Cysteine homeostasis plays an essential role in plant immunity.

• Cysteine is the metabolic precursor of essential biomolecules such as vitamins, cofactors, antioxidants and many defense compounds. The last step of cysteine metabolism is catalysed by O-acetylserine(thiol)lyase (OASTL), which incorporates reduced sulfur into O-acetylserine to produce cysteine. In Arabidopsis thaliana, the main OASTL isoform OAS-A1 and the cytosolic desulfhydrase DES1, which degrades cysteine, contribute to the cytosolic cysteine homeostasis. • Meta-analysis of the transcriptomes of knockout plants for OAS-A1 and for DES1 show a high correlation with the biotic stress series

Ca(2+)/calmodulin regulates salicylic-acid-mediated plant immunity.

Intracellular calcium transients during plant-pathogen interactions are necessary early events leading to local and systemic acquired resistance. Salicylic acid, a critical messenger, is also required for both of these responses, but whether and how salicylic acid level is regulated by Ca(2+) signalling during plant-pathogen interaction is unclear. Here we report a mechanism connecting Ca(2+) signal to salicylic-acid-mediated immune response through calmodulin, AtSR1 (also known as CAMTA3), a Ca(2+)/calmodulin-binding transcription factor, and EDS1, an established regulator of salicylic acid l

The conserved nematode pheromone ascr#18 primes plant immunity.

Plants sense a diverse array of small molecules and macromolecules derived from their natural environment, including diverse microbe-associated molecular patterns (MAMPs) that can trigger defense responses. Several MAMPs have been shown to prime plants for enhanced defense, providing extended protection against pathogens with minimal fitness costs. However, the extent to which conserved small molecule signatures of other phyla contribute to priming of plant defenses is unclear. Here, we demonstrate that exposure of seeds or plants to the ascaroside ascr#18, a pheromone secreted by plant-parasi

Papain-like cysteine proteases as hubs in plant immunity.

902 I. 902 II. 903 III. 903 IV. 903 V. 905 VI. 905 VII. 905 906 References 906 SUMMARY: Plants deploy a sophisticated immune system to cope with different microbial pathogens and other invaders. Recent research provides an increasing body of evidence for papain-like cysteine proteases (PLCPs) being central hubs in plant immunity. PLCPs are required for full resistance of plants to various pathogens. At the same time, PLCPs are targeted by secreted pathogen effectors to suppress immune responses. Consequently, they are subject to a co-evolutionary host-pathogen arms race. When activated, PLCPs

Pathogenicity of and plant immunity to soft rot pectobacteria.

Soft rot pectobacteria are broad host range enterobacterial pathogens that cause disease on a variety of plant species including the major crop potato. Pectobacteria are aggressive necrotrophs that harbor a large arsenal of plant cell wall-degrading enzymes as their primary virulence determinants. These enzymes together with additional virulence factors are employed to macerate the host tissue and promote host cell death to provide nutrients for the pathogens. In contrast to (hemi)biotrophs such as Pseudomonas, type III secretion systems (T3SS) and T3 effectors do not appear central to pathoge

Graft compatibility emerges from the balance between regeneration and immunity: lessons from plant-plant interactions.

Plant grafting is a significant horticultural technique that enables the combination of desirable traits such as enhanced resilience, disease resistance, and productivity. Despite its widespread application, the mechanisms underlying graft compatibility remain poorly understood. Because grafting is largely an anthropogenic process, plants are unlikely to have evolved mechanisms specifically to recognize graft partners. Here, we propose that graft compatibility is not controlled by a dedicated recognition system, but instead emerges from the balance between existing tissue regeneration and immu

Lapachol, a naphthoquinone identified through chemical screening, inhibits plant immunity.

Quinones, a ubiquitous class of organic compounds produced in all kingdoms of life, are recognized for their diverse biological functions. In plants, quinones are implicated in allelopathy, where they act as allelochemicals influencing the growth and development of neighboring plants, of which one well-documented role of quinone is in the induction of haustoria of parasitic plant roots. In this study, through an immune marker gene-based chemical biology screening, we identified Lapachol, a naphthoquinone originally isolated from species of the Bignoniaceae family. Lapachol suppresses elicitor-

GmCBP60b Plays Both Positive and Negative Roles in Plant Immunity.

CBP60b (CALMODULIN-BINDING PROTEIN 60b) is a member of the CBP60 transcription factor family. In Arabidopsis, AtCBP60b not only regulates growth and development but also activates the transcriptions in immune responses. So far, CBP60b has only been studied extensively in the model plant Arabidopsis and rarely in crops. In this study, Bean pod mottle virus (BPMV)-mediated gene silencing (BPMV-VIGS) was used to silence GmCBP60b.1/2 in soybean plants. The silencing of GmCBP60b.1/2 resulted in typical autoimmunity, such as dwarfism and enhanced resistance to both Soybean mosaic virus (SMV) and Pse

A general non-self response as part of plant immunity.

Plants, like other multicellular lifeforms, are colonized by microorganisms. How plants respond to their microbiota is currently not well understood. We used a phylogenetically diverse set of 39 endogenous bacterial strains from Arabidopsis thaliana leaves to assess host transcriptional and metabolic adaptations to bacterial encounters. We identified a molecular response, which we termed the general non-self response (GNSR) that involves the expression of a core set of 24 genes. The GNSR genes are not only consistently induced by the presence of most strains, they also comprise the most differ

A putative nuclear copper chaperone promotes plant immunity in Arabidopsis.

Copper is essential for many metabolic processes but must be sequestrated by copper chaperones. It is well known that plant copper chaperones regulate various physiological processes. However, the functions of copper chaperones in the plant nucleus remain largely unknown. Here, we identified a putative copper chaperone induced by pathogens (CCP) in Arabidopsis thaliana. CCP harbors a classical MXCXXC copper-binding site (CBS) at its N-terminus and a nuclear localization signal (NLS) at its C-terminus. CCP mainly formed nuclear speckles in the plant nucleus, which requires the NLS and CBS domai

Characterization of plant immunity-activating mechanism by a pyrazole derivative.

A newly identified chemical, 4-{3-[(3,5-dichloro-2-hydroxybenzylidene)amino]propyl}-4,5-dihydro-1H-pyrazol-5-one (BAPP) was characterized as a plant immunity activator. BAPP enhanced disease resistance in rice against rice blast disease and expression of a defense-related gene without growth inhibition. Moreover, BAPP was able to enhance disease resistance in dicotyledonous tomato and Arabidopsis plants against bacterial pathogen without growth inhibition, suggesting that BAPP could be a candidate as an effective plant activator. Analysis using Arabidopsis sid2-1 and npr1-2 mutants suggested t

The Arabidopsis PEPR pathway couples local and systemic plant immunity.

Recognition of microbial challenges leads to enhanced immunity at both the local and systemic levels. In Arabidopsis, EFR and PEPR1/PEPR2 act as the receptor for the bacterial elongation factor EF-Tu (elf18 epitope) and for the endogenous PROPEP-derived Pep epitopes, respectively. The PEPR pathway has been described to mediate defence signalling following microbial recognition. Here we show that PROPEP2/PROPEP3 induction upon pathogen challenges is robust against jasmonate, salicylate, or ethylene dysfunction. Comparative transcriptome profiling between Pep2- and elf18-treated plants points to

The quest for long-distance signals in plant systemic immunity.

Plants induce long-lasting systemic immunity after local pathogen attack by emitting resistance-priming signals from infection sites. A number of plant molecules have been proposed as mobile factors for this response, but many do not fully satisfy criteria for timing and action in systemic immunity. Azelaic acid has been identified as a pathogen-induced metabolite in Arabidopsis vascular sap that has several properties of a long-distance resistance-priming signal.

S-nitrosylation of AtSABP3 antagonizes the expression of plant immunity.

Changes in cellular redox status are a well established response across phyla following pathogen challenge. In this context, the synthesis of nitric oxide (NO) is a conspicuous feature of plants responding to attempted microbial infection and this redox-based regulator underpins the development of plant immunity. However, the associated molecular mechanism(s) have not been defined. Here we show that NO accretion during the nitrosative burst promotes increasing S-nitrosylation of the Arabidopsis thaliana salicylic acid-binding protein 3 (AtSABP3) at cysteine (Cys) 280, suppressing both binding

Sugars as Central Integrators of Plant Immunity and Stress Responses.

Sugars are fundamental metabolites that sustain plant growth and development, but accumulating evidence demonstrates that they also function as regulatory hubs in plant defence. In this review, we synthesise current knowledge on how carbohydrate dynamics coordinate metabolism, signalling, and structural responses to shape plant resistance against both pathogens and herbivores. We highlight how spatial and temporal regulation of sugar transport, mediated by SWEET, SUT/SUC, and STP transporters, controls carbon availability at plant-heterotroph interfaces, while sucrose-cleaving enzymes such as

Revisiting SA biosynthesis: new post-PAL route in plant immunity.

Salicylic acid (SA), the active component of aspirin, is a crucial defense phytohormone that coordinates both local and systemic acquired resistance. For decades, SA biosynthesis was attributed to two pathways: (i) the isochorismate synthase pathway, which is dominant in arabidopsis (Arabidopsis thaliana) and other Brassicaceae, and (ii) the phenylalanine ammonia-lyase (PAL) pathway, common in most other plants. However, the PAL pathway remained biochemically incomplete due to the lack of a molecularly identified benzoic acid 2-hydroxylase. Recent findings fill this gap by identifying a conser

The HEAT repeat protein ILITYHIA is required for plant immunity.

Plant innate immunity is mediated in part by resistance (R) proteins that detect pathogens and mount a robust defense response to fight against infection. We previously characterized proteins in the MOS4-associated complex (MAC) that function in the regulation of plant immune responses downstream of the autoactivated R protein snc1. The MAC is a highly conserved spliceosome-associated complex homologous to the nineteen complex (NTC) in yeast and human. The availability of proteomics data sets in these organisms allowed us to systematically test the biological function of additional putative MA

Systemic propagation of immunity in plants.

Systemic immunity triggered by local plant-microbe interactions is studied as systemic acquired resistance (SAR) or induced systemic resistance (ISR) depending on the site of induction and the lifestyle of the inducing microorganism. SAR is induced by pathogens interacting with leaves, whereas ISR is induced by beneficial microbes interacting with roots. Although salicylic acid (SA) is a central component of SAR, additional signals exclusively promote systemic and not local immunity. These signals cooperate in SAR- and possibly also ISR-associated signaling networks that regulate systemic immu

Innate immune memory in plants.

The plant innate immune system comprises local and systemic immune responses. Systemic plant immunity develops after foliar infection by microbial pathogens, upon root colonization by certain microbes, or in response to physical injury. The systemic plant immune response to localized foliar infection is associated with elevated levels of pattern-recognition receptors, accumulation of dormant signaling enzymes, and alterations in chromatin state. Together, these systemic responses provide a memory to the initial infection by priming the remote leaves for enhanced defense and immunity to reinfec

Long Noncoding RNA: An Uncharted Territory in Plant Diseases and Immunity.

Long noncoding RNAs (lncRNAs) are a class of transcripts longer than 200 nucleotides with little or no protein-coding capacity. They represent a substantial fraction of eukaryotic genomes and function as molecular architects that coordinate diverse biological processes and physiological plasticity, including multilayered plant defence systems, through versatile regulatory interactions at multiple levels of gene regulation. Recent advances in high-throughput RNA sequencing technologies have greatly expanded the identification of plant lncRNAs, revealing their diverse roles as scaffolds, decoys,
