Polyhydroxyalkanoates (PHA) are biopolymers with potential to replace conventional oil-based plastics. However, PHA high production costs limit their scope of commercial applications. Downstream processing is currently the major cost factor for PHA production but one of the least investigated aspects of the PHA production chain. In this study, the extraction of poly(3-hydroxybutyrate-co-3-hydroxyvalerate) produced at pilot scale by a mixed microbial culture was performed using sodium hydroxide (NaOH) or sodium hypochlorite (NaClO) as digestion agents of non-PHA cellular mass. Optimal condition

Poly(4-hydroxybutyrate) (P4HB) is a high-performance, well-recyclable, and biodegradable polyhydroxyalkanoate (PHA). However, conventional bioproduction of homopolymeric P4HB involves complex and costly processes with C4 feedstocks, particularly 1,4-butanediol (BDO), and enzyme-coenzyme systems in genetically engineered bacteria. An alternative extracellular chemical route utilizing aerial oxidation of BDO offers cost and energy benefits but struggle with conversion efficiency. Inspired by efficient intracellular oxidation of primary alcohols, we propose a ruthenium-phosphine synergistic catal

Polyhydroxyalkanoates (PHAs) are gaining more and more importance the world over due to their structural diversity and close analogy to plastics. Their biodegradability makes them extremely desirable substitutes for synthetic plastics. PHAs are produced in organisms under certain stress conditions. Here, we investigated 253 sequenced (completely and unfinished) genomes for the diversity and phylogenetics of the PHA biosynthesis. Discrepancies in the phylogenetic trees for phaA, phaB and phaC genes of the PHA biosynthesis have led to the suggestion that horizontal gene transfer (HGT) may be a m

This study investigated polyhydroxyalkanoates (PHA) production from carboxylic acids obtained by yeast-assisted cheese whey (CW) fermentation. A three-stage hybrid strategy was employed. First, a lactose-fermenting yeast (Kluyveromyces marxianus) was inoculated into raw or sterilized CW to produce ethanol and/or lactic acid. Second, these intermediates were converted to carboxylic acids, with an emphasis on caproic acid, using a chain-elongating microbial culture. Batch and fermenter experiments under different operating conditions produced four distinct fermentation products with total carbox

Palm oil is an attractive feedstock for bioproduction due to its high carbon content and low cost. However, its metabolism generates excess reducing power, leading to redox imbalances and reduced metabolic efficiency in industrial fermentations. Through a model-driven approach integrating flux balance analysis, we activated the Calvin-Benson-Bassham (CBB) cycle in Cupriavidus necator to recycle surplus reducing power and restore metabolic balance in polyhydroxyalkanoate (PHA) bioproduction. Computational simulations predicted that constitutive activation of the CBB cycle enhanced CO2 fixation

Benzene, toluene, ethylbenzene, xylene and styrene (BTEXS) are priority gaseous pollutants due to their widespread release and health risks. This study demonstrates an efficient BTEXS bioconversion process into polyhydroxyalkanoates (PHA) using a specialized mixed microbial culture dominated by Pseudonocardia and Rhodococcus. The consortium achieved simultaneous degradation rates of 15.1 ± 3.9 g m-3h-1 for toluene and 17.6 ± 5.7 g m-3h-1 for ethylbenzene, with removal efficiencies over 90 %. The operating strategy promoted PHA accumulation up to 21.4 % gPHA gDCW-1. A two-step process was succe

Polyhydroxyalkanoates (PHAs) are synthesized by microorganisms as cytoplasmic biopolymers in response to nutritional starvation. These biopolymers have diverse applications because of their non-toxic and biodegradable nature and can be an effective alternative to conventional petrochemical polymers as they offer similar qualities. For this purpose, PHA-producing bacterium LO1 was isolated from lubricating oil contaminated soil and identified as Bacillus subtilis (MK071733). Furthermore, LO1 was found to be the most prominent PHA accumulating strain on groundnut oil as carbon source under optim

Growing environmental concerns about conventional plastics have created a demand for alternative resources that are both truly biodegradable and cost-effective. Microbial Polyhydroxyalkanoates (PHAs) are drawing great attention due to their degradability and lower environmental impact on ecosystems. However, higher production cost of bacteria-derived PHAs has become a bottleneck for commercial applications, prompting the search for alternative microbial hosts. Among various microbes, yeasts have become an alternative host for PHAs production due to their stress tolerance, generally regarded as

Polyhydroxyalkanoates (PHAs) are intracellular microbial polyesters whose commercial deployment is strongly influenced by downstream processing costs and environmental burden. Here, we demonstrate a nearly solvent-free, mild aqueous strategy for the recovery of PHAs directly from wet bacterial biomass using crude enzyme systems derived from Tenebrio molitor. Under optimized conditions (0.6 wt% crude protein, pH 7.6, 40°C), near-quantitative (≥ 95%) recovery of PHB and PHB/HV and up to 60% recovery of mcl-PHA were achieved without prior biomass drying. Proteomic analysis identified abundant dig

Polyhydroxyalkanoates (PHAs) are a structurally diverse family of microbially synthesised, biodegradable polyesters that accumulate as intracellular carbon and energy reserves under conditions of nutrient imbalance. Their combination of adjustable mechanical performance with controlled hydrolytic and enzymatic degradation, and non-toxic degradation intermediates (ex. D-3-hydroxybutyrate) has made them a longstanding candidate biomaterial for human tissue engineering, drug delivery, and resorbable implants. Comparatively, their application in veterinary medicine remains an emerging and fragment

The persistence of fossil fuel-based plastics poses significant environmental challenges, prompting increased research into biodegradable polyhydroxyalkanoate (PHA) polymers derived from cost-effective and sustainable resources. Different microorganisms can produce PHA amongst carbon dioxide (CO2)-assimilating autotrophic organisms, particularly noteworthy in carbon capture and utilization (CCU). Autotrophic bacteria have evolved to utilize either light (photoautotrophy) or inorganic chemicals (chemolithoautotrophy) to capture CO2, which powers their primary and secondary metabolic activities.

INTRODUCTION: Among urologic diseases, ureteropelvic segment stenosis with hydronephrosis is a common indication for instrumental or surgical correction. The restriction of urine flow with dilatation proximal to obstruction develops in 6.5-37% of cases at different times after the ureteral reconstruction. All this urges to develop and improve stents and search for effective ways to place stents and control their function.
AIM: To investigate the effectiveness of polyhydroxyalkanoates based biodegradable stent compared with a commercial analogue in upper urinary tract drainage after ureteropel

BACKGROUND: Cyanide is one of the most toxic chemicals produced by anthropogenic activities like mining and jewelry industries, which generate wastewater residues with high concentrations of this compound. Pseudomonas pseudoalcaligenes CECT5344 is a model microorganism to be used in detoxification of industrial wastewaters containing not only free cyanide (CN(-)) but also cyano-derivatives, such as cyanate, nitriles and metal-cyanide complexes. Previous in silico analyses suggested the existence of genes putatively involved in metabolism of short chain length (scl-) and medium chain length (mc

Nitrous oxide (N2O) emission has been reported to be enhanced during denitrification when internally-stored compounds are used as carbon sources. However, negligible N2O emissions have been detected in the few studies where polyhydroxyalkanoates (PHA) were specifically used. This study investigated and compared the potential enhancement of N2O production, based on utilization of an internally-stored polymer and external carbon (acetate) by a denitrifying phosphorus removal culture. Results indicated that at relatively low chemical oxygen demand-to-nitrogen (COD/N) ratios, more nitrite was redu

We developed a new method for isolation and quantitation of polyhydroxyalkanoate (PHA) from culture broth. In this method, the cells were sonicated in sodium dodecyl sulfate (SDS) solution and centrifuged to recover PHA. The recovered PHA was rinsed with deionized water and ethanol, and then weighed after drying. Hazardous chemicals such as chloroform, methanol, and sulfuric acid were not used, and no expensive analytical instruments were needed. We applied this method to Cupriavidus necator culture broths that included various amounts of poly(3-hydroxybutyrate) (PHB) or poly(3-hydroxybutyrate

Polyhydroxyalkanoate (PHA) granule associated proteins from Pseudomonas oleovorans were purified and the N-terminal sequences of two major proteins migrating in sodium dodecyl sulfate polyacrylamide gels with a relative molecular mass of 18 and 43 kDa (GA1 and GA2, respectively) were analyzed. Radiolabeled degenerate probes deduced from these amino acid sequences were used to identify genomic DNA fragments from P. oleovorans and Pseudomonas putida encoding GA1 and GA2. DNA sequence analysis of the fragments obtained from P. putida revealed that the genes encoding these proteins were adjacent t

Ethanol has emerged as an attractive and sustainable feedstock for biomanufacturing because of its high atom economy and direct conversion to acetyl-CoA. However, its use for polyhydroxyalkanoate (PHA) production remains limited by the scarcity of microbial chassis. Here, we report a newly isolated wild-type strain, Cupriavidus sp. ZWJ01, obtained through ethanol-enrichment screening of soil samples collected from Pu'er City, Yunnan, China. This strain exhibits an exceptional innate capacity for ethanol-based PHA biosynthesis. Fermentation profiling under diverse carbon regimes revealed robust
