A case of 46,XX/69,XXX mixoploidy is described. The patient had a normal 46,XX diploid karyotype in lymphocytes but a triploid 69,XXX cell line in most of her fibroblasts. In order to learn more about the underlying mechanism resulting in mixoploidy, we studied short tandem repeat polymorphisms (STRPs) in lymphocyte DNA of the patient's parents and in both lymphocyte and fibroblast DNA of the proband. The findings showed maternal origin of the supernumerary chromosome complement and are best explained by second polar body incorporation into a blastomere.

In multi-ciliated cells, directed and synchronous ciliary beating in the apical membrane occurs through appropriate configuration of basal bodies (BBs, roots of cilia). Although it has been experimentally shown that the position and orientation of BBs are coordinated by apical cytoskeletons (CSKs), such as microtubules (MTs), and planar cell polarity (PCP), the underlying mechanism for achieving the patterning of BBs is not yet understood. In this study, we propose that polarity in bundles of apical MTs play a crucial role in the patterning of BBs. First, the necessity of the polarity was disc

The carotid body and its own nerve were removed from cats anesthetized with sodium pentobarbital and placed in an air gap system; the carotid body was bathed in modified Locke's solution equilibrated with 50% O2 in N2, pH 7.43 at 35 degrees C. The sensory discharges, changes in "resting" receptor polarization and the mass receptor potential evoked by ACh or NaCN were recorded with nonpolarizable electrodes placed across the gap. Receptor potentials and sensory discharges evoked by ACh showed an appreciable increase in amplitude and frequency when the preparation was bathed in eserinized Locke.

BACKGROUND: Inflammation has been implicated in numerous diseases. The treatment of inflammation-related diseases is a significant global burden. Recent studies have found that lactic acid and its signalling pathway can inhibit inflammatory responses through macrophage polarization without any toxic side effects.
OBJECTIVE: This study aimed to develop a rational drug delivery system that can release lactic acid into the desired area.
METHODS: We extracted the macrophage-derived apoptotic bodies (ABs) to prepare the AB-encapsulated nanoparticles (ABNs) and evaluated the amount of lactic acid

The development of oocytes is crucial in the development and fertility of embryos in In Vitro Fertilization (IVF). There are several internal and external factors that could affect the fertilization competency of oocytes, one of which is the first polar body (PBI). However, there is still a debate regarding the influence of PBI on the developmental competency of oocytes. This work aims to find the correlation of the successful fertilization in which the mature oocyte develops into the stage of two pronuclei (2PN) by using You Only Look Once (YOLO), which is a type of Convolutional Neural Netwo

Coordinated beating of cilia in the trachea generates a directional flow of mucus required to clear the airways. Each cilium originates from a barrel-shaped basal body, from the side of which protrudes a structure known as the basal foot. We generated mice in which exons 6 and 7 of Odf2, encoding a basal body and centrosome-associated protein Odf2/cenexin, are disrupted. Although Odf2(ΔEx6,7/ΔEx6,7) mice form cilia, ciliary beating is uncoordinated, and the mice display a coughing/sneezing phenotype. Whereas residual expression of the C-terminal region of Odf2 in these mice is sufficient for c

INTRODUCTION: Macrophages and their fused counterpart, foreign body giant cells (FBGCs), have a determining influence on the long-term functionality of implanted biomaterials, coordinating biomaterial resorption and fibrous encapsulation. Modulating macrophage phenotype into favorable polarization states is a widely adopted strategy to attempt steering the foreign body response. However, the links between macrophage phenotype and FBGC formation and function are unknown. The goal of this study was to determine the fusion potential of well-defined macrophage polarization states, and to explore w

Oocytes with excessively large first polar bodies (PB1) often occur in assisted reproductive procedures. Many times these oocytes are discarded without insemination and, as a result, the application of this portion of oocytes has scarcely been reported to date. Few studies have examined large PB1 oocytes in infertile women and have virtually entirely studied genetic variations for large PB1 oocyte abnormalities. Here, we describe an unusual case of a live birth from a remarkably large PB1 oocyte in a frozen embryo transfer (FET) cycle. This is the first instance of a successful live birth resu…

PURPOSE: Our objective was to identify a marker for oocyte aneuploidy in follicular fluid (FF) in women with an increased risk of oocyte aneuploidy after controlled ovarian hyperstimulation.
MATERIALS AND METHODS: Three groups of oocytes were constituted for polar body screening by FISH (chromosomes 13, 16, 18, 21 and 22): Group 1, advanced maternal age (n = 156); Group 2, implantation failure (i.e. no pregnancy after the transfer of more than 10 embryos; n = 101) and Group 3, implantation failure and advanced maternal age (n = 56). FSH and other proteins were assayed in the corresponding FF

BACKGROUND: This study was conducted to investigate the fertilization and embryo development of human oocytes injected at different time intervals after extrusion of the first polar body (PB) following in vitro maturation (IVM) in IVM cycles. Also, we evaluated whether spindle imaging could serve as a tool to determine the optimal ICSI time.
METHODS: Oocytes were collected from 43 women with polycystic ovary syndrome. Metaphase I (MI) oocytes after in vitro culture for 24 h from germinal vesicle stage were subjected to ICSI according to time after first PB extrusion. The intervals were: withi

Preimplantation genetic diagnosis (PGD) may help couples at risk to avoid pregnancies with known genetic diseases. In Germany, the only option to perform PGD is the analysis of polar bodies (PB). Mucopolysaccharidosis type I (MPS I) is an autosomal recessive lysosomal storage disorder. Q70X is one of the frequent diseases causing mutations of alpha-L-iduronidase (IDUA), leading to a severe phenotype with mental retardation and various somatic abnormalities, and making a request for PGD is understandable. Using five polymorphic DNA markers from the vicinity of IDUA, PGD on first PB was performe…

OBJECTIVE: To evaluate the effect of polar body biopsy (PBB) on pregnancy and pregnancy loss outcomes in intracytoplasmic sperm injection (ICSI) cycles.
METHODOLOGY: This is a retrospective case-control study that analyzed 147 ICSI embryo transfer (ET) cycles. The study included 82 subfertile patients (31 patients with ICSI and PBB therapy = case group; 51 patients with ICSI without the PBB therapy = control group). We conducted a statistical analysis of all pregnancies and births resulting from fresh and thawed ICSI cycles, with and without the use of PBB after a single embryo transfer (SET)

Cell motility is critical to biological processes from wound healing to cancer metastasis to embryonic development. The involvement of organelles in cell motility is well established, but the role of organelle positional reorganization in cell motility remains poorly understood. Here we present an automated image analysis technique for tracking the shape and motion of Golgi bodies and cell nuclei. We quantify the relationship between nuclear orientation and the orientation of the Golgi body relative to the nucleus before, during, and after exposure of mouse fibroblasts to a controlled change i

Our objective was to directly compare the diagnostic usefulness of lamellar body counting (LBC) and the TDx-FLM II assay (Abbott Laboratories, Abbott Park, IL) for predicting respiratory distress syndrome (RDS). This was a 5-year, retrospective, cohort study. A diagnosis of RDS was given to infants who received surfactant treatment and/or required ventilator support and/or continuous positive airway pressure for more than 24 hours. There were 172 infants without RDS and 12 with RDS included in the study. By using a TDx-FLM II cutoff of 55 mg/g or more for maturity, the sensitivity was 83%, spe

The polarity of the biological membrane, or lipid order, regulates many cellular events. It is generally believed that the plasma membrane polarity is regulated according to cell type and function, sometimes even within a cell. Neurons have a variety of functionally specialized subregions, each of which bears distinct proteins and lipids, and the membrane polarity of the subregions may differ accordingly. However, no direct experimental evidence of it has been presented to date. In the present study, we used a cell-impermeable solvatochromic membrane probe NR12A to investigate the local polari

Poly(ethylene glycol) (PEG) hydrogels hold promise for in vivo applications but induce a foreign body response (FBR). While macrophages are key in the FBR, many questions remain. This study investigates temporal changes in the transcriptome of implant-associated monocytes and macrophages. Proinflammatory pathways are upregulated in monocytes compared to control monocytes but subside by day 28. Macrophages are initially proinflammatory but shift to a profibrotic state by day 14, coinciding with fibrous capsule emergence. Next, this study assesses the origin of macrophages responsible for fibrou

BACKGROUND: Trophectoderm (TE) cell biopsy at the blastocyst stage is currently the most common method used in preimplantation genetic testing for monogenic disorders (PGT-M). However, this approach may result in the wasting of some genetically unaffected embryos because only a proportion of zygotes develop to the blastocyst stage. Unaffected embryos, which degenerated during blastomere-blastocyst transformation, may give birth if transferred before the blastocyst stage and may be of great value to women with a low oocyte count. This study sought to investigate the potential application of pol

Weight and skin-fold measurements were made at five-day intervals during a 47-day expedition by six men and three women from the edge of the sea ice to the South Pole. From these, together with detailed manual records of the nutrition for individual participants, the average daily energy expenditure was determined before and after a resupply at approximately mid-point of the expedition. For all participants body weight fell during the expedition with the overall loss being much smaller for the three female participants (-4.0, -4.0, -4.4kg) than for the male participants, (mean±sd) -8.6±2.0kg.

BACKGROUND: Parthenogenetic mosaicism is an extremely rare condition identified only in five subjects to date. The previous studies indicate that this condition is mediated by parthenogenetic activation and is free from a specific phenotype ascribed to unmaking of a maternally inherited recessive variant in the parthenogenetic cell lineage.
RESULTS: We examined a 28-year-old Japanese 46,XX female with Silver-Russell syndrome and idiopathic hypersomnia. The results revealed (1) predominance of maternally derived alleles for all the differentially methylated regions examined; (2) no disease-rel
