The effect of charge on cellular uptake and phototoxicity of polylysine chlorin(e6) conjugates.

The effect of charge on the cellular uptake, localization and phototoxicity of conjugates between chlorin(e6) (ce6) and poly-L-lysine was studied in vitro. These conjugates (average MW 35-55 kDa) were synthesized to have polycationic, polyanionic or neutral charges. Two human cell lines (A431 epidermoid carcinoma cells and EA.hy926 hybrid endothelial cells) were studied and the cellular uptake of ce6 delivered by the conjugates of varying charge and free ce6 was measured at conjugate ce6 equivalent concentrations up to 0.4 microM. Uptake was time and concentration dependent and temperature dep

Rapid Broad-Spectrum Colorimetric Detection of Bacteria Using Polylysine-Mediated Gold Nanoparticle Aggregation.

Rapid on-site detection of bacterial contamination remains an urgent need in public health and environmental monitoring. Herein, we report a one-step, label-free colorimetric sensor for broad-spectrum bacterial detection based on a competitive electrostatic mechanism. Cationic poly-l-lysine (PLL) induces the aggregation of anionic, citrate-capped gold nanoparticles (AuNPs), resulting in a distinct red-to-blue color change. In the presence of bacteria, their negatively charged surfaces adsorb PLL, thereby inhibiting AuNP aggregation and maintaining the red color of the dispersion. This strategy

Hyperbranched Polylysine-Based Lipopolyplexes Enable Efficient mRNA Delivery With Long-Term Storage Stability.

Clinical translation of messenger RNA (mRNA) therapeutics is often hampered by the poor storage stability of lipid-based delivery vectors. Many lipopolyplex (LPP) delivery systems struggle to achieve efficient nucleic acid release through the membrane fusion pathway. Herein, we developed a novel ternary lipopolyplex (LPP) platform of lipid/HBPL/mRNA LPPs by incorporating hyperbranched poly-L-lysine (HBPL) as a functional polymeric core. The highly branched architecture of HBPL enabled a unique balance between high mRNA encapsulation efficiency, efficient intracellular release, and exceptional

Polylysine enhances cationic liposome-mediated transfection of the hepatoblastoma cell line Hep G2.

Plasmid DNA condensed by polylysine enhanced cationic-liposome-mediated transfection of Hep G2 cells. The luciferase expression plasmid pCMVL was complexed with the polycation poly-L-lysine and mixed with liposomes that contained a 1:1 molar ratio of the cationic lipid 1,2-dioleoyloxy-3-trimethyl-ammoniumpropane, with the neutral phospholipid 1,2-di-(cis-9-octadecenoyl)-sn-glycero-3-phosphoethanolamine. Polylysine enhanced cationic-liposome-mediated transfection of the hepatoblastoma cell line Hep G2 9-fold compared with pCMVL complexed alone with liposomes. The ratio of cationic to anionic ch

Dietary ε-Polylysine Affects on Gut Microbiota and Plasma Metabolites Profiling in Mice.

Given the antibacterial effects of ε-polylysine acting on cell membranes, and that glycerol phospholipids are important components of the cell membrane, we hypothesized that ε-polylysine may regulate glycerophospholipid metabolism by modifying the gut microbiota. To test this hypothesis, we treated post-weaning C57 mice with different levels of ε-polylysine (0, 300, 600, and 1,200 ppm) in their basic diet. The growth performance and morphology of intestine were then determined. Modification of the gut microbiota and their function were analyzed using 16S rDNA sequencing. Metabolite identificat

Polylysine dendrigraft is able to differentially impact Cutibacterium acnes strains preventing acneic skin.

With a view to reducing the impact of Cutibacterium acnes (C. acnes) on acne vulgaris, it now appears interesting to modify the balance between acneic and non-acneic strains of C. acnes using moderate approach. In the present study, we identified that a G2 dendrigraft of lysine dendrimer (G2 dendrimer) was able to modify membrane fluidity and biofilm formation of a C. acnes acneic strain (RT5), whereas it appeared no or less active on a C. acnes non-acneic strain (RT6). Moreover, skin ex vivo data indicated that the G2 is able to decrease inflammation (IL1α and TLR-2) and improve skin desquama

Polylysine-based copolymer self-assemblies featuring acidity-activated structural transformation perceives and relieves sepsis.

Sepsis, a systemic inflammatory response syndrome, causes severe immune dysfunction and is associated with high mortality because of the lack of effective clinical interventions. To address the pathogenesis of sepsis, such as bacterial infection and the exacerbation of inflammation and oxidative stress, an acidity-activated polylysine (PLL)-based copolymer self-assembly (PPDD) was developed. This material was synthesized by conjugating polyethylene glycol-modified PLL (PEG-PLL) with 2,7-dichlorofluorescein diacetate (DCFH-DA). PPDD, with its PLL-derived antibacterial and antioxidant properties

A polyphenol and ε-polylysine functionalized bacterial cellulose/PVA multifunctional hydrogel for wound healing.

Hydrogels for wound dressings have recently attracted considerable attention in the field of biomedical materials. Developing hydrogel dressings with multiple functions, including good antibacterial, mechanical and adhesive properties, to enhance wound regeneration is significant for clinical applications. To this end, a novel hydrogel wound dressing (PB-EPL/TA@BC) was developed, which was prepared by incorporating bacterial cellulose (BC) modified with tannic acid and ε-polylysine (EPL) into a PVA and borax matrix through a simple method without introducing any other chemical reagents. Receptor-mediated gene transfer to cells of hepatic origin by galactosylated albumin-polylysine complexes.

To study whether we could enhance the liver targeting of DNA delivery via asialoglycoprotein receptors using a complex of poly-L-lysine (PLL)-condensed DNA and galactosylated bovine serum albumin (GalBSA) (GPD complex), DNA was first combined with PLL and then with GalBSA via charge interaction (GalBSA: PLL: DNA=3:0.5:1, w/w/w). This vector was characterized by dynamic laser light scattering, gel retardation assay, and electron microscopy to determine the particle size, electrostatic charge interaction, and 3-D structure. The inhibition of acetylcholinesterase by a brain-targeting polylysine-ApoE peptide: biochemical and structural characterizations.

The in-trans delivery of protein therapeutics across the blood-brain barrier by K16ApoE peptide carrier has been demonstrated to improve the neurological symptoms and increase the life-span of late-infantile neuronal ceroid lipofuscinosis (LINCL) mice. However, acute toxicity of K16ApoE was observed in LINCL mice resulting in a narrow therapeutic index, limiting the potential of translating the K16ApoE into a viable drug delivery system. This study aims to unravel the toxic mechanism of action. We hypothesized that the toxic response towards the peptide was induced by inhibition of acetylcholi

Biotinylated ε-Polylysine-Cyclodextrin-Coated Mesoporous Silica Nanoparticles for Targeted pH-Responsive Baicalin Delivery.

INTRODUCTION: Baicalin (BAI) is a natural flavonoid with antitumor potential, but its poor water solubility and low bioavailability limit clinical use. Mesoporous silica nanoparticles are promising drug carriers due to their large surface area and tunable pore structure, yet premature drug leakage remains a key challenge. METHODS: A multifunctional nanoplatform, designated as BPCD@BAI@BMSN, was constructed to enable efficient baicalin loading, pH-responsive gated release, and biotin-mediated active tumor targeting. A novel biotin-ε-polylysine-cyclodextrin (BPCD) conjugate was synthesized via

Enhancement by polylysine of transient, but not stable, expression of genes carried into cells by polyoma VP1 pseudocapsids.

Gene transfer to provide long-term expression of a therapeutic product, without introducing unwelcome genetic information, is a goal being sought for therapy of both hereditary and acquired diseases. Polyoma virus pseudocapsids, generated from a VP1-expressing recombinant baculovirus, lack viral DNA and have been successfully used to introduce small exogenous genes stably into cells in vitro by a process designated 'pseudofection'; although pseudocapsids protect only about 3 kbp of exogenous DNA, low efficiency transfer of a larger fragment (6.2 kbp) has been observed. Here, expression of a 7.

Alginate-polylysine-alginate (APA) microencapsulated transgenic human amniotic epithelial cells ameliorate fibrosis in hypertrophic scars.

BACKGROUND: Hypertrophic scar (HS) is a severe skin fibrosis. Transplanting stem cells carrying anti-fibrotic cytokine genes, like interferon-gamma (IFN-γ), is a novel therapeutic strategy. Human amniotic epithelial cells (hAECs) are ideal seed cells and gene vectors. Microencapsulation creates a favorable environment for transplanted cells. This study investigates the effect of alginate-polylysine-alginate (APA)-microencapsulated hAECs modified with IFN-γ on HS fibrosis. MATERIALS AND METHODS: hAECs were isolated from human placentas and characterized. The full-length IFN-γ gene was cloned i

Injectable polyethylene glycol/methacrylated polylysine double cross-linked hydrogel releases neuropeptides for infected wound healing.

Wound infections caused by microorganisms often give rise to extensive inflammation and vascular damage that compromise the wound healing process. Designing approaches to more effectively controlling wound infections and accelerating this healing process are urgently needed. This study was designed with the goal of synthesizing an injectable, double cross-linked hydrogel suitable for use when treating infected wounds. After initially synthesizing methacrylated polylysine (PLMA) through polylysine grafting with methacrylic anhydride, CGRP and PLMA were incorporated into a PEG hydrogel network t

Dietary Supplementation of ε-Polylysine Beneficially Affects Ileal Microbiota Structure and Function in Ningxiang Pigs.

Intestinal microbiota plays an important role in the health of animals. However, little is known about the gut microbiota in Ningxiang pigs. Thus, we investigated how dietary supplementation with different ε-polylysine concentrations (0, 20, 40, 80, and 160 ppm) affected the ileal microbiota in Ningxiang pigs using a replicated 5 × 5 Latin square method. Each experimental period included 10 days for diet adaptation, 3 days for feces collection and 2 days for digesta collection. The ileal contents were collected and used for sequencing of the V3-V4 hypervariable region of the 16S rRNA gene. The

Polylysine-mediated translocation of the diphtheria toxin catalytic domain through the anthrax protective antigen pore.

The protective antigen (PA) moiety of anthrax toxin forms oligomeric pores in the endosomal membrane, which translocate the effector proteins of the toxin to the cytosol. Effector proteins bind to oligomeric PA via their respective N-terminal domains and undergo N- to C-terminal translocation through the pore. Earlier we reported that a tract of basic amino acids fused to the N-terminus of an unrelated effector protein (the catalytic domain diphtheria toxin, DTA) potentiated that protein to undergo weak PA-dependent translocation. In this study, we varied the location of the tract (N-terminal
