Integrated proteomic analysis reveals differential remodelling of defense responses in potato to herbivory by Phthorimaea operculella.

Plants rely on both transcriptional reprogramming and post-translational machinery to regulate their responses to biotic stress. While transcriptome-wide dynamics underlying plant defense against insect herbivores have been extensively documented, systematic investigations into protein-level regulation and integrative analyses of these multi-layered regulatory networks remain limited. In this study, we employed quantitative proteomics and phospho-proteomics to characterize the protein-level responses of potato plants to attacks by Phthorimaea operculella. A total of 60 differentially expressed

Assessing the effects of a novel biostimulant to enhance leafminer resistance and plant growth on common bean.

The leafminer Liriomyza trifolii is one of the major insects that affect Phaseolus vulgaris production worldwide. Novel and safe biobased stimulator compound (BSTC) with micronutrient-amino acid chelated compounds was developed from natural compounds and was used for foliar spray of P. vulgaris. Treated plants showed significantly increased in quality and productivity as well as significant reduction in leafminer infestation by close the tunnel end resulting in larvae suffocation and death. BSTC contains chemical composition that has important function in inducing immunity and resistance again

Integrated transcriptomic and metabolomic analysis reveals differential molecular mechanisms underlying resistance of native and invasive Phytolacca americana against Spodoptera litura herbivory.

BACKGROUND: In response to herbivory from insects, plants have evolved complex defense strategies. Alien plants often escape control by specialist insects in their introduced range. This release from herbivory pressure may lead to a reallocation of the defensive strategies of alien plants, thereby promoting their invasiveness. However, the molecular mechanisms underlying this resource reallocation process remain unclear. In the present study, we used Phytolacca americana as a model species, and combined transcriptomic and metabolomic analyses to investigate the molecular basis of its invasion

A ShbHLH34-ShPUR Module Activates ShTPS1-mediated β-caryophyllene Biosynthesis for Insect Resistance in Sugarcane.

Terpenoids represent one of the largest and most structurally diverse classes of plant secondary metabolites, playing essential roles in defense against herbivorous insects. Despite their importance, the mechanisms regulating terpenoid biosynthesis remain largely unexplored. Here, in sugarcane, we identified and characterized a terpene synthase gene from Saccharum spp. Hybrids, ShTPS1, which is strongly induced by feeding from fall armyworm Spodoptera frugiperda and sugarcane stem borer Chilo infuscatellus. Enzymatic assays in vitro revealed that ShTPS1 catalyzes the biosynthesis of volatile t

Plant latex and other exudates as plant defense systems: roles of various defense chemicals and proteins contained therein.

Plant latex and other exudates are saps that are exuded from the points of plant damage caused either mechanically or by insect herbivory. Although many (ca. 10%) of plant species exude latex or exudates, and although the defensive roles of plant latex against herbivorous insects have long been suggested by several studies, the detailed roles and functions of various latex ingredients, proteins and chemicals, in anti-herbivore plant defenses have not been well documented despite the wide occurrence of latex in the plant kingdom. Recently, however, substantial progress has been made. Several la

Plant-to-plant defence induction in cotton is mediated by delayed release of volatiles upon herbivory.

Caterpillar feeding immediately triggers the release of volatile compounds stored in the leaves of cotton plants. Additionally, after 1 d of herbivory, the leaves release other newly synthesised volatiles. We investigated whether these volatiles affect chemical defences in neighbouring plants and whether such temporal shifts in emissions matter for signalling between plants. Undamaged receiver plants were exposed to volatiles from plants infested with Spodoptera caterpillars. For receiver plants, we measured changes in defence-related traits such as volatile emissions, secondary metabolites, p

Macroevolution of plant defence favours toxin potency over concentration.

Antagonistic coevolution has long been predicted to leave a macroevolutionary signature of escalating defences in host species. Opposing this classic theory, we previously reported phylogenetic declines in the concentrations of cardenolide toxins among milkweed species in the genus Asclepias. Here we tested the hypothesis of a trade-off between toxin concentration and potency, such that species with lower investment in toxin abundance show higher potency per unit toxin. We tested leaf extracts from more than 50 Asclepias species in vitro against the monarch butterfly's neural Na+/K+-ATPase, th

A salivary elicitor in Laedelphax stiatellus positively regulates plant defenses on optimal and suboptimal hosts.

While herbivorous insects often secrete effector proteins to suppress plant immunity, alternative adaptation strategies remain less explored. Here, we report a counterintuitive approach in the small brown planthopper, Laodelphax striatellus. SBPH downregulated a salivary gland-specific elicitor gene, LsG40, when transferred from the optimal host rice to the suboptimal host corn. When expressed in plants, including the model organism tobacco and the two host plants corn and rice, LsG40 acts as a defense elicitor triggering calcium-dependent reactive oxygen species burst, cell death, callose dep

Bottom-up effects of strawberry cultivar resistance on the demographic and predatory performance of Neoseiulus californicus (Phytoseiidae) against Phytonemus pallidus (Tarsonemidae).

The cyclamen mite, Phytonemus pallidus Banks, is a major threat to strawberries, causing significant damage. Chemical control is challenging due to its hidden feeding habits. Therefore, emphasizing host plant resistance is crucial, particularly in the tritrophic system. In the present study, the compatibility of the predatory mite Neoseiulus californicus McGregor with resistant and susceptible strawberry cultivars was evaluated for the management of the cyclamen mite, P. pallidus, under laboratory conditions. Leaves from two greenhouse-adapted cultivars, Sabrina (susceptible) and Albion (resis

Salivary Protein Sfapyrase of Spodoptera frugiperda Stimulates Plant Defence Response.

Plants have developed various resistance mechanisms against herbivorous insects through prolonged coevolution. Plant defence responses can be triggered by specific compounds present in insect saliva. Apyrase, a known enzyme that catalyzes the hydrolysis of adenosine triphosphate (ATP) and adenosine diphosphate (ADP) into adenosine monophosphate (AMP) and inorganic phosphorus, has recently been identified in some herbivorous insects. However, whether insect salivary apyrase induces or inhibits plant responses remains poorly understood. In this study, we identified an apyrase-like protein in the

Adaptation to Plant Defence in an Agricultural Insect Pest: Integrating Genome Scans and Gene Expression in the Soybean Aphid Reveals Multi-Genic Pathways.

In agroecosystems, intense selection pressures cause species to adapt and spread, often leading to the evolution and persistence of pests. Understanding how pests rapidly adapt can help develop sustainable strategies for their management and improve agroecosystem health. Pest adaptation involves stable variations in DNA sequence, as well as dynamic shifts in gene expression, often mediated by non-coding regulatory elements. We examined adaptation to plant defences in the soybean aphid, Aphis glycines, in which virulent aphids have overcome plant defences and avirulent aphids have not. Previous

Rhizobacteria regulate colonising Sitobion avenae aphid populations through induced host resistance and alter plant volatiles promoting early parasitoid recruitment on barley (Hordeum vulgare).

BACKGROUND: Soil rhizobacteria can enhance crop resistance to insect herbivores and influence higher trophic interactions, offering potential for sustainable pest management. Aphids are sap-feeding pests that alter plant physiology, reduce yields and transmit plant viruses. Previous work has shown that inoculation with specific rhizobacterial strains can reduce aphid fitness in short-term trials. Here, we extend this approach by testing effects across three plant growth stages in glasshouse experiments, monitoring natural colonisation by aphids and their natural enemies in an outdoor pot study

Application of methyl jasmonate and salicylic acid lead to contrasting effects on the plant's metabolome and herbivory.

Phytohormone applications are used to mimic herbivory and can induce plant defences. This study investigated (i) metabolomic changes in leaf tissues of Jacobaea vulgaris and J. aquatica after methyl jasmonate (MeJA) and salicylic acid (SA) applications and (ii) the effects on a leaf-chewing, a leaf-mining and a piercing-sucking herbivore. MeJA treated leaves showed clearly different metabolomic profiles than control leaves, while the differences in metabolomic profiles between SA treated leaves and control leaves were less clear. More NMR peaks increased than decreased after MeJA treatment whi

Assessing the Benefits and Costs of the Hydrogen Cyanide Antiherbivore Defense in Trifolium repens.

Understanding the evolution of plant defenses against herbivores requires identifying the benefits and costs of defense. Here, we tested the hypothesis that the benefits and costs of hydrogen cyanide (HCN) defense against herbivory on white clover (Trifolium repens) are temperature dependent. We first tested how temperature affected HCN production in vitro, and then examined how temperature influenced the efficacy of HCN defense of T. repens against a generalist slug (Deroceras reticulatum) herbivore using no-choice and choice feeding trial assays. To understand how temperature affected the co

Mechanisms of plant defense against insect herbivores.

Plants respond to herbivory through various morphological, biochemicals, and molecular mechanisms to counter/offset the effects of herbivore attack. The biochemical mechanisms of defense against the herbivores are wide-ranging, highly dynamic, and are mediated both by direct and indirect defenses. The defensive compounds are either produced constitutively or in response to plant damage, and affect feeding, growth, and survival of herbivores. In addition, plants also release volatile organic compounds that attract the natural enemies of the herbivores. These strategies either act independently

Genome-wide DNA methylation homeostasis influences jasmonate signaling and potato defenses against the potato tuber moth Phthorimaea operculella.

BACKGROUND: DNA methylation is an epigenetic mechanism involved in plant defense, yet its role in insect resistance remains unclear. Although herbivory remodels plant methylomes, the links between DNA methylation, resistance traits, and phytohormone signaling remain unresolved. RESULTS: We induced genome-wide hypomethylation in potato plants, Solanum tuberosum L., using the DNA methyltransferase inhibitor 5-azacytidine (5-azaC), and assessed their resistance to the potato tuber moth (PTM), Phthorimaea operculella Zeller. Foliar application of 100 μm 5-azaC significantly reduced global DNA met

The Cys2/His2-type zinc finger transcription factor CsZAT10 positively regulates Cucumis sativus resistance against Bemisia tabaci by activating the plant defense system.

BACKGROUND: Whiteflies (Bemisia tabaci) are globally invasive agricultural pests that currently lack effective control measures. Cys2/His2-type zinc finger proteins (ZFPs) are ubiquitous transcription factors that play critical roles in regulating plant resistance to various stresses. However, it remains unclear whether these proteins modulate plant defense responses during whitefly infestation. This study investigates the function and mechanisms of the Cys2/His2-type zinc finger transcription factor CsZAT10 in regulating cucumber resistance to B. tabaci. RESULTS: CsZAT10 contains two zinc fi

Exogenous Inhibitors Enhance Plant Resistance to Rice Planthoppers Through Boosting Phenylpropanoid Pathway Under Elevated CO2.

The damage caused by the Nilaparvata lugens to rice plants will become aggravated under elevated CO2 (eCO2), emphasizing the urgent necessity to enhance rice defences against N. lugens under eCO2 conditions. The phenylpropanoid pathway is composed of several branches and is known to enhance plant resistance. Inhibiting key enzymes in certain branches can compensatorily activate other branches, thereby strengthening plant defence. In this study, rice plants were treated with the cinnamic acid-4-hydroxylase (C4H) inhibitor piperonylic acid (PA) and the 4-coumarate:CoA ligase (4CL) inhibitor 3,4-

Feeding of pea leafminer larvae simultaneously activates jasmonic and salicylic acid pathways in plants to release a terpenoid for indirect defense.

The pea leafminer, Liriomyza huidobrensis, is an important pest species affecting ornamental crops worldwide. Plant damage consists of oviposition and feeding punctures created by female adult flies as well as larva-bored mines in leaf mesophyll tissues. How plants indirectly defend themselves from these two types of leafminer damage has not been sufficiently investigated. In this study, we compared the indirect defense responses of bean plants infested by either female adults or larvae. Puncturing of leaves by adults released green leaf volatiles and terpenoids, while larval feeding caused pl

Production of pyrrolizidine alkaloids and shikonins in Echium plantagineum L. in response to various plant stressors.

BACKGROUND: Echium plantagineum, a native of Europe and Africa, is a noxious invasive weed in Australia forming monocultural stands in pastures and rangelands. It produces a complex mixture of bioactive secondary metabolites, including toxic pyrrolizidine alkaloids (PAs), that protect the plant from insect and livestock herbivory and naphthoquinones (NQs), which suppress competition from weeds, insects and pathogens, and also influence invasion success. However, the extent to which allelochemical production is impacted by environmental factors, thereby influencing plant defense against pests,
