Differential regulation of genes transcribed by nucleus-encoded plastid RNA polymerase, and DNA amplification, within ribosome-deficient plastids in stable phenocopies of cereal albino mutants.

We isolated stable albino plants of barley and maize by inhibiting plastid protein synthesis with streptomycin and propagating bleached seedlings in the absence of antibiotics in vitro. Albino plants are deficient in plastid translation products and plastid ribosomal RNAs, and are stable phenocopies of the barley albostrians and maize iojap mutants, which contain ribosome-free plastids. Once plastid ribosomes are lost they cannot be re-synthesized because about one-third of plastid ribosomal proteins are themselves plastid encoded. The group II/subgroup IIA intron in plastid rpl2 transcripts w

Genetic engineering of dinoflagellate algae and the lethality of an introduced plastid terminal oxidase.

Dinoflagellate algae are a diverse group of single-celled eukaryotes, often living in marine environments. The majority of species are entirely free-living, but many can become symbionts with corals, jellyfish and other marine organisms. With rising sea temperatures, the function of the dinoflagellate photosynthetic machinery, and the redox state of the photosynthetic electron transport chain are impaired. This photosynthetic impairment is likely to be an important cause of coral bleaching. In the chloroplasts of plants and many algae, disturbance of the chloroplast redox state can be in part

Comparative plastid genomics of Gastrochilus (Orchidaceae) with insights into molecular markers and phylogeny.

BACKGROUND: Gastrochilus (Orchidaceae) is a genus of epiphytic orchids valued for horticultural and medicinal purposes. However, the structural and evolutionary features of its plastid genomes have not been systematically examined. Here, we characterize the complete plastid genomes of 23 Gastrochilus species, elucidating structural features and key variations to provide new insights into their evolutionary significance. RESULTS: The plastid genomes showed a typical quadripartite structure, ranging from 146,183 to 148,666 bp in length. Each genome encodes 120 genes, comprising 74 protein-codin

Plastid carbonic anhydrases are essential for growth of Arabidopsis on ambient air but not for photosynthesis.

The physiological role of chloroplast carbonic anhydrases (CAs) has long been debated, particularly in the context of photosynthesis. While early hypotheses proposed that CAs enhance CO2 assimilation by rapidly accessing the HCO3- pool, direct evidence has been lacking. In this study, we examined Arabidopsis (Arabidopsis thaliana) mutants lacking both chloroplast-localized βCA1 (AT3G01500) and βCA5 (AT4G33580) to assess their impact on plant growth and photosynthetic performance. Our results show that plants deficient in chloroplast CA activity are unable to grow under ambient CO2 conditions (

Monitoring of Lipid Fluxes Between Host and Plastid-Bearing Apicomplexan Parasites.

Apicomplexan parasites are unicellular eukaryotes responsible for major human diseases such as malaria and toxoplasmosis, which cause massive social and economic burden. Toxoplasmosis, caused by Toxoplasma gondii, is a global chronic infectious disease affecting ~1/3 of the world population and is a major threat for any immunocompromised patient. To date, there is no efficient vaccine against these parasites and existing treatments are threatened by rapid emergence of parasite resistance. Throughout their life cycle, Apicomplexa require large amount of nutrients, especially lipids for propagat

Birth of Four Chimeric Plastid Gene Clusters in Japanese Umbrella Pine.

Many genes in the plastid genomes (plastomes) of plants are organized as gene clusters, in which genes are co-transcribed, resembling bacterial operons. These plastid operons are highly conserved, even among conifers, whose plastomes are highly rearranged relative to other seed plants. We have determined the complete plastome sequence of Sciadopitys verticillata (Japanese umbrella pine), the sole member of Sciadopityaceae. The Sciadopitys plastome is characterized by extensive inversions, pseudogenization of four tRNA genes after tandem duplications, and a unique pair of 370-bp inverted repeat

Natural Etiolation in Spearmint (Mentha spicata L. var. crispa) Shoots Emerging From Rhizomes-Plastids, Glandular Hairs, and Phytochemical Profile.

Spearmint (Mentha spicata L. var. crispa) is a widely cultivated aromatic herb valued both as a culinary ingredient and as a source of bioactive compounds, such as essential oils and phenolics, which contribute to its traditional uses and potential medicinal applications. In this work, we provided the first evidence of natural etiolation with etioplast and prolamellar body formation in leaves and stems of spearmint shoots emerging in the soil from underground rhizomes. Soil-grown and hydroponically dark-forced shoots exhibited comparable etiolation symptoms, plastid ultrastructure, and chlorop

Plastid Phylogenomics of Elatostemateae (Urticaceae): Insights Into Structural Variation and Phylogenetic Relationships.

The tribe Elatostemateae, with approximately 1300 species, is the most species-rich group within Urticaceae, yet plastome evolution and several intergeneric relationships remain incompletely resolved. We analyzed 40 species representing the major extant lineages of Elatostemateae using complete plastomes and nuclear ribosomal DNA (nrDNA); Metapilea was excluded because suitable DNA material was unavailable. Plastomes retained the typical angiosperm quadripartite structure and ranged from 143,738 to 163,969 bp. Comparative analyses revealed large inversions in the large single-copy (LSC) region

Transcriptomic and enzymological evidence for plastid peptidoglycan synthesis in the gymnosperm Picea abies.

It is understood that a cyanobacterium was the progenitor of plastids and that the biosynthesis of cell wall peptidoglycan was lost during chloroplast evolution. However, accumulated data, especially from the moss Physcomitrium patens, suggest that peptidoglycan remains essential for plastid division in some land plants. A fundamental set of peptidoglycan biosynthesis (Mur) genes has been identified in the genomes of these land plants, while many angiosperms no longer encode some core Mur genes, including a bifunctional penicillin-binding protein (PBP). Ten incomplete Mur genes were previously

Dynamic changes in the plastid and mitochondrial genomes of the angiosperm Corydalis pauciovulata (Papaveraceae).

BACKGROUND: Corydalis DC., the largest genus in the family Papaveraceae, comprises > 465 species. Complete plastid genomes (plastomes) of Corydalis show evolutionary changes, including syntenic arrangements, gene losses and duplications, and IR boundary shifts. However, little is known about the evolution of the mitochondrial genome (mitogenome) in Corydalis. Both the organelle genomes and transcriptomes are needed to better understand the relationships between the patterns of evolution in mitochondrial and plastid genomes. RESULTS: We obtained complete plastid and mitochondrial genomes from

Understanding Plastid Vesicle Transport - Could it Provide Benefit for Human Medicine?

BACKGROUND: In plants, vesicle transport occurs in the secretory pathway in the cytosol, between the membranes of different compartments. Several protein components have been identified to be involved in the process and their functions were characterized. Both cargos and other molecules (such as hormones) have been shown to use vesicle transport, although the major constituents of vesicles are lipids which are transferred from donor to acceptor membranes. In humans, malfunction of the cytosolic vesicle transport system leads to different diseases. METHOD: To better understand and ultimately c

Was the Chlamydial Adaptative Strategy to Tryptophan Starvation an Early Determinant of Plastid Endosymbiosis?

Chlamydiales were recently proposed to have sheltered the future cyanobacterial ancestor of plastids in a common inclusion. The intracellular pathogens are thought to have donated those critical transporters that triggered the efflux of photosynthetic carbon and the consequent onset of symbiosis. Chlamydiales are also suspected to have encoded glycogen metabolism TTS (Type Three Secretion) effectors responsible for photosynthetic carbon assimilation in the eukaryotic cytosol. We now review the reasons underlying other chlamydial lateral gene transfers evidenced in the descendants of plastid en

Factors mediating plastid dependency and the origins of parasitism in apicomplexans and their close relatives.

Apicomplexans are a major lineage of parasites, including causative agents of malaria and toxoplasmosis. How such highly adapted parasites evolved from free-living ancestors is poorly understood, particularly because they contain nonphotosynthetic plastids with which they have a complex metabolic dependency. Here, we examine the origin of apicomplexan parasitism by resolving the evolutionary distribution of several key characteristics in their closest free-living relatives, photosynthetic chromerids and predatory colpodellids. Using environmental sequence data, we describe the diversity of the

Identification of Chemicals That Abrogate Folate-Dependent Inhibition of Starch Accumulation in Non-Photosynthetic Plastids of Arabidopsis.

Folate, also known as vitamin B9, is an essential cofactor for a variety of enzymes and plays a crucial role in many biological processes. We previously reported that plastidial folate prevents starch biosynthesis triggered by the influx of sugar into non-starch-accumulating plastids, such as etioplasts, and chloroplasts under darkness; hence the loss of plastidial folate induces the accumulation of starch in plastids. To understand the molecular mechanism underlying this phenomenon, we screened our in-house chemical library and searched their derivatives to identify chemicals capable of induc

RNA-Guided Engineering of the Chloroplast Genome Enabled by Plastid-Expressed Guide RNAs.

Our goal is to develop RNA-guided engineering of the chloroplast genome using the CRISPR/Cas9 system. We designed chloroplast minigenes to obtain properly sized single guide RNAs (sgRNAs) in tobacco chloroplasts. The sgRNA 5' end is defined by transcription from an rRNA operon promoter, and its 3' end by processing a downstream tRNA (trnG) or a hepatitis delta virus (HDV) ribozyme. Cas9 is expressed from a nuclear gene and is targeted to chloroplasts by fusion to a transit peptide. Cas9 incorporated the sgRNA and introduced double-strand breaks in the plastid DNA (ptDNA). We report here that t

Before import: N-terminal acetylation at the interface of plastid protein targeting and proteostasis.

Most chloroplast proteins are synthesized in the cytosol as precursor proteins carrying N-terminal transit peptides that mediate post-translational import into the organelle. During this transient cytosolic phase, precursor proteins are exposed to the cellular N-terminal processing machinery, including methionine aminopeptidases and N-terminal acetyltransferases (NATs). Recent proteomic studies have provided direct evidence that plastid precursor proteins undergo N-terminal acetylation prior to import, establishing a previously unrecognized link between co-translational processing and plastid

Comparative plastid genomics reveals phylogenetic relationships and candidate molecular markers in Salvia.

INTRODUCTION: Salvia is the largest genus in Lamiaceae and includes numerous medicinal, ornamental, and aromatic species. Species identification and rational resource utilization in this genus remain difficult because morphological characters are variable, infrageneric boundaries are sometimes ambiguous, and broad comparative genomic resources are still limited. In addition, plastid RNA editing has rarely been characterized in Salvia. METHODS: We assembled six new plastid genomes, compared complete plastid genomes representing major lineages of Salvia, and characterized plastid RNA editing in

Rampant hybridization underlies cytonuclear discordance and shallow plastid divergence in Juniperus (Cupressaceae).

Reconstructing plant evolutionary history is complicated by processes such as hybridization, incomplete lineage sorting and the interplay of ecological adaptation with morphological innovation. These generate reticulate patterns that challenge tree-like models and can bias inferences from single genomic compartments. These issues are pronounced in long-lived woody lineages. To examine these dynamics, we analysed Juniperus, a widely distributed Northern Hemisphere gymnosperm genus, using near-complete taxon sampling, complete plastomes and >3000 nuclear single-copy genes recovered via target-en

Changes in plastid biogenesis leading to the formation of albino regenerants in barley microspore culture.

BACKGROUND: Microspore embryogenesis is potentially the most effective method of obtaining doubled haploids (DH) which are utilized in breeding programs to accelerate production of new cultivars. However, the regeneration of albino plants significantly limits the exploitation of androgenesis for DH production in cereals. Despite many efforts, the precise mechanisms leading to development of albino regenerants have not yet been elucidated. The objective of this study was to reveal the genotype-dependent molecular differences in chloroplast differentiation that lead to the formation of green and
