Shedding light on the effects of hydraulic fracturing flowback and produced water on phototactic behavior in Daphnia magna.

The effluent produced during hydraulic fracturing (i.e. flowback and produced water; FPW), is a complex hyper-saline solution that is known to negatively impact the survival and the fitness of the water flea Daphnia magna, but to date effects on behavior are unstudied. In the current study, the effects of FPW on phototactic behavior of D. magna were examined. Exposure of naïve animals to FPW resulted in a dose-dependent increase in the speed of appearance of daphnids in the illuminated zone of the test apparatus (i.e. a faster positive phototaxis response). A similar dose-dependent response wa

Photoresponse in the heterotrophic marine dinoflagellate Oxyrrhis marina.

Expressed rhodopsins were detected by proteomic analysis in an investigation of potential signal receptors in the cell membrane of the marine heterotrophic dinoflagellate Oxyrrhis marina (CCMP604). We inferred these to be sensory rhodopsins, a type of G-protein-coupled receptor trans-membrane signaling molecule. Because phototactic behavior based on sensory rhodopsins has been reported in other protists, we investigated the photosensory response of O. marina. This dinoflagellate exhibited strongest positive phototaxis at low levels (2-3 μE/m(2)/s) of white light when the cells were previously

The role of serotonergic signaling on phototactic and locomotor behavior in Daphnia magna.

The role of serotonin in Daphnia magna phototactic and locomotor behavior was assessed using reverse genetics and pharmacological treatments with serotonin and fluoxetine. The study was conducted with four clones: the wild type clone and three CRISPR D. magna ones with mutations in the tryptophan hydrolase gene (TRH) that is involved in serotonin synthesis. These included clones TRHA- and TRHB- with mutations in both alleles that lack serotonin and the mono-allelic mutant TRH+, that has serotonin. Obtained results indicated that animals lacking serotonin showed an increased negative phototacti

Blue Light Is a Universal Signal for Escherichia coli Chemoreceptors.

Blue light has been shown to elicit a tumbling response in Escherichia coli, a nonphototrophic bacterium. The exact mechanism of this phototactic response is still unknown. Here, we quantify phototaxis in E. coli by analyzing single-cell trajectories in populations of free-swimming bacteria before and after light exposure. Bacterial strains expressing only one type of chemoreceptor reveal that all five E. coli receptors (Aer, Tar, Tsr, Tap, and Trg) are capable of mediating responses to light. In particular, light exposure elicits a running response in the Tap-only strain, the opposite of the

The fos homolog kayak is required for adult eye formation and function in Drosophila.

This study characterizes the requirements of the kayak (kay) gene in Drosophila melanogaster adult eye biology by examining mutant phenotypes in photoreceptor development, external eye morphology, corneal and bristle ultrastructure, and visually guided behaviors, such as phototaxis, in kay strong loss-of-function homozygous mutant clones. Despite previous studies on kay, there is a dearth of phenotypic characterization of the morphological and behavioral consequences of kay loss-of-function alleles in the adult eye. We find that kay is expressed in developing ommatidia in eye discs. The kay mu

Photo-tropotaxis based on projection through the cerebral commissure in the terrestrial slug Limax.

In the terrestrial slug, Limax, eyes are located at the tip of the superior tentacles. This animal has long been believed to show negative phototaxis through tropotaxis, i.e., it compares the two light intensities detected by bilateral eyes to move away from a light source. As one of the possible manifestations of such negative phototaxis, a circling movement has been observed: if one of the superior tentacles is removed, the slugs continuously move in the direction of the removed side. However, there has been no evidence demonstrating that this behavior is actually based on negative phototrop

Asymmetric distribution of type IV pili triggered by directional light in unicellular cyanobacteria.

The type IV pili (T4P) system is a supermolecular machine observed in prokaryotes. Cells repeat the cycle of T4P extension, surface attachment, and retraction to drive twitching motility. Although the properties of T4P as a motor have been scrutinized with biophysics techniques, the mechanism of regulation remains unclear. Here we provided the framework of the T4P dynamics at the single-cell level in Synechocystis sp. PCC6803, which can recognize light direction. We demonstrated that the dynamics was detected by fluorescent beads under an optical microscope and controlled by blue light that in

Infrared light photobiostimulation mediates periodicity in Dirofilaria immitis microfilariae.

Microfilariae (Mfs) of filarial nematode parasites exhibit nocturnal periodicity, with their numbers in peripheral blood peaking at night and decreasing during the day. However, the reason for their appearance at night remains unknown. In this study, in vitro photobiostimulation experiments showed that Mfs exhibited positive phototaxis toward infrared light with lower photon flux densities of infrared light at wavelengths of 890 and 700 nm, in particular, mediating paradoxically higher velocity than intense ones. Microarray analysis revealed that infrared light stimulation influenced gene expr

Why does the effectiveness of light trapping for tea leafhoppers decline sharply in late autumn?

Light trapping is a common monitoring and control method for Empoasca onukii Matsuda (Hemiptera: Cicadellidae), a major pest in tea gardens. However, in late autumn (late October), despite large populations of leafhoppers in tea gardens around 30°N latitude, the effectiveness of light trapping declines sharply. The factors causing this decline are unclear. In this study, field experiments revealed significant differences in ambient temperature and photoperiod corresponding to variation in light trapping effectiveness across different leafhopper peaks. Through a series of bioassays, we found th

Direct and Ambient Light Pollution Alters Recruitment for a Diurnal Plant-Pollinator System.

Artificial light at night (ALAN) functions as a novel environmental stimulus that has the potential to disrupt interactions among species. Despite recent efforts to explain nocturnal pollinators' responses to this stimulus, the likelihood and associated mechanisms of attraction toward artificial light and potential consequences on fitness for diurnal pollinators are still largely unclear. Here, we took advantage of the obligate mutualism between yucca moths (Tegeticula maculata maculata) and yucca plants (Hesperoyucca whipplei) to understand how direct light exposure and skyglow can influence

Photophobic and phototactic responses of Amoeba proteus in KCN and SHAM solutions.

The effects of the metabolic inhibitors KCN and SHAM on phototaxis and photophobic response in Amoeba proteus have been studied. Both drugs neither change amoebae photophobic response nor the phototactic reaction. The results indicate clearly that the negative phototactic orientation is not impaired by impediment of respiration thus it is not directly coupled to the differences in energy production in different parts of A. proteus body.

Evolution of Proteins of the DNA Photolyase/Cryptochrome Family.

Proteins of the cryptochrome/DNA photolyase family (CPF) are phylogenetically related and structurally conserved flavoproteins that perform various functions. DNA photolyases repair DNA damage caused by UV-B radiation by exposure to UV-A/blue light simultaneously or subsequently. Cryptochromes are photoreceptor proteins regulating circadian clock, morphogenesis, phototaxis, and other responses to UV and blue light in various organisms. The review describes the structure and functions of CPF proteins, their evolutionary relationship, and possible functions of the CPF ancestor protein.

Xylazine, veterinary anesthetic and recreational drug adulterant, reduces cephalic regeneration and alters defensive responding in planarians.

Xylazine is a veterinary anesthetic and α2-adrenoceptor agonist found in illicit drug supplies. Injection of opioids adulterated with xylazine is correlated with necrotic tissue injury and a withdrawal syndrome including anxiety. Planarians are flatworms with a simple nervous system and no circulatory system that exhibit several characteristics, including regenerative capacity and negative phototaxis (light avoidance). Because regeneration reflects tissue repair and negative phototaxis constitutes defensive behavior, planarians provide a simple in vivo system to explore possible effects of xyl

Psilocybin induces stereotyped movements and reduces defensive responding in planarians through 5-hydroxytryptamine mechanisms.

Psilocybin is a serotonergic 5-HT 2A R agonist that causes psychedelic and anxiolytic effects in human users. To delineate conservation of psilocybin pharmacology, we investigated behavioral effects of psilocybin in planarians ( Dugesia dorotocephala ), the simplest living animal with cephalization that also has a well defined serotonin (5-hydroxytryptamine [5-HT]) system. We quantified stereotyped movements (e.g. head bops, twists, scrunches, and C-shapes) and defensive responding (negative phototaxis) and probed a 5-HT 2A R mechanism for psilocybin using a selective 5-HT 2A R antagonist (vol

Reduction of coastal lighting decreases seabird strandings.

Artificial light at night (ALAN) is negatively impacting numerous species of nocturnally active birds. Nocturnal positive phototaxis, the movement towards ALAN, is exhibited by many marine birds and can result in stranding on land. Seabird species facing major population declines may be most at risk. Leach's Storm-Petrels (Hydrobates leucorhous) are small, threatened seabirds with an extensive breeding range in the North Atlantic and North Pacific Oceans. The Atlantic population, which represents approximately 40-48% of the global population, is declining sharply. Nocturnal positive phototaxis

Photoregulation in prokaryotes.

The spectroscopic identification of sensory rhodopsin I by Bogomolni and Spudich in 1982 provided a molecular link between the light environment and phototaxis in Halobacterium salinarum, and thus laid the foundation for the study of signal transducing photosensors in prokaryotes. In recent years, a number of new prokaryotic photosensory receptors have been discovered across a broad range of taxa, including dozens in chemotrophic species. Among these photoreceptors are new classes of rhodopsins, BLUF-domain proteins, bacteriophytochromes, cryptochromes, and LOV-family photosensors. Genetic and

Diet and energy-sensing inputs affect TorC1-mediated axon misrouting but not TorC2-directed synapse growth in a Drosophila model of tuberous sclerosis.

The Target of Rapamycin (TOR) growth regulatory system is influenced by a number of different inputs, including growth factor signaling, nutrient availability, and cellular energy levels. While the effects of TOR on cell and organismal growth have been well characterized, this pathway also has profound effects on neural development and behavior. Hyperactivation of the TOR pathway by mutations in the upstream TOR inhibitors TSC1 (tuberous sclerosis complex 1) or TSC2 promotes benign tumors and neurological and behavioral deficits, a syndrome known as tuberous sclerosis (TS). In Drosophila, neur

The effects of xylazine on locomotion and motor behaviour in a planarian model.

In recent years, the recreational use of xylazine has increased dramatically in the USA. Although xylazine has been used as an anesthetic in veterinary medicine for decades, little is known about its behavioral effects. We took advantage of the planarian's innate negative phototaxis, the reliable movement from the light side to the dark side of a Petri dish, to explore the organism's suitability as an animal model for investigating the preclinical pharmacology of xylazine. In two experiments, we tested the effects of several doses of xylazine on locomotion by recording the latency to transitio

Albino Xenopus laevis tadpoles prefer dark environments compared to wild type.

Tadpoles display preferences for different environments but the sensory modalities that govern these choices are not well understood. Here, we examined light preferences and associated sensory mechanisms of albino and wild-type Xenopus laevis tadpoles. We found that albino tadpoles spent more time in darker environments compared to the wild type, although they showed no differences in overall activity. This preference persisted when the tadpoles had their optic nerve severed or pineal glands removed, suggesting these sensory systems alone are not necessary for phototaxis. These experiments wer

The E3 ligase ube3a is required for learning in Drosophila melanogaster.

Angelman syndrome and autism are neurodevelopmental disorders linked to mutations and duplications of an E3 ligase called ube3a respectively. Since cognitive deficits and learning disabilities are hallmark symptoms of both these disorders, we investigated a role for dube3a in the learning ability of flies using the aversive phototaxis suppression assay. We show that down and up-regulation of dube3a are both detrimental to learning in larvae and adults. Using conditional gene expression we found that dube3a is required for normal brain development and during adulthood. Furthermore, we suggest t
