iTRAQ-based proteomic identification of cyclophilin as a candidate mediator for cyclosporin A-mediated immunomodulation in pearl oysters Pinctada fucata martensii.

Transplantation surgery induces severe immune stress in pearl oysters Pinctada fucata martensii, yet effective immunomodulatory strategies remain limited. This study establishes a proteomics-guided strategy for immunomodulation by identifying cyclophilin as a candidate mediator through iTRAQ-based quantitative analysis and validating cyclosporin A (CsA) through serum immune indicator and transcriptome analysis. Serum iTRAQ analysis identified 1069 differentially expressed proteins following transplantation, among which cyclophilin exhibited significant up-regulation and was selected as a candi

Enhancement of Anti-Melanogenic Activity of Pinctada martensii Nacre-Derived Peptide Through C-Terminal Amidation: Mechanism Investigation in B16F10 Cells.

Excessive melanin accumulation induced by UV exposure and hormonal changes poses significant challenges to skin health, necessitating safe and effective tyrosinase inhibitors. Pearl and nacre powder have long been utilized in traditional Chinese medicine for skin whitening. Our previous study identified a tyrosinase inhibitory peptide (Ala-His-Tyr-Tyr-Asp) from Pinctada martensii nacre in silico; however, chemical modification strategies for enhancing bioactive peptide efficacy remain underexplored. The present study investigated the structure-activity relationship of NP-TIP-1 (Tyrosinase Inhi

Field evidence of nylon fishing-net macroplastic embedded in the tissue of a wild pearl oyster (Pinctada fucata): A histological case observation.

The objective of this study was to document and characterise an opportunistic case of macroplastic embedding within the soft tissues of a wild pearl oyster (Pinctada fucata) and to describe the associated tissue-level responses. Most existing research on plastic pollution in bivalves has focused on microplastic (<5 mm) ingestion, whereas the physical incorporation of macroplastic (>5 mm) debris into bivalve tissues remains poorly documented under field conditions. Four adult P. fucata specimens, obtained as bycatch from artisanal gill-net fishing off the southeastern coast of India (Nagapattin

Tracing key genes associated with the Pinctada margaritifera albino phenotype from juvenile to cultured pearl harvest stages using multiple whole transcriptome sequencing.

BACKGROUND: Albino mutations are commonly observed in the animal kingdom, including in bivalves. In the black-lipped pearl oyster Pinctada margaritifera, albino specimens are characterized by total or partial absence of colouration resulting in typical white shell phenotype expression. The relationship of shell colour with resulting cultured pearl colour is of great economic interest in P. margaritifera, on which a pearl industry is based. Hence, the albino phenotype provides a useful way to examine the molecular mechanisms underlying pigmentation. RESULTS: Whole transcriptome RNA-sequencing

Lethal effects of ichthyotoxic raphidophytes, Chattonella marina, C. antiqua, and Heterosigma akashiwo, on post-embryonic stages of the Japanese pearl oyster, Pinctada fucata martensii.

The inimical effects of the ichthyotoxic harmful algal bloom (HAB)-forming raphidophytes Heterosigma akashiwo, Chattonella marina, and Chattonella antiqua on the early-life stages of the Japanese pearl oyster Pinctada fucata martensii were studied. Fertilized eggs and developing embryos were not affected following exposure to the harmful raphidophytes; however, all three algal species severely affected trochophores and D-larvae, early-stage D-larvae, and late-stage pre-settling larvae. Exposure to C. marina (5×102cellsml-1), C. antiqua (103cellsml-1), and H. akashiwo (5×103cellsml-1) resulted

A large-scale experiment finds no consistent evidence of change in mortality or commercial productivity in silverlip pearl oysters (Pinctada maxima) exposed to a seismic source survey.

High-intensity, impulsive sounds are used to locate oil and gas reserves during seismic exploration of the seafloor. The impacts of this noise pollution on the health and mortality of marine invertebrates are not well known, including the silverlip pearl oyster (Pinctada maxima), which comprises one of the world's last remaining significant wildstock pearl oyster fisheries, in northwestern Australia. We exposed ≈11,000 P. maxima to a four-day experimental seismic survey, plus one vessel-control day. After exposure, survival rates were monitored throughout a full two-year production cycle, and

A pearl protein self-assembles to form protein complexes that amplify mineralization.

The formation of the nacre pearl in marine invertebrates represents an on-demand production of mineralization in response to an irritant or parasite threat to the mantle organ. In the Japanese pearl oyster (Pinctada fucata), this process is mediated by a 12-member protein family known as PFMG (Pinctada fucata mantle gene). One of these proteins, PFGM1, has been implicated in modulating calcium carbonate crystal growth and has been reported to possess an EF-hand-like domain. In this report, we establish that the recombinant PFMG1 (rPFMG1) is an intrinsically disordered "imitator" EF-hand protei

Tidal flushing and wind driven circulation of Ahe atoll lagoon (Tuamotu Archipelago, French Polynesia) from in situ observations and numerical modelling.

Hydrodynamic functioning and water circulation of the semi-closed deep lagoon of Ahe atoll (Tuamotu Archipelago, French Polynesia) were investigated using 1 year of field data and a 3D hydrodynamical model. Tidal amplitude averaged less than 30 cm, but tide generated very strong currents (2 ms(-1)) in the pass, creating a jet-like circulation that partitioned the lagoon into three residual circulation cells. The pass entirely flushed excess water brought by waves-induced radiation stress. Circulation patterns were computed for climatological meteorological conditions and summarized with stream

[Progress in study on the role of metabolomics in toxic effects of environmental pollutants and the underlying mechanism].

Metabolomics methods were applied in the study of the toxicity of environmental pollutants. It has been shown that exposure to heavy metals such as mercury, arsenic, cadmium, chromium and lead could cause significant changes in energy, lipids, nucleic acids and amino acids in mammalian cells. After exposure to benzo(a)pyrene [B(a)P], the glands of Pinctada pumila could produce various changes, such as energy metabolic disorder, cell damage, signal transduction disorder, oxidative stress and osmotic disorder. Persistent organic compounds polychlorinated biphenyls (PCBs) could exert toxic effect

[The study of phosphorus in seawater cultured pearls using FTIR].

Seawater cultured pearl was studied by high resolution fourier transform infrared spectroscopy (FTIR) both before and after having been heated at 160 centigrade degree. The results of FTIR indicate that the main material in both samples is aragonite (CaCO3). But compared with the spectra of the seawater cultured pearl, there are some changes that happen in that of the heated sample. Some other peaks arise in the latter. And the intensities of these peaks increase with the heating time. Differential Thermal Analysis (DTA) curve of the seawater cultured pearl shows that seawater cultured pearl h

Whole-genome bisulfite sequencing reveals the function of DNA methylation in the allotransplantation immunity of pearl oysters.

INTRODUCTION: In the pearl culture industry, a major challenge is the overactive immunological response in pearl oysters resulting from allotransplantation, leading to shell-bead rejection and death. To better understand the molecular mechanisms of postoperative recovery and the regulatory role of DNA methylation in gene expression, we analyzed the changes in DNA methylation levels after allotransplantation in pearl oyster Pinctada fucata martensii, and elucidated the regulatory function of DNA methylation in promoter activity of nicotinic acetylcholine receptor (nAChR) gene. METHODS: We cons

The first bioactive (angiotensin-converting enzyme-inhibitory) peptide isolated from pearl matrix protein.

In this research, we unveil the medical potential of pearls by identifying a novel bioactive peptide within them for the first time. The peptide, termed KKCHFWPFPW, emerges as a pioneering angiotensin I-converting enzyme (ACE) inhibitor, originating from the pearl matrix of Pinctada fucata. Employing quadrupole time-of-flight mass spectrometry, this peptide was meticulously selected and pinpointed. With a molecular weight of 1417.5 Da and a theoretical isoelectric point of 9.31, its inhibitory potency was demonstrated through a half-maximal inhibitory concentration (IC50) of 4.17 μM, establish

Field Experiments, Behavioral Analyses, and Digestive Physiology Reveal the Selective Oyster-Feeding Strategy of Thais luteostoma.

Pearl oyster aquaculture is severely constrained by biofouling organisms, particularly fouling oysters, which substantially impair pearl oyster growth and farming efficiency. This study investigated the selective oyster-feeding behavior of the predatory gastropod Thais luteostoma and evaluated its potential as an ecological biofouling control agent in pearl oyster culture. Field co-culture experiments showed that T. luteostoma did not adversely affect the survival of Pinctada fucata martensii, while effectively reducing biofouling loads and significantly improving pearl oyster growth performan

A Spirochaete is suggested as the causative agent of Akoya oyster disease by metagenomic analysis.

Mass mortality that is acompanied by reddish browning of the soft tissues has been occurring in cultured pearl oyster, Pinctada fucata martensii. The disease is called Akoya oyster disease (AOD). Although spreading pattern of the disease and transmission experiments suggest that the disease is infectious, the causative agent has not yet been identified. We used shotgun and 16S rRNA-based metagenomic analysis to identify genes that are present specifically in affected oysters. The genes found only in diseased oysters were mostly bacterial origin, suggesting that the causative agent was a bacter

Cooperative roles of Pm-RORα splice variants in modulating the immune response in the pearl oyster P. f. martensii.

RAR-related orphan receptor alpha (RORα), a core member of the NR1F subgroup within the nuclear receptor superfamily, critically regulates lipid metabolism and immune responses. This study identified the Pm-RORα (Pm-RORα1 and Pm-RORα2) gene and its alternative splicing isoforms in Pinctada fucata martensii and investigated their functions through bioinformatic analysis, quantitative PCR (qPCR), and bimolecular fluorescence complementation (BiFC). Phylogenetic analysis revealed high homology between Pm-RORα and the Pacific oyster Crassostrea gigas ROR. Pm-RORα1_1 and Pm-RORα1_2 were generated b

Making Space for Red Tide: Discolored Water and the Early Twentieth Century Bayscape of Japanese Pearl Cultivation.

"Red tide" has become a familiar shorthand for unusual changes in the color of ocean waters. It is intimately related both to blooms of creatures like dinoflagellates and to the devastating effects they pose to coastal fisheries. This essay tracks the early twentieth century emergence of discolored water as an aquacultural problem, known in Japan as akashio, and its trans-oceanic transformation into the terms and practices of "red tide" in the post-World War II United States. For Japan's "Pearl King" Mikimoto Kōkichi and his contacts in diverse marine scientific communities, the years-long cyc

Ps19, a novel chitin binding protein from Pteria sterna capable to mineralize aragonite plates in vitro.

Mollusk shell is composed of two CaCO3 polymorphs (calcite and aragonite) and an organic matrix that consists of acetic acid- or ethylenediaminetetraacetic acid (EDTA)-soluble and insoluble proteins and other biomolecules (polysaccharides, β-chitin). However, the shell matrix proteins involved in nacre formation are not fully known. Thus, the aim of this study was to identify and characterize a novel protein from the acetic acid-insoluble fraction from the shell of Pteria sterna, named in this study as Ps19, to have a better understanding of the biomineralization process. Mussel- and nacre-inspired dual-bionic alginate-based hydrogel coating with multi-matrix applicability, high separation stability and antifouling performance for oil/water separation.

Natural hydrogel-modified porous matrices with superwetting interfaces are ideal for oil/water separation. In this study, inspired by two marine organisms, a novel hydrogel coating with multi-matrix suitability, high oil/water separation capability and antifouling properties was developed. Specifically, inspired by mussel byssus, hydrogel coating was successfully deposited on porous matrix surface based on the introduction of tannic acid (TA). Moreover, inspired by the "brick and mortar" microstructure of Pinctada nacre, silica particles were in-situ synthesized in the sodium alginate (SA)/Ca2
