Demographic Bottlenecks and Size-Dependent Competition Shape Populus euphratica Persistence.

Although Populus euphratica forests are key components of arid riparian ecosystems, the demographic and competitive processes associated with their population persistence remain insufficiently understood. We investigated population structure, demographic dynamics and intraspecific competition in a natural P. euphratica population within the Tarim River Basin, northwestern China. Field data from 2690 individuals were analyzed using diameter-class structure, population dynamic indices and the Hegyi competition index. The population exhibited an inverse J-shaped diameter distribution: Class I con

Isolated from Populus euphratica rhizosphere soil, and mining their metabolites.

INTRODUCTION: The microbial community in desert ecosystems is a vital and highly active component. Streptomyces, one of the dominant genera within this community, exhibits significant capabilities in metabolic degradation and the synthesis of secondary metabolites. METHODS: To investigate the metabolic potential of Streptomyces desertis, two strains of Streptomyces were isolated from rhizosphere soil samples of Populus euphratica in the Taklimakan Desert during the initial phase of this study, TRM 70351T and TRM 70361T. RESULTS: The identification of these strains as belonging to the genus S

Cd, Fe, and light sensitivity: interrelationships in Cd-treated populus.

Cadmium is a toxic heavy metal causing iron deficiency in the shoot and light sensitivity of photosynthetic tissues that leads to decreased photosynthetic performance and biomass production. Light intensity had strong impact on both photosynthetic activity and metal accumulation of cadmium-treated plants. At elevated irradiation, cadmium accumulation increased due to the higher dry mass of plants, but its allocation hardly changed. A considerable amount of iron accumulated in the roots, and iron concentration was higher in leaves developed at moderate rather than low irradiation. At the same t

Structural characterization of lignin from triploid of Populus tomentosa Carr.

To improve yields while minimizing the extent of mechanical action (just 2 h of planetary ball-milling), the residual wood meal obtained from extraction of milled wood lignin (MWL) was sequentially treated with cellulolytic enzyme and alkali, and the yields of MWL, cellulolytic enzyme lignin (CEL), and alkaline lignin (AL) were 5.4, 23.2, and 16.3%, respectively. The chemical structures of the lignin fractions obtained were characterized by carbohydrate analysis, gel permeation chromatography (GPC), Fourier transform infrared (FT-IR) spectroscopy, and various advanced NMR spectroscopic techniq

Biochemical properties of isoprene synthase in poplar (Populus x canescens).

Isoprene synthase (ISPS) catalyzes the elimination of pyrophosphate from dimethylallyl diphosphate (DMADP) forming isoprene, a volatile hydrocarbon emitted from many plant species to the atmosphere. In the present work, immunological techniques were applied to study and localize ISPS in poplar leaves (Populus x canescens). Immunogold labeling using polyclonal antibodies generated against His-tagged recombinant ISPS protein detected ca. 44% of ISPS in the stroma of the chloroplasts and ca. 56% of gold particles attached to the stromal-facing side of the thylakoid membranes. ISPS isolated from l

Populus euphratica PeIDD5 regulates PeNAC029 to suppress salt tolerance in poplars.

Populus euphratica downregulates the transcription factor PeNAC029 during prolonged salinity, thereby promoting long-term acclimation to salt stress. To elucidate this regulatory network, we identified PeIDD5, an INDETERMINATE DOMAIN (IDD) family member, as a direct transcriptional activator of PeNAC029. This study investigates the role of the PeIDD5-NAC029 transcriptional module in the salt tolerance of two contrasting poplars, P. euphratica (salt-tolerant) and P. × canescens (salt-sensitive). PeIDD5 directly binds the conserved cis-element TTTGTCC in the promoter regions of PeNAC029 and PcNA

Chromosome-level genome assembly of a triploid poplar Populus alba 'Berolinensis'.

Many recent studies have provided significant insights into polyploid breeding, but limited research has been carried out on trees. The genomic information needed to understand growth and response to abiotic stress in polyploidy trees is largely unknown, but has become critical due to the threats to forests imposed by climate change. Populus alba 'Berolinensis,' also known "Yinzhong poplar," is a triploid poplar from northeast China. This hybrid triploid poplar is widely used as a landscape ornamental and in urban forestry owing to its adaptation to adverse environments and faster growth than

Fibre-specific regulation of lignin biosynthesis improves biomass quality in Populus.

Lignin is a major component of cell wall biomass and decisively affects biomass utilisation. Engineering of lignin biosynthesis is extensively studied, while lignin modification often causes growth defects. We developed a strategy for cell-type-specific modification of lignin to achieve improvements in cell wall property without growth penalty. We targeted a lignin-related transcription factor, LTF1, for modification of lignin biosynthesis. LTF1 can be engineered to a nonphosphorylation form which is introduced into Populus under the control of either a vessel-specific or fibre-specific promot

Genetic architecture of the metabolic pathway of salicylic acid biosynthesis in Populus.

Salicylic acid (SA) is a vital hormone for adaptive responses to biotic and abiotic stresses, which facilitates growth-immunity trade-offs in plants. However, the genetic regulatory networks underlying the metabolic pathway of SA biosynthesis in perennial species remain unclear. Here, we integrated genome-wide association study (GWAS) with metabolite and expression profiling methodologies to dissect the genetic architecture of SA biosynthesis in Populus. First, we quantified nine intermediate metabolites of SA biosynthesis in 300 unrelated Populus tomentosa Carr. individuals. Then, we used a s

Inducing triploids and tetraploids with high temperatures in Populus sect. Tacamahaca.

This study is the first to report that triploids and tetraploids have been successfully produced through embryo sac and zygotic embryo chromosome doubling with high temperatures in P. simonii Carr. and its hybrid. A new synthetic polyploid induced by hybridization with unreduced gametes and heterozygotic embryo chromosome doubling can effectively combine polyploidy and heterosis, which can provide two major breeding advantages. In Populus, successfully creating and cultivating new polyploid varieties have economic and ecological production value. This was the first successful study in which em

Biosynthesis of phenolic glycosides from phenylpropanoid and benzenoid precursors in populus.

Salicylate-containing phenolic glycosides (PGs) are abundant and often play a dominant role in plant-herbivore interactions of Populus and Salix species (family Salicaceae), but the biosynthetic pathway to PGs remains unclear. Cinnamic acid (CA) is thought to be a precursor of the salicyl moiety of PGs. However, the origin of the 6-hydroxy-2-cyclohexen-on-oyl (HCH) moiety found in certain PGs, such as salicortin, is not known. HCH is of interest because it confers toxicity and antifeedant properties against herbivores. We incubated Populus nigra leaf tissue with stable isotope-labeled CA, benz

PcXTH22 as a core regulator enhancing aluminum toxicity tolerance in Populus cathayana.

Aluminum (Al) toxicity in acidic soils restricts plant growth. Xyloglucan endotransglucosylase/hydrolases (XTHs) modulate Al responses, but their mechanisms in poplar remain unknown. Here, we characterized PcXTH22-mediated Al tolerance in Populus cathayana. The PcXTH22OE lines showed negligible injury, whereas the pcxth22 mutants suffered severe damage, confirming the role of PcXTH22 as a positive Al regulator. The pcxth22 lines showed increased xyloglucan endotransglucosylase/hydrolase (XET/XTH) activities and xyloglucan content under Al toxicity. This offered more Al³ ⁺ binding sites and res

Molecular genetics of growth and development in Populus. I. Triploidy in hybrid poplars.

While constructing a genetic linkage map of a hybrid poplar genome (Populus trichocarpa x P. deltoides), we identified several restriction fragment length polymorphismus (RFLPs) for which the parental trees are heterozygous. Although 8 of the 11 F1 hybrid offspring inherited, as expected, single RFLP alleles from each parent, 3 F1 trees in the mapping pedigree inherited both maternal alleles along with a single paternal allele at some loci. Aneuploidy or polyploidy in these 3 F1 trees due to partial or complete nondisj unction during female gametogenesis is the simplest explanation for this fi

Lignin macromolecular structural stability drives sapwood-heartwood color homogeneity in Populus deltoides.

Radial xylem maturation in woody plants typically involves programmed cell death (PCD) and accumulation of colored secondary metabolites, leading to distinct color convergent between sapwood and heartwood. In fast-growing P. deltoides, however, the xylem exhibits remarkable color convergence along the radial gradient. Through the integration of anatomical observations, chemical analyses, and advanced spectroscopy (FTIR, Raman, 2D-HSQC NMR), color convergence was analyzed from two coordinated regulatory features: (1) a non-condensed lignin structure characterized with an elevated syringyl/guaia

Single Amino Acid Divergence Drives Functional Specialisation of Homologous GRFs in Populus.

Gene duplication with sub- and/or neo-functionalization is a key pathway in plant adaptive evolution. The duplication-degeneration-complementation (DDC) model explains sub-functionalization of paralogs, yet the relative contributions of coding sequence versus cis-regulatory changes remain contested. In this study, we show that a single amino acid substitution (Met30/Ile30) in the WRC domain drives sub-functionalization of poplar paralogs PagGRF29 and PagGRF10, demonstrating a key role for coding changes. Met30 confers high DNA-binding capacity and represents the ancestral state, whereas Ile30

Genome-wide identification, expression and functional analysis of Populus xylogen-like genes.

As an extracellular arabinogalactan protein (AGP) containing a non-specific lipid transfer protein (nsLTP) domain, xylogen mediates the local intercellular communication required for tracheary element (TE) differentiation in Zinnia cell culture. Although XYLP (xylogen-like protein) gene families have been reported in Arabidopsis and rice, no comprehensive analysis has been performed in woody plants. In this work, 31 XYLP genes in five phylogenetic groups were identified from Populus trichocarpa genome and a comprehensive bioinformatic analysis including gene and protein structures, chromosomal

PtoeIF5A1: A Pleiotropic Regulator of Development, PCD, and Salt Tolerance in Populus tomentosa.

Eukaryotic translation initiation factor 5A (eIF5A) is a highly conserved protein family unique to eukaryotes, yet its functional characterization in woody plants remains limited. In this study, we identified four eIF5A genes (PtoeIF5A1-PtoeIF5A4) from the genome of Populus tomentosa, a fast-growing tree species indigenous to China, and characterized their expression patterns and functional roles through bioinformatics analysis, quantitative real-time PCR, stable overexpression in Arabidopsis thaliana, and transient expression in Nicotiana benthamiana leaves.
