Influence of the diadinoxanthin pool size on photoprotection in the marine planktonic diatom Phaeodactylum tricornutum.

The pool size of the xanthophyll cycle pigment diadinoxanthin (DD) in the diatom Phaeodactylum tricornutum depends on illumination conditions during culture. Intermittent light caused a doubling of the DD pool without significant change in other pigment contents and photosynthetic parameters, including the photosystem II (PSII) antenna size. On exposure to high-light intensity, extensive de-epoxidation of DD to diatoxanthin (DT) rapidly caused a very strong quenching of the maximum chlorophyll fluorescence yield (F(m), PSII reaction centers closed), which was fully reversed in the dark. The no

Chemical crosslinking studies of the isolated light-harvesting B800-850 complex of Chromatium minutissimum.

The spatial relationship of polypeptides comprising the light-harvesting B800-850 complex of Chromatium minutissimum has been studied by means of chemical crosslinking of the isolated complex with cleavable, 1.2 nm-long dithiobis(succinimidyl propionate). The samples were analyzed by different types of electrophoresis and spectrophotometrically. No difference was shown between crosslinking of the B800-850 complex either solubilized or incorporated into proteoliposomes. It was found that two main polypeptides form only one type of heterodimer. The crosslinked complex was more thermostable. This

Effect of detergent concentration on the thermal stability of a membrane protein: The case study of bacterial reaction center solubilized by N,N-dimethyldodecylamine-N-oxide.

We report on the response of reaction center (RC) from Rhodobacter sphaeroides (an archetype of membrane proteins) to the exposure at high temperature. The RCs have been solubilized in aqueous solution of the detergent N,N-dimethyldodecylamine-N-oxide (LDAO). Changes in the protein conformation have been probed by monitoring the variation in the absorbance of the bacteriochlorine cofactors and modification in the efficiency of energy transfer from tryptophans to cofactors and among the cofactors (through fluorescence measurements). The RC aggregation taking place at high temperature has been i

Expression of the psbA gene during photoinhibition and recovery in Synechocystis PCC 6714: inhibition and damage of transcriptional and translational machinery prevent the restoration of photosystem II activity.

The D1 reaction center protein of the photosystem II complex is very sensitive to light. It is continuously being damaged, degraded and resynthesized. Under high light, photosystem II inactivation is observed. This is because the rate of D1 damage is faster than that of its replacement. This process can be reversed if exposure to high light is not too long. In this work we study the changes that occur in the transcriptional and translational machinery that could lead to irreversible photoinhibition in Synechocystis PCC 6714. In the first minutes of photoinhibition, high light induced an accumu

The cyanobacterium Synechococcus resists UV-B by exchanging photosystem II reaction-center D1 proteins.

Current ambient UV-B levels can significantly depress productivity in aquatic habitats, largely because UV-B inhibits several steps of photosynthesis, including the photooxidation of water catalyzed by photosystem II. We show that upon UV-B exposure the cyanobacterium Synechococcus sp. PCC 7942 rapidly changes the expression of a family of three psbA genes encoding photosystem II D1 proteins. In wild-type cells the psbAI gene is expressed constitutively, but strong accumulations of psbAII and psbAIII transcripts are induced within 15 min of moderate UV-B exposure (0.4 W/m2). This transcription

Requirement of phosphatidylglycerol for maintenance of photosynthetic machinery.

Phosphatidylglycerol (PG) is a ubiquitous component of thylakoid membranes. Experiments with the pgsA mutant of the cyanobacterium Synechocystis sp. PCC6803 defective in biosynthesis of PG have demonstrated an indispensable role of PG in photosynthesis. In the present study, we have investigated the light susceptibility of the pgsA mutant with regard to the maintenance of the photosynthetic machinery. Growth of the mutant cells without PG increased the light susceptibility of the cells and resulted in severe photoinhibition of photosynthesis upon a high-light treatment, whereas the growth in t

Cyclotriphosphazene appended porphyrins and fulleropyrrolidine complexes as supramolecular multiple photosynthetic reaction centers: steady and excited states photophysical investigation.

New multiple photosynthetic reaction centers were constructed from cyclophosphazene decorated multiporphyrin chromophores and a fulleropyrrolidine having a pyridine ligand (FPY). The excited state electron transfer in the self-assembled donor-acceptor assembly was investigated by using steady state absorption and emission, time-resolved emission spectroscopy and nanosecond laser flash photolysis. The effect of metal (Zn(2+)) coordination to porphyrin units in the multiporphyrin arrays on cyclophosphazine scaffold (P3N3Zn) was studied by comparing with metal free porphyrin assembly on a cycloph

Photoperiod and temperature influence cadmium's effects on photosynthesis-related gene transcription in Chlorella vulgaris.

Routine metal compound toxicity tests are performed at one constant photoperiod and temperature. There is little knowledge about the interactions between metal compound toxicity and photoperiod or temperature. The purpose of this study was to analyze the effects of photoperiod and temperature on cadmium (Cd) toxicity in the fresh alga, Chlorella vulgaris, both singly and in combination. Exposure to 2 or 4 microM Cd alone significantly decreased the transcription of the photosynthesis-related genes psbA, psaB and rbcL. Three-way ANOVA analysis showed that both temperature and photoperiod intera

Evaluation of selected benzoquinones, naphthoquinones, and anthraquinones as replacements for phylloquinone in the A1 acceptor site of the photosystem I reaction center.

Selected substituted 1,4-benzoquinones, 1,4-naphthoquinones, and 9,10-anthraquinones were investigated as possible replacement quinones in spinach photosystem I (PSI) preparations that had been depleted of endogenous phylloquinone by extraction with hexane/methanol. As a criterion for successful biochemical reconstitution, the restoration of electron transfer was determined by measuring P-430 turnover at room temperature from flash-induced absorbance transients. Restoration of complete electron transfer between A0- and P-430 (terminal iron-sulfur centers, FAFB) was demonstrated by using phyllo

Comparative FTIR analysis of the microenvironment of in cyanide-treated, high pH-treated and iron-depleted photosystem II membrane fragments.

FTIR difference spectra due to light-induced formation were measured in control PS II membrane fragments and in samples where the magnetic interaction with the non-heme iron center in its high-spin Fe2+ state is eliminated by three different methods, i.e., extraction of the non-heme iron center, treatment with cyanide, and incubation at high pH (pH = 11). The results obtained reveal that (i) the most pronounced band at 1479 cm-1 reflecting the C../--O stretching mode of in the semiquinone anion radical remains invariant to "iron depletion" while it shifts by 4 and 2 cm-1 to lower frequencies u

Molecular assembly of artificial photosynthetic antenna core complex on an amino-terminated ITO electrode.

Bacterial photosynthetic membrane proteins, light-harvesting antenna complex (LH1), reaction center (RC), and their combined 'core' complex (LH1-RC) are functional elements in the primary photosynthetic events, i.e., capturing and transferring light energy and subsequent charge separation. These photosynthetic units (PSUs) isolated from Rhodospirillum rubrum (Rs. rubrum) were assembled onto an ITO electrode modified with 3-aminopropyltriethoxysilane (APS-ITO). The near IR absorption spectra of PSUs on the assembled electrodes were identical to those of solutions, indicating that the LH1 and LH

Photosynthetic reaction centers/ITO hybrid nanostructure.

Photosynthetic reaction center proteins purified from Rhodobacter sphaeroides purple bacterium were deposited on the surface of indium tin oxide (ITO), a transparent conductive oxide, and the photochemical/-physical properties of the composite were investigated. The kinetics of the light induced absorption change indicated that the RC was active in the composite and there was an interaction between the protein cofactors and the ITO. The electrochromic response of the bacteriopheophytine absorption at 771 nm showed an increased electric field perturbation around this chromophore on the surface

The 110-kDa reaction center protein of photosystem I, P700-chlorophyll a-protein 1, is an iron-sulfur protein.

Germination and growth of barley (Hordeum vulgare L.) in the presence of 59Fe2+ or 35SO4(2-) allows heavy incorporation of both isotopes into the thylakoid membranes and into isolated photosystem I particles. Analysis of 59Fe-labeled preparations by sodium dodecyl sulfate-polyacrylamide gel electrophoresis under mild conditions demonstrates that a minimum of four iron atoms/P700 is carried on P700-chlorophyll a-protein 1. When isolated from 35S-labeled preparations, P700-chlorophyll a-protein 1 binds zero valence 35S, which is converted into acid-labile [35S]sulfide by dithiothreitol reduction

Moderate overexpression of PROTON GRADIENT REGULATION 5 improves photosynthetic performance and plant growth under fluctuating low light in Arabidopsis thaliana.

Cyclic electron transport around photosystem I (PSI) plays an important role in regulating photosynthesis by balancing ATP/NADPH production and maintaining PSI function under fluctuating light. Although PGR5-dependent photosynthetic regulation has been extensively studied under high or fluctuating light, its physiological significance under prolonged low irradiance interrupted by transient increases in light remains less well understood. Here, we analyzed Arabidopsis thaliana lines with different levels of PGR5 accumulation to investigate how PGR5 abundance influences photosynthetic regulation

Immobilization results in sustained calcium transport in Nostoc calcicola Bréb.

The uptake pattern of Ca2+ by the cyanobacterium Nostoc calcicola Bréb in its freely suspended and immobilized form is comprised of two distinct phages; (a) rapid uptake for 1st 10 min followed by (b) slower transport at least up to 60 min. Entrapment of cyanobacterial cells in polyvinyl foam always maintained a higher Ca2+ profile over freely suspended cells. Also, the intracellular Ca2+ concentration was three times more in the former under similar experimental conditions. Whereas, illumination supported maximum Ca2+ transport in all the sets, darkness resulted in drastic reduction (90%) of

Unexpected changes in photosystem I function in a cytochrome c6-deficient mutant of the cyanobacterium Synechocystis PCC 6803.

Cytochrome c6, the product of the petJ gene, is a photosynthetic electron carrier in cyanobacteria, which transfers electrons to photosystem I and which is synthesised under conditions of copper deficiency to functionally replace plastocyanin. The photosystem I photochemical activity (energy storage, photoinduced P700 redox changes) was examined in a petJ-null mutant of Synechocystis PCC 6803. Surprisingly, photosystem I activity in the petJ-null mutant grown in the absence of copper was not much affected. However, in a medium with a low inorganic carbon concentration and with NH4+ ion as nitr

Generation of Bacteriochlorophyll Triplets by the Mutant Reaction Center of Cereibacter sphaeroides G(M71)L/A(M260)W.

In the photosynthetic membrane of the purple non-sulfur bacterium Cereibacter sphaeroides, light energy is absorbed by the pigments of light-harvesting complexes and transferred to the photoreaction center (RC), where it is converted into the electrochemical energy of separated charges. Molecules of the bacteriochlorophyll (BChl) dimer P, monomeric BChl, bacteriopheophytin, and ubiquinone are involved in the process of photoinduced electron transfer in RCs. At all stages of the photochemical process, triplet states of pigments are produced with a certain probability, which in native RCs are qu

Silver Island Film for Enhancing Light Harvesting in Natural Photosynthetic Proteins.

The effects of combining naturally evolved photosynthetic pigment-protein complexes with inorganic functional materials, especially plasmonically active metallic nanostructures, have been a widely studied topic in the last few decades. Besides other applications, it seems to be reasonable using such hybrid systems for designing future biomimetic solar cells. In this paper, we describe selected results that point out to various aspects of the interactions between photosynthetic complexes and plasmonic excitations in Silver Island Films (SIFs). In addition to simple light-harvesting complexes, l

Regulatory plasticity balances photosynthetic electron flow with enhanced pH-dependent cytochrome b6f control in Arabidopsis.

To avoid photodamage of photosystem I under fluctuating light, plants have evolved multiple photoprotective mechanisms. One key mechanism is photosynthetic control, in which acidification of the thylakoid lumen downregulates electron transport through the cytochrome b6f complex, thereby preventing overreduction of photosystem I. The Arabidopsis proton gradient regulation 5 (pgr5) mutant, which is defective in cyclic electron transport around photosystem I, fails to induce photosynthetic control and consequently suffers severe photosystem I photodamage under fluctuating light. Previously, we sh
