Drimys winteri J.R. (Winteraceae) produce drimane sesquiterpenoids with activity against Candida yeast. In this work, drimenol, polygodial (1), isotadeonal (2), and a new drimane α,β-unsaturated 1,4-dialdehyde, named winterdial (4), were purified from barks of D. winteri. The oxidation of drimenol produced the monoaldehyde drimenal (3). These four aldehyde sesquiterpenoids were evaluated against six Candida species isolated from candidemia patients in Chilean hospitals. Results showed that 1 displays fungistatic activity against all yeasts (3.75 to 15.0 µg/mL), but irritant effects on eyes and

Sepsis is a clinical condition resulting from a dysregulated immune response to an infection that leads to organ dysfunction. Despite numerous efforts to optimize treatment, sepsis remains to be the main cause of death in most intensive care units. The endogenous cannabinoid system (ECS) plays an important role in inflammation. Cannabinoid receptor 2 (CB2R) activation is immunosuppressive, which might be beneficial during the hyper-inflammatory phase of sepsis. Beta-caryophyllene (BCP) is a non-psychoactive natural cannabinoid (phytocannabinoid) found in Cannabis sativa and in essential oils o

It is known that the first osmotic phase affects the growth rates of roots immediately upon addition of salt; thus, dissecting metabolites profiling provides an opportunity to throw light into the basis of plant tolerance by searching for altered signatures that may be associated with tolerance at this organ. This study examined the influence of salt treatment on fatty acid composition and chemical composition of the essential oil of C. aurantium roots. Results proved that, under salt treatment, an increase of double bond index and linoleic desaturation ratio was pointed out. On the other hand

UNLABELLED: (-)-Ambrox, the smelling principle in ambergris of sperm whales, is a saturated polycyclic terpenoid ether with multiple tertiary and quaternary centers. Despite these structural features, it is readily biodegradable and thus represents an interesting case to understand the degradation pathway of a complex, saturated polycyclic terpene. We isolated two Sphingomonas spp. strains from activated sewage sludge capable of rapidly mineralizing (-)-Ambrox. The structures of seven isolated metabolites were determined by 2D-NMR, and further metabolite structures were proposed based on their

The main challenge in schistosomiasis control has been the emergence of drug-resistant parasites. Since the 1970's, praziquantel (PZQ) is the single drug for treatment. This fact highlights the importance to research news chemotherapeutic agents. In the last years, S. mansoni excretory system and tegument have been major targets for drug development. The purpose of this study was to evaluate the effect of sesquiterpenes, alpha-humulene and trans-caryophyllene on S. mansoni survival, excretory system and membrane integrity, after in vitro exposure. The in vitro studies, showed that sesquiterpen

We herein disclose that the epimerization of yahazunol can lead to an antifungal enhancement. Yahazunol-related drimane meroterpenoids and pseudonatural mimics were synthesized based on an improved synthesis of 9-epi-borono-sclareolide and the concomitant Suzuki coupling. Several 9-epi-drimane meroterpenoids presented antifungal activity. The new stereoisomer 9-epi-yahazunol (EC50 = 4.32 μM) and its analog S-T5 (EC50 = 2.8 μM) showed much better antifungal activity than yahazunol (EC50 >150 μM) against Rhizoctonia solani. Compound S-T5 possesses a promising in vivo efficacy against R. solani,

Lactone-containing natural products are important candidates for drug discovery. Drimane-type sesquiterpenes (DTSs), characterized by a bicyclic trans-decalin scaffold, can bear both β- and γ-lactone moieties. While γ-lactone-containing DTSs have frequently been reported, β-lactone-containing derivatives are rare, and their biosynthesis remains unexplored. Here, we identified a biosynthetic gene cluster (dri) in Aspergillus ustus and confirmed ustidrimane A (1), a β- and γ-lactone-containing DTS, as its product. Heterologous gene expression, precursor feeding, and enzymatic investigation provi

Mikania micrantha is a perennial liana of the genus Mikania of the Asteraceae family. It is a commonly used medicine in South America for treating fever, malaria, dysentery, snake bites, etc. Because of its strong adaptability and ability to inhibit the growth of its associated plants, Mikania micrantha is considered an invasive species in China and is known as a plant killer. Preliminary studies have shown that Mikania micrantha has an antipruritic effect, but the antipruritic active substance is not yet clear. In this study, a 4-aminopyridine-induced itching model in mice was used to determi

The effect of exogenous application of jasmonic acid (JA) on the concentration of main terpenes and density of glandular trichomes was investigated in the Mexican oregano, propagated from seeds from 3 localities. JA 1 mM was applied locally and to the whole plant. JA locally applied increased the number of trichomes, with a mean of 20 trichomes more with respect to the controls in plants from Tecomavaca and Zapotitlán Salinas, and significantly increased the thymol concentration by 185% systemically and 255% locally, compared to the control. JA applied to the whole plant decreased the number o

The sesquiterpene (E)-β-caryophyllene is emitted from maize (Zea mays) leaves and roots in response to herbivore attack. This compound serves as a signal for the attraction of herbivore enemies and is present in most European maize varieties. However, most North American maize lines have lost the ability to produce (E)-β-caryophyllene. Previously, we showed that restoring the ability to synthesize (E)-β-caryophyllene in a non-producing maize line improved its resistance against the root herbivore Diabrotica virgifera virgifera. However, it is largely unknown whether this modification affects t

The first enantioselective total synthesis of 10-isocyano-4-cadinene, a marine sesquiterpene isolated from nudibranchs of the family Phyllidiidae, and determination of its absolute stereochemistry were achieved. 10-Isocyano-4-cadinene is expected to be a novel nontoxic antifouling agent. In the synthesis, intermolecular Diels-Alder reaction and samarium diiodide induced Barbier-type cyclization were employed as key steps. The absolute configuration of 10-isocyano-4-cadinene was determined as (1S,6S,7R,10S) by comparison of the optical rotations between natural and synthetic samples. In additio

A total of 118 Japanese marine invertebrates were extracted with methanol, and their chloroform-soluble materials tested for antifouling activity as well as toxicity against barnacle (Balanus amphitrite) larvae. Among 86 species that showed more than 80% larval settlement inhibition, 13 species, including 8 sponges were weakly toxic to the barnacle larvae. The sponge Acanthella cavernosa, which showed most promising activity, was extracted with ethanol and fractionated by solvent partitioning, silica gel column chromatography, gel filtration, and ODS HPLC to give two active compounds which on

The Asian Citrus Psyllid (ACP), Diaphorina citri, severely threatens citrus production worldwide by transmitting the greening (= Huanglongbing)-causing bacterium Candidatus Liberibacter asiaticus. There is growing evidence that the push-pull strategy is suitable to partially mitigate HLB by repelling ACP with transgenic plants engineered to produce repellents and attracting the vector to plants with a minimal disease transmission rate. Species that pull ACP away from commercial citrus plants have been identified, and transgenic plants that repel ACP have been developed. The concept of a repell

Whiteflies pose a major threat to crops worldwide, primarily because they transmit begomoviruses with which they have evolved intricate mutualistic relationships. The mutualisms are known to exacerbate whitefly invasions and drive widespread plant virus pandemics. Yet, certain plant genotypes are able to resist both the whiteflies and the viruses and a good understanding of the underlying mechanisms could help to develop more resistant varieties. Here, we show that the viruliferous whitefly Bemisia tabaci induces an early and strong release of the sesquiterpene β-caryophyllene in cultivated to

Experiments were carried out to test the capacity for a laboratory-scale biofilter operated at an elevated temperature level (∼50°C) to remove an air stream containing β-caryophyllene, a naturally occurring sesquiterpene of environmental concern emitted from wood-related industrial facilities. A water jacket was used to maintain high temperatures in a laboratory-scale biofilter. Inocula, pollutant loading and nutrient supply rate effects were evaluated over 84 days of biofilter operation. The start-up process took over two months when citrus peels were used as inocula while a relatively short

The ability of the plant-derived compound nootkatone to control nymphs of the blacklegged tick, Ixodes scapularis Say, was evaluated at lawn perimeter plots at homes in Lyme disease endemic areas of Connecticut. Three formulations of nootkatone ranging from 0.05 to 0.84% (0.06 - 1.03 g AI/m2) were applied by a hydraulic sprayer from 2008 to 2010. In 2008, the 0.84% emulsifiable nootkatone formulation provided 100% control of I. scapularis through week 1, but declined to 49 and 0% by 2 and 3 wk posttreatment, respectively. A combination of 0.05% nootkatone and entomopathogenic fungus, Metarhizi

A 63-year-old male school teacher with itchy depigmented macules on his left dorsum manus, left shoulder and abdomen presented at our clinic on 8 July 1986. He had practiced the incense ceremony for about 15 years, and had burnt several incenses and sandalwood. 48 h closed patch testing revealed perfume in the incenses was the cause. We assumed that perfume in the incense was volatized in air when incense was burnt; skin surface contact occurred with airborne particle, which dissolved in sebum; thus allergic contact dermatitis accompanied by depigmentation might arise.
