DNA Hypomethylation Is Not Cell Intrinsically Toxic to Polycomb Repressive Complex 2 Deficient Malignant Peripheral Nerve Sheath Tumors.

Malignant peripheral nerve sheath tumors (MPNSTs) are aggressive soft tissue sarcomas and the most common cause of disease-associated death for neurofibromatosis type 1 (NF1) patients. In the context of NF1, MPNSTs develop from benign premalignant precursors and the transition to malignancy is typically accompanied by loss of the polycomb repressive complex 2 (PRC2), which results in aberrant upregulation of over 1200 genes due to global depletion of histone H3 lysine 27 trimethylation (H3K27me3). Previous studies suggest cells compensate for the loss of this repressive histone mark via hyperm

Deregulation of Polycomb Repressive Complex-2 in Mantle Cell Lymphoma Confers Growth Advantage by Epigenetic Suppression of cdkn2b.

The polycomb repressive complex 2 (PRC2) maintains the transcriptional repression of target genes through its catalytic component enhancer of zeste homolog 2 (EZH2). Through modulating critical gene expression, EZH2 also plays a role in cancer development and progression by promoting cancer cell survival and invasion. Mutations in EZH2 are prevalent in certain B-cell lymphoma subtypes such as diffuse large cell lymphoma and follicular lymphoma; while no EZH2 mutation has been reported in the mantle cell lymphoma (MCL). Here we demonstrate that the PRC2 components EZH2, EED and SUZ12 are upregu

Aberrant differential expression of EZH1 and EZH2 in Polycomb repressive complex 2 among B- and T/NK-cell neoplasms.

The Polycomb repressive complex-2 members (EZH2, EED, SUZ12 and EZH1) are important regulators of haematopoiesis, cell cycle and differentiation. Over-expression of EZH2 has been linked to cancer metastases and poor prognosis. Detailed information on the expression of other members in normal and neoplastic lymphoid tissue remains to be elucidated. Immunohistochemical and immunofluorescent analyses of 156 samples from haematopoietic neoplasms patients and 27 haematopoietic cell lines were used. B-cell neoplasms showed a significant over-expression of EZH2, EED and SUZ12 in the aggressive subtyp

Enhancer of zeste 2 polycomb repressive complex 2 subunit overexpression suppresses apoptosis in canine T-cell lymphoma: an in vitro functional analysis.

Lymphoma is one of the most common neoplastic diseases in dogs, and T-cell lymphoma in particular has frequently been reported to show limited sensitivity to conventional chemotherapy from the onset of treatment when compared with B-cell lymphoma. Owing to its high-grade and aggressive clinical behavior, there is a strong need to explore novel therapeutic approaches for canine T-cell lymphoma, including strategies targeting aberrant gene regulation. In this study, we focused on the epigenetic enzyme enhancer of zeste 2 polycomb repressive complex 2 subunit (EZH2) and investigated its functiona

Chromobox 4 interacts with pregnane X receptor to repress CYP3A4 expression through polycomb repressive complex 1/2-mediated epigenetic regulation.

Polycomb repressive complexes (PRCs) 1 and 2 are epigenetic regulators that suppress gene transcription through histone H3 lysine K27 trimethylation. This modification, catalyzed by enhancer of zeste homolog 2 (EZH2) within PRC2, is recognized by chromobox (CBX) proteins within PRC1, leading to chromatin condensation and transcriptional repression. Although EZH2 and CBX inhibitors have been developed for cancer therapy, the role of PRC1/2 inhibition in the regulation of drug metabolism-related genes remains unclear. In this study, we investigated the regulatory mechanisms of CYP3A4 expression

Polycomb repressive complexes 1 and 2 independently and dynamically regulate euchromatin during cerebellar neurodevelopment.

Polycomb Repressive Complexes (PRCs) are known for chemically modifying histones to compact chromatin structure and repress transcription. Broadly speaking, PRC1 monoubiquitinates histone 2A at lysine 119 (H2AK119ub), and PRC2 methylates histone H3 lysine 27 (H3K27me3, H3K27me2 and H3K27me1), but the scope and functions of these activities are complicated by a multiplicity of factors involving distinct cellular contexts and compositions of both complexes. Because epigenetic dysregulation is associated with neurodevelopmental disorders, but little is known about normal PRC activities in neurons

Epigenetically regulated transcriptional repressor, BASIC PENTACYSTEINE1 (BPC1) acts together with polycomb repressive complex 2 (PRC2) to suppress camalexin biosynthesis in Arabidopsis.

Camalexin is a phytoalexin produced by plants of the Brassicaceae family, including the model species Arabidopsis thaliana, and its biosynthesis is tightly regulated to balance growth and defense. Here, we identify BASIC PENTACYSTEINE 1 (BPC1) as a key negative regulator of camalexin biosynthesis. Transcriptome analyses of bpc1-1 and bpc1-1;bpc2-1 mutants revealed both distinct and overlapping functions of BPC1 and BPC2, with BPC1-specific differentially expressed genes being highly enriched for camalexin biosynthetic processes. Consistently, multiple camalexin pathway genes were upregulated i

Deregulated expression of polycomb repressive complex 2 target genes in a NF1 patient with microdeletion generating the RNF135-SUZ12 chimeric gene.

Neurofibromatosis type I (NF1) microdeletion syndrome, accounting for 5-11% of NF1 patients, is caused by the heterozygous deletion of NF1 and a variable number of flanking genes in the 17q11.2 region. This syndrome is characterized by more severe symptoms than those shown by patients with intragenic NF1 mutation and by variable expressivity, which is not fully explained by the haploinsufficiency of the genes included in the deletions. We here reevaluate an 8-year-old NF1 patient, who carries an atypical deletion generating the RNF135-SUZ12 chimeric gene, previously described when he was 3 yea

Epigenetic changes mediated by polycomb repressive complex 2 and E2a are associated with drug resistance in a mouse model of lymphoma.

BACKGROUND: The genetic origins of chemotherapy resistance are well established; however, the role of epigenetics in drug resistance is less well understood. To investigate mechanisms of drug resistance, we performed systematic genetic, epigenetic, and transcriptomic analyses of an alkylating agent-sensitive murine lymphoma cell line and a series of resistant lines derived by drug dose escalation. METHODS: Dose escalation of the alkylating agent mafosfamide was used to create a series of increasingly drug-resistant mouse Burkitt's lymphoma cell lines. Whole genome sequencing, DNA microarrays,

ZINC FINGER PROTEIN 1 and 8 interact with polycomb repressive complex 2 to repress class B and C floral organ identity genes.

Precise control of gene expression is essential for establishing and maintaining cell identities in response to endogenous signals and environmental cues in multicellular organisms. An example is the regulation of the class B and C floral organ identity genes-APETALA3 (AP3), PISTILLATA (PI), and AGAMOUS (AG)-which are repressed in leaves and activated in floral meristems. Their repression in leaves is associated with CURLY LEAF (CLF), a core component of the polycomb repressive complex 2 (PRC2). However, since CLF is expressed in both leaf and floral primordia, its target specificity is likely

Druggability assessments of small peptides as protein-protein interaction inhibitors targeting EED-EZH2 binding within the Polycomb Repressive Complex 2 (PRC2), an epigenetic regulator.

Histone methylation is a key epigenetic modification involved in gene silencing and plays a crucial role in chromatin remodelling. Polycomb repressive complex 2 (PRC2), a histone-modifying complex, has been implicated in various diseases, including cancers and genetic disorders. Since PRC2 is a multi-protein complex, the structural interactions among its component proteins are essential for its proper function. Consequently, inhibiting the formation of this complex is considered an effective strategy to block PRC2 activity. This research evaluated small peptides as protein-protein interaction

Long noncoding RNA MALAT1 releases epigenetic silencing of HIV-1 replication by displacing the polycomb repressive complex 2 from binding to the LTR promoter.

Long noncoding RNAs (lncRNAs) may either repress or activate HIV-1 replication and latency; however, specific mechanisms for their action are not always clear. In HIV-1 infected CD4+ T cells, we performed RNA-Sequencing (RNA-Seq) analysis and discovered an up-regulation of MALAT1 (metastasis-associated lung adenocarcinoma transcript 1), an lncRNA previously described in cancer cells that associate with cancer pathogenesis. Moreover, we found that MALAT1 promoted HIV-1 transcription and infection, as its knockdown by CRISPR/Cas9 markedly reduced the HIV-1 long terminal repeat (LTR)-driven gene

Polycomb Repressive Complex 2 Enacts Wnt Signaling in Intestinal Homeostasis and Contributes to the Instigation of Stemness in Diseases Entailing Epithelial Hyperplasia or Neoplasia.

Canonical Wnt/β-catenin signaling regulates the homeostasis of intestinal epithelium by controlling the balance between intestinal stem cell self-renewal and differentiation but epigenetic mechanisms enacting the process are not known. We hypothesized that epigenetic regulator, Polycomb Repressive Complex-2 (PRC2), is involved in Wnt-mediated epithelial homeostasis on the crypt-villus axis and aberrancies therein are implicated both in celiac disease and in intestinal malignancies. We found that PRC2 establishes repressive crypt and villus specific trimethylation of histone H3 lysine 27 (H3K27

PRC1 suppresses a female gene regulatory network to ensure testicular differentiation.

Gonadal sex determination and differentiation are controlled by somatic support cells of testes (Sertoli cells) and ovaries (granulosa cells). In testes, the epigenetic mechanism that maintains chromatin states responsible for suppressing female sexual differentiation remains unclear. Here, we show that Polycomb repressive complex 1 (PRC1) suppresses a female gene regulatory network in postnatal Sertoli cells. We genetically disrupted PRC1 function in embryonic Sertoli cells after sex determination, and we found that PRC1-depleted postnatal Sertoli cells exhibited defective proliferation and c

Loss of BAP1 function leads to EZH2-dependent transformation.

The tumor suppressors BAP1 and ASXL1 interact to form a polycomb deubiquitinase complex that removes monoubiquitin from histone H2A lysine 119 (H2AK119Ub). However, BAP1 and ASXL1 are mutated in distinct cancer types, consistent with independent roles in regulating epigenetic state and malignant transformation. Here we demonstrate that Bap1 loss in mice results in increased trimethylated histone H3 lysine 27 (H3K27me3), elevated enhancer of zeste 2 polycomb repressive complex 2 subunit (Ezh2) expression, and enhanced repression of polycomb repressive complex 2 (PRC2) targets. These findings co

Insight into the Inhibitory Mechanism of Embryonic Ectoderm Development Subunit by Triazolopyrimidine Derivatives as Inhibitors through Molecular Dynamics Simulation.

Inhibition of the Embryonic Ectoderm Development (EED) subunit in Polycomb Repressive Complex 2 (PRC2) can inhibit tumor growth. In this paper, we selected six experimentally designed EED competitive Inhibitors of the triazolopyrimidine derivatives class. We investigated the difference in the binding mode of the natural substrate to the Inhibitors and the effects of differences in the parent nuclei, heads, and tails of the Inhibitors on the inhibitory capacity. The results showed that the binding free energy of this class of Inhibitors was close to or lower compared to the natural substrate, p

PRC2-RNA interactions: Viewpoint from Tom Cech, Chen Davidovich, and Richard Jenner.

Diverse biochemical, structural, and in vivo data support models for the regulation of polycomb repressive complex 2 (PRC2) activity by RNAs, which may contribute to the maintenance of epigenetic states. Here, we summarize this research and also suggest why it can be difficult to capture biologically relevant PRC2-RNA interactions in living cells.

H3K27me3 spreading organizes canonical PRC1 chromatin architecture to regulate developmental programs.

Polycomb repressive complex 2 (PRC2)-mediated histone H3 K27 trimethylation (H3K27me3) recruits canonical PRC1 (cPRC1) to maintain heterochromatin. In early development, Polycomb-regulated genes can display long-range three-dimensional interactions, many of which resolve during lineage differentiation. Here we report that Polycomb-anchored looping is controlled by H3K27me3 spreading and regulates target gene silencing to influence cell fate specification. Using glioma-derived H3 Lys27-to-Met (H3K27M) mutations as tools to restrict H3K27me3 spreading, we show that H3K27me3 confinement concentra

Drosophila Rumpelstiltskin regulates Polycomb cis and trans interactions and Homie barrier activity.

BACKGROUND: Three-dimensional genome organization is maintained in part by chromatin insulators, DNA-protein complexes that create boundaries between active and inactive chromatin and regulate enhancer-promoter interactions. In Drosophila, the Homing insulator at eve (Homie) sequence prevents repressive Polycomb (PcG) chromatin at even-skipped (eve) from spreading into the neighboring essential gene, TER94. The exact mechanisms controlling Homie barrier activity are understudied. RESULTS: Here, use of an in vivo reporter assay identified the requirement of the hnRNP M homolog, Rumpelstiltskin
