We have studied the intracellular fate of exchangeable cholesterol in a model system with lipid vesicles (cholesterol/phospholipid mole ratio 1:1) and cultured human lung fibroblasts. Exchangeable [3H]cholesterol in lipid vesicles was readily incorporated into cellular plasma membranes and transported to intracellular esterification sites. The formation of [3H]cholesteryl esters was not affected by cytoskeleton-disrupting drugs. The shape and distribution of lysosomes in the ciliary epithelium of rat eyes were examined by electron microscopy combined with acid phosphatase (ACPase) cytochemistry and three-dimensional observation of 2 microns-thick sections. This is a transmission electron microscopy study of peritoneal mesothelium and surface cells covering the ovary. One thousand two hundred cells derived from 22 samples of peritoneum and 1,200 derived from 22 samples of ovary were compared for cell size, length-to-diameter ratio, and distribution and complexity of surface villi. Evidence suggesting that diphtheria toxin reaches the cytoplasm of susceptible mammalian cells by two independent mechanisms is presented. The internalization of antibody via pinocytosis and its pH-dependent interaction with the neonatal Fc receptor (FcRn) play crucial roles in antibody recycling within circulation. To achieve an optimal half-life, the binding of antibodies to FcRn at acidic pH and their release at physiological pH must be carefully characterized. A preliminary injection of triton WR 1339 protectively influenced the chronic toxic CCl4-hepatitis. The ultrastructural peculiarities were close to the normal ones in hepatocytes, in addition to the increased triglyceride rejection from cells by means of autophagy. Essential fatty acid (EFA) deficiency has been shown to protect against the glomerulonephritis in a murine model of systemic lupus erythematosus. Qualitative and quantitative determinations of immunoglobulins (Ig's) and beta-1-C globulin were carried out in fluid vesicles of hydatidiform moles and carrier mothers. The mean concentration of IgG was 9.9 per cent, IgA had 14.3 per cent, in relation to maternal values. No IgM concentrations were detected in molar fluid. Beta-1-C globulin showed 60.5 per cent concentration in relation to mother's serum. It is suggested that in trophoblast of molar vesicles a selective transudate migration of Ig's by a mechanism of pinocytosis takes place to the vesicular fluid.

Macrophage membrane cholesterol is present in two subcellular cholesterol pools, a rapidly exchanging compartment comprising about two-thirds of the total cholesterol, and a slowly exchanging compartment comprising one-third of the total.
