Insights Into the Role of Platelet Factor 4 in Neurodegenerative Diseases.

BACKGROUND: As population aging accelerates worldwide, the prevalence and burden of neurodegenerative diseases (NDs) continue to rise, underscoring the urgent need to identify key molecular mechanisms driving disease progression and novel therapeutic targets. Chronic neuroinflammation is a hallmark of multiple NDs, including Alzheimer's disease and Parkinson's disease. Platelet factor 4 (PF4) is a chemokine primarily released by activated platelets and has traditionally been investigated for its roles in hemostasis, thrombosis, and peripheral inflammation. RESULTS: Growing evidence has implic

Cellular contributions to the pathogenesis of anti-platelet factor 4 disorders.

PURPOSE OF REVIEW: Anti-platelet factor 4 (PF4) disorders, including heparin-induced thrombocytopenia (HIT) and vaccine-induced immune thrombocytopenia and thrombosis (VITT), and emerging disorders such as VITT-like monoclonal gammopathy of thrombotic significance (MGTS), are monoclonal antibody-mediated and characterized by thrombocytopenia and thrombosis. Understanding the cellular and molecular mechanisms among these anti-PF4 disorders can help explain the variability in clinical presentations. RECENT FINDINGS: Recent work demonstrated that beyond platelets, immune and vascular cells serve

Anti-platelet Factor 4 Antibody-Mediated Disorders: An Updated Narrative Review.

Anti-platelet factor 4 (PF4) antibody-mediated disorders are a heterogeneous group of diseases characterized by the presence of highly pathogenic immunoglobulins G directed against PF4 and/or PF4/heparin complexes. These antibodies are able to activate platelets, neutrophils, and monocytes, thus resulting in thrombocytopenia and a hypercoagulable state. Five different forms of anti-PF4 antibody-mediated disorders have been identified: (1) classic heparin-induced thrombocytopenia (HIT) mediated by heparin and certain polyanionic drugs; (2) autoimmune HIT characterized by the presence of anti-PF

Interactions of marine sulfated glycans with antithrombin and platelet factor 4.

The molecular interactions of sulfated glycans, such as heparin, with antithrombin (AT) and platelet factor 4 (PF4) are essential for certain biological events such as anticoagulation and heparin induced thrombocytopenia (HIT). In this study, a library including 84 sulfated glycans (polymers and oligomers) extracted from marine algae along with several animal-originated polysaccharides were subjected to a structure-activity relationship (SAR) study regarding their specific molecular interactions with AT and PF4 using surface plasmon resonance. In this SAR study, multiple characteristics were c

[Thromboxane, prostacyclin and platelet factor 4 in patients with atrial fibrillation].

Thromboxane++ (TX), prostacyclin (PC) and platelet factor IV (PF) were assayed using radioimmune kits in 46 patients with cardiac fibrillation secondary to coronary heart disease and rheumatic fever. The study established elevated TX beta 2 levels, low concentrations of 6-ketoPGF1 alpha, enhanced activity of PF. Restoration of the sinus rhythm by electroimpulse treatment (EIT) brought about augmentation of the Tx-Pc-PF unbalance on day 2-3. This may give rise to "normalization" thromboembolism. On days 5-7 after EIT the intensity of platelet aggregation and PF secretion diminish. It is conclud

Anti-Platelet factor 4 immunothrombosis-not just heparin and vaccine triggers.

Derailments at the tightly regulated interface of blood coagulation and innate inflammatory immune responses can lead to pathologic immunothrombosis. A special subset of immunothrombosis is caused by antibodies against platelet-factor 4 (PF4). Anti-PF4 antibodies triggered by heparin treatment in heparin-induced thrombocytopenia (HIT) are known for more than 50 years. Interest in anti-PF4 disorders rekindled when first cases of vaccine-induced immune thrombocytopenia and thrombosis (VITT) occurred during the worldwide COVID-19 vaccination campaign. During this time new diagnostic procedures we

Platelet factor 4 regulation of monocyte KLF4 in experimental cerebral malaria.

Cerebral malaria continues to be a difficult to treat complication of Plasmodium falciparum infection in children. We have shown that platelets can have major deleterious immune functions in experimental cerebral malaria (ECM). One of the platelet derived mediators we have identified as particularly important is platelet factor 4/CXCL4. Our prior work demonstrated that PF4(-/-) mice are protected from ECM, have reduced plasma cytokines, and have reduced T-cell trafficking to the brain. We now show that PF4 drives monocyte cytokine production in a Kruppel like factor 4 (KLF4) dependent manner.

Recombinant platelet factor 4, an angiogenic marker for human breast carcinoma.

The present study was designed to test the hypothesis that rhPF4 binds with high specificity to the neovasculature of breast cancer carcinoma. To achieve this goal, we used intravital microscopy to study the binding characteristics of systemically injected fluorescently labeled rhPF4 (FITC-rhPF4) to the microvasculature of dorsal skinfold chambers in nude mice implanted with tumor spheroids prepared from the human breast cancer cell line MCF-7. Our results show that intravenously as well as intra-arterially injected FITC-rhPF4 exclusively labeled, with high intensity and specificity, the endot

Antiphospholipid syndrome (APS) is a platelet factor 4 (PF4)-centric immunothrombotic disorder.

Antiphospholipid syndrome (APS) is an immunothrombotic disorder, frequently attributed to autoantibodies that bind β2-glycoprotein I (β2GPI). A study showed that the platelet-specific chemokine, platelet factor 4 (PF4), binds to β2GPI, enhancing recognition of β2GPI by APS antibodies. APS antibodies induce the release of neutrophil extracellular traps (NETs), webs of decondensed chromatin that bind both PF4 and b2GPI. We propose that PF4 bridges β2GPI to NETs (and other PF4-targeted polyanions), leading to the formation of prothrombotic PF4:b2GPI:NET immunotargets in APS. Dynamic light-scatter

Anti-Platelet Factor 4 Antibody Clonal Heterogeneity and MGUS Status in HIT.

BACKGROUND: Monoclonal gammopathy of thrombotic significance (MGTS) is a recently described chronic prothrombotic condition characterized by monoclonal anti-PF4 antibodies that are detected above the polyclonal antibody background in patient sera (i.e. present as monoclonal gammopathy of undetermined significance, MGUS). Due to conflicting data in the published literature on antibody clonality in heparin-induced thrombocytopenia (HIT), we evaluated clonality and abundance of anti-PF4 antibodies in HIT, including investigating whether an MGUS, if present in HIT, represents the causative anti-PF

Novel adenovirus vaccine vectors lacking thrombosis-associated interactions with platelet factor 4.

The adenoviral vector-based AstraZeneca and Janssen COVID-19 vaccines have been associated with rare cases of thrombosis, believed to be triggered, among other factors, by vector binding to the blood protein platelet factor 4 (PF4). To identify vectors with lower thrombosis risk, we screened 50 natural and hexon-modified adenoviruses (Ads). Unlike the applied COVID-19 vaccines and most tested vectors, Ad34 and Ad80, as well as Ad5 vectors with deleted or chemically shielded hexon hyper-variable region 1 (HVR1), did not bind to PF4. Furthermore, interactions with PF4 substantially modified Ad5

Platelet factor 4 regulates T cell effector functions in malignant pleural effusions.

Malignant pleural effusion (MPE) is defined as the presence of tumor cells in pleural fluid and it is a fatal complication of advanced lung adenocarcinoma (LAC). To understand the immune response to the tumor in MPE, we compared the concentration of immunomodulatory factors in MPE of LAC and pleural effusion of heart failure (HF) patients by ELISA, and the proliferation and cytotoxic phenotype of T cells stimulated in the presence of LAC and HF pleural fluids by cytometry. Increased platelet factor 4 and aberrant permeability of follicular fluid in PCOS.

BACKGROUND/PURPOSE: Abnormal folliculogenesis is one of the cardinal presentations of polycystic ovarian syndrome (PCOS) and permeability of follicular wall has been proposed to be involved in the normal follicular growth. However, whether or not there is a change in intrafollicular permeability underlies PCOS is unknown. METHODS: This was a tertiary center-based case-control study. From 2014 to 2015, thirteen patients with PCOS who underwent in vitro fertilization-embryo transfer (IVF-ET) were enrolled. Eleven normo-ovulatory patients who underwent IVF-ET due to male factor and/or tubal fact

Anti-platelet Factor 4/Heparin Antibody Production in a Mouse Immunization Model.

Heparin-induced thrombocytopenia (HIT) is the most prevalent form of drug-induced, immune-mediated thrombocytopenia. It involves the production of antibodies that target platelet factor 4 (PF4) in complex with heparin, leading to platelet activation. The pathogenesis of HIT is not fully understood, primarily due to the limitations of studying human patients. To bridge this gap and gain a deeper understanding of the mechanisms behind HIT antibody generation, researchers developed a murine model using immunization with mouse PF4 (mPF4)/heparin complexes. Here, we provide a detailed methodology f

Platelet factor 4 protects kidney allograft in a rat kidney transplantation model.

Platelets are the cellular mediator of thrombosis, but it is becoming increasingly evident that platelets actively participate in inflammation and immune responses. A recent paper indicated that platelet factor 4 (PF4) alleviated cardiac allograft rejection in mice. But the role of PF4 on kidney transplantation has never been investigated. In our current experiment, PF4 administration alleviates immune responses to kidney transplantation. PF4 significantly alleviates vascular and glomerular changes, as well as interstitial inflammation, fibrosis, and tubular atrophy at day 56 after transplanta

Platelet factor 4 localization in carotid atherosclerotic plaques: correlation with clinical parameters.

Emerging evidence supports a role for platelets in the progression of atherosclerosis in addition to an involvement in thrombotic vascular occlusion. Platelet Factor 4 (PF4), a chemokine released by activated platelets, stimulates several pro-atherogenic processes. Therefore, we examined the localization of PF4 and the homologous protein, Neutrophil Activating Protein-2 (NAP-2) in lesions representing the evolution of human atherosclerotic plaques. Carotid plaques from 132 patients with critical carotid stenosis and 6 autopsy specimens were studied. Clinical, histologic and immunohistochemical

Alcohol-induced protein folding transitions in platelet factor 4: the O-state.

Platelet factor 4 (PF4) (7800 daltons) is an anti-parallel beta-sheet, alpha/beta class protein whose tertiary structure is stabilized by the presence of two disulfide bonds. Titration of PF4 with 2-propanol or similar low molecular weight, aliphatic alcohols induces reversible protein folding transitions which are observed to be in slow exchange on the 600-MHz 1H NMR time scale. Line fitting of resolved resonances assigned to ring protons of Y60, H35, H23, and alpha H of K50 in native and alcohol-induced states (O-states) allows derivation of folding equilibrium constants and exchange kinetic

Saline whole-blood impedance aggregometry for detection of heparin-independent platelet-activating anti-platelet factor 4 antibodies.

OBJECTIVES: This study evaluates the practice of incorporating a saline testing condition into the heparin-induced thrombocytopenia (HIT) functional assay using whole-blood impedance aggregometry (WBIA) to identify a subgroup of highly pathogenic anti-platelet factor 4 (PF4) antibodies with heparin-independent platelet-activating properties. METHODS: From 2023 to 2025, we analyzed 150 evaluable patients (65 WBIA Negative, 85 WBIA Positive). Positive cases were subdivided into Classic-Positive (low-dose heparin positive only; n = 46) and Saline-Positive (positive under both low-dose heparin an
