Plant stanols induce intestinal tumor formation by up-regulating Wnt and EGFR signaling in Apc Min mice.

The rate of APC mutations in the intestine increases in middle-age. At the same period of life, plant sterol and stanol enriched functional foods are introduced to diet to lower blood cholesterol. This study examined the effect of plant stanol enriched diet on intestinal adenoma formation in the Apc(Min) mouse. Apc(Min) mice were fed 0.8% plant stanol diet or control diet for nine weeks. Cholesterol, plant sterols and plant stanols were analyzed from the caecum content and the intestinal mucosa. Levels of β-catenin, cyclin D1, epidermal growth factor receptor (EGFR) and extracellular signal-re

New nortriterpenoid isolated from anti-rheumatoid arthritic plant, Tripterygium wilfordii, modulates tumor growth and neovascularization.

Preparations of Tripterygium wilfordii, "Thunder God vine", have been used in China to treat rheumatoid arthritis. Rheumatoid arthritis, as well as solid tumors, is closely associated with neovascularization. Antiarthritic drugs therefore may modulate tumor growth as well as neovascularization. We found that a compound purified from T. wilfordii, the nortriterpenoid, demethylzeylasteral (TZ-93), inhibited the proliferation of vascular endothelial cells approximately 30 times more effectively than it did for the proliferation of human tumor cells. In in vitro assays using bovine aortic endothel

Platycodon grandiflorum, as a medicinal and food homologous plant: a comprehensive review of anti-tumor components, mechanisms, modern applications, and preventive healthcare.

Platycodon grandiflorum (Jacq.) A. D C. (P. grandiflorum), as a medicinal and edible plant, is widely distributed in China. In terms of Chinese medical theories, Traditional Chinese Medicine (TCM) believes that the root of P. grandiflorum has the functions of relieving sore throat, eliminating phlegm, and draining pus, and is widely used in lung diseases and respiratory system disorders. Modern pharmacological studies have shown that Platycodon grandiflorum saponin, flavonoids, polysaccharides, and phenolic acids are the main active components of P. grandiflorum, which have significant antitus

Fish Oil, Plant Polyphenols, and Their Combinations Have No Tumor Growth Promoting Effects on Human Lung and Colon Carcinoma Xenograft Mice.

The goal of this study was to evaluate if combinations of ingredients with known anti-cachexia benefits (Fish oil-FO with either curcumin or Green tea extract-GTE), have adverse effects on tumor growth, using human carcinoma xenograft mice models. FO (EPA/DHA 360 mg/kg bw), GTE (90 mg/kg bw), and curcumin (180 mg/kg bw) were administered orally, alone or in combination, to nude mice bearing either A549 human non-small cell lung carcinoma or SW620 human colon carcinoma tumors. Bodyweight, tumor growth, survival time, and other clinical endpoints were assessed. The ingredients either alone or in

Simple plant-based production and purification of the assembled human ferritin heavy chain as a nanocarrier for tumor-targeted drug delivery and bioimaging in cancer therapy.

Nanoparticles are used as carriers for the delivery of drugs and imaging agents. Proteins are safer than synthetic nanocarriers due to their greater biocompatibility and the absence of toxic degradation products. In this context, ferritin has the additional benefit of inherently targeting the membrane receptor transferrin 1, which is overexpressed by most cancer cells. Furthermore, this self-assembling multimeric protein can be loaded with more than 2000 iron atoms, as well as drugs, contrast agents, and other cargos. However, recombinant ferritin currently costs ~3.5 million € g-1 , presumabl

Plant-derived extracellular vesicles as a natural drug delivery platform for glioblastoma therapy: A dual role in preserving endothelial integrity while modulating the tumor microenvironment.

Glioblastoma (GBM) is the most aggressive primary brain tumor, with limited treatment options due to the restrictive blood-brain barrier (BBB) and the heterogeneity of the blood-tumor barrier (BTB). Temozolomide (TMZ), the standard chemotherapy, suffers from poor BBB permeability, rapid degradation, and systemic toxicity. Plant-derived extracellular vesicles (PDEVs) have emerged as promising natural nanocarriers, offering biocompatibility, stability, and the ability to cross biological barriers. This study investigates the use of extracellular vesicles from Citrus limon L. (LDEs) to encapsulat

Explicit water-ligand docking, drug-likeness and molecular dynamics simulation analysis to predict the potency of Boerhavia diffusa plant extract against mutant wilms tumor-1 protein responsible for type 4 nephrotic syndrome.

Thestructure and function of a protein are closely connected. Changes in a protein structure can impact on its function. Nephrotic syndrome type 4 (NPHS4) is an uncommon genetic condition caused by mutations in the WT1 gene, which codes for the wilms tumor-1 protein. Several studies have discovered that patients with nephrotic syndromes are resistant to steroid therapy and are likely to develop end-stage renal failure. The use of phytochemicals-based therapeutics is in demand due to their high potential and low toxicity. Based on this context, we employed the Autodock raccoon to screen 67 dist

Induction of breast and lung neoplastic lesions in mice by alpha-ecdysone.

Alpha ecdysone is naturally present in the leaves of some plants, including the carcinogenic plant bracken fern (Pteris aquilina). Feeding mice with alpha-ecdysone at a dose level of 3 ppm/25 g body weight, 2 times per week for 22 months induced a significant increase in the body weight of the animals. Breast and lung tumors developed in the experimental mice. Other pathological changes such as fibrocystic disease of the breast and bronchocentric granulomas appeared in these animals. The results of this study proved that alpha-ecdysone may be one of the active carcinogens in bracken fern plant

A cohort mortality study of petrochemical workers.

A historical prospective cohort mortality study was conducted for a cohort of 6,588 white male employees of a Texas petrochemical plant because of a suspected increased incidence of malignant brain tumors. Mortality experience from 1941 to 1977 was determined and compared with that of the general U.S. white male population adjusting for age and time period. Overall and cause-specific standardized mortality ratios were calculated for various subgroups of the population defined by length of employment, latency and payroll status. Significant deficits in total cohort mortality were found for all

Induction Medium to Analyze In Vitro Morphological Changes and Biotrophic Features in Sporidia of Ustilago maydis.

Biotrophic fungal pathogens such as Ustilago maydis rely on a host to complete their lifecycle. U. maydis has the advantage, in contrast to obligate biotrophs such as rust fungi, that the saprophytic yeast-like sporidia can be cultivated in artificial media, while the dikaryotic filamentous and infectious stage depends on the host to complete the sexual phase of the lifecycle. Key phenotypes during plant infection include filamentous growth after mating, an increase of fungal biomass in developing tumors, production of an extracellular matrix (ECM) around fungal cells, and melanin formation du

Angioprevention of Urologic Cancers by Plant-Derived Foods.

The number of cancer cases worldwide keeps growing unstoppably, despite the undeniable advances achieved by basic research and clinical practice. Urologic tumors, including some as prevalent as prostate, bladder or kidney tumors, are no exceptions to this rule. Moreover, the fact that many of these tumors are detected in early stages lengthens the duration of their treatment, with a significant increase in health care costs. In this scenario, prevention offers the most cost-effective long-term strategy for the global control of these diseases. Although specialized diets are not the only way to

Microbe Profile: Plasmodiophora brassicae, the gall-forming protist behind clubroot disease.

Plasmodiophora brassicae is a soil-borne intracellular obligate biotrophic protist and the causal agent of clubroot disease in Brassicaceae. Phylogenetically placed within the Rhizaria supergroup, it exhibits a complex two-stage life cycle leading to the formation of club-shaped galls, driven by phytohormone and effector-mediated host reprogramming. A recently obtained telomere-to-telomere genome assembly of the Canadian pathotype Pb3A revealed a compact genome structure encoding a complex secretome. Despite its agronomic relevance, key aspects of its biology, including the existence of sexual
