Nasal trigeminal inputs release the A5 inhibition received by the respiratory rhythm generator of the mouse neonate.

Experiments were performed on neonatal mice to analyze why, in vitro, the respiratory rhythm generator (RRG) was silent and how it could be activated. We demonstrated that in vitro the RRG in intact brain stems is silenced by a powerful inhibition arising from the pontine A5 neurons through medullary alpha(2) adrenoceptors and that in vivo nasal trigeminal inputs facilitate the RRG as nasal continuous positive airway pressure increases the breathing frequency, whereas nasal occlusion and nasal afferent anesthesia depress it. Because nasal trigeminal afferents project to the A5 nuclei, we appli

Effects of alpha-adrenoceptor agonists and antagonists in a maze-exploration model of 'fear'-motivated behaviour.

An elevated X-maze with alternating open and enclosed arms was investigated as a model for the study of fear-induced behaviour. As predicted, the anxiolytics diazepam and amylobarbitone increased, and the putative anxiogenics ACTH and picrotoxin decreased the proportion of open arm entries. The alpha 1-adrenoceptor agonists phenylephrine and ST587, and the alpha 2-adrenoceptor antagonists idazoxan, piperoxane, RS-21361 and yohimbine decreased relative open-arm entries, thus resembling the putative anxiogenics. On the other hand, azepexole, clonidine and guanabenz, agonists at alpha 2-adrenocep

Counteractive effects of norepinephrine and amphetamine on quabain-induced amnesia.

Ouabain, administered 5 min prior to learning induces amnesia for a single-trial passive avoidance task in day-old chickens by inhibiting memory formation during the labile sodium-pump dependent phase. Amphetamine and norepinephrine (NE) successfully counteract ouabain-induced amnesia when administered immediately after learning. The actions of these drugs on ouabain-induced amnesia parallel that of diphenylhydantoin (DPH), and within similar time constraints. It is concluded that amphetamine (through release of NE) and norepinephrine exert their effects through stimulation of Na+/K+ ATP'ase a

Reversal of opioid-induced muscular rigidity in rats: evidence for alpha-2 adrenergic involvement.

Compounds from several different pharmacological classes were tested for their ability to reverse the muscular rigidity induced by an intravenous dose of fentanyl that also caused loss of the righting reflex (LOR). Opioid antagonists reversed the entire syndrome--LOR and rigidity but, generally, rigidity could be reversed nonspecifically by doses of compounds that caused LOR by themselves (e.g., CNS depressants). Muscle relaxants and agonists of histamine, which appeared to be acting peripherally, were also effective. On the other hand, serotonergic drugs and dopamine agonists were not. Howeve

Differentiation of drugs acting centrally upon the cardiovascular system by means of sympathetic and vagal responses.

The response pattern of the autonomic nervous system was investigated after central administration (intra-cisternal, vertebral artery) of amphetamine, morphine, fentanyl, dextromoramide and the substance R 28935, chemically related to the neuroleptic agent pimozide. Effects on the sympathetic system were measured by recording electrical discharges of fibres of the (preganglionic) major splanchnic nerve in anaesthetized cats; those on the vagal system by recording the heart rate in anaesthetized dogs under beta-adrenoceptor blockade; the baroreceptor reflex was elicited by the blood pressure in

[Action of delta 9 tetrahydrocannabinol on the central cardiovascular regulation : mechanism and localization].

Delta9-tetrahydrocannabinol (30-300 mug.kg-1 i.v.) induced in cats and dogs a decrease in blood pressure and heart rate. This decrease appears to be centrally mediated. In fact, the splanchnic and cardiac discharges were reduced in intact animals as well as in debuffered cats ruling out a reflexly mediated action. The mechanism of this central decrease in the sympathetic tone appears to be different from the mechanism of the reduction induced by clonidine or by narcotic analgesics agents. In fact, piperoxan (1 mg.kg-1 i.v.), an alpha adrenoceptor blocking agent, antagonized or reversed the cen

Hypotensive properties of benzodioxane derivatives structurally related to R 28935. Comparison of activity with some receptor affinities.

Hypotensive activities were determined of some derivatives of erythro 1-(1-[2-(1,4-benzodioxane-2-yl)-2-hydroxyethyl]-4-piperidyl)-2-benzimidazolinone (R 28935) following intravenous administration to anaesthetized normotensive rats. Introduction of chlorine into the benzimidazolinone part of R 28935 negatively influenced the hypotensive potency as did opening of the dioxane ring. An appropriate substituent restored the hypotensive effectiveness of the "opened" structures. In general, the compounds were weak inhibitors of the specific binding of (3H)-prazosin (alpha 1-adrenoceptors), (3H)-clon

Presence of opioid receptors in mesencephalic nucleus dorsalis raphe concerned in cardiovascular regulation in cats.

The effects of microinjection of opioid receptor agonist and antagonist into mesencephalic nucleus dorsalis raphe, were studied on mean arterial pressure and heart rate to elucidate the nature and role of these opioid receptors in cardiovascular regulation. Microinjection of morphine (5 micrograms and 10 micrograms) into nucleus dorsalis raphe elicited both inhibitory and excitatory cardiovascular responses respectively, while microinjection of opioid receptor antagonist, naloxone (10 micrograms) failed to produce any significant cardiovascular responses. However, local pretreatment with nalox

Biochemical actions of sympathomimetic drugs which overcome cycloheximide-induced amnesia.

Earlier investigations of sympathomimetic drugs overcoming the amnesic action of cycloheximide (CXM) in day-old chickens were extended to biochemical studies in vitro. The effects of amphetamine, norepinephrine, alpha and beta noradrenergic stimulants and receptor blockers on Na+/K+ ATP'ase activity in total homogenate of chicken forebrain were investigated. Norepinephrine and the beta stimulant, isoprenaline significantly stimulated the activity of this enzyme, while the beta blocker, propranolol inhibited activity. Amphetamine, the alpha stimulant, methoxamine and the alpha receptor blocker,

Bronchopulmonary effects of clonidine on the bronchomotor responses of the guinea-pig.

In conscious guinea pigs, clonidine (10 and 100 micrograms/kg i.v.) lowered diastolic (-7.9 +/- 3.5 and -12.4 +/- 5.2%) and systolic (-8.6 +/- 3.0 and -11.9 +/- 4.2%) arterial pressure and reduced heart rate (-14.5 +/- 3.9 and -27.7 +/- 3.8%), but did not significantly modify pulmonary airway resistance. Hypotension was suppressed by yohimbine and bradycardia was partially suppressed by atropine and yohimbine, which demonstrates in this animal an alpha 2-adrenergic effect for hypotension and a mixed cholinergic and alpha 2-adrenergic effect for bradycardia. Noradrenergic regulation of growth hormone secretion in the baboon.

We have investigated the effects of iv administered noradrenergic agonists and antagonists on plasma GH concentration in the adolescent baboon, Papio papio, with the aim of defining the relative roles of adrenergic receptor subtypes (alpha 1, alpha 2, beta 1, and beta 2) in the regulation of GH release. Clonidine (0.02 mg/kg) or UK-14,304 (0.02 mg/kg), potent centrally acting alpha 2 noradrenergic agonists, were infused into 24 animals pretreated with either saline, or selective alpha 1 and alpha 2 noradrenergic antagonists. Effects of central alpha-2 adrenergic compounds on canine narcolepsy, a disorder of rapid eye movement sleep.

Recent experiments have demonstrated that pharmacological activation of central noradrenergic systems by monoaminergic stimulators or uptake blockers or through the stimulation of alpha-1 adrenergic receptors improved cataplexy, a major symptom of narcolepsy. In order to further the understanding of the control of cataplexy by noradrenergic mechanisms, the involvement of central alpha-2 adrenoceptors was examined in genetically narcoleptic Doberman pinschers using in vivo pharmacology. Yohimbine (1.5-96.0 micrograms/kg i.v.) and seven other selective and centrally acting alpha-2 adrenoceptor a

Blockade of alpha-2 adrenergic receptors in the rostral ventrolateral medulla attenuates the sympathoinhibitory response to cocaine.

The purpose of this study was to determine whether neurons in the rostral ventrolateral medulla play a role in the sympathoinhibitory response elicited by i.v. administration of cocaine and, if so, to identify the type(s) of receptors involved. Adrenergic antagonists were microinjected bilaterally into the rostral ventrolateral medulla in pentobarbital-anesthetized rats in an attempt to block the decrease in sympathetic nerve discharge (SND) elicited by cocaine (1 mg/kg i.v.). After the bilateral microinjection of saline, cocaine elicited a -56 +/- 5% (mean +/- S.E.) decrease in SND lasting 36

The role of alpha- and beta- adrenergic receptors in some actions of catecholamines on intestinal smooth muscle.

1. Experiments were carried out to determine the type of adrenergic receptor concerned in the action of noradrenaline in increasing the rate of exchange of potassium ions in the taenia of the guinea-pig caecum. The preparations were bathed in potassium-rich solutions in order to depolarize the muscle fibres and so eliminate changes in potassium flux secondary to alterations in membrane potential.2. Noradrenaline was more effective than isoprenaline in increasing both the influx and efflux of (42)K, suggesting that the alpha-receptors were involved. This action on potassium permeability was abo

Local regulation of transmitter release from rodent sympathetic nerve terminals?

1. Electrophysiological techniques were used to observe the release of transmitter from single release sites in the sympathetic neuro-effector junction of the rodent vas deferens. Transmitter release produces transient peaks in the rate of depolarization of the smooth muscle cells, known as 'discrete events'. 2. The amplitude distributions of stimulus-evoked discrete events in both mouse and guinea-pig vas are multi-modal. In the mouse, the distribution fits a Poisson with a 'quantal content' of about two. There are too many zeros in the amplitude distributions of guinea-pig discrete events to

Chronic treatment with the α2-adrenoceptor antagonist fluparoxan prevents age-related deficits in spatial working memory in APP×PS1 transgenic mice without altering β-amyloid plaque load or astrocytosis.

Locus coeruleus degeneration and reduced central noradrenaline content is an early feature of Alzheimer's disease. In transgenic mouse models of Alzheimer's disease-like pathology, lesioning the locus coeruleus exacerbates β-amyloid (Aβ) pathology, neuroinflammation and memory deficits. Here we aimed to determine whether chronic treatment with the α(2)-adrenoceptor antagonist fluparoxan, that enhances noradrenaline release, can prevent the onset of Alzheimer's-like pathology and memory deficits in APP/PS1 transgenic mice (TASTPM). Fluparoxan (1mg/kg/day) was administered to TASTPM and wild typ
