Application of Simultaneous Active and Passive Fluorescence Observations: Extending a Fluorescence-Based qL Estimation Model.

The fraction of open Photosystem II (PSII) reaction centers (qL) is critical for connecting broadband PSII fluorescence (ChlFPSII) with the actual electron transport from PSII to Photosystem I. Accurately estimating qL is fundamental for determining ChlFPSII, which, in turn, is vital for mechanistically estimating the actual electron transport rate and photosynthetic CO2 assimilation. Chlorophyll fluorescence provides direct physiological insights, offering a robust foundation for qL estimation. However, uncertainties in the ChlFPSII-qL relationship across different plant functional types (PFT

Detergent Choice Shapes the Solution Structures of Photosystems I and II: Implications for Crystallization and High-Resolution Studies.

Photosystems I (PSI) and II (PSII) are pigment-protein complexes that perform the light-driven charge separation necessary to convert solar energy into a biochemically usable form in a fundamental process called photosynthesis. Small-angle X-ray scattering provides unique structural insights into PSI and PSII in solution under near-physiological conditions. Here, we study the solubilization of PSI and PSII with different detergents, the octaethylene glycol monododecyl ether (C12E8) and the most commonly used n-dodecyl-β-D-maltoside (DDM). It is noteworthy that the volume of the C12E8 detergent

Daily Heat Stress Induces Donor-Side Limitation of PSI Via Downregulation of the Cyt b6f Complex.

The rapidly warming climate is driving increasingly frequent and intense heat waves worldwide, posing major challenges to plant metabolism and growth. Despite the central role of photosynthesis in plant growth, the effects of prolonged heat stress on the primary reactions of photosynthesis remain poorly understood. Here, we studied how long-term heat exposure affects growth and photosynthetic performance in the model plant Arabidopsis thaliana. Plants were exposed daily to high temperature (38°C) for 4 h throughout the growth period. Conflicting to earlier studies, we show that PSII-LHCII rema

Differences between barley and maize revealed in limitations of photosystems I and II under high temperature and low air humidity.

Non-photochemical quenching and limitations of the photosystem I and photosystem II activities were studied in C3 -plant barley and C4 -plant maize. Plants were exposed to prolonged heat stress under high and low air humidity. Both species decreased non-photochemical quenching at 37-42°C, which increased at 46°C. A decrease of photosystem II activity at 46°C in lower air humidity was achieved through different mechanisms. In barley, photosystem II was downregulated by the increase of non-photochemical quenching. In maize, photosystem II was downregulated by the increase of acceptor-side limita

Photochemistry of Photosystems II and I in Rice Plants Grown under Different N Levels at Normal and High Temperature.

Although N levels affect leaf photosynthetic capacity, the effects of N levels on the photochemistry of photosystems II and I (PSII and PSI, respectively) are not well-understood. In the present study, we examined this aspect in rice (Oryza sativa L. 'Hitomebore') plants grown under three different N levels at normal or high temperatures that can occur during rice culture and do not severely suppress photosynthesis. At both growth temperatures, the quantum efficiency of PSII [Y(II)] and the fraction of the primary quinone electron acceptor in its oxidized state were positively correlated with

Electron Transfer on the Donor Side of Manganese-Depleted Photosystem 2.

After removal of manganese ions responsible for light-driven water oxidation, redox-active tyrosine YZ (tyrosine 161 of the D1 subunit) still remains the dominant electron donor to the photooxidized chlorophyll P680 (P680+) in the reaction center of photosystem 2 (PS2). Here, we investigated P680+ reduction by YZ under single-turnover flashes in Mn-depleted PS2 core complexes in the presence of weak acids and NH4Cl. Analysis of changes in the light-induced absorption at 830 nm (reflecting P680 redox transitions) at pH 6.0 showed that P680+ reduction is well approximated by two kinetic componen

[Effect of Methylmercury on the Light Dependence Fluorescence Parameters in a Green Alga Chlamydomonas moewusii].

The effect of a dangerous toxic substance, methylmercury, on light dependence curves of chlorophyll fluorescence in Chlamydomonas moewusii was studied. We found low concentration of methylmercury (10(-7) M) to cause a decrease in the relative rate of the non-cyclic electron transport activity of PS 2, a decline in the maximum utilization of light energy (α), and a decline in the saturation light intensity (E(s)). Non-photochemical fluorescence quenching increased after short-term exposure and decreased in the course of prolonged incubation. These parameters were more sensitive to the action of

Trade-offs between photosynthetic capacity and structural economy underlie the rapid proliferation of floating Ulva prolifera.

The green tide alga Ulva prolifera shifts from an attached to a floating state during bloom formation, yet the physiological basis of this transition remains unclear. Here, using attached and floating thalli derived from a single parental clonal line and cultured under identical controlled conditions, we combined physiological measurements, protein-fraction analysis, and transcriptomics to compare growth and resource-allocation patterns between the two states. Despite similar bulk carbon and nitrogen contents, floating thalli reached significantly greater thallus length than attached thalli. A

Hydrogen sulfide improves photosynthetic efficiency by regulating light energy dissipation and reversible phosphorylation of thylakoid proteins in rice under salt stress.

Hydrogen sulfide (H2S) has been regarded as a small gasotransmitter associated with physiological and biochemical regulation in plant responses to environmental stresses. However, the regulatory mechanisms of H2S in photosynthesis under adverse conditions remains poorly understood in plants. Here, the role of H2S in the regulation of photosystem I (PSI) and photosystem II (PSII) was investigated in rice seedlings subjected to salt stress. Our results showed that NaHS (H2S donor) pretreatment significantly enhanced photosynthetic pigment content, gas exchange parameters, and the photochemical c

Psb28 protein is involved in the biogenesis of the photosystem II inner antenna CP47 (PsbB) in the cyanobacterium Synechocystis sp. PCC 6803.

The role of the Psb28 protein in the structure and function of the photosystem II (PSII) complex has been studied in the cyanobacterium Synechocystis sp. PCC 6803. The protein was localized in the membrane fraction and, whereas most of the protein was detected as an unassembled protein, a small portion was found in the PSII core complex lacking the CP43 antenna (RC47). The association of Psb28 with RC47 was further confirmed by preferential isolation of RC47 from the strain containing a histidine-tagged derivative of Psb28 using nickel-affinity chromatography. However, the affinity-purified fr

Metabolic Capacity of the Antarctic Cyanobacterium Phormidium pseudopriestleyi That Sustains Oxygenic Photosynthesis in the Presence of Hydrogen Sulfide.

Sulfide inhibits oxygenic photosynthesis by blocking electron transfer between H2O and the oxygen-evolving complex in the D1 protein of Photosystem II. The ability of cyanobacteria to counter this effect has implications for understanding the productivity of benthic microbial mats in sulfidic environments throughout Earth history. In Lake Fryxell, Antarctica, the benthic, filamentous cyanobacterium Phormidium pseudopriestleyi creates a 1-2 mm thick layer of 50 µmol L-1 O2 in otherwise sulfidic water, demonstrating that it sustains oxygenic photosynthesis in the presence of sulfide. Light-Harvesting Protein LHCB4 Is Important for Stable Dimeric Organization of Photosystem II.

Light-harvesting protein LHCB4 is a monomeric antenna of photosystem II (PSII) that connects the PSII core (C) with strongly (S) and moderately (M) bound LHCII trimers, forming the stable dimeric supercomplex C2S2M2. LHCB4 is known to stabilize LHCII attachment to the PSII core dimer (C2). Here, we show that LHCB4 is essential for the intrinsic stability of the PSII core dimer itself, a fundamental structural unit required for efficient photochemistry. In the lhcb4 mutant, clear-native PAGE revealed the absence of C2S2M(2) and only small amounts of C2S2 and C2S supercomplexes. Single-particle

From proton microdomains to protein migration: hypothesis for ΔpH-controlled PSII relocation.

Thylakoid membranes are highly organized yet dynamic systems in which Photosystem II (PSII) complexes undergo continual assembly, repair and lateral redistribution. Here a hypothesis is proposed that local lumenal pH, generated by the trans-thylakoidal proton gradient and potentially exhibiting transient spatial heterogeneity, may contribute to the regulation of protein movement and PSII maturation within the thylakoid membrane. Five mechanistically distinct but potentially coupled routes by which ΔpH could influence protein dynamics are outlined: (i) pH-dependent modulation of lumenal protein

Plant sample integrity is important for efficient photosystem II repair and thylakoid remodelling in Arabidopsis.

The thylakoid is a dynamic membrane that houses the protein complexes involved in the light-dependent reactions of photosynthesis. Although light provides the energy that drives these reactions, it also damages the photosynthetic apparatus, particularly photosystem II. This damage can be repaired via the photosystem II repair cycle, a process thought to be accompanied by structural rearrangements of the thylakoid membrane. When the light-induced damage exceeds the capacity for repair, photoinhibition occurs, resulting in a decline in photosynthetic capacity. While photoinhibition has been exte

Limited Responsiveness of Chloroplast Gene Expression during Acclimation to High Light in Tobacco.

Acclimation to changing light intensities poses major challenges to plant metabolism and has been shown to involve regulatory adjustments in chloroplast gene expression. However, this regulation has not been examined at a plastid genome-wide level and for many genes, it is unknown whether their expression responds to altered light intensities. Here, we applied comparative ribosome profiling and transcriptomic experiments to analyze changes in chloroplast transcript accumulation and translation in leaves of tobacco (Nicotiana tabacum) seedlings after transfer from moderate light to physiologica

Compensatory respiration supports survival during extended heat stress in Microcystis aeruginosa.

Phototrophic organisms must balance energy between photosynthesis and respiration to survive. In cyanobacteria, this coordination is especially critical as both processes share the same membrane and electron transport complexes. Although electron exchange between the two pathways is known, photosynthesis is often presumed to dominate under many conditions. Here, under an extended heat-stress condition, our data indicate that respiration can compensate for reduced photosynthetic activity. We compared two Microcystis aeruginosa strains (PCC7806 and Lake Kinneret isolated strain C-1004). PCC7806

Mechanisms underpinning natural variation in non-photochemical quenching kinetics.

Plants use light as an energy source to reduce carbon dioxide into carbohydrates during photosynthesis. However, when the incident light exceeds the photosynthesis rate, the excess energy must be dispersed, or it can result in the unregulated formation of harmful reactive oxygen species, especially in plants exposed to very high light or abiotic stress conditions that compromise photosynthetic efficiency. The excess energy is typically dispersed harmlessly as heat, which can be measured as non-photochemical quenching (NPQ) of chlorophyll fluorescence. NPQ kinetics vary within plant populations

High light intensity aggravates latent manganese deficiency in maize.

Manganese (Mn) plays an important role in the oxygen-evolving complex, where energy from light absorption is used for water splitting. Although changes in light intensity and Mn status can interfere with the functionality of the photosynthetic apparatus, the interaction between these two factors and the underlying mechanisms remain largely unknown. Here, maize seedlings were grown hydroponically and exposed to two different light intensities under Mn-sufficient or -deficient conditions. No visual Mn deficiency symptoms appeared even though the foliar Mn concentration in the Mn-deficient treatm

Effects of cadmium on photosynthesis of Schima superba young plant detected by chlorophyll fluorescence.

Contamination by heavy metals has become a serious environmental pollution issue today due to its potential threat to plant, wildlife, and human health. Photosynthesis, a process in which light energy is used to produce sugar and other organic compounds, is sensitive to heavy metals. In the present study, the response of photosynthetic process and carbon assimilation of Schima superba was investigated under cadmium (Cd) stress. Three Cd concentrations (0, 300, and 600 mg kg-1) were used designated as control (CK), low Cd (L1), and high Cd treatment (L2) of plants. Results showed that photosyst
