Structural Insights and Challenges in the Supercomplexes Formed by Photosystem I in Phototrophs.

Photosystem I (PSI) is a central component of photosynthesis, driving essential processes such as light energy conversion and energy metabolism. This review addresses several knowledge gaps regarding PSI by providing comprehensive insights into the structural diversity of PSI supercomplexes across various evolutionary groups, including cyanobacteria, algae and land plants. It clarifies the oligomerization states of PSI and its interactions with light-harvesting complexes (LHCs) and other protein complexes such as NDH and Cyt b6f. Environmental factors, including light intensity, iron availabil

Photosystem I: towards stable photovoltage generation in a semi-artificial system.

The generation of voltage (ΔV) by purified photosynthetic protein-pigment complex of photosystem I (PS I) from the cyanobacterium Synechocystis sp. PCC 6803 immobilized onto a porous Millipore nitrocellulose membrane filter (MF) and contacting with indium tin oxide (ITO) semiconductor glass slides, termed ITO|PS I - MF|ITO have been directly measured. In the presence of the ascorbate/2,6-dichlorophenolindophenol redox couple and disaccharide trehalose, photoinduced generation of ΔV with a maximum amplitude was observed upon light irradiation for more than 1 h. After storing the ITO|PS I - MF|I

Selective Stabilization of PSI-Associated Electron Transport Network Underlies Cytokinin-Mediated Delay of Leaf Senescence in Barley.

Leaf senescence progressively remodels the photosynthetic apparatus, leading to impaired electron transport and declining carbon assimilation. Here, we investigated how dark-induced senescence (DIS) and exogenous 6-benzyladenine (BA) affect photosystem function, cyclic electron flow (CEF), photosynthetic protein remodeling and CO2 assimilation in two barley (Hordeum vulgare L.) cultivars differing in their senescence characteristics, Carina (spring) and Lomerit (winter). DIS markedly reduced the chlorophyll content, PSI and PSII photochemistry, electron transport and CO2 assimilation in both c

Canthaxanthin substitution of β-carotene yields structurally stable yet spectrally robust photosystem I cores in Synechocystis sp. PCC 6803.

Carotenoids (Cars) are essential cofactors in photosystem I (PSI), where they stabilize the pigment-protein framework and support excitation-energy transfer and photoprotection. Although metabolic engineering has enabled the production of diverse keto-carotenoids (keto-Cars) across oxyphototrophs, the structural and photophysical consequences of installing a single engineered Car into purified PSI cores remain unclear. Here, we generated a Synechocystis sp. PCC 6803 strain expressing only the crtW gene (CrtW strain), which converts β-carotene to canthaxanthin, and isolated PSI trimers to evalu

Coordination of cyclic electron flow and alternative electron sinks adjusts photosynthetic regulation in maize under fluctuating light during combined heat and drought stress.

Field-grown maize is frequently exposed to fluctuating light (FL) combined with heat and drought during summer, yet the underlying photosynthetic regulatory mechanisms remain poorly understood. We examined chlorophyll fluorescence, the P700 redox state, and electrochromic shift signals in maize leaves subjected to FL under individual and compound stresses of moderate heat and drought. Following short-term FL treatment, photosystem I (PSI) photoinhibition was not exacerbated by any stress treatment, whereas photosystem II (PSII) photoinhibition was significantly aggravated by heat and compound

Ocean acidification exacerbates UVR-induced inhibition of photosystem II and I in Corallina officinalis.

Ocean acidification (OA) exerts diverse effects on marine macroalgae, with calcified species being particularly vulnerable. Due to calcified skeletons can contribute to physical screening against solar ultraviolet radiation (UVR), OA-driven calcification loss may increase exposure of the photosynthetic apparatus to UVR. Here, we cultured Corallina officinalis under ambient CO₂ (∼420 μatm) or elevated CO₂ (∼1000 μatm), with or without UVR, under natural solar radiation. Our results confirmed that OA reduced calcification and, under UVR, enhanced donor-side impairment of photosystem II (PSII), a

Structural insights into assembly and environmental adaptation of photosystem I from a lichen green alga.

Photosystem I (PSI) of terrestrial plants typically contains an antenna system smaller than that of aquatic algae, presumably an adaptation to terrestrial high-light conditions. Moreover, green plants face multiple other stresses including drought when colonizing terrestrial environments and have evolved effective protective mechanisms. However, how exactly PSI antenna size is reduced and the precise nature of these photoprotective strategies remain to be elucidated. Lichens, composed of a mycobiont and a phycobiont, represent a symbiotic life form that thrives under terrestrial-like stress, w

Function of iron-stress-induced protein A in cyanobacterial cells with monomeric and trimeric photosystem I.

The acclimation of cyanobacteria to iron deficiency is crucial for their survival in natural environments. In response to iron deficiency, many cyanobacterial species induce the production of a pigment-protein complex called iron-stress-induced protein A (IsiA). IsiA proteins associate with photosystem I (PSI) and can function as light-harvesting antennas or dissipate excess energy. They may also serve as chlorophyll storage during iron limitation. In this study, we examined the functional role of IsiA in cells of Synechocystis sp. PCC 6803 grown under iron limitation conditions by measuring t

Removal of Photosystem I Subunit PsaE Leads to Photosynthetic Hydrogen Production in a Cyanobacterial PsaF-HoxYH Fusion Strain.

Photosynthetic hydrogen (photoH2) production is a promising approach for the utilisation of solar energy. Fusion of a hydrogenase (H2ase) to photosystem I (PSI) provides a direct light-driven route for photoH2 generation. We established the lateral transmembrane subunit PsaF of PSI as a new fusion partner for the NiFe-hydrogenase (HoxYH) in the cyanobacterium Synechocystis sp. PCC 6803. Gradual reduction of PsaC expression in a PsaF-HoxYH strain was established to access the most redox negative FeS-cluster FX of PSI, but this did not result in photoH2 production. In another approach PSI subuni

Acclimation of Chlamydomonas reinhardtii to different growth irradiances.

We report on the changes the photosynthetic apparatus of Chlamydomonas reinhardtii undergoes upon acclimation to different light intensity. When grown in high light, cells had a faster growth rate and higher biomass production compared with low and control light conditions. However, cells acclimated to low light intensity are indeed able to produce more biomass per photon available as compared with high light-acclimated cells, which dissipate as heat a large part of light absorbed, thus reducing their photosynthetic efficiency. This dissipative state is strictly dependent on the accumulation o

Comprehensive analysis of the effects on photosynthesis and energy balance in tomato leaves under magnesium deficiency.

Magnesium (Mg), as an essential and central mineral element for chlorophyll biosynthesis, plays a crucial role in plant photosynthesis. Magnesium deficiency inevitably affects the photosynthetic ability of leaves, thereby impairing the yield and quality of crops. However, few studies have revealed the intrinsic mechanisms by which Mg deficiency hinders growth and photosynthesis, particularly by analyzing the processes of light capture, dissipation, absorption, and utilization in tomato. The experiment studied the effects of Mg deficiency on internal structure of leaves, light absorption, elect

Microdomains heterogeneity in the thylakoid membrane proteins visualized by super-resolution microscopy.

The investigation of spatial heterogeneity within the thylakoid membrane (TM) proteins has gained increasing attention in photosynthetic research. The recent advances in live-cell imaging have allowed the identification of heterogeneous organisation of photosystems in small cyanobacterial cells. These sub-micrometre TM regions, termed microdomains in cyanobacteria, exhibit functional similarities with granal (Photosystem II dominant) and stromal (Photosystem I dominant) regions observed in TM of higher plants. This study delves into microdomain heterogeneity using super-resolution Airyscan-bas

Structural basis for energy and electron transfer of the photosystem I-IsiA-flavodoxin supercomplex.

Under iron-deficiency stress, which occurs frequently in natural aquatic environments, cyanobacteria reduce the amount of iron-enriched proteins, including photosystem I (PSI) and ferredoxin (Fd), and upregulate the expression of iron-stress-induced proteins A and B (IsiA and flavodoxin (Fld)). Multiple IsiAs function as the peripheral antennae that encircle the PSI core, whereas Fld replaces Fd as the electron receptor of PSI. Here, we report the structures of the PSI3-IsiA18-Fld3 and PSI3-IsiA18 supercomplexes from Synechococcus sp. PCC 7942, revealing features that are different from the pr

Iron allocation to chloroplast proteins depends on the DNA-binding protein WHIRLY1.

The DNA-binding protein WHIRLY1, sharing structural similarities with ferritin, plays a role in the formation of iron cofactor proteins within chloroplasts. Previous studies indicated that barley plants with a knockdown of HvWHIRLY1 containing a minimal amount of the protein are compromised in chloroplast development and photosynthesis, and get chlorotic leaves when grown at high irradiance. Thereby, the leaves display signs of iron deficiency. Metal determination revealed, however, that leaves of WHIRLY1-deficient plants had a regular iron content. Nevertheless, WHIRLY1-deficiency affected th

Light intensity dependent photosynthetic electron transport in eelgrass (Zostera marina L.).

Responses of electron transport to three levels of irradiation (20, 200, and 1200 μmol photons m-2 s-1 PAR; exposures called LL, ML and HL, respectively) were investigated in eelgrass (Zostera marina L.) utilizing the chlorophyll a fluorescence technique. Exposure to ML and HL reduced the maximum quantum yield of photosystem II (PSII) (Fv/Fm) and the maximum slope decrease of MR/MRO (VPSI), indicating the occurrence of photoinhibition of both PSII and photosystem I (PSI). A comparatively slow recovery rate of Fv/Fm due to longer half-life recovery time of PSII and 40% lower descending amplitud

Light quantity and photosystem function mediate host susceptibility to Turnip mosaic virus via a salicylic acid-independent mechanism.

Evidence going as far back as the early part of the 20th century suggests that both light and chloroplast function may play key roles in host susceptibility to viruses. Despite the long history of such work, confirmation of these phenomena and a determination of the underlying mechanisms remain elusive. Here, we revisited these questions using modern imaging technologies to study the susceptibility of Nicotiana benthamiana to Turnip mosaic virus (TuMV). We found that both light deficiency and photosystem impairment increased the susceptibility of N. benthamiana to TuMV infection. Time-lapse ph

Physiological characterisation of magnesium deficiency in sugar beet: acclimation to low magnesium differentially affects photosystems I and II.

Magnesium deficiency in plants is a widespread problem, affecting productivity and quality in agriculture, yet at a physiological level it has been poorly studied in crop plants. Here, a physiological characterization of Mg deficiency in Beta vulgaris L., an important crop model, is presented. The impact of Mg deficiency on plant growth, mineral profile and photosynthetic activity was studied. The aerial biomass of plants decreased after 24 days of hydroponic culture in Mg-free nutrient solution, whereas the root biomass was unaffected. Analysis of mineral profiles revealed that Mg decreased m

Structural and spectral adaptation of the seagrass Posidonia oceanica photosystem I to seabed light.

Seagrasses are marine angiosperms re-adapted to underwater life, forming productive ecosystems and long-term carbon sinks. Posidonia oceanica thrives up to 50 m depth, where light is scarce and spectrally shifted; yet, the molecular basis of its photosynthetic adaptation remains unclear. Here, we report that P. oceanica genetically adapts for highly efficient photon use under dim light by enhancing photosystem antenna size and reducing exciton trapping time. We determine the structures of P. oceanica PSI supercomplexes by cryo-electron microscopy, revealing an expanded antenna system composed

Excitonic energy transfer in red algal Photosystem I reveals an evolutionary bridge between cyanobacteria and plants.

Photosystem I (PSI) converts light into chemical energy with near-unity quantum efficiency, yet its energy-transfer and charge-separation mechanisms remain debated. Evolution has diversified PSI architectures: Cyanobacterial PSI trimers confine red-shifted pigments to the core, whereas plant PSI-Light Harvesting Complex Isupercomplexes incorporate extensive peripheral red and charge-transfer states that reshape trapping. The unicellular red alga Cyanidioschyzon merolae exemplifies functional diversification across distinct evolutionary branches, combining a photosystem II and plant-like monome

Moderate heat stress has been reported to increase PSI cyclic electron flow (CEF). We subjected leaves of Arabidopsis (Arabidopsis thaliana) mutants disrupted in the regulation of one or the other pathway of CEF flow-crr2 (chlororespiratory reduction, deficient in regulation of chloroplast NAD(P)H dehydrogenase-dependent CEF) and pgr5 (proton gradient regulation, proposed to have reduced efficiency of antimycin-A-sensitive-CEF regulation) to moderate heat stress.
