Plant Lectin, MoMo30, Pressures HIV-1 to Select for Variants with Deleted N-Linked Glycosylation Sites.

Momordica balsamina, a plant traditionally used in African medicine, contains a 30 kDa protein, MoMo30, previously identified by our group as an anti-HIV agent that binds glycan residues on the gp120 envelope protein, thereby acting as an entry inhibitor. In this study, we investigated whether prolonged exposure to MoMo30 exerts selective pressure on HIV-1 and induces mutations in the viral envelope (env) gene. T-lymphocyte cells were infected with HIV-1NL4-3 and continuously treated with MoMo30 over a 24-day period. Viral RNA was isolated at regular intervals, and env genes were sequenced usi

Transgenic plants expressing ω-ACTX-Hv1a and snowdrop lectin (GNA) fusion protein show enhanced resistance to aphids.

Recombinant fusion proteins containing arthropod toxins have been developed as a new class of biopesticides. The recombinant fusion protein Hv1a/GNA, containing the spider venom toxin ω-ACTX-Hv1a linked to snowdrop lectin (GNA) was shown to reduce survival of the peach-potato aphid Myzus persicae when delivered in artificial diet, with survival <10% after 8 days exposure to fusion protein at 1 mg/ml. Although the fusion protein was rapidly degraded by proteases in the insect, Hv1a/GNA oral toxicity to M. persicae was significantly greater than GNA alone. A construct encoding the fusion protein

Cratylia argentea seed lectin, a possible defensive protein against plant-eating organisms: effects on rat metabolism and gut histology.

This present work was undertaken to answer two basic questions (a) is C. argentea lectin part of the general defensive strategy of the plant against predation by animals? (b) if so, how does it act on them? To achieve these goals the lectin from C. argentea seeds was purified to homogeneity and included at a 2% level in a diet containing 10% total protein and given to growing rats for 10 days. In vivo it was noted that the lectin from C. argentea is resistant to gut proteolysis, binds to the cells lining the small intestine and induces enlargement in the small intestine, caecum and colon, kidn

Enhanced anti-proliferative activity of Jacalin, a plant lectin in combination with taxol in MDA-MB-468 triple-negative breast cancer cells.

Plant lectins derived from a variety of plant sources can inhibit the proliferation of certain types of human cancer cells and have been studied extensively. In the present study, the effects of Jacalin, a Thomsen–Friedenreich disaccharide-binding lectin isolated from jackfruit seeds, on the triple-negative breast cancer (TNBC) cell line MDA-MB-468 was investigated in combination with Taxol. It was observed that Jacalin inhibited the proliferation of these cancer cells. The antiproliferative effect of Jacalin on MDA-MB-468 was found to be reversible. However, when Jacalin was removed from the

Metabolic inhibitors as tools to delineate participation of distinct intracellular pathways in enhancement of lactose-induced dissociation of neutrophil and thymocyte aggregates formed by mediation of a plant lectin.

Signaling processes in the course of the formation of the lectin-mediated aggregates may partake in conveying enhanced stability to the cell clusters. To prove the validity of this reasoning in a model, we have studied the impact of addition of three metabolic inhibitors (N-ethylmaleimide, nordihydroguaiaretic acid, and trifluoperazine) on lactose-dependent dissociation of cell aggregates, formed in the presence of the galactoside-binding mistletoe lectin. Using both human neutrophils and rat thymocytes to avoid measurement of responses restricted to a single cell type, an enhanced dissociatio

The plant lectin wheat germ agglutinin inhibits the binding of pemphigus foliaceus autoantibodies to desmoglein 1 in a majority of patients and prevents pathomechanisms of pemphigus foliaceus in vitro and in vivo.

Pemphigus foliaceus (PF) is a life-threatening autoimmune blistering skin disease caused by pathogenic IgG autoantibodies against desmoglein 1 (dg1), a desmosomal cadherin-type adhesion glycoprotein. Using lectins and glycosidases, we have shown that dg1 displays an N-glycosylation pattern of the complex triantennary type. We have found that lectins and glycosidases interfere with N-bound sugar residues on the amino-terminal ectodomain of dg1 and completely abolish, in vitro, the antigenicity of dg1 in most of the patients' sera. Moreover, in an ex vivo model using punch biopsies from normal h

Insulin-like activity of concanavalin A and wheat germ agglutinin--direct interactions with insulin receptors.

Concanavalin A and wheat germ agglutinin are as effective as insulin in enhancing the rate of glucose transport and in inhibiting epinephrine-stimulated lipolysis in isolated adipocytes. These lectins, also like insulin, inhibit basal as well as epinephrine-stimulated adenylate cyclase activity of membranes obtained from homogenates of fat cells. Low concentrations of wheat germ agglutinin enhance the specific binding of insulin to receptors of fat cells and liver membranes. Higher concentrations of this plant lectin, as well as of concanavalin A, competitively displace the binding of insulin

Reappraisal of the 'lectin hypothesis' in the aetiopathogenesis of coeliac disease.

The agglutinating properties of a crude gluten digest, purified gliadin fractions and established plant lectins were investigated using mammalian erythrocytes, rat enterocytes and normal and coeliac human enterocytes as the target systems. Gliadin preparations failed to cause agglutination of any of the cells tested, whereas established pure plant lectins were active cell agglutinins. These studies indicate that gliadin peptides do not interact with intestinal cells in a polyvalent, lectin-like manner and as such cannot be regarded as true lectins. Mucosal damage in coeliac disease is unlikely

Effects of metabolic inhibitors and lectins on the menadione-dependent generation of H2O2 by rat thymocytes.

The capacity of oxidative metabolism and its regulation is an important factor in disease control. Using scopoletin as a fluorescent substrate of peroxidase the extent of menadione-dependent production of H2O2 by rat thymocytes was determined. The reaction was inhibited by 2,4-dinitrophenol, papaverine, nordihydroguaiaretic acid and iodacetamide. The membrane-penetrating SH reagent N-ethylmaleimide primed the reaction, probably due to an inhibition of glutathione peroxidase. To delineate an influence of cell-surface protein-carbohydrate interactions by exogenous lectins, the impact of cell bin

Augmenting hepatitis C E1E2 glycoprotein antibody detection via an alternative lectin-assisted mannose-binding ELISA.

In this study we compared several plant lectins for use in a lectin capture ELISAs to enrich HCV glycoproteins from cellular extracts for subsequent detection of HCV glycoprotein-specific antibodies. We selected various high-mannose plant lectins and assessed for their properties to capture HCV glycoproteins. Three lectins, Galanthus nivalis lectin (GNL), Narcissus Pseudonarcissus Lectin (NPL) and Hippeastrum hybrid lectin (HHL) demonstrated effective capturing properties. These were subsequently tested for specificity and sensitivity against viral isolates spanning diverse HCV genotypes. Furt

In vitro effect of fucose-specific lectins on rat erythrocyte membranes under normal conditions and during toxic stress.

We studied the effect of various fucose-specific lectins on lipid peroxidation in rat erythrocyte membranes under normal conditions and during exposure to the toxic agent. Under normal conditions parameters of lipid peroxidation in erythrocyte membranes increased after treatment with lectins or cadmium nitrate. Lectins possess biological activity modulated lipid peroxidation in membranes of erythrocytes from experimental animals, which depended on the time of adaptation. Differences were revealed in the effect of bacterial and plant lectins on lipid peroxidation in erythrocytes treated with th

The synthetic kappa-opioid agonist (-)U50,488 does not affect calcium transport into R1.1 mouse thymoma cell line.

In this paper, the effect of the synthetic kappa-opioid agonist (-)U50,488 on 45Ca2- transport into R1.1 mouse thymoma cells is presented. This thymoma cell line expresses selectively the kappa-opioid class of receptors. 45Ca2+ transport into R1.1 cells was not affected by the kappa-opioid agonist (-)U50,488 (10(-10) M-10(-4) M) alone, or in the presence of the plant lectins: PHA (250 microg/ml) and Con A (800 microg/ml), after a 60 min treatment. The plant lectins PHA and Con A stimulated 45Ca2+ transport into R1.1 cells, in high concentrations (100-800 microg/ml) and (200-1000 microg/ml) res

Legume Lectins with Different Specificities as Potential Glycan Probes for Pathogenic Enveloped Viruses.

Pathogenic enveloped viruses are covered with a glycan shield that provides a dual function: the glycan structures contribute to virus protection as well as host cell recognition. The three classical types of N-glycans, in particular complex glycans, high-mannose glycans, and hybrid glycans, together with some O-glycans, participate in the glycan shield of the Ebola virus, influenza virus, human cytomegalovirus, herpes virus, human immunodeficiency virus, Lassa virus, and MERS-CoV, SARS-CoV, and SARS-CoV-2, which are responsible for respiratory syndromes. The glycans are linked to glycoprotein

Structure-function inferences based on molecular modeling, sequence-based methods and biological data analysis of snake venom lectins.

Lectins are a structurally and functionally diverse group of proteins from different sources, capable to recognize and bind specifically carbohydrates. Several snake venoms contain calcium-dependent true lectins (SVLs) that recognize galactose. Herein, in order to enlighten some of the structure-function relationships of snake venom lectins (SVLs), we constructed theoretical models for 10 SVLs based on the Crotalus atrox lectin (CaL), the only SVL crystal structure available, and compared with other animal and plant lectins, and C-type lectin-like proteins (CLPs) that do not bind carbohydrates

[Comparative analysis of membrane glycoproteins of normal and chronic lymphatic leukemia B lymphocytes by lectins].

Eight plant lectins were used to investigate membrane alterations in lymphocytes from patients with chronic lymphocytic leukemia (CLL). By rosetting with lectins attached to latex particles, the cell percentages with the abundance of each lectin receptor were compared in B normal and leukemic lymphocytes. Comparing these data with the number of lectin molecules bound to each cell and the affinity, which are values calculated with 125I-labeled lectins, it was possible to deduce differences in the composition of glycoproteins in B normal and B-CLL lymphocytes membrane. Compared to B normal, B-CL

Effects of lectin ingestion on animal growth and internal organs.

Lectins are essential and omnipresent plant constituents. As many foods are of plant origin, the daily ingestion of lectins by both humans and animals is appreciable. For example, in an ad hoc survey, 53 edible plants were shown to contain lectins and approx 30% of fresh and processed food regularly consumed by humans had significant hemagglutinating activity (1). The situation is potentially even more acute in animal nutrition because animal diet is less diverse than that of humans, and in most instances foodstuffs are not thoroughly heat-treated. This is particularly significant in the light

Mitogenic potential of endogenous GlcNAc-specific lectins from rat brain cell nuclear membranes.

BACKGROUND: Lectins are carbohydrate-binding proteins that participate in diverse biological processes, including cell adhesion, immune modulation, cell proliferation. While plant-derived lectins are well characterized for their proliferative effects, these properties of animal-derived lectins, particularly from neural tissues, remain poorly understood. This study aimed to evaluate the proliferative potential of a GlcNAc-specific lectin isolated from the cell nuclear membrane of rat brain cortex cells using the MTT assay. METHODS: GlcNAc-specific lectins were purified via affinity chromatogra
