Antioxidant capacity, polyphenol content and iron bioavailability from algae (Ulva sp., Sargassum sp. and Porphyra sp.) in human subjects.

Marine algae are easily produced and are good sources of Fe. If this Fe is bioavailable, algae consumption could help to combat Fe deficiency and anaemia worldwide. The objective of the present study was to evaluate Fe bioavailability, polyphenol content and antioxidant capacity from three species of marine algae distributed worldwide. A total of eighty-three subjects received maize- or wheat-based meals containing marine algae (Ulva sp., Sargassum sp. and Porphyra sp.) in different proportions (2.5, 5.0 and 7.5 g) added to the water to prepare the dough. All meals administered contained radio

Sulfated polysaccharides from Laminaria japonica and Porphyra inhibit SARS-CoV-2 main protease: mechanistic insights into molecular interactions.

The replication of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) in host cells relies on its non-structural protein, the main protease (Mpro), a cysteine protease that serves as a primary target for antiviral drug design. Here, the inhibitory effects of sulfated polysaccharides from Laminaria japonica (IJSPS) and Porphyra (PSPS) on Mpro activity were investigated. IJSPS displayed a marked inhibitory effect on Mpro activity, whereas PSPS showed no effect. The half maximal inhibitory concentration (IC50) and inhibition constant (Ki) of the IJSPS for Mpro activity are 1.17 ± 0.34 n

Comparative functional evaluation of the atypically modified GlcHMS326 and porphyra-334, Two structurally distinct mycosporine-like amino acids.

Mycosporine-like amino acids (MAAs) are natural compounds widely studied for their photoprotective and antioxidant properties. Typical MAAs consist of one or two amino acids attached to a cyclohexenone or cyclohexenimine ring, whereas atypical MAAs possess unique chemical modifications such as glycosylation and methylation. Recently, we identified an atypical MAA, GlcHMS326, from the cyanobacterium Gloeocapsa sp. BRSZ, characterized by glycosylation, methylation, and hydroxylation. In this study, we compared the chemical stability and biological activities of GlcHMS326 with those of a typical

Integrative metabolomic profiling reveals comparable nutritional quality and diminished sensory qualities in Porphyra yezoensis from January versus April harvest.

Although early-harvested Porphyra yezoensis has greater economic value, its nutritional variation from late-harvests remains unclear. Non-targeted UPLC-MS/MS and targeted LC-MS/MS analyses was used to characterize metabolic and nutritional differences between early (January) and late (April) harvests (n = 6 per group). The late-harvests exhibited significant enrichment (P < 0.05) in amino acid, lipid, organic acid, and carbohydrate metabolic pathways, accompanied by higher levels of crude protein, free amino acids, lipids, polyunsaturated fatty acids, vitamins, and crude fiber. Peptide identification from a Porphyra dioica protein hydrolysate with antioxidant, angiotensin converting enzyme and dipeptidyl peptidase IV inhibitory activities.

A Porphyra dioica protein extract was enzymatically hydrolysed and then fractionated using semi-preparative reverse-phase high performance chromatography. The hydrolysate and its fractions were tested for their oxygen radical absorbance capacity (ORAC) along with their angiotensin converting enzyme (ACE) and dipeptidyl peptidase IV (DPP-IV) inhibitory activities. The most potent fraction was analysed by liquid chromatography mass spectrometry. Eight peptide sequences were selected for synthesis based on their structure-activity criteria for bioactivity. Asp-Tyr-Tyr-Lys-Arg showed the highest O

Biogeographic Population Structure of Chimeric Blades of Porphyra in the Northeast Atlantic Reveals Southern Rich Gene Pools, Introgression and Cryptic Plasticity.

The genus Porphyra sensu lato (Bangiaceae, Rhodophyta), an important seaweed grown in aquaculture, is the most genetically diverse group of the Class Bangiophyceae, but has poorly understood genetic variability linked to complex evolutionary processes. Genetic studies in the last decades have largely focused on resolving gene phylogenies; however, there is little information on historical population biogeography, structure and gene flow in the Bangiaceae, probably due to their cryptic nature, chimerism and polyploidy, which render analyses challenging. This study aims to understand biogeograph

In vitro-simulated intestinal flora fermentation of Porphyra haitanensis polysaccharides obtained by different assisted extractions and their fermented products against HT-29 human colon cancer cells.

Herein, we studied the in vitro-simulated intestinal flora fermentation of Porphyra haitanensis polysaccharides (PHPs) with microwave, ultrasonic, ultra-high pressure-assisted extraction and the protective effect of their fermented products against HT-29 human colon cancer cells. The results showed that PHPs were largely degraded at the 18 h stage of ascending colon fermentation, further greatly increasing the contents of reducing sugars and short-chain fatty acids (p < 0.05). Particularly, the PHPs subjected to ultra-high pressure-assisted extraction (UHP-PHP) showed the highest reducing suga

Region-specific carbon metabolism underlies floridean starch accumulation in rhizoid-like filament cells of Pyropia yezoensis.

BACKGROUND: Pyropia yezoensis thalli exhibit pronounced cellular differentiation throughout their growth and development. In particular, basal rhizoid-like filament cells (R-region cells) differ significantly from other thallus cells in cell morphology, photosynthetic physiology and carbon metabolism. Previous studies have shown that abundant floridean starch granules accumulate in R-region cells, whereas only few floridean starch granules are present in neighbouring vegetative cells (V-region cells). However, the specific mechanism underlying floridean starch accumulation in R-region cells re

In vitro protein digestibility assessment in edible seaweeds: Methodological approach and species comparison.

Seaweeds are promising sustainable alternative protein sources, however, their complex cell wall structures may limit protein bioaccessibility. This study evaluated the in vitro protein digestibility of three seaweeds, Ulva lacinulata (green), Porphyra dioica (red), and Saccharina latissima (brown), applying the INFOGEST protocol specifically adapted for seaweed matrices. The influence of preservation methods (freezing vs freeze-drying) was also assessed. Compositional and multi-scale structural characterization of the seaweeds before and after digestion was performed to understand their diges

Algal and Cyanobacterial Hydrolysates as Serum-Reducing Supplements Across Mammalian, Avian and Fish Cells with Identification of Bioactive Chromopeptides.

Maintaining sustainable production of healthy and nutritious food represents a major global challenge. Cultivated meat could reduce the environmental impact of livestock production, but it remains costly, largely due to the expense of fetal bovine serum, a common cell culture media supplement. Algal and cyanobacterial proteins, particularly phycobiliproteins, represent promising sustainable sources of potentially bioactive peptides that may contribute to the development of serum-reduced or serum-free culture media. This study investigated the potential of hydrolysates prepared using the protei

Anti-inflammation activities of mycosporine-like amino acids (MAAs) in response to UV radiation suggest potential anti-skin aging activity.

Certain photosynthetic marine organisms have evolved mechanisms to counteract UV-radiation by synthesizing UV-absorbing compounds, such as mycosporine-like amino acids (MAAs). In this study, MAAs were separated from the extracts of marine green alga Chlamydomonas hedleyi using HPLC and were identified as porphyra-334, shinorine, and mycosporine-glycine (mycosporine-Gly), based on their retention times and maximum absorption wavelengths. Furthermore, their structures were confirmed by triple quadrupole MS/MS. Their roles as UV-absorbing compounds were investigated in the human fibroblast cell l

Exudates of different marine algae promote growth and mediate trace metal binding in Phaeodactylum tricornutum.

Phaeodactylum tricornutum was grown in filtered natural seawater enriched with nitrate, phosphate, and silicate only (control) or with exudates from itself, from Emiliania huxleyi (a coccolithophore micro-alga), Porphyra spp. (a red macro-alga) or Enteromorpha spp. (a green macro-alga). Cathodic (and anodic) stripping voltammetry (C(A)SV) were used to determine the concentrations of trace metals, both in the medium and in the algae, as well as total Cu-complexing organic ligands in the medium and, among these, some thiols (compounds identified as cysteine- or as glutathione by CSV). Functional characterization and periplasmic localization of a phosphorus starvation-induced alkaline phosphatase involved in phosphate acquisition in Pyropia yezoensis.

Expression of the alkaline phosphatase gene in Pyropia yezoensis is transcriptionally induced under phosphorus deprivation. To elucidate its physiological role in phosphate acquisition, we purified the enzyme to homogeneity from phosphorus-starved thalli and characterized its biochemical properties. The alkaline phosphatase exhibited broad substrate specificity toward glycerophosphate, sugar phosphates, and nucleoside phosphates, and its activity was enhanced by Na+, Ca2+, and Mg2+ at concentrations comparable to those in natural seawater. Mechanism of seaweed polyphenols interacting with intestinal flora to regulate blood glucose.

Seaweed polyphenols possess hypoglycemic biological functions, but the specific mechanisms remain unclear, which limits their further application. In this study, the area under the blood glucose curve (AUC) of the fourth-period Porphyra haitanensis polyphenols extract (FPPE)-fed mice was significantly reduced by 17.12 ± 0.87%. Moreover, FPPE exhibited α-glucosidase inhibitory activity at 64.39 ± 4.05%. In vitro fermentation experiments showed that FPPE modulated the composition of the gut microbiota, thereby enhancing the production of short-chain fatty acids (SCFAs). Analysis of intestinal fl
