The sequence of regulatory events in the sporulation control network of Physarum polycephalum analysed by time-resolved somatic complementation of mutants.

The developmental decision for sporulation of Physarum polycephalum plasmodia is under sensory control by environmental factors like visible light or heat shock and endogenous signals like glucose starvation. Several hours after perceiving an inductive stimulus, plasmodia become committed to sporulation; thereby, they lose their unlimited replicative potential and execute a developmental program that involves differentiation into various cell types required to form a mature fruiting body. Plasmodia are multinuclear single cells which spontaneously fuse upon physical contact. Fusion of mutant p

circRNA03358-directed metabolic reprogramming confers Physarum flavicomum GD217 with specific resistance to TiO2 nanoparticles: Moving toward the path of combating nanoparticle pollution/exposure.

The increasing use of titanium dioxide (TiO2) nanoparticles (NPs) has raised concerns related to their environmental accumulation and the associated ecological risks. Understanding the key biomolecular responses of TiO₂ NP-tolerant organisms like Physarum flavicomum GD217 is essential for combating the pollution of and exposure to these NPs. In this study, we employed multi-omics approaches combined with molecular biology techniques to investigate the stress responses of GD217 to mixed-phase TiO₂ NPs (M-TiO₂ NPs). We found that GD217's response is primarily metabolism-related and identified ci

Consequences of impeding in mitochondrial function in Physarum polycephalum. I. Reversible effects of anoxia, KCN, and influences of the Ca2+ ionophore A-23187.

5 mM KCN as well as anoxia have corresponding effects on Physarum plasmodia: Both induce a disintegration of the plasmalemma invaginations, an increase in cytoplasmic vacuoles, formation of a thick cortical actomyosin layer and an increase of cytoplasmic actomyosin fibrils. Both KCN treatment and anoxia cause a temporary increase in the level of the force oscillations and a reversible prolongation of the periods of the contraction-relaxation cycle of cytoplasmic actomyosin. The normal pattern of oscillation can be restored by the addition of 10 mM alpha-ketoglutarate +5 mM AMP to the solution

Cell cycle inhibitory effects of leaf extract from Curcuma vamana M. Sabu & Mangaly on mitotically synchronous cultures of Physarum polycephalum Schw.

Leaf extracts of C. vamana, endemic to Kerala state in India, were found to inhibit cell cycle progression in synchronous cultures of P. polycephalum in a concentration and phase-specific manner. Crude alkaloid extract (CAE) elicited maximum cell cycle delays in comparison to soxhletted chloroform, acetone and aqueous extracts. Total alkaloid content of CAE was found to be 64.9 mg/g. CAE showed lowest DPPH radical scavenging activity. Other extracts with higher free radical scavenging activity exhibited lesser cell cycle inhibiting potential. Upto 21% decrease in nuclear DNA was observed in CA

Distinct actin architectures across a giant syncytial cell.

The syncytial slime mold Physarum polycephalum exhibits cytosolic streaming, cell migration, and cell network remodeling within a single cell. It is unclear how cytoskeletal elements are spatiotemporally organized to mediate these processes. Here, we combine live and fixed imaging of Physarum F-actin to uncover organizational and dynamic signatures of the actin cytoskeleton across cell structures. We find circumferential actin filaments constrict and relax to drive peristaltic contractions in feeding tubes, while migratory fans are characterized by actin patches and comets. Our work presents u

Coexistence of trapped and flow-transported nuclei enables fast pigeon post communication across multinucleated cell.

Multi-nucleated cells exist in all domains of life, ranging from animals, plants, and fungi to single-celled organisms such as the slime mold Physarum polycephalum. The large cell size, in the case of Physarum reaching centimeters and more, challenges the coordination of nuclei activity as signals need to cross large distances. In search of a mechanism for fast long-ranged communication among nuclei, we quantify nuclei dynamics and cytoplasmic flows in Physarum's tubular network. We observe nuclei in two interchangeable, dynamic states: mobile, flowing within the cytoplasmic shuttle flow, or t

Slime moulds use heuristics based on within-patch experience to decide when to leave.

Animals foraging in patchy, non-renewing or slowly renewing environments must make decisions about how long to remain within a patch. Organisms can use heuristics ('rules of thumb') based on available information to decide when to leave the patch. Here, we investigated proximate patch-departure heuristics in two species of giant, brainless amoeba: the slime moulds Didymium bahiense and Physarum polycephalum. We explicitly tested the importance of information obtained through experience by eliminating chemosensory cues of patch quality. In P. polycephalum, patch departure was influenced by the

Learning Without Neurons: A Review of Associative Memory in Protists.

For decades, the capacity for associative learning was considered a hallmark of multicellular organisms possessing complex nervous systems. However, a growing body of evidence in the field of basal cognition is challenging this neuro-centric paradigm, demonstrating that unicellular eukaryotes, or protozoans, exhibit sophisticated behavioral plasticity. This review synthesizes historical and contemporary research into associative learning-specifically classical and operant conditioning-in prominent protozoan models such as Paramecium, Stentor, and the acellular slime mold Physarum polycephalum.

Zingerone protects against stannous chloride-induced and hydrogen peroxide-induced oxidative DNA damage in vitro.

In this paper, we report the dose-dependent antioxidant activity and DNA protective effects of zingerone. At 500 μg/mL, the DPPH radical scavenging activity of zingerone and ascorbic acid as a standard was found to be 86.7 and 94.2 % respectively. At the same concentration, zingerone also showed significant reducing power (absorbance 0.471) compared to that of ascorbic acid (absorbance 0.394). The in vitro toxicity of stannous chloride (SnCl2) was evaluated using genomic and plasmid DNA. SnCl2-induced degradation of genomic DNA was found to occur at a concentration of 0.8 mM onwards with compl

Tubulin evolution: an electrophoretic and immunological analysis.

This paper summarizes a survey of the electrophoretic behavior of the tubulins of 23 species (mostly protists) as well as their reactivity towards 4 anti-tubulin antibodies (raised against two ciliate tubulins and two vertebrate ones). Some generalizations concerning the relative migration rates of alpha VS beta tubulin could be made, in particular the alpha/beta inversion, first described in Physarum was extended to several ciliates. Antivertebrate tubulin antibodies displayed a very broad spectrum of reactions, reacting with virtually all the species tested. They appear to correspond to auto

Isolation and characterization of a novel gene with differential expression in benign and malignant human breast tumours.

We report the identification of a novel cDNA representing an mRNA showing significantly higher levels of expression in benign breast lesions than in carcinomas. This cDNA was identified by differential screening of a cDNA library generated from a breast carcinoma, and shows consistently higher expression in fibroadenomas than in carcinomas. The expression in both benign and malignant tissues is highest in epithelial cells as determined by in situ hybridization to tissue sections. The nucleotide sequence of the full-length cDNA has been determined, and the deduced protein is highly basic with n

Two-carba cyclic phosphatidic acid treatment promotes phenotypic switch from M1 to M2 microglia and prevents behavioral abnormalities in a mouse model of neuropsychiatric systemic lupus erythematosus.

Systemic lupus erythematosus (SLE) is an autoimmune disease associated with the production of double-stranded DNA (dsDNA) antibodies and other antibodies that predominantly affects women with a wide range of lesions. Although neuropsychiatric lupus erythematosus (NPSLE), characterized by neuropsychiatric symptoms related to cerebrovascular diseases or depression, ranks high in severity, no specific treatment has been defined. Two-carba cyclic phosphatidic acid (2ccPA), a derivative of cyclic phosphatidic acid, was isolated from the true slime mold Physarum polycephalum in 1992. 2ccPA treatment

Amoeba-inspired Tug-of-War algorithms for exploration-exploitation dilemma in extended Bandit Problem.

The true slime mold Physarum polycephalum, a single-celled amoeboid organism, is capable of efficiently allocating a constant amount of intracellular resource to its pseudopod-like branches that best fit the environment where dynamic light stimuli are applied. Inspired by the resource allocation process, the authors formulated a concurrent search algorithm, called the Tug-of-War (TOW) model, for maximizing the profit in the multi-armed Bandit Problem (BP). A player (gambler) of the BP should decide as quickly and accurately as possible which slot machine to invest in out of the N machines and

Drastic rise of intracellular adenosine(5')tetraphospho(5')adenosine correlates with onset of DNA synthesis in eukaryotic cells.

An assay of adenosine(5')tetraphospho(5')adenosine (Ap4A), based on the luciferin/luciferase method for ATP measurement, was developed, which allows one to determine picomolar amounts of unlabeled Ap4A in cellular extracts. In eukaryotic cells this method yielded levels of Ap4A varying from 0.01 microM to 13 microM depending on the growth, cell cycle, transformation, and differentiation state of cells. After mitogenic stimulation of G1-arrested mouse 3T3 and baby hamster kidney fibroblasts the Ap4A pools gradually increased 1000-fold during progression through the G1 phase reaching maximum Ap4

How does academia respond to the burden of infectious and parasitic disease?

BackgroundAcademic research is one of the main avenues through which humans can fight the threat of infectious diseases. However, there have been concerns regarding whether the academic system has provided sufficient efforts to fight infectious diseases we potentially face. Answering these questions could contribute to evidence-based recommendations for setting research priorities and third-mission policies.MethodsWith a focus on one of the most common categories of communicable diseases, infectious and parasitic diseases (IPDs), we searched Web of Science for articles and reviews relevant to
