Nodulation Signaling Pathway 2: A Proposed Multifunctional Signaling Hub for Coordinating Plant Architecture, Rhizosphere Chemistry, and Symbiotic Associations Under Nutrient Stress.

Under nutrient stress, plants adjust shoot branching, root remodeling, and symbiotic associations to optimize survival and growth. The GRAS-type transcription factor NODULATION SIGNALING PATHWAY 2 (NSP2) participates in these responses through context-dependent interactions with partners: NSP1, MYB40, and SLR1, and is indirectly repressed by SMAX1 via NSP1 and SLR1. NSP2 functions differ among species: it directly binds DNA in rice (Oryza sativa) strigolactone (SL) biosynthesis, whereas in M. truncatula it acts as a co-regulator in nodulation and flavonoid regulation, with NSP1 or MYB40 provid

Which steps are essential for the formation of functional legume nodules?

Nodulation is reviewed in terms of the phenotypes proposed by Vincent (1980). Individual legumes may be infectible by one or more of the three bacterial genera (collectively known as rhizobia) Rhizobium, Bradyrhizobium, or Azorhizobium. The type of infection process by which rhizobia gain entry is largely governed by the host genotype. In addition to the widely studied root-hair pathway, infections may be associated with lateral root emergence or occur between root epidermal cells. The exact chemical and physical nature of the root hair/epidermal cell wall is likely to be a critical factor in

Nodulation Induces Systemic Resistance of Medicago truncatula and Pisum sativum Against Erysiphe pisi and Primes for Powdery Mildew-Triggered Salicylic Acid Accumulation.

Plants encounter beneficial and detrimental microorganisms both above- and belowground and the health status of the plant depends on the composition of this pan-microbiome. Beneficial microorganisms contribute to plant nutrition or systemically or locally protect plants against pathogens, thus facilitating adaptation to a variety of environments. Induced systemic resistance, caused by root-associated microbes, manifests as aboveground resistance against necrotrophic pathogens and is mediated by jasmonic acid/ethylene-dependent signaling. By contrast, systemic acquired resistance relies on sali

Toxicity testing of heavy metals with the Rhizobium-legume symbiosis: High sensitivity to cadmium and arsenic compounds.

Legume root nodules are the site of biological nitrogen fixation in the Rhizobium-legume symbiosis. Nodules are structures unique to this symbiosis and they are morphologically as well as physiologically distinct from other plant organs. Organic substances affecting the macro- or microsymbionts vitality, such as PAHs (WETZEL: et al., 1991), reduce nodulation even before visible damage to the plant can be detected. We present data that the formation of nodules (nodulation) may also serve for ecotoxicological evaluation of heavy metals in different binding states. Tests were performed in petri d

Glucomannan-mediated attachment of Rhizobium leguminosarum to pea root hairs is required for competitive nodule infection.

The Rhizobium leguminosarum biovar viciae genome contains several genes predicted to determine surface polysaccharides. Mutants predicted to affect the initial steps of polysaccharide synthesis were identified and characterized. In addition to the known cellulose (cel) and acidic exopolysaccharide (EPS) (pss) genes, we mutated three other loci; one of these loci (gmsA) determines glucomannan synthesis and one (gelA) determines a gel-forming polysaccharide, but the role of the other locus (an exoY-like gene) was not identified. Mutants were tested for attachment and biofilm formation in vitro a

Lotus japonicus CLV1-Like Receptor HAR1 Promotes Nitrogen Utilisation and Growth Under Non-Symbiotic Conditions.

Legumes establish mutualistic symbiosis with nitrogen (N)-fixing bacteria, which allows them to utilise atmospheric N2. Because the maintenance of symbiosis requires abundant carbon (C) sources, legumes regulate the balance between carbon consumption and nitrogen acquisition by systemically controlling the nodule number through CLAVATA1 (CLV1)-like receptors. In Lotus japonicus, the CLV1-like receptor HYPERNODULATION ABERRANT ROOT FORMATION1 (HAR1) acts in shoots to regulate root nodulation and contributes to symbiotic C/N coordination. This raises the possibility that HAR1 may also influence

Single-Cell RNA-Sequencing in Nodulated Roots of Medicago truncatula.

This section details the optimized methods for single-cell RNA sequencing (scRNA-seq) in Medicago truncatula roots and nodules. We describe a detailed experimental workflow, comprising plant growth and bacteria application, protoplast isolation, quality control, and library processing according to 10× Genomics®, suitable for both long- and short-read sequencing. This protocol can serve as a starting point for establishing robust, high-quality scRNA-seq workflows to study nodulation and root development in legumes.

Genome-wide identification of MtTCTPs and functional analysis of MtTCTP3 as a positive regulator of nodulation in Medicago truncatula.

The Translationally Controlled Tumor Protein (TCTP) is a conserved eukaryotic regulator of cell proliferation and stress responses, but its function in legume-rhizobium symbiosis remains unclear. Here, we identified five MtTCTP genes in the model legume Medicago truncatula and characterized the role of MtTCTP3 in symbiotic nodulation. MtTCTP3 is highly expressed in root nodules, particularly in the nodule apical meristem. Overexpression of MtTCTP3 significantly enhanced nodule formation, nitrogenase activity, and biomass accumulation, while silencing of MtTCTP3 caused reduced nodule number and

Systemic Effects of Nitrate, Asparagine, and Glutamine on Nodulation and Biological Nitrogen Fixation in Soybean.

Although it is well established that nitrate exerts an inhibitory effect on nodulation and biological nitrogen fixation in soybean, the underlying mechanism remains unclear. In soybean plants, nitrate is assimilated into asparagine (Asn) and glutamine (Gln); their systemic circulation within the plant may contribute to the reduced N-fixation capacity of nodules. To investigate the effects of nitrate, Asn, and Gln on soybean nodulation and biological N fixation, a unilateral nodulated double-root soybean system was used. The non-nodulated side roots were supplied with nitrate (14 mM), Asn (20 m

Molecular cartography of root nodule organogenesis: insights into the role of transcription factors.

Nodule organogenesis is a complex developmental process which is induced post-embryonically following rhizobia infection and relies on coordinated action of a suite of transcription factors at multiple stages starting from its initiation, de novo meristem establishment and differentiation. The rewiring of nitrate-responsive NIN-LIKE PROTEIN (NLP) homolog, NODULE INCEPTION (NIN) transcription factor through genetic adaptations in the common ancestor of the nitrogen-fixing clade is crucial to the origin of nodulation trait. Moreover, nodule formation emerged through the co-option of evolutionary

MtGA2ox10 encoding C20-GA2-oxidase regulates rhizobial infection and nodule development in Medicago truncatula.

Gibberellin (GA) plays a controversial role in the legume-rhizobium symbiosis. Recent studies have shown that the GA level in legumes must be precisely controlled for successful rhizobial infection and nodule organogenesis. However, regulation of the GA level via catabolism in legume roots has not been reported to date. Here, we investigate a novel GA inactivating C20-GA2-oxidase gene MtGA2ox10 in Medicago truncatula. RNA sequencing analysis and quantitative polymerase chain reaction revealed that MtGA2ox10 was induced as early as 6 h post-inoculation (hpi) of rhizobia and reached peak transcr

Exo-oligosaccharide signaling in Lotus japonicus roots promotes synthesis and secretion of symbiotic phenylpropanoids resulting in increased production of Nod factors in rhizobia.

Besides lipo-chitooligosaccharidic Nod factors (NFs), rhizobial exo-oligosaccharides (EOS), derived from exo-polysaccharides (EPS), are symbiotic signals that promote nodule formation on legume roots. However, plant genes regulated by EOS signaling remain unidentified. Here, we used purified EPS and the rhizobial glycanase ExoK to produce EOS in vitro. EOS was applied to Lotus japonicus roots to identify genome-wide gene expression changes. Mutants of Sinorhizobium sp. NGR234 and L. japonicus were used to investigate the symbiotic roles of EOS and NFs. Transcriptomic analysis showed that EOS i

A rhizobium-induced FT-FD module locally activates nodule stem cell gene to promote nodulation.

Functional divergence of gene homologs enables species to evolve unique physiological processes, such as nitrogen fixation of nodules in legumes. Nodulation is initiated via rhizobia-induced reactivation of root cortical cells into stem cells, yet the molecular mechanism underlying this process has remained elusive. There are eight florigen homologs in the soybean genome, including GmFT2a and GmFT5a, which are produced in leaves and possibly translocated to roots to promote nodulation. Here, we report the identification of a distinct florigen homolog, GmFT5b, and its partner GmFDL23, both of w

Nitrogen Fertilization Modulates Soybean Nodulation and Nitrogen Fixation via NO-Mediated S-Nitrosylation.

Soybean, a symbiotic nitrogen-fixing crop, experiences suppressed nodule nitrogen fixation under excessive nitrogen fertilizer. Nitric oxide (NO) is a key signaling molecule regulating development and stress, primarily via protein S-nitrosylation, although its role in soybeans is unclear. Using a unilateral nodulation system, treatments with nitrogen, an NO scavenger, and an NO donor were applied. Results showed that nitrogen application increased the NO content in the nodules and reduced the nitrogen fixation capacity. Conversely, the NO scavenger lowered the NO levels but enhanced fixation.

Reactive Sulfur Species Produced by Cystathionine γ-lyase Function in the Establishment of Mesorhizobium loti-Lotus japonicus Symbiosis.

Reactive sulfur species (RSS) are present in root nodules; however, their role in symbiosis and the mechanisms underlying their production remain unclear. We herein investigated whether RSS produced by the cystathionine γ-lyase (CSE) of microsymbionts are involved in root nodule symbiosis. A cse mutant of Mesorhizobium loti exhibited the decreased production of hydrogen sulfide and other RSS. Although the CSE mutation of M. loti did not affect the early stages of symbiosis, i.e., infection and nodulation, with Lotus japonicus, it reduced the nitrogenase activity of nodules and induced their ea

The Compact Root Architecture 2 systemic pathway is required for the repression of cytokinins and miR399 accumulation in Medicago truncatula N-limited plants.

Legume plants can acquire mineral nitrogen (N) either through their roots or via a symbiotic interaction with N-fixing rhizobia bacteria housed in root nodules. To identify shoot-to-root systemic signals acting in Medicago truncatula plants at N deficit or N satiety, plants were grown in a split-root experimental design in which either high or low N was provided to half of the root system, allowing the analysis of systemic pathways independently of any local N response. Among the plant hormone families analyzed, the cytokinin trans-zeatin accumulated in plants at N satiety. Cytokinin applicati

Spatiotemporal dynamics of ethylene biosynthesis shape infection and nodule initiation in Medicago truncatula.

Ethylene is a well-established negative regulator of nodulation, yet how ethylene biosynthesis and perception are spatially coordinated during early symbiotic signaling remains unresolved. Here, we investigate the dynamics of ethylene responses in Medicago (Medicago truncatula) using transcriptomics, promoter-reporter analyses, loss-of-function approaches and a synthetic reporter. We show that the activity of the ethylene-responsive EBSn reporter shifts from inner root tissues under nonsymbiotic conditions to the outer cortex and epidermis following rhizobial inoculation, revealing a spatial r

Identification of growth-promoting microorganisms and their impact on chickpea growth in tropical regions.

The study of plant growth-promoting microorganisms is crucial for developing new agricultural strategies aimed at increasing productivity and resilience in semi-arid environments, where water scarcity and soil degradation pose critical challenges. Therefore, this study aimed to identify and relate the effects of inoculation of growth-promoting or nodulating microorganisms in isolates from chickpea roots grown in a semiarid region. The nodules were washed with distilled water, 95% ethanol and 3% NaClO. They were then crushed, and the resulting suspension was diluted six times in saline solution

Leguminous plants nodulated by selected strains of Cupriavidus necator grow in heavy metal contaminated soils amended with calcium silicate.

Increasing concern regarding mining area environmental contamination with heavy metals has resulted in an emphasis of current research on phytoremediation. The aim of the present study was to assess the efficiency of symbiotic Cupriavidus necator strains on different leguminous plants in soil contaminated with heavy metals following the application of inorganic materials. The application of limestone and calcium silicate induced a significant increase in soil pH, with reductions in zinc and cadmium availability of 99 and 94 %, respectively. In addition, improved nodulation of Mimosa caesalpini
