ABSTRACT: The control of small ruminant lentivirus (SRLV) infections - Maedi, Visna, and caprine arthritis and encephalitis (CAE) - is based on the establishment of SRLV-free closed herds, achieved by serologically testing of all small ruminants aged one year and older for antibodies against SRLV. After four completely negative serological test results within a period of at least 24 months, a farm is recognized as SRLV-free. Maintaining SRLV-free status is based on regular serological testing of all small ruminants in the herd over one year of age. To reduce the administrative and financial bu

This review describes the pathogenesis of a slowly progressive disease complex caused by naturally occurring nononcogenic retroviruses in sheep and goats. In nature, infections are usually clinically silent, but disease may manifest itself after prolonged incubation periods. Clinically, this is seen as dyspnea, progressive paralysis, and/or progressive arthritis. In all organs the basic lesion is inflammatory with infiltration and proliferation of lymphocytes, plasma cells, and macrophages. Other organ-specific pathologic changes such as primary demyelination in the central nervous system and

Fetuses of 20 pregnant ewes at 4 gestational periods (45, 55, 85, and 100 days) were inoculated with ovine progressive pneumonia virus. Fourteen of 16 fetuses exposed to virus before gestational day 80 were either resorbed or expelled, whereas 10 of 15 fetuses exposed to virus after day 80 were normal at birth. Three of the 9 expelled fetuses and 1 of 2 newborn lambs had accumulations of lymphoid cells in the lungs. Virus was readily isolated from the tissues of expelled fetuses and newborn lambs. Lambs did not have precipitating antibody to the virus at birth, but 3 to 5 lambs had specific an

There is only limited information on sheep interferon available. Recent publications have reported on: 1. an interferon (IFN) alpha subtype, which is secreted by the fetal trophectoderm into the lumen of the uterus between the 10th and 21st day of gestation. It was therefore named ovine trophoblast protein (oTP-1), and is responsible for signalling pregnancy to the ewe via high affinity receptors in the endometrium. It is thought that oTP-1 acts by directly influencing prostaglandin metabolism. 2. the role of lentivirus-induced interferon (LV-IFN) in the pathogenesis of Maedi/Visna. The result

In Europe, Maedi-Visna disease has high prevalence rates at the individual and flock levels, respectively, and is regarded as one of the most significant infectious disease in sheep. The lack of treatment or a commercial vaccine underscores the need for accurate and reliable diagnostic tools to support control programs. Conventional methods, including ELISA and qPCR, provide useful but incomplete information due to the genetic variability of small ruminant lentiviruses (SRLVs) and the heterogeneous host immune response. In this work, third-generation sequencing was assessed as a diagnostic str

BACKGROUND: Lentivirus ovivismae and Lentivirus capartenc (formerly maedi-visna and caprine arthritis-encephalitis virus) are highly contagious, chronic viral infections that cause significant economic losses in small ruminant farming. This study aimed to determine the current seroprevalence of these infections in the Yozgat region, where sheep and goat farming is widespread and clinical signs are frequently observed.
MATERIALS AND METHODS: Blood samples were collected from 160 sheep and 160 goats in districts with high small ruminant populations, including Sorgun, Sarıkaya, Saraykent, Boğazl

The biological properties of the ruminant animal lentiviruses, visna and caprine arthritis-encephalitis viruses, closely resemble those of their human counterparts, the human immunodeficiency viruses (HIV). All of these viruses are morphologically identical and are disseminated from host to host in nature during exchange of body fluids. Artificial conditions that favor excess exchange of such fluids precipitate epidemics by these viruses. The strategy of replication of the animal viruses in tissue culture and in vivo are very similar to that of the human virus. Virus replication is highly prod

Leucomyelitis was the predominant feature in four North American adult sheep (cases 1-4) with ovine lentivirus (OvLV) infection. All four animals were OvLV-seropositive and a syncytogenic virus consistent with OvLV was isolated from the brain of case 3 and the lungs of case 4. Clinically, the sheep had dyspnoea and neurologic signs of varying severity. Changes in the central nervous system included asymmetrical meningoleucomyelitis with white matter degeneration in all four sheep and scattered foci of leucoencephalitis in periventricular, subependymal and other white matter areas of the brain

Experiments were performed to determine whether visna/maedi virus (VMV), a small ruminant lentivirus (SRLV), could infect sheep via ocular tissues. The EV1 strain of VMV was administered into the conjunctival space of uninfected sheep, and the animals monitored for the presence of provirus DNA and anti-VMV antibodies in blood. The results showed that provirus DNA appeared in peripheral blood mononuclear cells of all animals within a few weeks of receiving either 10(6) TCID50 or 10(3) TCID50 of VMV. Of the animals receiving the higher dose of virus via the conjunctival space, two seroconverted

Maedi Visna Virus (MVV) is the etiological agent of a systemic disease of sheep, which causes lesions in lungs, the central nervous system, joints, and mammary glands. It has been speculated that the association with Brucella ovis may lead to the venereal shedding of the virus. In this work, samples of epididymis from ten rams positive for MVV and infected experimentally with Brucella ovis, were subjected to liquid-phase PCR, immunohistochemistry (IHC) and in situ PCR tests, aimed at identifying the pathogens in a tissue context. IHC was carried out using a monoclonal antibody raised against p

Several animal and human demyelinating diseases of the central nervous system (CNS) are associated with RNA or DNA viruses. These viruses infect CNS cells lytically or persistently. They mainly belong to the group of envelope viruses which derive their envelope partly from the host cell membrane. The process of virus release may result in the appearance of new antigens of virus-infected cells or the incorporation of cell membrane material into the viral envelope. These changes may lead to an immune response which selectively injures the CNS. These alterations of host cell membranes and host ce

While the incidence of the human immunodeficiency virus (HIV) infection has leveled off somewhat in homosexual men, infection in women, children and adolescents is rising. Unless effective preventive measures are introduced, the number of pediatric patients with HIV and related illnesses will continue to increase. Animal models play a key role in the understanding of the pathogenesis and in the establishment of therapeutic approaches of infectious diseases. Ovine lentivirus (OvLV) comprises a subgenus of the lentivirus genus in the family Retroviridae, that shares genotypic, phenotypic and pat

BACKGROUND: Respiratory diseases significantly affect small ruminants, leading to considerable illness and mortality. These conditions are often multifactorial, resulting from interactions among pathogens, host immunity and environmental stressors.
OBJECTIVE: This study aimed to detect antibodies against major respiratory viruses, bluetongue virus (BTV), Maedi-Visna virus (MVV), parainfluenza virus type 3 (PI-3) and Peste des petits ruminants virus (PPRV) in sheep from the North Shewa Zone, Ethiopia.
METHODS: A cross-sectional serological survey was conducted between September and November 2

Maedi-visna is a systemic disease of sheep caused by a lentivirus, maedi-visna virus (MVV), which mainly affects the lungs and central nervous system but may also affect the mammary glands, joints and other tissues. The aim of the present study was to determine whether the third eyelid was affected in cases of systemic infection. Third eyelid and lung samples from sheep naturally infected with maedi were used. Total DNA was extracted from paraffin-wax-embedded tissues, and a nested polymerase chain reaction (PCR) was performed to amplify MVV proviral DNA. The samples were also tested by in-sit

Importation of animals or animal products cannot take place without some element of risk. Risk analysis is a blend of art and science and is a tool intended to provide decision-makers with an objective, repeatable and defensible assessment of the risks posed by a particular import proposal. Risk analysis comprises risk identification, risk assessment, risk management and risk communication. Examples are presented of risk analysis involving anthrax in green hides, slow virus diseases and sheep embryos, and Office International des Epizooties List A diseases and embryos. The author proposes that

Aminoglycosides are concentration-dependent antibiotics exerting a bactericidal effect when concentrations at the site of infection are equal to or greater than 5 times the minimum inhibitory concentrations (MIC). When administered intravenously, they exhibit poor lung penetration and high systemic renal and ototoxicity, imposing to restrict their administration to 5 days. Experimental studies conducted in anesthetized and mechanically ventilated sheep and pigs provide evidence that high doses of nebulized aminoglycosides induce a rapid and potent bacterial killing in the infected lung parench

This paper reviews a notion that the prevalence of multiple sclerosis is high in global areas where sheep populations are concentrated. Pilot studies are reported to serum antibodies in humans to three sheep diseases; focal symmetrical encephalomalacia (FSE), maedi visna and sarcocystis. In MS patients and controls antibodies were not found to the epsilon neurotoxin of the FSE organism, Clostridium welchii type D and to a caprine retrovirus that is closely related to maedi-visna virus. However, 34% of MS and control patients had antibodies to the protozoan parasite Sarcocystis spp., tissue cys

One of the arguments in favor of a viral pathogenesis for multiple sclerosis is the existence of several experimental and natural animal models of virus-induced primary demyelination. This review deals comprehensively with such models. Well-known examples of demyelinating viral infections in their natural host are JHM, Theiler, visna, and canine distemper encephalomyelitides. Recent reports of experimental murine infections with pathogens such as vesicular stomatitis, Chandipura, herpes simplex, Venezuelan equine encephalomyelitis, and Semliki Forest viruses are also discussed. The thrust of t
