Induction of quinone reductase (QR) by withanolides isolated from Physalis pubescens L. (Solanaceae).

In the present study, it was demonstrated that the dichloromethane extract of Physalis pubescens L. (DEPP) had weak potential quinone reductase (QR) inducing activity, but an UPLC-ESI-MS method with glutathione (GSH) as the substrate revealed that the DEPP had electrophiles (with an α,β-unsaturated ketone moiety). These electrophiles could induce quinone reductase (QR) activity, which might be attributed to the modification of the highly reactive cysteine residues in Keap1. Herein, four withanolides, including three new compounds physapubescin B (2), physapubescin C (3), physapubescin D (4), t

PgZHD18-PgWDR5b increases basal thermotolerance in Physalis grisea by lignin biosynthesis dependent on H3K4me3.

Physalis grisea is a widely valued orphan crop with medicinal properties and edible functional fruits. Under global warming, high temperature stress (HTS) severely restricts the growth and development of P. grisea, while the epigenetic regulatory mechanism underlying its HTS response remains largely unclear. In this study, we identified the ZINC FINGER HOMEODOMAIN (ZHD) transcription factor PgZHD18 as a positive regulator of basal thermotolerance in P. grisea. Functional assays showed that PgZHD18 enhances basal thermotolerance by improving ROS scavenging capacity, alleviating membrane damage,

Establishing Physalis as a Solanaceae model system enables genetic reevaluation of the inflated calyx syndrome.

The highly diverse Solanaceae family contains several widely studied models and crop species. Fully exploring, appreciating, and exploiting this diversity requires additional model systems. Particularly promising are orphan fruit crops in the genus Physalis, which occupy a key evolutionary position in the Solanaceae and capture understudied variation in traits such as inflorescence complexity, fruit ripening and metabolites, disease and insect resistance, self-compatibility, and most notable, the striking inflated calyx syndrome (ICS), an evolutionary novelty found across angiosperms where sep

Physalis angulata Calyces Modulate Macrophage Polarization and Alleviate Chemically Induced Intestinal Inflammation in Mice.

As part of the search for new bioactive plants from the Colombian Caribbean, the dichloromethane fraction of the calyces of Physalis angulata L. (PADF) was selected for its anti-inflammatory activity. In this work, we investigated the immunomodulatory effect of PADF in activated macrophages and during dextran sulfate sodium (DSS)-induced colitis. PADF displayed a low content of withanolides or phenolic compounds, and a higher content of sucrose esters, representative anti-inflammatory metabolites of the Physalis genus. The PADF fraction at 12.5 μg/mL prevented the induction of interleukin (IL)

STING Agonist-Loaded Physalis Mottle Virus-like Particles as a Potent Prophylactic for Metastatic Melanoma.

Stimulator of interferon genes (STING) agonists have been presented as a compelling cancer therapeutic for many years yet have been unable to break past clinical trials. To date, the best use cases for STING agonists are as intratumoral therapeutics for solid tumors, but in human trials, rapid wash-out and low efficacy hamper success. Systemic administrations of STING agonists have also been explored, but hurdles include enzymatic instability, leading to rapid clearance and immune toxicity. To overcome these challenges, nanoparticle delivery technologies have been proposed, and in the present
