAC-phen

New insights into structure and function of bis-phosphinic acid derivatives and implications for CFTR modulation.

C407 is a compound that corrects the Cystic Fibrosis Transmembrane Conductance Regulator (CFTR) protein carrying the p.Phe508del (F508del) mutation. We investigated the corrector effect of c407 and its derivatives on F508del-CFTR protein. Molecular docking and dynamics simulations combined with site-directed mutagenesis suggested that c407 stabilizes the F508del-Nucleotide Binding Domain 1 (NBD1) during the co-translational folding process by occupying the position of the p.Phe1068 side chain located at the fourth intracellular loop (ICL4). After CFTR domains assembly, c407 occupies the positi

Copper-Catalyzed Phosphorylation of 2,3-Allenoic Acids and Phosphine Oxide: Access to Phosphorylated Butenolides.

We investigated a novel Cu-catalyzed annulation of 2,3-allenoic acids with diphenylphosphine oxide, leading to the formation of 4-phosphate butenolides in up to 88% yield. The formation of the C-P bond provides new avenues for the functionalization of different furan-2(5H)-ones, with favorable features such as suitable functional group tolerance and mild synthesis conditions.

Investigating the phosphinic acid tripeptide mimetic DG013A as a tool compound inhibitor of the M1-aminopeptidase ERAP1.

ERAP1 is a zinc-dependent M1-aminopeptidase that trims lipophilic amino acids from the N-terminus of peptides. Owing to its importance in the processing of antigens and regulation of the adaptive immune response, dysregulation of the highly polymorphic ERAP1 has been implicated in autoimmune disease and cancer. To test this hypothesis and establish the role of ERAP1 in these disease areas, high affinity, cell permeable and selective chemical probes are essential. DG013A 1, is a phosphinic acid tripeptide mimetic inhibitor with reported low nanomolar affinity for ERAP1. However, this chemotype

Non-metal Lewis Acid-Catalyzed Reduction of Phosphine Oxides with Silanes: Mechanistic Study and Reaction Optimization.

Non-metal Lewis acid (NM LA)-catalyzed silane-mediated reductions represent an important strategy for the conversion of phosphine oxides to phosphines. Although a variety of catalytic systems have been developed, their underlying reaction mechanisms remain insufficiently understood. Herein, we have elucidated two unified mechanistic manifolds for these transformations based on combined computational and experimental studies: a push-pull mechanism and a silane-activation mechanism. Guided by these mechanistic insights, we further improve the conventional Brønsted acid-catalyzed silane-mediated

Transition Metal Complexes of Phosphinous Acids Featuring a Quasichelating Unit: Synthesis, Characterization, and Hetero-bimetallic Complexes.

Diorganophosphane oxides were employed as preligands for the synthesis of catalytically active transition metal complexes of the phosphinous acids (CF3)2POH and (C2F5)2POH. Their reactions with solid PtCl2 and PdCl2 led to the formation of mononuclear phosphinous acid complexes [Cl2M{P(R(f))2OH}2] (M = Pd, Pt; R(f) = C2F5, CF3), which can be crystallized, for example, as its pyridinium salts, 2[HPy](+)[Cl2Pd{P(CF3)2O}2](2-). In vacuo HCl is liberated from the neutral palladium complexes affording mixtures of di- and polynuclear complexes. Moreover, (C2F5)2POH was reacted with several β-diketon

Synthesis of phosphonate and phosphinate analogs of 2-oxo acids for site-specific inhibition of thiamine-diphosphate-dependent dehydrogenases of 2-oxo acids in vivo.

In this work, we describe the synthesis of the phosphonate and phosphinate analogs of pyruvate, as well as phosphonate analogs of 2-oxoglutarate and its homologs 2-oxoadipate and 2-oxopimelate. Arbuzov reaction and condensation of triethyl orthoacetate with the hypophosphorous acid or methyl dichlorophosphine is presented as efficient methods for preparation of 2-oxo organophosphorus esters. Subsequent transformations of the esters to free acids are achieved by treatment with trimethylbromosilane, or sodium iodide, or hydrogenation of the corresponding benzyl esters. The site-specific inhibiti

8:8 Perfluoroalkyl phosphinic acid affects neurobehavioral development, thyroid disruption, and DNA methylation in developing zebrafish.

Recent studies have reported potential neurotoxicity and epigenetic alteration associated with exposure to several per- and polyfluoroalkyl substances (PFASs). However, such information is limited to a few compounds (e.g., perfluorooctane sulfonate), primarily based on rodent experiments, and the underlying toxicological mechanism(s) for many PFAS in the environment remain poorly understood. In the present study, we investigated 8:8 perfluoroalkyl phosphinic acid (8:8 PFPiA), an under-studied PFAS with high persistency in the environment and biota, using the zebrafish model. We exposed zebrafi

Air-stable platinum and palladium complexes featuring bis[2,4-bis(trifluoromethyl)phenyl]phosphinous acid ligands.

Secondary phosphane oxides, R(2)P(O)H, are commonly used as preligands for transition-metal complexes of phosphinous acids, R(2)P-OH (R=alkyl, aryl), which are relevant as efficient catalysts in cross-coupling processes. In contrast to previous work by other groups, we are interested in the ligating properties of an electron-deficient phosphinous acid, (R(f))(2)P-OH, bearing the strongly electron-withdrawing and sterically demanding 2,4-bis(trifluoromethyl)phenyl group towards catalysis-relevant metals, such as palladium and platinum. The preligand bis[2,4-bis(trifluoromethyl)phenyl]phosphane

Synthesis of Unsymmetrical Tertiary Phosphine Oxides via Sequential Substitution Reaction of Phosphonic Acid Dithioesters with Grignard Reagents.

A facile synthetic method for unsymmetrical tertiary phosphine oxides is reported. Sequential treatment of phosphonodithioic acid S,S-di(p-tolyl) esters with two Grignard reagents enabled the stepwise introduction of different carbon substituents on the phosphorus atom. The chemical stability of dithioesters and monosubstituted thioesters has enhanced the utility of this method, rendering a wide range of organophosphorus compounds easily available.

Palladium(II)-Catalyzed Synthesis of Sulfinates from Boronic Acids and DABSO: A Redox-Neutral, Phosphine-Free Transformation.

A redox-neutral palladium(II)-catalyzed conversion of aryl, heteroaryl, and alkenyl boronic acids into sulfinate intermediates, and onwards to sulfones and sulfonamides, has been realized. A simple Pd(OAc)2 catalyst, in combination with the sulfur dioxide surrogate 1,4-diazabicyclo[2.2.2]octane bis(sulfur dioxide) (DABSO), is sufficient to achieve rapid and high-yielding conversion of the boronic acids into the corresponding sulfinates. Addition of C- or N-based electrophiles then allows conversion into sulfones and sulfonamides, respectively, in a one-pot, two-step process.

Atroposelective Bromination for the Synthesis of Chiral Biaryl Phosphines via Cross-Assembled Catalysis with Chiral Phosphoric Acid and Achiral Phenol.

Enantioselective electrophilic halogenation using organocatalysts has emerged as a powerful tool in asymmetric synthesis, yet its application has been largely concentrated on substrates bearing hydrogen bond donors. This prerequisite aligns with the design of conventional bifunctional catalysts, which operate through complementary hydrogen bond interactions with both the substrate and N-haloamide reagents. Chiral Buchwald-type ligands are useful monophosphine ligands, but their preparation mainly relies on kinetic resolution. Catalytic atroposelective halogenation of achiral Buchwald-type liga

Enantioselective Preparation of P-Chirogenic SPOs from Phosphinous Acid Boranes Applied in Non-Atropoisomeric Biarylphosphine and Derivative Syntheses.

An efficient enantioselective synthesis of P-chirogenic phosphine(oxide)s with a non-atropoisomeric biaryl substituent or bridge was reported. The method was based on two key steps. First, the enantioselective synthesis of P-chirogenic o-halogenophenylphosphine oxides was achieved with complete retention of configuration at the P atom, by one-pot reaction of secondary phosphine oxides (SPOs) with benzyne, in situ generated from 1,2-dibromo- or -diiodobenzene. The P-chirogenic SPOs were enantioselectively prepared by successive acid hydrolysis of aminophosphine boranes derived from (+)- or (-)-

Synthesis of P,N-heterocycles from omega-amino-H-phosphinates: conformationally restricted alpha-amino acid analogs.

P,N-heterocycles (3-hydroxy-1,3-azaphospholane and 3-hydroxy-1,3-azaphosphorinane-3-oxide) are synthesized in moderate yield from readily available omega-amino-H-phosphinates and aldehydes or ketones via an intramolecular Kabachnik-Fields reaction. The products are conformationally restricted phosphinic analogs of alpha-amino acids. The multigram-scale syntheses of the H2N(CH2)(n)PO2H2 phosphinic precursors (n = 1, 2, 3) and some derivatives are also described.

Efficient synthesis of sterically crowded biaryls by palladium-phosphinous acid-catalyzed cross-coupling of aryl halides and aryl grignards.

A series of sterically hindered biaryls have been obtained by palladium- and nickel-phosphinous acid-catalyzed Kumada-Corriu cross-coupling of ortho-substituted aryl halides and Grignard reagents. This method allows formation of di- and tri-ortho-substituted biaryls in 87-98% yield under mild reaction conditions even when electron-rich aryl chlorides are used. The reaction also proceeds with aryl iodides at -20 degrees C, and under these conditions, functional groups that are generally not compatible with Grignard reagents are tolerated.

Phosphine-Catalyzed Dehydrative Annulation of 2-Picolylamines with Carboxylic Acids to Access Imidazo[1,5-a]pyridines by Sequential C-N Bond Formation.

Imidazo[1,5-a]pyridines constitute a privileged class of N-fused heterocycles widely found in bioactive molecules and functional materials, yet their synthesis often relies on harsh, stoichiometric activating reagents. Herein, we report a phosphine-catalyzed condensation-annulation strategy for the synthesis of imidazo[1,5-a]pyridines from readily available 2-picolylamines and carboxylic acids under mild and neutral conditions. The method exhibits broad functional group tolerance and achieves chemoselective dehydrative coupling, proceeding efficiently in the presence of sensitive functionaliti

Cooperative Effects between Chiral Cpx -Iridium(III) Catalysts and Chiral Carboxylic Acids in Enantioselective C-H Amidations of Phosphine Oxides.

An enantioselective C-H amidation of phosphine oxides by using an iridium(III) catalyst bearing an atropchiral cyclopentadienyl (Cpx ) ligand is reported. A very strong cooperative effect between the chiral Cpx ligand and a phthaloyl tert-leucine enabled the transformation. Matched-mismatched cases of the different acid enantiomers are shown. The amidated P-chiral arylphosphine oxides are formed in yields of up to 95 % and with excellent enantioselectivities of up to 99:1 er. Enantiospecific reduction provides access to valuable P-chiral phosphorus(III) compounds.
