A 1.5 kb repeat sequence flanks the suppressor of forked gene at the euchromatin-heterochromatin boundary of the Drosophila melanogaster X chromosome.

A 1.5 kilobasepair repeated DNA sequence is duplicated in direct orientation so as to flank the suppressor of forked gene in the euchromatin-heterochromatin transition region on the X chromosome of Drosophila melanogaster. These two copies are almost identical, but DNA blotting, analysis of cloned sequences and database searches show that elsewhere in the genome, homologous sequences are poorly conserved. They are often associated with other repeats, suggesting that they may belong to a scrambled and clustered middle repetitive DNA family. The sequences do not appear to be related to transposa

[Triploidy in populations of three species of black flies of the genus Prosimulium roubaud (Diptera, Simuliidae)].

Karyotypes of 94 larvae belonging to species of black flies of the genus Prosimulium Rouband were studied on aceto-orcein stained squashes of 94 larvae belonging to three species of Prosimulium: P. isos Rubzov, P. pecticrassum Rubzov, and P. pamiricum Chubareva et Petrova. All the larvae appeared to be triploid parthenogenetic females, with no males being found. The structural peculiarities of these polytene chromosomes suggest a heterozygous nature of the examined larvae.

The distribution and chromosome polymorphism of Simulium dieguerense (Diptera: Simuliidae).

Simulium dieguerense has been identified from Senegal, Mali and Guinea, in the Western Zone of the World Health Organization/Onchocerciasis Control Programme thus considerably extending its known distribution and indicating the need to reassess its possible importance as a vector of human onchocerciasis. A distribution map based on these identifications has been drawn. Examination of the larval polytene chromosomes revealed the presence of a hitherto unreported sex chromosome system. About 90% of males and 17% of females were found to be heterozygous for an altered segment of the centromere re

Development of a method for the cytological identification of man-biting sibling species within the Simulium damnosum complex.

A method is described for the preparation of adult Malpighian tubule polytene chromosomes from members of the Simulium damnosum complex. This method was used to show that the banding pattern is conserved between adult and larval tissues, and consequently it has been possible to demonstrate that within the S. sanctipauli subcomplex both S. soubrense 'B' and S. sanctipauli bite humans. The degree tubule development is correlated with the gonotrophic cycle and appears to be dependent on food source. This suggests that there is a physiological stage which is optimum for obtaining maximum polyteny

Heterochromatin and sibling species of Simulium praelargum s.l. (Diptera: Simuliidae).

Cytological descriptions are given for two sympatric sibling species, Simulium praelargum IIIL-st and Simulium praelargum IIIL-1.2, from Darjeeling, West Bengal, India. Sibling IIIL-1.2 differs from IIIL-st by a two step fixed included inversion (IIIL-1.2) found on the long arm of chromosome III. Both siblings possess heterochromatinized IIIS polytene chromosome ends, while IIIL-1.2 is unique within the Simuliidae in that it exhibits a very large enhanced chromocentre that persists in normal somatic tissue, contrary to other chromocentre-containing taxa. In IIIL-1.2, the chromocentre occurs as

Coordination of larval and prepupal gene expression by the DHR3 orphan receptor during Drosophila metamorphosis.

The DHR3 orphan receptor gene is induced directly by the steroid hormone ecdysone at the onset of Drosophila metamorphosis. DHR3 expression peaks in early prepupae, as the early puff genes are repressed and betaFTZ-F1 is induced. Here we provide evidence that DHR3 directly contributes to both of these regulatory responses. DHR3 protein is bound to many ecdysone-induced puffs in the polytene chromosomes, including the early puffs that encode the BR-C and E74 regulatory genes, as well as the E75, E78 and betaFTZ-F1 orphan receptor loci. Three DHR3 binding sites were identified downstream from th

Chromosomal evidence for sibling species of the malaria vector Anopheles (Cellia) culicifacies Giles.

The ovarian polytene chromosomes of adult Anopheles culicifacies from natural populations in Pakistan, India and Sri Lanka, and from four laboratory colonies (one from Pakistan and three from India) are compared with the published cytological map for this taxon. Two types of X-chromosome exist. One of these is identical with the published map; the other differs by two inversions. No autosome arm rearrangements are detected. A photomap is presented which documents the X-chromosome inversions. Both types of Xs are found in a natural population near New Delhi in the apparent absence of heterozygo

Comparative physical genome mapping of malaria vectors Anopheles sinensis and Anopheles gambiae.

BACKGROUND: Anopheles sinensis is a dominant natural vector of Plasmodium vivax in China, Taiwan, Japan, and Korea. Recent genome sequencing of An. sinensis provides important insights into the genomic basis of vectorial capacity. However, the lack of a physical genome map with chromosome assignment and orientation of sequencing scaffolds hinders comparative analyses with other genomes to infer evolutionary changes relevant to the vector capacity. RESULTS: Here, a physical genome map for An. sinensis was constructed by assigning 52 scaffolds onto the chromosomes using fluorescence in situ hyb

Cytotaxonomy of Sirba form populations of the Simulium damnosum complex in west Africa: amendments to sex chromosomes and sibling status.

The banding pattern of larval salivary polytene chromosomes was analysed to re-examine the distribution, by sex and geography, of the sex linked inversion (IS-3) in "Sirba" form which was previously described as consisting of Simulium sirbanum and S. sudanense (Vajime and Dunbar, 1975). West African larval samples from seven countries within the Onchocerciasis Control Programme area and some adjacent locations were used. It was concluded that "Sirba" consists of two indisputable cytoforms, one distinguished by X0Y0 males and X0X0 females, the other by X1Y0 males and X1X1 females; and that the

Field studies on inversion polymorphism in Anopheles superpictus from southern Italy.

Polytene chromosome studies were carried out on various population samples of Anopheles superpictus from different localities in Southern Italy. More than 7,000 female specimens, mostly obtained from daytime collections of indoor resting mosquitoes, were successfully scored for nurse cell polytene chromosomes. Night biting samples were also examined in some localities. Only one chromosomal polymorphism, due to a paracentric inversion involving the central third of the 2L arm, was recorded in all samples. In all localities, the inverted 2La arrangement showed remarkably stable frequency althoug

An integrated linkage, chromosome, and genome map for the yellow fever mosquito Aedes aegypti.

BACKGROUND: Aedes aegypti, the yellow fever mosquito, is an efficient vector of arboviruses and a convenient model system for laboratory research. Extensive linkage mapping of morphological and molecular markers localized a number of quantitative trait loci (QTLs) related to the mosquito's ability to transmit various pathogens. However, linking the QTLs to Ae. aegypti chromosomes and genomic sequences has been challenging because of the poor quality of polytene chromosomes and the highly fragmented genome assembly for this species. METHODOLOGY/PRINCIPAL FINDINGS: Based on the approach develop

Tissue-specific differences in the spatial interposition of X-chromosome and 3R chromosome regions in the malaria mosquito Anopheles messeae Fall.

Spatial organization of a chromosome in a nucleus is very important in biology but many aspects of it are still generally unresolved. We focused on tissue-specific features of chromosome architecture in closely related malaria mosquitoes, which have essential inter-specific differences in polytene chromosome attachments in nurse cells. We showed that the region responsible for X-chromosome attachment interacts with nuclear lamina stronger in nurse cells, then in salivary glands cells in Anopheles messeae Fall. The inter-tissue differences were demonstrated more convincingly in an experiment of

Determination of Complex Formation between Drosophila Nrf2 and GATA4 Factors at Selective Chromatin Loci Demonstrates Transcription Coactivation.

Nrf2 is the dominant cellular stress response factor that protects cells through transcriptional responses to xenobiotic and oxidative stimuli. Nrf2 malfunction is highly correlated with many human diseases, but the underlying molecular mechanisms remain to be fully uncovered. GATA4 is a conserved GATA family transcription factor that is essential for cardiac and dorsal epidermal development. Here, we describe a novel interaction between Drosophila Nrf2 and GATA4 proteins, i.e., cap'n'collar C (CncC) and Pannier (Pnr), respectively. Using the bimolecular fluorescence complementation (BiFC) ass

Tissue-specific features of the X chromosome and nucleolus spatial dynamics in a malaria mosquito, Anopheles atroparvus.

Spatial organization of chromosome territories is important for maintenance of genomic stability and regulation of gene expression. Recent studies have shown tissue-specific features of chromosome attachments to the nuclear envelope in various organisms including malaria mosquitoes. However, other spatial characteristics of nucleus organization, like volume and shape of chromosome territories, have not been studied in Anopheles. We conducted a thorough analysis of tissue-specific features of the X chromosome and nucleolus volume and shape in follicular epithelium and nurse cells of the Anophel

Drosophila is an inclusive model for human diseases, growth and development.

Cytogenetic studies over the last century have led to the complete mapping of the Drosophila polytene chromosomes. The resulting data and the analysis of puffing at specific gene sites, manifestations of enhanced transcriptional activity, have led to the use of the fruit fly as the most well-understood animal model for a plethora of cellular mechanisms and genetic defects. In recent years the fly data base has contributed greatly to the use of Drosophila as a remarkable model for the functional genomics of many human genes. Here I review briefly the diversity of "model genes" studied in this d

Interspecific transgenic analysis of basal versus heat-shock-induced expression of a Drosophila pseudoobscura hsp82-neo fusion gene in D. melanogaster.

Drosophila melanogaster transformants containing a D. pseudoobscura hsp82-neo fusion gene were used to examine the relationship between chromosome structure and its variation to transcriptional activation and gene expression. At normal temperatures (25° C) transgenic hsp82-neo was transcribed in diffuse polytene chromosomal bands encoding antibiotic G418-resistance without intensive puff formation. Substantial basal expression of the transgene was observed in all tissues examined: salivary glands, brain, ventral ganglion, foregut, gastric caeca, midgut, imaginal discs, nurse cells and oocytes.

Structural divergence of chromosomes between malaria vectors Anopheles lesteri and Anopheles sinensis.

BACKGROUND: Anopheles lesteri and Anopheles sinensis are two major malaria vectors in China and Southeast Asia. They are dramatically different in terms of geographical distribution, host preference, resting habitats, and other traits associated with ecological adaptation and malaria transmission. Both species belong to the Anopheles hyrcanus group, but the extent of genetic differences between them is not well understood. To provide an effective way to differentiate between species and to find useful markers for population genetics studies, we performed a comparative cytogenetic analysis of t

Identification of the Drosophila interband-specific protein Z4 as a DNA-binding zinc-finger protein determining chromosomal structure.

The subdivision of polytene chromosomes into bands and interbands suggests a structural chromatin organization that is related to the formation of functional domains of gene expression. We made use of the antibody Z4 to gain insight into this level of chromosomal structure, as the Z4 antibody mirrors this patterning by binding to an antigen that is present in most interbands. The Z4 gene encodes a protein with seven zinc fingers, it is essential for fly development and acts in a dose-dependent manner on the development of several tissues. Z4 mutants have a dose-sensitive effect on w(m4) positi

A physically anchored genetic map and linkage to avirulence reveals recombination suppression over the proximal region of Hessian fly chromosome A2.

Resistance in wheat (Triticum aestivum) to the Hessian fly (Mayetiola destructor), a major insect pest of wheat, is based on a gene-for-gene interaction. Close linkage (3 +/- 2 cM) was discovered between Hessian fly avirulence genes vH3 and vH5. Bulked segregant analysis revealed two DNA markers (28-178 and 23-201) within 10 cM of these loci and only 3 +/- 2 cM apart. However, 28-178 was located in the middle of the short arm of Hessian fly chromosome A2 whereas 23-201 was located in the middle of the long arm of chromosome A2, suggesting the presence of severe recombination suppression over i

Progress in mapping the yellow fever mosquito genome.

Mosquito-borne diseases cause significant problems for the human health. For this reason, the genomes of three most dangerous species of mosquitoes, including the yellow fever mosquito Aedes aegypti, were sequenced in last decade. The efficient vector of arboviruses.
