The Crd1 gene encodes a putative di-iron enzyme required for photosystem I accumulation in copper deficiency and hypoxia in Chlamydomonas reinhardtii.

Chlamydomonas reinhardtii adapts to copper deficiency by degrading apoplastocyanin and inducing Cyc6 and Cpx1 encoding cytochrome c(6) and coproporphyrinogen oxidase, respectively. To identify other components in this pathway, colonies resulting from insertional mutagenesis were screened for copper- conditional phenotypes. Twelve crd (copper response defect) strains were identified. In copper-deficient conditions, the crd strains fail to accumulate photosystem I and light-harvesting complex I, and they contain reduced amounts of light-harvesting complex II. Cyc6, Cpx1 expression and plastocyan

The copper economy response is partially conserved in rice (Oryza sativa L.).

Copper (Cu) is an essential element for plants, especially in photosynthesis, as it is required for plastocyanin function in electron transfer reactions at thylakoid membranes. In Arabidopsis thaliana, Cu deficiency leads to the Cu economy response, in which plants prioritize Cu usage by plastocyanin in detriment of non-essential cupric proteins. In rice (Oryza sativa), however, this response has not been characterized. Rice OsHMA5 is a Cu xylem-loading transporter involved in Cu translocation from roots to shoots, as suggested by the analysis of oshma5 mutant plants. Aiming to understand how

Genetic dissection of nutritional copper signaling in chlamydomonas distinguishes regulatory and target genes.

A genetic screen for Chlamydomonas reinhardtii mutants with copper-dependent growth or nonphotosynthetic phenotypes revealed three loci, COPPER RESPONSE REGULATOR 1 (CRR1), COPPER RESPONSE DEFECT 1 (CRD1), and COPPER RESPONSE DEFECT 2 (CRD2), distinguished as regulatory or target genes on the basis of phenotype. CRR1 was shown previously to be required for transcriptional activation of target genes like CYC6, CPX1, and CRD1, encoding, respectively, cytochrome c(6) (which is a heme-containing substitute for copper-containing plastocyanin), coproporphyrinogen III oxidase, and Mg-protoporphyrin I

Compensatory respiration supports survival during extended heat stress in Microcystis aeruginosa.

Phototrophic organisms must balance energy between photosynthesis and respiration to survive. In cyanobacteria, this coordination is especially critical as both processes share the same membrane and electron transport complexes. Although electron exchange between the two pathways is known, photosynthesis is often presumed to dominate under many conditions. Here, under an extended heat-stress condition, our data indicate that respiration can compensate for reduced photosynthetic activity. We compared two Microcystis aeruginosa strains (PCC7806 and Lake Kinneret isolated strain C-1004). PCC7806

Copper Delivery to Chloroplast Proteins and its Regulation.

Copper is required for photosynthesis in chloroplasts of plants because it is a cofactor of plastocyanin, an essential electron carrier in the thylakoid lumen. Other chloroplast copper proteins are copper/zinc superoxide dismutase and polyphenol oxidase, but these proteins seem to be dispensable under conditions of low copper supply when transcripts for these proteins undergo microRNA-mediated down regulation. Two ATP-driven copper transporters function in tandem to deliver copper to chloroplast compartments. This review seeks to summarize the mechanisms of copper delivery to chloroplast prote

Conserved structural features on protein surfaces: small exterior hydrophobic clusters.

The extent to which side-chains at the surface of globular proteins adopt well-defined conformations is a matter of some controversy and, in turn, the idea that specific interactions amongst them might make a significant contribution to defining tertiary structures would be generally seen as questionable. In at least some cases, however, there is evidence for organisation of the surface to form discrete, tightly packed clusters. In this paper we examine the role of such clusters in accommodating large, hydrophobic residues on the exterior of protein structures. Taking poplar plastocyanin as a

Large-scale purification of active cytochrome b6/f complex from spinach chloroplasts.

A preparation is described through which large quantities of pure, active cytochrome b6/f complex can be isolated from spinach chloroplasts. The resulting complex is at least 90% pure with respect to the maximum content of redox centers, consists of four polypeptides according to polyacrylamide gel electrophoresis, and lacks both ferredoxin: NADP+ oxidoreductase and the high molecular weight form of cytochrome f seen in some other preparations. The complex contains 2 mol b6 and 2 atoms of nonheme iron per mole of cytochrome f, and possesses a high plastoquinol-plastocyanin oxidoreductase activ

Further evidence for the electron pool hypothesis. The effect of KCN and DSPD on the photophobic reaction in the filamentous blue-green alga Phormidium uncinatum.

In continuation of experiments with photo-system II inhibitors [3-(3,4-dichlorophenyl)-1,1-dimethylurea and 2,5-dibromo-3-methyl-6-isopropylbenzo-quinone] the effect of photosystem I inhibitors was studied. 1. Neither the plastocyanin inhibitor, potassium cyanide, nor the ferredoxin antagonist, disalicyliden propandiamin, markedly affected those phobic reactions which are mediated by the electron transport via photosystem II into the electron pool. 2. On the other hand those phobic reactions, which are triggered by an increased flow of electrons out of the pool, are specifically inhibited by b

The 520 nm absorbance changes in Scenedesmus obliquus and its relation to photosystem I.

The kinetics (region of seconds) of the light-induced 520 nm absorbance changes and its dark reversal have been studied in detail in the wild type and in some pigment and photosynthetic mutants of Scenedesmus obliquus. The following 5 lines of evidence led us to conclude that the signal is entirely due to the photosystem I reaction modified by electron flow from Photosystem II. Gradual blocking of the electron transport with 3(3,4-dichlorophenyl)-1,1-dimethylurea resulted in diminution and ultimate elimination of the biphasic nature of the signal without reducing the extent of the absorbance c

The PSI-E subunit of photosystem I binds ferredoxin:NADP+ oxidoreductase.

A photosystem I complex containing the polypeptides PSI-A to PSI-L, light-harvesting complex I and ferredoxin:NADP+ oxidoreductase has been isolated from barley using the non-ionic detergent n-decyl-beta-D-maltopyranoside. The ratio between bound ferredoxin:NADP+ oxidoreductase and P700 is 0.4 +/- 0.2. The complex is highly active in catalyzing light-induced transfer of electrons from plastocyanin to NADP+ at rates of 280 +/- 150 and 1800 +/- 800 mumol NADPH/(mg chl.h), without and in the presence of saturating amounts of exogenously added ferredoxin:NADP+ oxidoreductase, respectively. SQUAMOSA Promoter Binding Protein-Like7 Is a Central Regulator for Copper Homeostasis in Arabidopsis.

Expression of miR398 is induced in response to copper deficiency and is involved in the degradation of mRNAs encoding copper/zinc superoxide dismutase in Arabidopsis thaliana. We found that SPL7 (for SQUAMOSA promoter binding protein-like7) is essential for this response of miR398. SPL7 is homologous to Copper response regulator1, the transcription factor that is required for switching between plastocyanin and cytochrome c(6) in response to copper deficiency in Chlamydomonas reinhardtii. SPL7 bound directly to GTAC motifs in the miR398 promoter in vitro, and these motifs were essential and suf

Unexpected changes in photosystem I function in a cytochrome c6-deficient mutant of the cyanobacterium Synechocystis PCC 6803.

Cytochrome c6, the product of the petJ gene, is a photosynthetic electron carrier in cyanobacteria, which transfers electrons to photosystem I and which is synthesised under conditions of copper deficiency to functionally replace plastocyanin. The photosystem I photochemical activity (energy storage, photoinduced P700 redox changes) was examined in a petJ-null mutant of Synechocystis PCC 6803. Surprisingly, photosystem I activity in the petJ-null mutant grown in the absence of copper was not much affected. However, in a medium with a low inorganic carbon concentration and with NH4+ ion as nitr

Manganese deficiency alters photosynthetic electron transport in Marchantia polymorpha.

Manganese (Mn) is considered as an essential element for plant growth. Mn starvation has been shown to affect photosystem II, the site of the Mn4CaO5 cluster responsible for water oxidation. Less is known on the effect of Mn starvation on photosystem I. Here we studied the effects of Mn deficiency in vivo on redox changes of P700 and plastocyanin (Pc) in the liverwort Marchantia polymorpha using the KLAS-NIR spectrophotometer. Far-red illumination is used to excite preferentially photosystem I, thus facilitating cyclic electron transport. Under Mn starvation, we observed slower oxidation of P7

How do plants respond to copper deficiency?

The transition metal copper is essential for all organisms yet excess copper is toxic because of production of free radicals via its free form. Therefore, the levels of copper are precisely regulated in a cell. Under copper depleted conditions, the expression of Cu/Zn superoxide dismutase (SOD) is downregulated and its function is compensated by Fe SOD in chloroplasts of higher plants. We presented evidence that a microRNA, miR398, is involved in this downregulation of Cu/Zn SOD genes in Arabidopsis thaliana when grown at low copper levels, corresponding to less than 1 microM Cu in tissue cult

Modelling the impact of geometric parameters on the redox potential of blue copper proteins.

The synthesis and structure of a homologous series of cationic N(2)S(2) copper(I) Schiff base complexes constructed using o-tert-butylthiobenzaldehyde and a series of terminal diamines (ethane, propane, butane) are reported. The complexes differ only in the length of the methylene chain between the imine groups. This simple modification forces the copper centre to shift geometry from a planar (1,2-diaminoethane) to a more distorted tetrahedral motif (1,4-diaminobutane). The redox potentials of the three cations were measured using cyclic voltammetry in donor (acetonitrile) and non-donor solven

Flash photolysis-electron spin resonance studies of photosystem I. A fast reduction of component of P-700+.

A 300 mus decay component of ESR Signal I (P-700+) in chloroplasts is observed following a 10 mus actinic xenon flash. This transient is inhibited by treatments which block electron transfer from Photosystem II to Photosystem I (e.g. 3-(3,4-dichlorophenyl)-1,1-dimethylurea (DCMU), 2,5-dibromo-3-methyl-6-isopropyl-p-benzoquinone (DBMIB), KCN and HgCl2). The fast transient reduction of P-700+ can be restored in the case of DCMU or DBMIB inhibition by addition of an electron donor couple (2,6-dichlorophenol indophenol (Cl2Ind)/ascorbate) which supplies electrons to cytochrome f. However, this don

Deficiency and toxicity of nanomolar copper in low irradiance-A physiological and metalloproteomic study in the aquatic plant Ceratophyllum demersum.

Essential trace elements (Cu(2+), Zn(2+), etc) lead to toxic effects above a certain threshold, which is a major environmental problem in many areas of the world. Here, environmentally relevant sub-micromolar concentrations of Cu(2+) and simulations of natural light and temperature cycles were applied to the aquatic macrophyte Ceratophyllum demersum a s a model for plant shoots. In this low irradiance study resembling non-summer conditions, growth was optimal in the range 7.5-35nM Cu, while PSII activity (Fv/Fm) was maximal around 7.5nM Cu. Damage to the light harvesting complex of photosystem

Molecular underpinnings and biogeochemical consequences of enhanced diatom growth in a warming Southern Ocean.

The Southern Ocean (SO) harbors some of the most intense phytoplankton blooms on Earth. Changes in temperature and iron availability are expected to alter the intensity of SO phytoplankton blooms, but little is known about how these changes will influence community composition and downstream biogeochemical processes. We performed light-saturated experimental manipulations on surface ocean microbial communities from McMurdo Sound in the Ross Sea to examine the effects of increased iron availability (+2 nM) and warming (+3 and +6 °C) on nutrient uptake, as well as the growth and transcriptional

Novel pyridinium salt derivatives as potential inhibitors for photosystem I.

BACKGROUND: Photosystem I (PS I) is a plastocyanin-ferredoxin oxidoreductase within the photosynthetic electron transport chain and represents a key target for herbicide development. RESULTS: Pyridinium salt compounds A and B were designed and synthesized with active substructural insertion and combination strategies. Bioassays revealed that most compounds exhibited > 70% inhibition of growth in Portulaca oleracea and Echinochloa crusgalli when applied to germinated seeds at a concentration of 10 μg mL-1. Compound B8 demonstrated 90-100% postemergence herbicidal activity on E. crusgalli, Eleu
