Mitochondrial Resilience: Unraveling the Triadic Interplay of Phosphocreatine, Cyclophilin D, and STAT3 in Heart Failure.

Heart failure remains a major global health burden, with mitochondrial dysfunction recognized as a key contributor to its onset and progression. This review highlights three critical regulators of mitochondrial integrity phosphocreatine (PCr), cyclophilin D (CypD), and signal transducer and activator of transcription 3 (STAT3) and their coordinated roles in cardiac function. PCr is vital for sustaining myocardial energy balance, particularly under metabolic stress. CypD controls the mitochondrial permeability transition pore, regulating cell death pathways that contribute to cardiac injury. Be

Sodium Nitrate Improves Oxidative Energy Contribution and Reduces Phosphocreatine Contribution During High-Intensity Intermittent Exercise.

The present study aimed to verify the effect of acute nitrate supplementation on oxidative, phosphocreatine, and glycolytic energy contribution (COXI, CPCr, and CLAC, respectively) during a high-intensity intermittent exercise (HIIE). Fifteen physically active subjects were submitted to incremental running test on a treadmill and two random HIIE (10 × 1 min at maximal aerobic speed with 1 min of passive recovery) in the following conditions: sodium nitrate (SN) or Placebo (PL). Repeated measure ANOVA was used to compare COXI, and CPCr within the 10 efforts. Student t test was used to compare t

Slower recovery of muscle phosphocreatine in malignant hyperthermia-susceptible individuals assessed by 31P-MR spectroscopy.

Our aim was to develop an exercise protocol using 31P-magnetic resonance spectroscopy (31P-MRS), which can discriminate between malignant hyperthermia-susceptible (MHS) individuals and controls. MRS spectra of the forearm muscles were recorded at rest, during and after a standardized exercise protocol in 10 MHS patients and compared with spectra obtained in 10 controls. There was no difference in resting intracellular pH (pHi) or PCr/(Pi+PCr) ratio between the groups (PCr = phosphocreatine, Pi = inorganic phosphorus). At the end of the exercise and during the initial recovery phase, the pHi an

Phosphorus magnetic resonance spectroscopy of patients with mitochondrial cytopathies demonstrates decreased levels of brain phosphocreatine.

The hypothesis that brain mitochondria are directly affected in several phenotypes associated with disordered oxidative phosphorylation was tested using phosphorus 31 (31P) magnetic resonance spectroscopy. Abnormal phosphorylation potentials in skeletal muscle have been demonstrated by 31P magnetic resonance spectroscopy in patients with mitochondrial cytopathies (heritable disorders of oxidative phosphorylation), but abnormalities of phosphorylation potentials in other organs have not been documented. Several lines of evidence suggest that these mutations may affect mitochondria in nonmuscle

Effects of hyperventilation on phosphocreatine kinetics and muscle deoxygenation during moderate-intensity plantar flexion exercise.

The effects of controlled voluntary hyperventilation (Hyp) on phosphocreatine (PCr) kinetics and muscle deoxygenation were examined during moderate-intensity plantar flexion exercise. Male subjects (n = 7) performed trials consisting of 20-min rest, 6-min exercise, and 10-min recovery in control [Con; end-tidal Pco(2) (Pet(CO(2))) approximately 33 mmHg] and Hyp (Pet(CO(2)) approximately 17 mmHg) conditions. Phosphorus-31 magnetic resonance and near-infrared spectroscopy were used simultaneously to monitor intramuscular acid-base status, high-energy phosphates, and muscle oxygenation. Resting i

Phosphocreatine attenuates doxorubicin-induced hepatotoxicity via activation of Nrf2/HO-1 and PGC-1α pathways in mice.

Phosphocreatine (PCr), an energy-buffering molecule that supports mitochondrial function, may protect against chemotherapy-induced liver injury. Doxorubicin (DOX)-induced hepatotoxicity, driven by oxidative stress, mitochondrial dysfunction, and apoptosis, remains a major challenge in cancer therapy. This study evaluated the protective effects of PCr against DOX-induced liver injury in mice, focusing on mitochondrial function and apoptosis via the Nrf2/HO-1 and PGC-1α signaling pathways. Male mice were treated with DOX to induce hepatotoxicity, followed by PCr administration. Key apoptosis-rel

Simultaneous monitoring of glycogen, creatine, and phosphocreatine in type II glycogen storage disease using saturation transfer MRI.

PURPOSE: There is a need for non-invasive approaches to assess the progression of glycogen storage diseases (GSD). Here, we use saturation transfer (ST) MRI via relayed nuclear Overhauser effects (glycoNOE) to detect abnormal changes in muscle glycogen of a GSD II mouse model. In addition to glycogen, the energy metabolites phosphocreatine (PCr) and creatine (Cr) were studied to assess the muscle disease. METHODS: Water saturation (Z-spectra) and 1H MRS were acquired at 9.4 T on the skeletal muscle of healthy control mice and homozygous acid α $$ \upalpha $$ -glucosidase (GAA) knock-out mic

Effect of exogenous phosphocreatine on cardiomycytic apoptosis and expression of Bcl-2 and Bax after cardiopulmonary resuscitation in rats.

BACKGROUND: Ischemia-reperfusion injury in the myocardium after cardiac arrest (CA) and cardiopulmonary resuscitation (CPR) is an important pathologic basis of post-cardiac arrest of syndrome (PCAS), and apoptosis is one of the major mechanisms in myocardial ischemia-reperfusion injury. To lessen myocardial ischemia-reperfusion injury after cardiac arrest and CPR, it is important to reduce energy consumption and to increase energy supply in the myocardium. This study aimed to observe changes of cell apoptosis and expression of Bcl-2 and Bax protein on the myocardium after CPR in rats, and the

Phosphorylated guanidinoacetate partly compensates for the lack of phosphocreatine in skeletal muscle of mice lacking guanidinoacetate methyltransferase.

The effects of creatine (Cr) absence in skeletal muscle caused by a deletion of guanidinoacetate methyltransferase (GAMT) were studied in a knockout mouse model by in vivo (31)P magnetic resonance (MR) spectroscopy. (31)P MR spectra of hindleg muscle of GAMT-deficient (GAMT-/-) mice showed no phosphocreatine (PCr) signal and instead showed the signal for phosphorylated guanidinoacetate (PGua), the immediate precursor of Cr, which is not normally present. Tissue pH did not differ between wild-type (WT) and GAMT-/- mice, while relative inorganic phosphate (P(i)) levels were increased in the latt

Reduced maximum shortening velocity in the absence of phosphocreatine observed in intact fibres of Xenopus skeletal muscle.

1. ADP inhibits the maximum shortening velocity (V0) in skeletal muscle. [ADP] may increase considerably during contractions and reduce V0 in the absence of energy buffering by phosphocreatine (PCr). We have tested this hypothesis by comparing V0 in long and short tetani produced in situations where PCr buffering is absent. 2. Single, intact muscle fibres were dissected from toe muscles of Xenopus and stimulated by current pulses at 20 degrees C. The test sequence consisted of a 400 ms tetanus, followed after 3 s by a 1400 ms tetanus and after an additional 4 s by a 400 ms tetanus. V0 was meas

The concentration of N-acetyl aspartate, creatine + phosphocreatine, and choline in different parts of the brain in adulthood and senium.

The fully relaxed water signal was used as an internal standard in a STEAM experiment to calculate the concentrations of the metabolites: N-acetyl aspartate (NAA), creatine + phosphocreatine (Cr + PCr), and choline (Cho) containing compounds in four different parts of the brain in two age groups of healthy volunteers (20-30 yr, n = 8) and (60-80 yr, n = 8). Furthermore, T1 and T2 relaxation time of the metabolites and signal ratios: NAA/Cho, NAA/Cr + PCr, and Cho/Cr + PCr at TE = 272 msec were calculated. The experiments were carried out using a Siemens Helicon SP 63/84 wholebody MR-scanner at

Astrocyte volume regulation and ATP and phosphocreatine concentrations after exposure to salicylate, ammonium, and fatty acids.

Cellular volume regulation following swelling in hypo-osmotic phosphate-buffered saline (PBS) and ATP and phosphocreatine concentrations of cells incubated in iso-osmotic or hypo-osmotic PBS were measured in primary cultured rat cerebral astrocytes exposed for 30 min to NH4Cl, salicylate, hexanoate, octanoate, and/or dodecanoate. These compounds have been implicated in the pathogenesis of cerebral edema in Reye's Syndrome. NH4Cl (0.10 - 10 mM) had no effect on astrocyte volume regulation or ATP concentration. Salicylate significantly reduced ATP concentrations at 3.0 mM and 10 mM but had no ef

Enhanced Quantitative Phosphocreatine MR Imaging of Skeletal Muscle Using a Global-Local Two-Branch Deep Learning Model.

PURPOSE: Phosphocreatine (PCr) is an essential marker of muscle metabolism, and accurate quantification of its (fs) and its exchange rate (ksw) is essential for diagnosing various muscular and neuromuscular diseases. Although chemical exchange saturation transfer (CEST) MRI can detect the saturation transfer effect from PCr, quantification of the underlying PCr fs and ksw, particularly at low fields, remains challenging due to significant overlapping confounding effects in tissues when using conventional fitting approaches. Deep learning (DL) presents a promising alternative, yet traditional D

Reduced Adenosine Triphosphate-to-Phosphocreatine Ratios in Neuropsychiatric Post-COVID Condition: Evidence From 31P Magnetic Resonance Spectroscopy.

BACKGROUND: Post-COVID condition (PCCo) affects 5% to 10% of individuals following SARS-CoV-2 infection, with cognitive disturbances being a major feature. Central hypotheses regarding its pathophysiology include disturbed cell energy metabolism and oxidative stress pointing to mitochondrial dysfunction. However, brain energy metabolism remains unexplored. METHODS: We investigated cerebral high-energy phosphate metabolism in 27 patients with PCCo and 23 fully recovered control participants using whole-brain 31P-magnetic resonance spectroscopic imaging at 3T. Phosphocreatine attenuates diabetes-exacerbated kidney fibrosis via TGF-β/Smad and PI3K/Akt pathways in a dual rat model.

Diabetes accelerates kidney fibrosis through chronic inflammation, oxidative stress, and activation of profibrotic signaling pathways, including TGF-β/Smad and PI3K/Akt. This study investigated the renoprotective effects of phosphocreatine (PCr) in a dual rat model combining STZ-induced diabetes with unilateral ureteral obstruction (UUO)-induced renal fibrosis. PCr was administered intraperitoneally at 20 or 50 mg/kg for eight weeks, starting one-week post-UUO. Renal function, oxidative stress markers, mitochondrial bioenergetics, and inflammatory cytokines (TNF-α, IL-6) were evaluated. Histop

Dynamic asymmetry of phosphocreatine concentration and O(2) uptake between the on- and off-transients of moderate- and high-intensity exercise in humans.

The on- and off-transient (i.e. phase II) responses of pulmonary oxygen uptake (V(O(2))) to moderate-intensity exercise (i.e. below the lactate threshold, theta;(L)) in humans has been shown to conform to both mono-exponentiality and 'on-off' symmetry, consistent with a system manifesting linear control dynamics. However above theta;(L) the V(O(2)) kinetics have been shown to be more complex: during high-intensity exercise neither mono-exponentiality nor 'on-off' symmetry have been shown to appropriately characterise the V(O(2)) response. Muscle [phosphocreatine] ([PCr]) responses to exercise,

Phosphocreatine attenuates isoproterenol-induced cardiac fibrosis via activation of the Nrf2/ARE antioxidant pathway and preservation of endothelial integrity.

BACKGROUND: Phosphocreatine (PCr), a high-energy phosphate donor with established cytoprotective and antioxidative properties, is known to support endothelial function. However, its role in the regulation of cardiac fibrogenesis remains poorly defined. This study aimed to determine whether PCr attenuates isoproterenol (ISO)-induced cardiac fibrosis and to examine the potential involvement of the nuclear factor erythroid 2-related factor 2/antioxidant response element (Nrf2/ARE) signaling pathway. METHODS: A murine model of cardiac fibrosis was established by subcutaneous administration of ISO

31P-MRS-Measured Phosphocreatine Recovery Kinetics in Human Muscles in Health and Disease-A Systematic Review and Meta-Analysis.

The noninvasive, in vivo measurement of postexercise phosphocreatine (PCr) recovery kinetics using 31-phosphorus magnetic resonance spectroscopy (31P-MRS) is a highly prevalent method for assessing skeletal muscle energetics. However, 31P-MRS methodology is notoriously laboratory-specific, leading to uncertainty about the normal range of PCr recovery kinetics among healthy individuals, as well as relationships with disease and demographic factors. This systematic review and meta-analysis characterized the normal range of PCr recovery kinetics from 31P-MRS in human skeletal muscles across the l
