There are several possible protocols for the treatment of ano-genital condylomata. In this study, the efficacy of podophyllotoxin (0.5%) was assessed. The authors observed a recovery rate at 1 month of 81% in cases of condylomata of the balano-preputial and vulvar mucous membranes, a rate of 67% in condylomata of the keratinized tissues and 63% in those of the anal margin. Response to treatment is also dependent on the morphology of the lesion; regression was observed in 78% of cases of condylomata acuminata and in 57% of cases of endophytic condylomata. The side effects were local and consist

The increasing incidence of genital warts has led to more public awareness of this infection and its possible sequelae. Currently available treatment remains unsatisfactory, and there is pressure to develop effective and convenient alternatives. Podophyllotoxin is standardised and stable, whereas podophyllin has a variable composition. In an open comparison of self applied podophyllotoxin 0.5% versus podophyllin 20% applied by a doctor to treat external penile warts, podophyllotoxin was more effective and gave quicker resolution than podophyllin. Side effects were similar for both preparations

A 16-week double-blind study of parallel groups was carried out in the treatment of 152 patients suffering from stable psoriasis vulgaris. Podophyllotoxin was applied once daily at three different concentrations in an ointment base, namely 0.1, 0.25 and 0.5%. One stable psoriatic lesion was selected and monitored during the study course. All three concentrations of podophyllotoxin induced a statistically highly significant improvement (p less than 0.001) of the selected lesions. There was statistically highly significant difference between the treated and control lesions already after 2 weeks

Podophyllotoxin (PPT) is commonly used for genital warts due to its antimitotic properties and relatively good accessibility since it can be extracted from plants in low-economy countries. However, due to relatively high toxicity, it cannot be used in a systematic way (intravenously). Thus, there is a need to find or create an equally effective derivative of PPT that will be less toxic. Natural PPT is a suitable and promising scaffold for the synthesis of its derivatives. Many of them have been studied in clinical and preclinical models. In this systematic review, we comprehensively assess the

Sixty-five patients with various types of genital warts were treated with 0.5% podophyllotoxin ethanol solution provided by Conpharm AB Sweden including 49 male and 16 female patients. The patients were treated topically bid, for 3 days as one treatment. If the desired effect was not achieved following one treatment, it was repeated in weekly intervals for no more than 3 treatments. Cumulative cure rates of 73.85%, 80% and 81.54% were obtained with podophyllotoxin treatment after the 1st, 2nd and 3rd treatments respectively. The genital warts cured by podophyllotoxin relapsed after one month a

NK611 is a novel water-soluble podophyllotoxin derivative that has comparable antitumour activity but higher potency and better bioavailability in animals as compared with etoposide. The primary objectives of this study were to determine, after both oral and intravenous administration in the same patient, the bioavailability and the pharmacokinetic profile of NK611. Secondary objectives involved evaluation of the toxicity and the antitumor activity. Patients were randomly assigned to receive oral or intravenous (30-min infusion) doses of 5, 10, and 20 mg/m2 on day 1, when pharmacokinetic studi

Plants containing podophyllotoxin and its analogues have been used as folk medicines for centuries. The characteristic chemical structures and strong biological activities of this class of compounds attracted attention worldwide. Currently, more than ninety natural podophyllotoxins were isolated, and structure modifications of these molecules were performed to afford a variety of derivatives, which offered optimized anti-tumor activity. This review summarized up to date reports on natural occurring podophyllotoxins and their sources, structural modification and biological activities. Special a

BACKGROUND: Anogenital warts are a common sexually transmitted disease caused by human papillomaviruses. Despite the fact, that imiquimod and podophyllotoxin represent common topical agents, direct comparative studies lack. This work compares the effectiveness and safety of self-applied imiquimod 5% cream and podophyllotoxin 0.5% solution.
METHODS: Within 2 years, consecutive patients presenting with untreated anogenital warts were included in a randomized, open label trial. The primary endpoint was complete clearance at the end of treatment (4 weeks after the start with podophyllotoxin, 16 w

Podophyllotoxin (PTOX), a plant-derived aryltetralin lignan, has long been appreciated for its strong cytotoxic and antimitotic properties. Various derivatives of PTOX, including etoposide and teniposide, are well-established chemotherapeutics, although their use is largely hampered by systemic toxicity, low solubility, and drug resistance. Recent developments in modern chemistry and cancer biology have led to growing interest in developing novel PTOX-derived compounds with improved efficacy and reduced toxicity. This review presents the molecular mechanisms underlying the antitumor activity o

This study systematically evaluated the therapeutic effects of podophyllotoxin in a DSS-induced mouse model of ulcerative colitis. A total of 374 podophyllotoxin-related targets were identified through database screening, and by intersecting them with 1,741 UC-related targets, 120 potential therapeutic targets were obtained. Subsequent GO and KEGG enrichment analyses revealed that these targets are primarily involved in biological processes such as the positive regulation of protein kinase B signaling, cellular response to lipopolysaccharide, and inflammatory responses, with significant enrich

Podophyllotoxin (POD) is one of the most characterized lignans that is commonly found in podophyllum, and its preparations and derivatives are widely used in clinical treatment due to strong antitumor and antivirus activities. POD has been reported for its neurotoxicity, liver toxicity, and potential reproductive toxicity. In the present study, we investigated the effects of POD on the organelles of mouse oocytes during meiosis. Our results showed that exposure to POD significantly reduced the developmental competence of mouse oocytes. Further analysis revealed that the endoplasmic reticulum (

Podophyllotoxin is used as medical cream which is widely applied to genital warts and molluscum contagiosum. Although previous study showed that podophyllotoxin had minimal toxicity, it was forbidden to use during pregnancy since it might be toxic to the embryos. In present study we used mouse as the model and tried to examine whether podophyllotoxin exposure was toxic to oocyte maturation, which further affected embryo development. Our results showed that podophyllotoxin exposure inhibited mouse oocyte maturation, showing with the failure of polar body extrusion, and the inhibitory effects of

To address the systemic toxicity of podophyllotoxin (PPT), two series of mitochondria-targeted PPT derivatives were designed and synthesized by conjugating PPT with delocalized lipophilic cations (DLCs) TPP+ and F16 via alkyl linkers. Biological evaluation revealed that the F16-conjugated compound 6d exhibited the most potent antiproliferative activity against HepG2 cells with an IC50 value of 0.66 μM, which was significantly superior to the positive control etoposide with an IC50 of 10.82 μM. Additionally, 6d demonstrated improved selectivity toward normal hepatocytes (QSG-7701) with a select

BACKGROUND: Genital warts are a common sexually transmitted disease caused by human papillomaviruses. Podophyllotoxin 0.5%, approved for patient self-administration, has been used most extensively in the treatment of genital warts. Imiquimod, a novel immune response modifier capable of inducing interferon-alpha and a variety of cytokines, has been examined as a potential treatment for genital warts. But 0.5% podophyllotoxin and 5% imiquimod have not been compared in any extensive and formal studies, although they are the common topical agents for genital warts.

OBJECTIVE: This study investigated the relationship between mitotic catastrophe (MC) and apoptosis in oral squamous cell carcinoma (OSCC) using podophyllotoxin (PPT), a natural compound with antimitotic properties.
DESIGN: We evaluated the concentration-dependent effects of PPT on cell proliferation (CCK-8 and soft agar assays) and morphology (transmission electron microscopy). Mechanistic insights were obtained by assessing DNA damage (western blotting), cell cycle progression (sub-G1 analysis), and apoptosis-related protein activation in both 2D and 3D spheroid models of HSC-3 oral squamous

UDP-glucuronosyltransferases (UGTs) are involved in the clearance of many important drugs and endogenous substances, and inhibition of UGTs' activity by herbal components might induce severe herb-drug interactions or metabolic disturbances of endogenous substances. The present study aims to determine the inhibition of UGTs' activity by podophyllotoxin derivatives, trying to indicate the potential herb-drug interaction or metabolic influence towards endogenous substances' metabolism. Recombinant UGT isoforms (except UGT1A4)-catalyzed 4-methylumbelliferone (4-MU) glucuronidation reaction and UGT
