Lung deposition and cellular uptake behavior of pathogen-mimicking nanovaccines in the first 48 hours.

Pulmonary immunization poses the unique challenge of balancing vaccine efficacy with minimizing inflammation in the respiratory tract. While previous studies have shown that mice immunized intranasally with F1-V-loaded polyanhydride nanoparticles are protected from a lethal challenge with Yersinia pestis, little is known about the initial interaction between the nanoparticles and immune cells following intranasal administration. Here, the deposition within the lung and internalization by phagocytic cells of polyanhydride nanovaccines encapsulating F1-V are compared with that of soluble F1-V al

Pulsatile release of parathyroid hormone from an implantable delivery system.

Intermittent (pulsatile) administration of parathyroid hormone (PTH) is known to improve bone micro-architecture, mineral density and strength. Therefore, daily injection of PTH has been clinically used for the treatment of osteoporosis. However, this regimen of administration is not convenient and is not a favorable choice of patients. In this study, an implantable delivery system has been developed to achieve pulsatile release of PTH. A well-defined cylindrical device was first fabricated with a biodegradable polymer, poly(l-lactic acid) (PLLA), using a reverse solid-free form fabrication te

Nanotherapeutic delivery of antibiotic cocktail enhances intra-macrophage killing of Mycobacterium marinum.

Mycobacterium marinum is a waterborne pathogen responsible for tuberculosis-like infections in cold-blooded animals and is an opportunistic pathogen in humans. M. marinum is the closest genetic relative of the Mycobacterium tuberculosis complex and is a reliable surrogate for drug susceptibility testing. We synthesized and evaluated two nanoparticle (NP) formulations for compatibility with rifampicin, isoniazid, pyrazinamide, and ethambutol (PIRE), the front-line antimycobacterial drugs used in combination against active tuberculosis infections. Improved in vitro antimicrobial activity was obs

Drug delivery system of curcumin to the lungs based on poly(3-alloxyloxy-1,2-propylene succinate)-sebacic acid copolymers.

Polyanhydrides are attractive materials for drug delivery matrices as a result of their cytocompatibility and fast degradation rate. Here, we synthesized and characterized copolymers of poly(3-allyloxy-1,2-propylene succinate) (PSAGE) and sebacic acid (SBA). The successful polymerization was confirmed by proton nuclear magnetic resonance (1H NMR) and Fourier transform infrared (FTIR) spectroscopy analyses. The material with PSAGE and 60% of SBA copolymer (PSAGE-SBA60) was more hydrophilic than the PSAGE and 80% of SBA copolymer (PSAGE-SBA80) (water contact angle 82.2° ± 11.6° vs. 98.6° ± 8.9°,

Effect of nanovaccine chemistry on humoral immune response kinetics and maturation.

Acute respiratory infections represent a significant portion of global morbidity and mortality annually. There is a critical need for efficacious vaccines against respiratory pathogens. To vaccinate against respiratory disease, pulmonary delivery is an attractive route because it mimics the route of natural infection and can confer both mucosal and systemic immunity. We have previously demonstrated that a single dose, intranasal vaccine based on polyanhydride nanoparticles elicited a protective immune response against Yersinia pestis for at least 40 weeks after immunization with F1-V. Herein,

Single-dose combination nanovaccine induces both rapid and durable humoral immunity and toxin neutralizing antibody responses against Bacillus anthracis.

Bacillus anthracis, the causative agent of anthrax, continues to be a prominent biological warfare and bioterrorism threat. Vaccination is likely to remain the most effective and user-friendly public health measure to counter this threat in the foreseeable future. The commercially available AVA BioThrax vaccine has a number of shortcomings where improvement would lead to a more practical and effective vaccine for use in the case of an exposure event. Identification of more effective adjuvants and novel delivery platforms is necessary to improve not only the effectiveness of the anthrax vaccine

Effect of Particle Synthesis Method on Protein Release and Stability.

Polyanhydride particle-based vaccines overcome several limitations of current vaccines owing to their ability to encapsulate different types of antigenic payloads, provide tunable release kinetics of payloads, and induce protective immunity against multiple respiratory infections. In this work, two particle synthesis methods were compared by analyzing the structure and antigenicity of released proteins from particles made by these methods. Flash nanoprecipitation is a lab-scale method to synthesize protein-loaded particles. Spray drying is a scalable method that allows for the production of pr

Applying biodegradable particles to enhance cancer vaccine efficacy.

One of the primary goals of our group and our collaborators here at the University of Iowa is to develop therapeutic cancer vaccines using biodegradable and biocompatible polymer-based vectors. A major advantage of using discretely packaged immunogenic cargo over non-encapsulated vaccines is that they promote enhanced cellular immunity, a key requirement in achieving antitumor activity. We discuss the importance of co-encapsulation of tumor antigen and adjuvant, with specific focus on the synthetic oligonucleotide adjuvant, cytosine-phosphate-guanine oligodeoxynucleotides. We also discuss our

Intravitreal devices for the treatment of vitreous inflammation.

The eye is a well-suited organ for local delivery of therapeutics to treat vitreous inflammation as well as other pathologic conditions that induce visual loss. Several conditions are particularly challenging to treat and often require chronic courses of therapy. The use of implantable intravitreal devices for drug delivery is an emerging field in the treatment of vitreous inflammation as well as other ophthalmologic diseases. There are unique challenges in the design of these devices which include implants, polymers, and micro- and nanoparticles. This paper reviews current and investigational

Investigation into the erosion mechanism of salicylate-based poly(anhydride-esters).

Salicylate-based poly(anhydride-esters), collectively referred to as PolyAspirin, hydrolytically degrade into salicylic acid, a nonsteroidal anti-inflammatory drug (NSAID). The variations of the poly(anhydride-ester) investigated in this article are linked by adipic acid, suberic acid, or sebacic acid. To elucidate the erosion mechanism of these polymers, water uptake, mass loss, contact angle, and changes in device thickness were monitored as a function of in vitro degradation time. The polymers examined here appeared to primarily undergo surface erosion. The adipate homopolymer absorbed the

A nanovaccine induces durable and functional MUC4-targeted T cell responses against pancreatic cancer.

Vaccination against cancer for prophylaxis or treatment is becoming a viable approach owing to new developments in the identification of novel biomarkers and advances in our understanding of cancer immunology. Furthermore, nanotechnology has resulted in multifunctional delivery platforms with versatile chemistries and tunable release properties. However, despite this impressive progress, immunotherapy with the goal of inducing antigen-specific T cell immunity against pancreatic cancer (PC) remains challenging. Herein, we report the characterization of the long-lived humoral and cellular immuni

Next-generation nanovaccine induces durable immunity and protects against SARS-CoV-2.

While first generation SARS-CoV-2 vaccines were effective in slowing the spread and severity of disease during the COVID-19 pandemic, there is a need for vaccines capable of inducing durable and broad immunity against emerging variants of concern. Nanoparticle-based vaccines (i.e., "nanovaccines") composed of polyanhydride nanoparticles and pentablock copolymer micelles have previously been shown to protect against respiratory pathogens, including influenza A virus, respiratory syncytial virus, and Yersinia pestis. In this work, a nanovaccine containing SARS-CoV-2 spike and nucleocapsid antige

Tailored dual release of retinol and salicylic acid from salicylate-based poly(anhydride-ester) microspheres with tunable degradation profiles.

Retinol and salicylic acid are potent bioactive agents commonly used to treat skin pathologies such as acne, psoriasis, and photodamage. Their combined use offers synergistic therapeutic benefits. However, effective delivery of retinol remains challenging due to its poor aqueous solubility and rapid degradation under physiological conditions. In this study, a dual-delivery microsphere platform based on salicylic acid-derived poly(anhydride-ester)s (SAPAEs) was designed to co-release retinol and salicylic acid in a controlled and pH-responsive manner. Four SAPAE compositions with distinct degra

Synthesis and cytotoxicity of salicylate-based poly(anhydride esters).

This paper describes the synthesis and cytotoxicity of poly(anhydride esters) that are composed of several salicylate derivatives, including halogenated salicylates, aminosalicylates, salicylsalicylic acid, and thiolsalicylic acid. The incorporation of these nonsteroidal antiinflammatory drugs (NSAIDs) into a biodegradable polymer backbone yields drug-based polymers that have potential for a variety of applications. The poly(anhydride esters) were synthesized by melt condensation polymerization. The halogenated salicylate derivatives yielded the highest molecular polymers as well as the highes

Salicylic acid-based poly(anhydride-ester) nerve guidance conduits: Impact of localized drug release on nerve regeneration.

Nerve guidance conduits (NGCs) can serve as physical scaffolds aligning and supporting regenerating cells while preventing scar tissue formation that often interferes with the regeneration process. Numerous studies have focused on functionalizing NGCs with neurotrophic factors, for example, to support nerve regeneration over longer gaps, but few directly incorporate therapeutic agents. Herein, we fabricated NGCs from a polyanhydride comprised of salicylic acid (SA), a nonsteroidal anti-inflammatory drug, then performed in vitro and in vivo assays. In vitro studies included cytotoxicity, anti-i

Single-dose combination nanovaccine induces both rapid and long-lived protection against pneumonic plague.

Yersinia pestis, the causative agent of pneumonic plague, induces a highly lethal infection if left untreated. Currently, there is no FDA-approved vaccine against this pathogen; however, USAMRIID has developed a recombinant fusion protein, F1-V, that has been shown to induce protection against pneumonic plague. Many F1-V-based vaccine formulations require prime-boost immunization to achieve protective immunity, and there are limited reports of rapid induction of protective immunity (≤ 14 days post-immunization (DPI)). The STimulator of INterferon Genes agonists cyclic dinucleotides (CDNs) have

Iodinated salicylate-based poly(anhydride-esters) as radiopaque biomaterials.

Poly(anhydride-esters) based on iodinated versions of salicylic acid were synthesized via both melt-condensation and solution polymerization techniques to generate radiopaque biomaterials. The poly(anhydride-esters) from iodinated salicylates were highly X-ray opaque compared to poly(anhydride-esters) from salicylic acid. Molecular weight and Young's modulus of polymers prepared by melt-condensation were typically two-to-three times higher than polymers prepared by solution methods. The glass transition temperatures of the polymers were dependent on the iodine concentration; polymers containin

Design of a protective single-dose intranasal nanoparticle-based vaccine platform for respiratory infectious diseases.

Despite the successes provided by vaccination, many challenges still exist with respect to controlling new and re-emerging infectious diseases. Innovative vaccine platforms composed of adaptable adjuvants able to appropriately modulate immune responses, induce long-lived immunity in a single dose, and deliver immunogens in a safe and stable manner via multiple routes of administration are needed. This work describes the development of a novel biodegradable polyanhydride nanoparticle-based vaccine platform administered as a single intranasal dose that induced long-lived protective immunity agai

Sustained local anesthetic release from bioerodible polymer matrices: a potential method for prolonged regional anesthesia.

Polyanhydride polymer matrices have been used successfully for sustained release of a number of drugs in vitro and in vivo. Dibucaine free base, dibucaine HCl, and bupivacaine HCl were incorporated into polymer matrices with copolymer 1,3-bis(p-carboxyphenoxy)propane-sebacic acid anhydride (1:4). Drug release was measured in vitro following incubation of the drug-polymer matrices in phosphate buffered solution, pH 7.4, at 37 degrees C, to approximate in vivo conditions. Local anesthetics were released in a sustained manner yielding 90% cumulative drug release over periods ranging from 3 to 14
