The science of alternative proteins: Comparing animal and plant-based sources.

The very basic concepts of components regarding dietary food upholds providing reinforcement to the cells, tissues, organs for physiological activities. Each dietary food type gives unique nutrition, strength and serves chemical compound required for the organism to perform any different function such as metabolism (catabolism and anabolism), growth, development and senescence. Protein is one of the dietary foods among dietary food types i.e. carbohydrates, lipids, fats and essential oil. Plant proteins consumption rate are very less when compared to the consumption of animal protein as food.

A synthetic biomolecular condensate from plant proteins with controlled colloidal properties.

A synthetic biomolecular condensate (sBC) consisting of a prolamin-rich, plant protein, zein, has been engineered. These artificial condensates were prepared from the liquid-liquid phase (LLP) separation of a protein-rich liquid phase in water. To ensure the colloidal stability of the separated condensate, the protein was chemically modified either via quaternization with glycidyl trimethyl ammonium chloride (GTMAC) or covalently connecting poly(ethylene glycol) by reductive amination, respectively. The modified protein condensates, termed QZs and PZs (for quaternized and PEG-conjugated zein,

Flavor-protein interactions for four plant proteins with ketones and esters.

The interaction between flavors and proteins results in a reduced headspace concentration of the flavor, affecting flavor perception. We analyzed the retention of a series of esters and ketones with different chain lengths (C4, C6, C8, and C10) by protein isolates of yellow pea, soy, fava bean, and chickpea, with whey as a reference. An increase in protein concentration led to a decrease in flavor compound in the headspace as measured with atmospheric pressure chemical ionization time-of-flight mass spectroscopy (APCI-TOF-MS). Flavor retention was described with a flavor-partitioning model. It

LC-MS determination of Nicotiana benthamiana host plant proteins in the drug products of recombinant plant-produced pembrolizumab.

Host proteins from Nicotiana benthamiana can remain in the recombinant biologic products after undergoing multiple steps of purification. These plant protein impurities may induce immunogenicity upon use. Therefore, controlling and monitoring host cell proteins is necessary throughout the process of recombinant protein production. Liquid chromatography-mass spectrometry (LC-MS) has been successfully used to characterize protein species down to nanogram levels in various types of samples. In this study, LC-MS was applied to detect N. benthamiana plant proteins in plant-produced pembrolizumab an

Tracking newly synthesised proteins in plants.

Protein synthesis rate is distinct from steady-state protein abundance and must be measured independently to understand how cells regulate protein levels and respond to environments. Technologies used to predict or directly quantify protein synthesis include ribosome profiling, radioactive and stable isotope labelling, and bioorthogonal noncanonical amino acid tagging (BONCAT). Here, we emphasise BONCAT's potential in plants, which, when combined with click-chemistry, enables the detection, enrichment, and characterisation of newly synthesised proteins themselves. We summarise lessons from BON

Protein Persulfidation in Plants: Function and Mechanism.

As an endogenous gaseous transmitter, the function of hydrogen sulfide (H2S) has been extensively studied in plants. Once synthesized, H2S may be involved in almost all life processes of plants. Among them, a key route for H2S bioactivity occurs via protein persulfidation, in which process oxidizes cysteine thiol (R-SH) groups into persulfide (R-SSH) groups. This process is thought to underpin a myriad of cellular processes in plants linked to growth, development, stress responses, and phytohormone signaling. Multiple lines of emerging evidence suggest that this redox-based reversible post-tra

Electrophoretic Approaches for Investigating Plant-Derived Compound Interactions With DNA and Proteins.

Plant-derived compounds are increasingly recognized as promising therapeutic agents due to their broad biological activities and structural diversity, making their interactions with DNA and proteins central to understanding mechanisms of action and guiding drug discovery. Electrophoretic techniques probe these interactions by tracking changes in electrophoretic mobility, conformation, complex formation, and biopolymer integrity. This review surveys electrophoretic strategies for characterizing plant-derived compound-DNA and -protein interactions, covering fundamental DNA-binding modes (interca

Plant vernalization proteins contain unusual PHD superdomains without histone H3 binding activity.

PHD fingers are modular domains in chromatin-associated proteins that decode the methylation status of histone H3 tails. A PHD finger signature is found in plant vernalization (VEL) proteins, which function as accessory factors of the Polycomb system to control flowering in Arabidopsis through an epigenetic silencing mechanism. It has been proposed that VEL PHD fingers bind to methylated histone H3 tails to facilitate association of the Polycomb silencing machinery with target genes. Here, we use structural analysis by X-ray crystallography to show that the VEL PHD finger forms the central mod

The plant apoplasm is an important recipient compartment for nematode secreted proteins.

Similarly to microbial pathogens, plant-parasitic nematodes secrete into their host plants proteins that are essential to establish a functional interaction. Identifying the destination of nematode secreted proteins within plant cell compartment(s) will provide compelling clues on their molecular functions. Here the fine localization of five nematode secreted proteins was analysed throughout parasitism in Arabidopsis thaliana. An immunocytochemical method was developed that preserves both the host and the pathogen tissues, allowing the localization of nematode secreted proteins within both org

CFEM proteins: unique to fungi with essential roles in plant host interactions.

Fungal diseases pose a serious threat to agriculture, forestry, and human health sectors and are major contributors to global economic losses. Plant-pathogenic fungi rely on a wide array of molecules that contribute to pathogenicity and virulence; thus, it is critical to understand their roles in plant-fungal interactions to propose control strategies. Among these, CFEM domain-containing proteins, which are unique to fungi primarily within the phyla Ascomycota and Basidiomycota, have diverse functions in the interaction with the host plant. The CFEM domain is characterized by the presence of e

Lipid transfer proteins involved in plant-pathogen interactions and their molecular mechanisms.

Nonspecific lipid transfer proteins (LTPs) are small, cysteine-rich proteins that play numerous functional roles in plant growth and development, including cutin wax formation, pollen tube adhesion, cell expansion, seed development, germination, and adaptation to changing environmental conditions. LTPs contain eight conserved cysteine residues and a hydrophobic cavity that provides a wide variety of lipid-binding specificities. As members of the pathogenesis-related protein 14 family (PR14), many LTPs inhibit fungal or bacterial growth, and act as positive regulators in plant disease resistanc

Utilizing Proteomic Approach to Analyze Potential Antioxidant Proteins in Plant against Irradiation.

Gamma-ray irradiation is an effective and clean method of sterilization by inactivating microorganisms. It can also be applied to induce anti-oxidants for future application. In this study, the mung bean (Vigna radiata) was exposed to gamma-ray irradiation under the dose of 0, 5 or 10 kGy. With increasing irradiation doses, the concentrations of malondiadehyde decreased while the levels of total flavonoids and DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity increased. It has been shown that consuming flavonoids can provide protective effects. In addition, proteomic analysis id

Meat, dairy and plant proteins alter bacterial composition of rat gut bacteria.

Long-term consumption of red meat has been considered a potential risk to gut health, but this is based on clinic investigations, excessive intake of fat, heme and some injurious compounds formed during cooking or additions to processed meat products. Whether intake of red meat protein affects gut bacteria and the health of the host remains unclear. In this work, we compared the composition of gut bacteria in the caecum, by sequencing the V4-V5 region of 16S ribosomal RNA gene, obtained from rats fed with proteins from red meat (beef and pork), white meat (chicken and fish) and other sources (

Plant protein and animal proteins: do they differentially affect cardiovascular disease risk?

Proteins from plant-based compared with animal-based food sources may have different effects on cardiovascular disease (CVD) risk factors. Numerous epidemiologic and intervention studies have evaluated their respective health benefits; however, it is difficult to isolate the role of plant or animal protein on CVD risk. This review evaluates the current evidence from observational and intervention studies, focusing on the specific protein-providing foods and populations studied. Dietary protein is derived from many food sources, and each provides a different composite of nonprotein compounds th

Digestion-resistant proteins support the healthy metabolite profiles associated with plant-based diets.

Plant-based diets are associated with both positive health outcomes and a diverse gut microbiome. Such diets alter the microbiome's metabolic outputs, including increasing phenylalanine-derived phenols associated with beneficial health outcomes (hippuric acid and 3-phenylpropionate), while decreasing tyrosine-derived phenols considered uremic toxins (phenol sulfate and p-cresol sulfate). The mechanisms linking plant eating to these phenol metabolites are not known. Plant-based foods are fiber and phytochemical rich. They also contain proteins that are resistant to host digestion and thus reach

A multi-modal workflow identifies non-γ-glutamyl kokumi peptides in plant proteins.

Plant proteins are increasingly incorporated into sustainable foods, yet their sensory performance falls short of consumer expectations. Kokumi sensation is typically associated with γ-glutamyl peptides, which enhance mouthfulness and flavour continuity via activation of the calcium-sensing receptor (CaSR). This study develops a multi-modal workflow to discover plant-derived CaSR-active peptides beyond γ-glutamyl motifs. Hemp, quinoa, and black gram were profiled using untargeted proteomics and in silico cryptide prediction. Black gram protein concentrate demonstrated the highest kokumi-genera
