The golden orb-weaver spider Nephila clavipes, known for its sexual size dimorphism, is abundant and widespread in the New World. The first annotated genome of orb-weaver spiders, exploring N. clavipes, has recently been reported. The study, focused primarily on the diversity of silk specific genes, shed light into the complex evolutionary history of spiders. Furthermore, a robust transcriptome analysis provided a massive resource for N. clavipes RNA survey. Here, I present evidence of viral sequences corresponding to the first 10 extant virus species associated to N. clavipes and indeed, neph

Acanthocystis turfacea chlorella virus 1 (ATCV-1) is a giant virus that is part of the human oral microvirome. Previously we showed that ATCV-1 infects mouse macrophages, stimulates production of inflammatory cytokines, and accelerates motor neuron disease in the Amyotrophic Lateral Sclerosis (ALS) model SOD1-G93A transgenic mice. This, coupled with significantly elevated levels of serum IgG1 antibody to ATCV-1 in ALS patients compared with healthy controls, suggests involvement of ATCV-1 in ALS. Herein, using serum and CSF from a different ALS cohort we again show elevated antibodies to ATCV-

BACKGROUND: Plant viruses have evolved adaptations that enable them to alter host cues, thereby facilitating their replication and efficient transmission by insect vectors. Satellite RNAs (satRNAs), which accompany certain plant RNA viruses and are dependent on them for replication and transmission, can change the progression of pathogenesis and the expression of disease symptoms. This study aimed to analyse how the change in the course of infection by satRNA (exacerbation or mitigation of pathogenesis) impacts the subsequent stages of virus transmission. We hypothesised that satRNAs influence

Grapevine (Vitis vinifera) is severely impacted by viral diseases, with grapevine leafroll disease (GLRD) being the most economically damaging. Grapevine leafroll-associated virus 2 (GLRaV-2) is a prevalent virus associated with GLRD, yet the molecular mechanisms underlying GLRaV-2 infection remain poorly understood. Here, we demonstrate that the grapevine transcription factor (TF) VvWRKY51 positively autoregulates its transcription and directly represses salicylic acid (SA)-responsive defense genes VvPR1 (pathogenesis-related gene 1), VvPR2, and VvPR5. Through in vivo and in vitro assays, we

In this study, we investigated unclassified picorna-like viruses in Culex (Melanoconion) mosquitoes from São Paulo, Brazil, an area of high mosquito biodiversity and arbovirus activity. Two mosquito pools were processed using next-generation sequencing (NGS), and datasets were analyzed via de novo assembly to reconstruct viral genomes and assess evolutionary relationships. We identified two highly similar viral genomes, named Culex (Melanoconion) picorna-like virus, CmV_B38 and CmV_B39, exhibiting 99.93% nucleotide identity, both of which encode a three-domain replication block characteristic

In this study, VP1 genes from duck hepatitis A virus (DHAV) genotypes 1 (DHAV-1) and 3 (DHAV-3) were amplified through reverse transcription polymerase chain reaction (RT-PCR) by using viral RNA as the template. The amplified genes were cloned into the plasmid pNZ8149 through homologous recombination, yielding the recombinant plasmids pNZ8149-DHAV-1 VP1 and pNZ8149-DHAV-3 VP1. These plasmids were electroporated into competent L. lactis NZ3900 cells to generate the recombinant strains L. lactis pNZ8149-DHAV-1 VP1/NZ3900 and L. lactis pNZ8149-DHAV-3 VP1/NZ3900, respectively. Cannabis sativa L. produces a wide range of bioactive metabolites, including phytocannabinoids such as tetrahydrocannabinol (THC), cannabidiol (CBD) and cannabigerol (CBG), which exhibit antiviral activity and modulate innate immune responses through Toll-like receptors (TLRs), particularly TLR4 and TLR7. The cross-regulation between cannabinoids and TLRs can influence the production of cytokines and antimicrobial peptides. Given their key role in orchestrating innate immunity, particularly inflammatory responses and antiviral activity, understanding these processes in bovine immune cells is e

UNLABELLED: Sea ice is vital to polar ecosystems and serves as a natural laboratory for studying microbial adaptations in extreme environments. Nevertheless, the diversity and ecology of Antarctic marine RNA viruses have not been sufficiently explored. Here, 11 metatranscriptomes from Antarctic sea ice and ice-associated environments were analyzed to characterize RNA viral communities and their ecological differentiation between sea ice and seawater. Diverse RNA viral lineages were identified, of which Cryppavirales and Picornavirales were dominant. Lenarviricota was predominant in sea ice, hi

Picornaviruses cause substantial human and veterinary disease, yet direct-acting antivirals remain limited. The nonstructural protein 2C is among the most conserved picornaviral proteins and has ATPase and helicase-like activities. 2C is involved in membrane remodeling, RNA replication, encapsidation, morphogenesis, and multiple virus-host interactions. In this review, we summarize the domain architecture and structural biology of 2C, highlighting the N-terminal membrane-binding region, the ATPase core, the C-terminal helical domain, and accessory motifs that regulate oligomerization and ligan

The expression of the BNYVV silencing suppressor (VSR) p14 protein, together with the production of a viral non-coding RNA (ncRNA3), affects the long-distance movement on natural host Beta species. Through immunoprecipitation of Flag-p14 and expression of TurboID-p14 coupled to mass spectrometry, we identified several potential p14 cellular partners. These include proteins related to RNA metabolism, proteasome activation, and notably, SGS3-a key player in the siRNA transitivity pathway-and DRB4, the DCL4 cofactor described in A. thaliana silencing pathways. The interaction between p14 and DRB4

Viral infection is a major cause of ill health in wild chimpanzees (Pan troglodytes), but most evidence to date has come from conspicuous disease outbreaks with high morbidity and mortality. To examine the relationship between viral infection and ill health during periods not associated with disease outbreaks, we conducted a longitudinal study of wild eastern chimpanzees (P. t. schweinfurthii) in the Kanyawara and Ngogo communities of Kibale National Park, Uganda. We collected standardized, observational health data for 4 years and then used metagenomics to characterize gastrointestinal virome

RNA domains within viral IRESs are crucial for initiating cap-independent translation of the genome in many positive-sense RNA viruses. However, the structures and mechanisms of these IRES domains remain unclear. Here, we present the 3 Å resolution crystal structure of the coxsackievirus B3 (CVB3) IRES domain V (dV) as a model for type I IRESs. The crystal structure revealed an elongated architecture of dV, with two sets of coaxially stacked stems forming an H-type four-way junction (4WJ) organized by an A-rich motif. Despite sequence dissimilarities, this dV from a type I IRES exhibits remark

Picornaviruses are small viruses with a plus-strand RNA genome in which RNA secondary structures bind cellular proteins to support viral translation and replication. Here, we characterize tRNA anticodon stem-loop-like structures in the 5'- and 3' untranslated regions (UTRs) of the RNA of Mengovirus, a member of the Cardiovirus group in the Picornaviridae family. These RNA elements specifically bind cellular Glycyl-tRNA synthetase (GARS). Mutation of the conserved CCA motifs in the loops of these GARS binding elements (GBEs) impairs binding, as does deletion of the anticodon binding domain of G

Avian species play a key role in the ecology of viruses, acting as reservoirs, sentinels, and biological carriers across diverse environments. Here, we describe the detection and genomic characterization of a novel iflavirus identified in fecal samples from the Chiroxiphiapareola("blue-backed manakin") collected in areas of the Middle North region of Brazil, Maranhão state. Viral RNA was extracted from pool fecal samples and subjected to next-generation sequencing. De novo assembly and comparative analyses enabled the recovery of a complete picorna-like viral genome of 9043 nucleotides, compri

Both autophagy and heat shock proteins (HSPs) play dual roles in viral infections, yet their coordination in antiviral defense remains unclear. Here, we show that a cytosolic small heat shock protein (AcsHSP) and a type II J-domain protein (AcDNAJB13) from areca palm interact with the coat protein (CP) of areca palm velarivirus 1 (APV1) independently of HSP70 chaperones. Their closest Nicotiana benthamiana homologs also bind CP. Both APV1 infection and CP expression induces the transcription of sHSP and DNAJB13, which in turn inhibits CP accumulation. Silencing of these genes enhances APV1 inf

The complex epidemic profile of swine infectious diseases hinders effective disease control, as separate multiple-vaccine inoculation causes excessive body stress, leading to insufficient immune responses, vaccine failure, reduced production performance and increased breeding costs. Based on our pre-established FMDV/SVA nanoparticle vaccine platform, this study screened and optimized nanoparticle antigens FMDV T3D-LS-LOOP and SVA VP1(B)-β-VP2, developed a bivalent FMDV/SVA nanoparticle vaccine via Escherichia coli prokaryotic expression system, and systematically evaluated its immune protectiv

Phosphatidylserine (PS) receptors facilitate entry of diverse enveloped viruses and have been implicated in enhancing coronavirus attachment, including for SARS-CoV-2, yet their precise contribution remains unclear. We examined this pathway using EjCoV-3, a recently identified bat merbecovirus. Exogenous PS, but not phosphatidylcholine, reduced viral infectivity in a concentration-dependent manner, suggesting competitive inhibition of receptor binding by virion-associated PS. Genome replication in positive-stranded RNA (ssRNA+) viruses is mediated by cognate enzymes, including ubiquitous RNA-dependent RNA polymerase (RdRp). In ssRNA+ viruses with multiple open reading frames (ORFs) in their genomes, replication often is accompanied by synthesis of subgenomic RNAs (transcription) for expression of 3'-proximal ORFs. In addition, all ssRNA+ viruses with genomes larger than ~7 kb encode helicases, linking helicases to RNA genome expansion. Helicases are essential ATPases that unwind nucleic acids and are classified into six recognized superfamilies (SF1-SF6). In the ph

Increasing data indicate that insects serve as major reservoirs and vectors of viruses, which account for the continuously increasing ecological burden and infectious disease outbreaks. Uncovering the hidden diversity of viruses in insects will further the understanding of the ecological and evolutionary perspectives in the emergence of insect-associated virus diseases. In this study, we queried transcriptome sequencing (RNA-Seq) data from more than 600 species across 32 insect orders dwelling in different ecological habitats and recovered more than 1,213 RNA viruses that were recapitulated in

UNLABELLED: Senecavirus A (SVA), also known as Seneca Valley virus (SVV), is an important small RNA virus that causes swine vesicular disease, posing a significant threat to the global swine industry. To date, the molecular mechanism of SVA replication remains poorly understood. Heat shock proteins (HSPs) are a family of molecular chaperone proteins that are widely present in organisms. More and more studies have shown that HSPs are associated with viral infections. Here, we screened HSPs and found that overexpression of HSPA5 significantly inhibited SVA replication, while knockout of HSPA5 pr
