Glucose-TOR-PDCB signaling restricts cell-to-cell communication through callose deposition.

Plant cells are connected by plasmodesmata (PD), membrane-lined channels that facilitate cell-to-cell transport. Forward genetic screens to uncover regulators of PD transport identified mutants with increased (ise1 to ise4) or decreased (dse1) PD trafficking during embryogenesis. Despite their opposite effects on PD transport, we found that the transcriptional profiles of dse1, ise3, and ise4 were notably similar with one notable exception: the set of genes controlled by the conserved kinase TARGET OF RAPAMYCIN (TOR) and ABI5, a bZIP transcription factor that acts downstream of TOR. We then sh

From a Movement-Deficient Grapevine Fanleaf Virus to the Identification of a New Viral Determinant of Nematode Transmission.

Grapevine fanleaf virus (GFLV) and arabis mosaic virus (ArMV) are nepoviruses responsible for grapevine degeneration. They are specifically transmitted from grapevine to grapevine by two distinct ectoparasitic dagger nematodes of the genus Xiphinema. GFLV and ArMV move from cell to cell as virions through tubules formed into plasmodesmata by the self-assembly of the viral movement protein. Five surface-exposed regions in the coat protein called R1 to R5, which differ between the two viruses, were previously defined and exchanged to test their involvement in virus transmission, leading to the i

A binding-activated CRISPR/dCas13X imaging system enables precise RNA tracking in plants.

Visualizing the spatiotemporal dynamics of native RNA molecules with single-transcript-level sensitivity in living plant cells remains a formidable challenge due to the high background noise caused by the unique cellular architecture of plants. Here, using transiently transformed Nicotiana benthamiana cells and stable Arabidopsis thaliana lines, we present a high-signal-to-noise RNA imaging platform that couples CRISPR-dCas13X with a binding-associated fluorescence stabilization strategy. We identified a novel plant-specific degron that selectively targets unbound probes for proteasomal degrad

A P-type pentatricopeptide repeat-containing protein interacts with the Japanese soil-borne wheat mosaic virus movement protein and modulates susceptibility to infection.

As obligate intracellular pathogens, plant viruses interact with host factors to achieve efficient replication and spread. Host factors also play important roles in virus detection and antiviral defence. Furoviruses cause substantial losses in cereals and efficient control measures are lacking. We therefore used Japanese soil-borne wheat mosaic furovirus movement protein (MPJSBWMV) in immunoprecipitation experiments to identify host interaction partners in the model host plant Nicotiana benthamiana. We identified a P-type pentatricopeptide repeat-containing protein (NbPPR) as the interaction p

Nicotiana tabacum Kunitz Peptidase Inhibitor-like Protein Regulates Intercellular Transport.

A coordinated and generalized plant response to adverse environmental factors largely depends on the proper and finely-tuned regulation of intercellular transport via plasmodesmata (PD). However, the knowledge of the whole network of PD-controlling mechanisms is far from complete. Earlier, a cellular factor, Kunitz peptidase inhibitor-like protein (KPILP), that affects PD gating and plays a proviral role, was identified in Nicotiana benthamiana plants. Here we characterized its homolog from N. tabacum, NtKPILP, which is hardly detectable in leaves of intact plants, in contrast to roots, flower

Phytohormones and emerging plant growth regulators in tailoring plant immunity against viral infections.

Viral infections are major contributors to crop yield loss and represent a significant threat to sustainable agriculture. Plants respond to virus attacks by activating sophisticated signalling cascades that initiate multiple defence mechanisms. Notably, several phytohormones, including salicylic acid (SA), jasmonic acid (JA), abscisic acid (ABA), and ethylene (ET), are known to shape these defence responses. In recent years, various plant growth regulators (PGRs) such as melatonin, carrageenans, sulfated fucan oligosaccharides, nitric oxide (NO), brassinosteroids (BRs), and hydrogen sulfide (H

Tomato Brown Rugose Fruit Virus Evades Tm-22-Mediated Resistance by Avoiding the Induction of Tm-22 Self-Association.

Tomato brown rugose fruit virus (ToBRFV) overcomes Tm-22-mediated resistance, posing a serious threat to global tomato production. We previously identified that key residues in the movement protein (MP)-H67, N125, K129, A134, I147 and I168 are essential for ToBRFV evasion of Tm-22 resistance. However, the underlying mechanisms remain unclear. In Tm-22-transgenic Nicotiana benthamiana plants at later infection stages, MP mutations H67C, N125A, K129Q and A134N abolished ToBRFV-GFP cell-to-cell and systemic movement, whereas I168N abolished ToBRFV-GFP systemic but not cell-to-cell movement, and I

Electrical signalling in Nitellopsis obtusa: potential biomarkers of biologically active compounds.

The Nitellopsis obtusa (N.A.Desvaux) J.Groves cell provides a model system for complex investigation of instantaneous effects of various biologically active compounds (BC) on the generation of plant bioelectrical signals in vivo. Experimental evidence using multiple electrical signals as biomarkers of the effects of BC (acetylcholine, asparagine, glutamate, nicotine, aluminium, nickel and cadmium ions) is provided. The effect of BC on membrane transport systems involved in the cell excitability were tested by current clamp, voltage clamp and patch clamp methods. Membrane potential (MP) alterat

The Melon Sterol Transporter Niemann-Pick C1 Protein Is a New Interactor of Cucumber mosaic virus Movement Protein.

Plant viruses need to use many host factors to establish infection. During the viral cycle, intracellular transport is fundamental to reach the plasmodesmata to enable cell-to-cell transport. Cucumovirus CMV (cucumber mosaic virus, CMV) can infect plants from most economically important crops. To identify additional host proteins involved in CMV movement in melon, we used the MP as a bait to screen a Yeast two-hybrid cDNA library from CMV-infected plants and identified a Niemann-Pick C1 (NPC1) protein as a novel MP interactor. NPC1 is a transmembrane protein involved in cholesterol transport i

PlantCCC prioritizes context-specific candidate ligand-receptor communication patterns in plant spatial transcriptomics.

Intercellular communication supports plant development and environmental responses, but its analysis in plant tissues is complicated by cell walls, plasmodesmata, and local tissue architecture. Spatial proximity therefore does not necessarily indicate effective communication. Plant ligand-receptor (L-R) resources also contain expanded gene families, homology-derived mappings, and uneven levels of experimental support. We developed PlantCCC, a spatially aware graph-learning framework that uses a plant L-R database as a candidate search space and combines residual spatial expression enhancement,

Re-assignment of Gongrosira leptotricha, a newly-recorded species in China, to Stephanosphaerinia clade (Chlamydomonadales, Chlorophyceae): insights from morphological and phylogenetic analyses.

A newly-recorded green algal species in China, identified as Gongrosira leptotricha Raineri, exhibits distinctive morphological characteristics. The thallus of this specimen is spherical or slightly irregular, calcified and firmly adheres to stones in running water. The prostrate part consists of loose, thin-walled tissue, with extended filaments being cylindrical and sparsely branched. The terminal sporangia are only slightly larger than the vegetative cells, with smaller cell dimensions and a higher length-to-width ratio. Biflagellate zoospores were formed within terminal sporangia. Each cel

Mobile small RNAs as mediators of stress-induced epigenomic remodeling and systemic stress adaptation.

Plants rely on sophisticated long-distance communication networks to coordinate adaptive responses to changing environmental conditions. Among these, mobile small RNAs (sRNAs), including microRNAs (miRNAs) and small interfering RNAs (siRNAs), function as critical systemic signaling molecules that move between tissues through plasmodesmata and the phloem. While their roles in post-transcriptional gene regulation are well established, emerging evidence indicates that mobile sRNAs also mediate stress-induced epigenomic remodeling in recipient tissues. Specifically, 24-nt siRNAs can direct RNA-dep

Peanut stunt virus movement protein is the limiting factor in Capsicum annuum infection.

Cucumber mosaic virus (CMV) is one of the most devastating plant viruses, with more than 1,200 species of host plants. The host range and economic importance of peanut stunt virus (PSV) are mostly limited to legumes, despite the similar taxonomy and genome structure with CMV. Since no data are available on the background of the limited host range of PSV, RNA 3 recombinant and reassortant viruses were generated (C12P3, P12C3, C12CP3, C12PC3, C12PΔC3) to study their infection phenotype on a common host (Nicotiana benthamiana) and on a selective host (Capsicum annuum cv. Brody). The PSV movement

Receptor-like kinases BIR1 and BIR3 modulate antiviral resistance by different mechanisms.

BRASSINOSTEROID INSENSITIVE1-ASSOCIATED RECEPTOR KINASE 1 (BAK1)-INTERACTING RECEPTOR-LIKE KINASE (BIR) proteins are negative regulators of cell death and defense against microbes in Arabidopsis thaliana. Here, we demonstrate that the members of the BIR family function as negative regulators of antiviral resistance in Arabidopsis. We show that during tobacco rattle virus (TRV) infection, BIR1 and BIR3 gene expression is antagonistically regulated by salicylic acid and jasmonic acid signaling pathways. BIR1 and BIR3 negatively regulate TRV resistance via distinct mechanisms. Our data indicate t

Myosotis scorpioides L. (Boraginaceae) floral nectary: Micromorphology, fluorescence, and ultrastructure.

The small flowers of Myosotis scorpioides are pollinated by various groups of insects feeding on their nectar accumulating at the base of the corolla tube. To date, only few studies have focused on the anatomy and ultrastructure of nectaries in plants from the family Boraginaceae. The aim of this study was to analyse the structure of the M. scorpioides nectary at different levels of organisation, with special emphasis on its ultrastructure, which has not been investigated so far. The studies were carried out with the use of light, fluorescence, scanning electron, and transmission electron micr

Subcellular Localization of Iron in Rhizophora mangle Leaves Revealed by Integrated Perls Reaction, TEM, STEM-HAADF, and EDS Analyses.

Mangrove ecosystems are frequently exposed to high concentrations of iron (Fe) in sediments, resulting in Fe accumulation in plant tissues. Although Fe is an essential micronutrient involved in several metabolic processes, its excess requires efficient mechanisms of compartmentalization and storage to maintain cellular homeostasis. Histochemical detection using the Perls reaction has usually been applied to identify ferric iron (Fe3+) in biological tissues; however, the combination of this technique with ultrastructural and elemental analyses remains relatively unexplored in plant cells. Graft union formation in artichoke grafting onto wild and cultivated cardoon: an anatomical study.

In order to develop a non-chemical method such as grafting effective against well-known artichoke soil borne diseases, an anatomical study of union formation in artichoke grafted onto selected wild and cultivated cardoon rootstocks, both resistant to Verticillium wilt, was performed. The cardoon accessions Belgio (cultivated cardoon) and Sardo (wild cardoon) were selected as rootstocks for grafting combinations with the artichoke cv. Romolo. Grafting experiments were carried out in the autumn and spring. The anatomical investigation of grafting union formation was conducted by scanning electro

Sub-optimal temperature leads to tighter coupling between photosynthetic electron transport and CO2 assimilation under fluctuating light in maize.

The C4 carbon concentrating pathway promotes high CO2 assimilation rates. To keep C4 photosynthesis energetically efficient, electron transport reactions and downstream biochemistry need to be carefully balanced. Here we use a combination of noninvasive measurements and metabolic profiling to study the efficiency of C4 photosynthesis in maize (Zea mays) under 2 conditions that can lead to decoupling between electron transport and carbon assimilation: fluctuating light and suboptimal temperature. Measurements were performed under 3 fluctuating light regimes and at 3 temperatures, providing the

Molecular characterization and subcellular localization of Arabidopsis class VIII myosin, ATM1.

Land plants possess myosin classes VIII and XI. Although some information is available on the molecular properties of class XI myosins, class VIII myosins are not characterized. Here, we report the first analysis of the enzymatic properties of class VIII myosin. The motor domain of Arabidopsis class VIII myosin, ATM1 (ATM1-MD), and the motor domain plus one IQ motif (ATM1-1IQ) were expressed in a baculovirus system and characterized. ATM1-MD and ATM1-1IQ had low actin-activated Mg(2+)-ATPase activity (Vmax = 4 s(-1)), although their affinities for actin were high (Kactin = 4 μM). The actin-sli
