Developing a simple method to enhance the generation of cone and rod photoreceptors in pluripotent stem cell-derived retinal organoids.

Cell replacement therapy is a promising treatment for irreversible retinal cell death in diverse diseases such as Stargardt's disease, age-related macular degeneration, and retinitis pigmentosa. The final impact of all retinal dystrophies is the loss of photoreceptors; hence, there is a pressing need for research into replacement. Seminal work has shown that a simple three-dimensional culture system enables differentiation of human pluripotent stem cells to retinal organoids containing large numbers of photoreceptors developing alongside retinal neurons and Müller glia cells in a laminated str

Misexpression of Xsiah-2 induces a small eye phenotype in Xenopus.

Recent data demonstrate a structural and functional conservation of factors crucial for the development of the insect and the vertebrate eye. We isolated Xenopus siah-2, a protein with 67% identity to Drosophila sina (seven in absentia) and 85% identity to the mouse and human siah-2 proteins. Sina is required in Drosophila for the R7 photoreceptor cell formation during eye development, because it down regulates proteins that inhibit R7 differentiation via the ubiquitin/proteasome pathway. Nothing is known about the developmental function of the siah protein in vertebrates. We show that in Xeno

Ordered Chaos: Photoreceptor Mosaics in the Larval and Adult Turquoise Killifish (Nothobranchius furzeri).

An organism's vision is dependent on the photoreceptor types in the eye and how these cells are organized across the retina. The turquoise killifish (Nothobranchius furzeri) is an annual killifish species that lives in the turbid waters of ephemeral pools. Although turquoise killifish are gaining popularity as a model, we know little about their photoreceptors. Here, the turquoise killifish retina was investigated for photoreceptor subtype identity and mosaic organization at the adult and larval stages to assess specialization for their light environment. Turquoise killifish were determined to

Rhodopsin promoter-EGFP fusion transgene expression in photoreceptor neurons of retina and pineal complex in mice.

Light detection in vertebrate eyes is mediated through retinal photoreceptor rod and cone cells that transduce light signals into electrical responses. The differentiation and synaptogenesis of photoreceptor cells are especially important since these cells are the main targets of degeneration in retinitis pigmentosa. We produced transgenic mice that express enhanced green fluorescent protein (EGFP) under the control of the mouse rhodopsin gene promoter. In Western blot analyses of transgenic retinal homogenates, expression of the endogenous rhodopsin gene was detected from post-natal day (P)8;

Nomen omen: Euglena gracilis possesses a rhodopsin-based photoreceptor.

The unicellular microalga Euglena gracilis has always been considered the ideal alga to investigate photoreceptive responses and systems, and it has been the subject of hundreds of articles. Moreover, because of its detectable photoreceptor, it has been given a key role in the evolution of photoreception, from single and simple cells to complex visual system of higher organisms. In this article, we report the Raman spectra recorded in vivo on photoreceptors of E. gracilis and Bos taurus retina. The almost perfect superimposability (correlation coefficient r = 0.955) of these spectra states tha

Analysis of cilia structure and function in zebrafish.

The cilium, a previously little studied cell surface protrusion, has emerged as an important organelle in vertebrate cells. This tiny structure is essential for normal embryonic development, including the formation of left-right asymmetry, limb morphogenesis, and the differentiation of sensory cells. In the adult, cilia also function in a variety of processes, such as the survival of photoreceptor cells, and the homeostasis in several tissues, including the epithelia of nephric ducts. Human ciliary malfunction is associated with situs inversus, kidney cysts, polydactyly, blindness, mental reta

Phototransduction in vertebrate rod and cone cells.

Vertebrate photoreceptor cells operate under very dim and bright illumination regimes. A protein machinery in rod and cone cells underlying the light response mediates photoexcitation, the return to the dark state, and adaptation processes. The machinery controls the homeostasis and mutual dependence of two cytoplasmic messengers, cGMP and Ca2+. The signaling pathway starts with light absorption by visual pigments (rhodopsin in rods or cone opsin in cones), which triggers an amplified signaling cascade, leading to the hydrolysis of cGMP and closure of cyclic nucleotide-gated channels in the ph

Exploring the molecular makeup of support cells in insect camera eyes.

Animals generally have either compound eyes, which have evolved repeatedly in different invertebrates, or camera eyes, which have evolved many times across the animal kingdom. Both eye types include two important kinds of cells: photoreceptor cells, which can be excited by light, and non-neuronal support cells (SupCs), which provide essential support to photoreceptors. Despite many examples of convergence in eye evolution, similarities in the gross developmental plan and molecular signatures have been discovered, even between phylogenetically distant and functionally different eye types. A single Musashi gene allele is sufficient to maintain mouse photoreceptor cells.

In vertebrates, two genes, Musashi1 (Msi1) and Musashi2 (Msi2), encode for highly similar Musashi protein paralogs. The Musashi proteins are known to bind to 3'-UTRs and control translation. In photoreceptor cells, the Musashi proteins promote the inclusion of photoreceptor-specific alternative exons by binding to the proximal downstream of their introns. While the Musashi proteins are expressed in various cell types, their role in regulating splicing appears to be confined to photoreceptor cells, where the two proteins have exceptionally high expression levels. To test if the photoreceptor-sp

Nanowire-based artificial photoreceptors with multi-hyperuniformity inspired by chicken retina.

Retinal cells of chicken possess a hyperuniform distribution enabling exceptional spatial sampling. Recently, interest has grown in mimicking biological imaging systems for optoelectronics technology. However, since biological retinal cells and current imaging pixels have a significant scale mismatch such as super-micron and sub-micron, the applicability of hyperuniform distribution must be investigated at sub-micron scale, considering wave-optics phenomena. In this study, we present theoretical analyses and design strategies for nanowire-based artificial photoreceptors with hyperuniformity. F

Structure and function of the interphotoreceptor matrix surrounding retinal photoreceptor cells.

The interphotoreceptor matrix (IPM) is a highly organized structure with interconnected domains surrounding cone and rod photoreceptor cells and extends throughout the subretinal space. Based on known roles of the extracellular matrix in other tissues, the IPM is thought to have several prominent functions including serving as a receptor for growth factors, regulating retinoid transport, participating in cytoskeletal organization in surrounding cells, and regulation of oxygen and nutrient transport. In addition, a number of studies suggest that the IPM also may play a significant role in the e

The pineal organ as a folded retina: immunocytochemical localization of opsins.

The most simple pineal complex (the pineal and parapineal organs of lampreys), consists of saccular evaginations of the diencephalic roof, and has a retina-like structure containing photoreceptor cells and secondary neurons. In more differentiated vertebrates, the successive folding of the pineal wall multiplies the cells and reduces the lumen of the organ, but the pattern of the histological organization remains similar to that of lampreys; therefore, we consider the histological structure of the pineal organ of higher vertebrates as a 'folded retina'. The cell membrane of several pineal phot

Photoreceptor replacement: a disease-agnostic approach for the treatment of advanced retinal degeneration.

Retinal dystrophies such as retinitis pigmentosa (RP) and age-related macular degeneration (AMD) affect millions of individuals worldwide and remain leading causes of irreversible blindness. Photoreceptor transplantation has emerged as a promising and disease-agnostic therapeutic approach for restoring vision in advanced degenerative retinal diseases characterised by the loss of rods and/or cones. This brief review summarises some key areas of progress and future directions for photoreceptor replacement therapy, with consideration given to translating pre-clinical findings to the clinic.

Retinal photoreceptor cilia and ciliopathies: Molecular mechanisms and therapeutic strategies.

Photoreceptor outer segments are specialized sensory cilia that are filled with flattened membranous discs responsible for transducing light into neural signals. These cilia maintain a dynamic system in which daily shedding of outer segments at distal tips balances continuous membrane synthesis, ensuring photoreceptor homeostasis. Mutations in ciliary genes impair the biogenesis or shedding of outer segments, leading to retinal ciliopathies. Recent studies have elucidated molecular mechanisms governing photoreceptor ciliary formation, maintenance, and renewal. In this review, we summarize curr

Nonvisual photoreceptors of the deep brain, pineal organs and retina.

The role of the nonvisual photoreception is to synchronise periodic functions of living organisms to the environmental light periods in order to help survival of various species in different biotopes. In vertebrates, the so-called deep brain (septal and hypothalamic) photoreceptors, the pineal organs (pineal- and parapineal organs, frontal- and parietal eye) and the retina (of the "lateral" eye) are involved in the light-based entrain of endogenous circadian clocks present in various organs. In humans, photoperiodicity was studied in connection with sleep disturbances in shift work, seasonal d

Analysis of rod/cone gap junctions from the reconstruction of mouse photoreceptor terminals.

Electrical coupling, mediated by gap junctions, contributes to signal averaging, synchronization, and noise reduction in neuronal circuits. In addition, gap junctions may also provide alternative neuronal pathways. However, because they are small and especially difficult to image, gap junctions are often ignored in large-scale 3D reconstructions. Here, we reconstruct gap junctions between photoreceptors in the mouse retina using serial blockface-scanning electron microscopy, focused ion beam-scanning electron microscopy, and confocal microscopy for the gap junction protein Cx36. Systemic immunosuppression promotes survival and integration of subretinally implanted human ESC-derived photoreceptor precursors in dogs.

Regenerative therapies aimed at replacing photoreceptors are a promising approach for the treatment of otherwise incurable causes of blindness. However, such therapies still face significant hurdles, including the need to improve subretinal delivery and long-term survival rate of transplanted cells, and promote sufficient integration into the host retina. Here, we successfully delivered in vitro-derived human photoreceptor precursor cells (PRPCs; also known as immature photoreceptors) to the subretinal space of seven normal and three rcd1/PDE6B mutant dogs with advanced inherited retinal degen

Production of clinical grade patient iPSC-derived 3D retinal organoids containing transplantable photoreceptor cells.

BACKGROUND: Neurodegenerative conditions that affect the retina are currently the leading cause of incurable blindness in the developed world. Although gene and drug therapies are being developed to slow disease progression in some cases, restorative cell replacement approaches are needed for patients with significant vision impairment due to retinal degeneration. While a variety of different cell types have been evaluated in the context of retinal cell replacement, induced pluripotent stem cells (iPSCs), which can be generated and delivered as an autologous therapeutic, are in many ways the m

Explaining Retinal Susceptibility to Diabetes Through Photoreceptor Biology.

While diabetic retinal disease (DRD) has classically been viewed as a microvascular complication, emerging evidence places the photoreceptor at the center of its pathogenesis. Recognizing this central role provides a critical framework for resolving a major clinical paradox in diabetes: why the retina exhibits profound susceptibility to hyperglycemic damage, whereas closely related neural tissues like the brain are mostly spared. In this review, we synthesize the evidence for photoreceptor-driven DRD pathology by evaluating two primary mechanistic paradigms. In the first, hyperglycemia-induced

Assessment of Scotopic Function in Rod-Cone Inherited Retinal Degeneration With the Scotopic Macular Integrity Assessment.

PURPOSE: The scotopic macular integrity assessment (S-MAIA) can perform scotopic assessment to detect localized changes to scotopic rod and cone function. This study is an exploratory investigation of the feasibility of using the S-MAIA in a rod-cone dystrophy population to identify the pattern of loss in scotopic photoreceptor function. METHODS: Twenty patients diagnosed with a rod-cone dystrophy underwent visual acuity testing, full-field stimulus threshold assessment, and multiple S-MAIA tests after dark adaptation periods of 20 minutes and 45 minutes performed separately.
