Occurrence and phase distribution of lipophilic marine algal toxins in the bottom boundary layer and sediment-porewater system of two mariculture sites.

To date, understanding the fate of lipophilic marine algal toxins (LMATs) in benthic environments on which cultivated shellfish depend is still limited. In this work, the occurrence, concentration levels, and phase distributions of LMATs in the benthic environments of two mariculture sites (Sishili and Rongcheng Bays) in China were investigated for the first time. Five LMATs: okadaic acid (OA), pectenotoxin-2 (PTX2), gymnodimine, 13-desmethyl spirolide C, and azaspiracid-2 (AZA2) and three derivatives: dinophysistoxin-1 isomer (DTX1-iso), pectenotoxin-2 seco acid, and 7-epi- pectenotoxin-2 sec

Cytotoxic effect of palytoxin on mussel.

Palytoxin is a large and complex polyhydroxylated molecule with potent neurotoxic activity. Dinoflagellates from the Ostreopsis genera were demonstrated to be producers of this compound and analogues. Even though initially palytoxin appearance was restricted to tropical areas, the recent occurrence of Ostreopsis outbreaks in Mediterranean Sea point to a worldwide dissemination probably related to climatic change. Those dinoflagellates can bioaccumulate in shellfish, especially in filter-feeding mollusks and have been involved in damaging effects in seafood or human toxic outbreaks. The present

Simplifying marine biotoxin data analysis: Advancing the Neuro-2a cell-based assay using R.

Marine biotoxins (MBs) pose a significant public health risk as seafood contaminants. Testing seafood for MBs before consumption can protect consumers and further provide quality assurances. The in vitro Neuro-2a (N2a) cell-based assay (CBA), is a sensitive, high-throughput, and cost-effective method for the semi-quantification of MBs such as ciguatoxins (CTXs) and brevetoxins (PbTxs), serving as an alternative to animal-based testing. However, the N2a CBA has not been validated, limiting its utility. High data variability based on several potential sources including inter-software inconsisten

Initial evaluation of the effects of aerosolized Florida red tide toxins (brevetoxins) in persons with asthma.

Florida red tides annually occur in the Gulf of Mexico, resulting from blooms of the marine dinoflagellate Karenia brevis. K. brevis produces highly potent natural polyether toxins, known as brevetoxins, that activate voltage-sensitive sodium channels. In experimental animals, brevetoxins cause significant bronchoconstriction. A study of persons who visited the beach recreationally found a significant increase in self-reported respiratory symptoms after exposure to aerosolized Florida red tides. Anecdotal reports indicate that persons with underlying respiratory diseases may be particularly su

Harmful algal toxins of the Florida red tide (Karenia brevis): natural chemical stressors in South Florida coastal ecosystems.

The Florida red tide is a descriptive name for high concentrations of the harmful marine alga, Karenia brevis. Although most prevalent along the south-west Florida coast, periodic blooms have occurred throughout the entire US and Mexico Gulf coasts and the Atlantic coast to North Carolina. This dinoflagellate produces a suite of polyether neurotoxins, called brevetoxins, that cause severe impacts to natural resources, as well as public health. These naturally produced biotoxins may represent one of the most common chemical stressors impacting South Florida coastal and marine ecosystems. Impact

First Identification of Palytoxin-Like Molecules in the Atlantic Coral Species Palythoa canariensis.

Palytoxin (PLTX) is a complex marine toxin produced by Zoanthids (Palyhtoa), dinoflagellates (Ostreopsis), and cyanobacteria (Trichodesmium). Contact with PLTX-like compounds present in aerosols or marine organisms has been associated with adverse effects on humans. The worldwide distribution of producer species and seafood contaminated with PLTX-like molecules illustrates the global threat to human health. The identification of species capable of palytoxin production is critical for human safety. We studied the presence of PLTX analogues in Palythoa canariensis, a coral species collected in t

Brevetoxicosis in loggerhead (Caretta caretta) and Kemp's ridley (Lepidochelys kempii) sea turtles: Plasma proteomics for Florida red tide biomarkers.

Sea turtles are among the many wildlife species adversely affected by Florida red tide, Karenia brevis. This marine dinoflagellate blooms almost annually along Florida's west coast and produces brevetoxins, a suite of potent neurotoxins. Brevetoxins can reach carnivorous loggerhead and Kemp's ridley sea turtles via aerosols and marine food webs, leading to multi-system physiological effects. Sea turtles stranded during red tide can be rescued, transported to rehabilitation facilities, and given palliative care with the goal of return to the wild. However, there are no definitive diagnostic cri

Cross-shelf hydrography differentially shapes phycotoxin distribution across the outer Patagonian continental margin (SW Atlantic).

Microalgal toxins have been studied for decades, yet their occurrence and ecological role in offshore environments remain poorly understood due to the scarcity of observations. The external margin of the Patagonian continental shelf (Southwest Atlantic, Argentina) is a highly productive region supporting intensive fisheries. Phytoplankton phenology and biomass have been assessed largely through remote sensing, while direct assessment of toxin-producing microalgae remains limited, despite recent reports of exceptionally intense blooms. Here, we investigated the co-occurrence of multiple phycoto

Molecular properties of the sodium channel: a receptor for multiple neurotoxins.

Sodium channels are the receptors for more than 10 distinct classes of biological toxins which act at five or more receptor sites. Brevetoxins and ciguatoxins act at neurotoxin receptor site 5. This receptor site is located on the alpha subunit of the sodium channel, and peptide segments in domain IV of that subunit are important in brevetoxin binding.

Lipo-protein deposition strategy towards smart microspheres with bacterial toxin triggered "adsorption-pore formation-immune regulation" cascade reaction for sepsis treatment.

Sepsis induced by Gram-positive bacteria causes immune disorder and multi-organ damage driven by pore-forming toxins (PFTs), which directly form pores on host cell membrane and exacerbate cell damage. Traditional drug therapy cannot remove the existing bacterial toxins, and exacerbate inflammatory storm by increasing toxin release during bacterial lysis. Meanwhile, current materials for toxin clearance lack the capability to regulate the microenvironment and pose systemic retention risks. This study proposes smart microspheres with bacterial toxin triggered "adsorption-pore formation-immune re

Toxicity screening of 13 Gambierdiscus strains using neuro-2a and erythrocyte lysis bioassays.

Species in the epi-benthic dinoflagellate genus Gambierdiscus produce ciguatoxins (CTXs) and maitotoxins (MTXs), which are among the most potent marine toxins known. Consumption of fish contaminated with sufficient quantities of CTXs causes Ciguatera Fish Poisoning (CFP), the largest cause of non-bacterial food poisoning worldwide. Maitotoxins, which can be found in the digestive system of fish, could also contribute to CFP if such tissues are consumed. Recently, an increasing number of Gambierdiscus species have been identified; yet, little is known about the variation in toxicity among Gambi

Direct assembly of multiply oxygenated carbon chains by decarbonylative radical-radical coupling reactions.

Pentoses and hexoses contain more than three oxygen-bearing stereocentres and are ideal starting materials for the synthesis of multiply oxygenated natural products such as sagittamide D, maitotoxin and hikizimycin. Here we demonstrate new radical-radical homocoupling reactions of sugar derivatives with minimal perturbation of their chiral centres. The radical exchange procedure using Et3B/O2 converted sugar-derived α-alkoxyacyl tellurides into α-alkoxy radicals via decarbonylation and rapidly dimerized the monomeric radicals. The robustness of this process was demonstrated by a single-step pr

Occurrence and sequestration of toxins in food chains.

Animals may acquire toxicity by absorbing toxic compounds from their food, e.g. from plants or other animals. Sequestration and accumulation of toxins may provide protection from predators, which learn to avoid this prey because of unpleasant experiences such as bitter taste. This is a common phenomenon in marine as well as in terrestrial ecosystems. Moreover, toxins may enter food chains where they accumulate reaching high, often lethal concentrations. Palytoxin which had been primarily detected in marine zoanthids (Palythoa sp.), occurs also in a wide range of other animals, e.g. in sponges,

Polyketide synthesis genes associated with toxin production in two species of Gambierdiscus (Dinophyceae).

BACKGROUND: Marine microbial protists, in particular, dinoflagellates, produce polyketide toxins with ecosystem-wide and human health impacts. Species of Gambierdiscus produce the polyether ladder compounds ciguatoxins and maitotoxins, which can lead to ciguatera fish poisoning, a serious human illness associated with reef fish consumption. Genes associated with the biosynthesis of polyether ladder compounds are yet to be elucidated, however, stable isotope feeding studies of such compounds consistently support their polyketide origin indicating that polyketide synthases are involved in their

The interaction of spongistatin 1 with tubulin.

A tritiated derivative of the sponge-derived natural product spongistatin 1 was prepared, and its interactions with tubulin were examined. [3H]Spongistatin 1 was found to bind rapidly to tubulin at a single site (the low specific activity of the [3H]spongistatin 1, 0.75 Ci/mmol, prevented our defining an association rate), and the inability of spongistatin 1 to cause an aberrant assembly reaction was confirmed. Spongistatin 1 bound to tubulin very tightly, and we could detect no significant dissociation reaction from tubulin. The tubulin-[3H]spongistatin 1 complex did dissociate in 8 M urea, s

Cultures of Dinophysis sacculus, D. acuminata and pectenotoxin 2 affect gametes and fertilization success of the Pacific oyster, Crassostrea gigas.

Harmful algal blooms (HABs) of toxic species of the dinoflagellate genus Dinophysis are a threat to human health as they are mainly responsible for diarrheic shellfish poisoning (DSP) in the consumers of contaminated shellfish. Such contamination leads to shellfish farm closures causing major economic and social issues. The direct effects of numerous HAB species have been demonstrated on adult bivalves, whereas the effects on critical early life stages remain relatively unexplored. The present study aimed to determine the in vitro effects of either cultivated strains of D. sacculus and D. acum

Characterisation of Two Toxic Gambierdiscus spp. (Gonyaulacales, Dinophyceae) from the Great Barrier Reef (Australia): G. lewisii sp. nov. and G. holmesii sp. nov.

Ciguatera fish poisoning (CFP) is a human illness caused via consumption of seafood contaminated with neurotoxins produced by some species from the epiphytic dinoflagellate genus Gambierdiscus. In this study, we describe two new species of Gambierdiscus isolated from Heron Island in the Southern Great Barrier Reef, Queensland, Australia. These new species were analysed using light microscopy, scanning electron microscopy, and phylogenetic analyses of nuclear encoded ribosomal ITS, SSU as well as D1-D3 and D8-D10 of the LSU gene regions. Development of an LC-MS/MS method to simultaneously monitor maitotoxins and selected ciguatoxins in algal cultures and P-CTX-1B in fish.

Ciguatera fish poisoning is a serious human health issue that is highly localized to tropical and sub-tropical coastal areas, affecting many of the indigenous island communities intrinsically linked to reef systems for sustenance and trade. It is caused by the consumption of reef fish contaminated with ciguatoxins and is reported as the most common cause of non-bacterial food poisoning. The causative toxins bioaccumulate up the food web, from small herbivorous fish that graze on microalgae of the genus Gambierdiscus into the higher trophic level omnivorous and carnivorous fish predating on the

The effect of brevenal on brevetoxin-induced DNA damage in human lymphocytes.

Brevenal is a nontoxic short-chain trans-syn polyether that competes with brevetoxin (PbTx) for the active site on voltage-sensitive sodium channels. The PbTxs are highly potent polyether toxins produced during blooms of several species of marine dinoflagellates, most notably Karenia brevis. Blooms of K. brevis have been associated with massive fish kills, marine mammal poisoning, and are potentially responsible for adverse human health effects such as respiratory irritation and airway constriction in beach-goers. Additionally, the consumption of shellfish contaminated with PbTxs results in ne

Brevetoxicosis in Florida manatees (Trichechus manatus latirostris): Plasma proteomics for Florida red tide biomarkers.

Florida manatees face recurring exposure to brevetoxins, a suite of neurotoxins produced by the dinoflagellate Karenia brevis, through inhalation and forage. Near annual blooms in the Gulf of Mexico lead to repeated epizootics and mass strandings. Although stranded manatees are often rescued and receive palliative care, plasma biomarkers for health surveillance and diagnostic decision making remain limited. To investigate mechanisms and identify biomarkers, we applied TMT plasma proteomics comparing exposed and unexposed manatees. The relative abundances of multiple manatee plasma proteins, es
