Investigating the interactions of endornaviruses with each other and with other viruses in common bean, Phaseolus vulgaris.

BACKGROUND: Plant viruses of the genus Alphaendornavirus are transmitted solely via seed and pollen and generally cause no apparent disease. It has been conjectured that certain plant endornaviruses may confer advantages on their hosts through improved performance (e.g., seed yield) or resilience to abiotic or biotic insult. We recently characterised nine common bean (Phaseolus vulgaris L.) varieties that harboured either Phaseolus vulgaris endornavirus (PvEV1) alone, or PvEV1 in combination with PvEV2 or PvEV1 in combination with PvEV2 and PvEV3. Here, we investigated the interactions of thes

Development of Comprehensive Serological Techniques for Sensitive, Quantitative and Rapid Detection of Soybean mosaic virus.

Soybean is an important grain and oil crop worldwide; however, the yield and seed quality of which are seriously affected by Soybean mosaic virus (SMV). As efficient detection technology is crucial for the field management of SMV, novel immunological detection methods were developed in the present study. According to the phylogenetic analysis, the CP coding sequence of SMV-SC7 was selected for the prokaryotic expression of the recombinant SMV-CP. Purified SMV-CP was used for the development of polyclonal antibodies (PAb) against the SMV-CP (PAb-SMV-CP) and monoclonal antibodies (MAb) against S

Occurrence of passion fruit woodiness disease in the coastal lowlands of Kenya and screening of passion fruit genotypes for resistance to passion fruit woodiness disease.

BACKGROUND: Passion fruit (Passiflora edulis [Sims]) is an important economic fruit crop in Kenya, grown for domestic, regional and international markets. However, passion fruit production is constrained by both biotic and abiotic stresses. Passion fruit woodiness disease (PWD) complex is the most injurious viral disease responsible for yield losses of up to 100%. In East Africa, it is caused by potyviruses. The most effective way to manage PWD is by using resistant cultivars. The objectives of this study were to determine the occurrence of passion fruit woodiness disease in selected counties

Triplex Immunostrip Assay for Rapid Diagnosis of Tobacco Mosaic Virus, Tobacco Vein Banding Mosaic Virus, and Potato Virus Y.

Mixed virus infection has increasingly become a problem in the production of Solanaceae crops in recent years; therefore, a fast and accurate detection method is needed. In this study, a novel triplex immunostrip assay was developed for the simultaneous detection of tobacco mosaic virus (TMV), tobacco vein banding mosaic virus (TVBMV), and potato virus Y (PVY). The limits of detection of this novel immunostrip reached 200 ppb (ng/ml), 1 ppm (µg/ml), and 2 ppm for TMV, PVY, and TVBMV particles, respectively. Importantly, no cross-reactivity was observed among TMV, TVBMV, and PVY or to a nontarg

First Report of Freesia sneak virus Associated with Foliar Necrosis of Freesia refracta in Bulgaria.

In the early spring of 2011 and 2012, severe necrotic leaf symptoms were observed on freesia (Freesia refracta, family Iridaceae) in several greenhouses around Plovdiv (south central Bulgaria). The disease spread and symptom severity in several cultivars (Medeo, Calvados, and Pink Fountain) led to nearly complete production failure for some growers. Initial symptoms consisted of scattered pale, chlorotic, interveinal lesions that coalesced. Later, irregular brown to black necrotic blotches partially covered the leaves. Flower break was also observed. Diseased plants were collected in late Apri

Experimental Confirmation of the Role of the α1-α2 Hairpin PVY VPg in eIF4E Interaction.

In recent years, the eIF4E protein family has been actively studied as one of the major susceptibility factors for Solanaceae plants in relation to potyvirus infections. This makes eIF4E an attractive target for genome editing to generate resistant varieties. The viral protein VPg interacts with eIF4E family proteins. Measuring the in vitro affinity of VPg for different eIF4E isoforms has traditionally been difficult. The main reason for this is the high aggregation propensity of VPg. To overcome this, we used protein engineering to generate a chimeric construct based on a fluorescent protein.

Virtual screening of potato virus Y coat protein for discovering lead inhibitors of viral intercellular traffic.

Potato virus Y (PVY) is one of the most economically detrimental phytoviruses affecting global Solanaceae, possessing challenges in agrochemical control. The structural elucidation of PVY coat protein (CP) offers opportunities for the rational design of CP-targeted antivirals; however, the feasibility of identifying lead compounds via virtual screening remains largely unexplored. Herein, we report the successful case of structure-based virtual screening leveraging PVY CP, enabling the identification of a structurally novel lead with a unique mechanism of action. Combining quantitative analysis

Chitosan-loaded copper oxide nanocomposite as a promising antiviral alleviates Zucchini yellow mosaic virus infection in squash plants.

BACKGROUND: Chitosan has been reported to induce host defense responses against plant viruses, while copper oxide nanoparticles can directly inhibit viral infection. However, the combined effect of chitosan-loaded copper oxide (CS-CuO) nanocomposites, particularly across different nanosizes, remains unexplored against Zucchini yellow mosaic virus (ZYMV). Therefore, this study investigates the antiviral potential of CS-CuO nanocomposites of varying mean particle sizes (70.33, 30.20, and 17.18 nm) to determine whether size-dependent enhancement of induced resistance occurs in squash plants. Differential Infection by Barley Yellow Mosaic Disease and Its Impact on Key Agronomic Traits in Barley.

Barley yellow mosaic disease (BYMD), a soilborne viral disease, severely compromises winter barley productivity. This disease is caused by barley yellow mosaic virus (BaYMV) and the barley mild mosaic virus (BaMMV), occurring in either single or mixed infections. To elucidate pathogenesis, 10 barley varieties with differential resistance were systematically analyzed. Virus-specific RT-qPCR revealed distinct accumulation patterns of BaYMV and BaMMV in infected tissues. Crucially, transmission requires temperatures persistently below 10°C for 40 to 54 days, facilitating viral entry into roots fo

Efficient artificial microRNA-mediated resistance against zucchini yellow mosaic virus in zucchini via agroinfiltration.

Zucchini yellow mosaic virus (ZYMV) infects cucurbits causing yellow mosaic in leaves, malformations in fruits, and degradation of the product quality. RNA interference (RNAi) is a cellular mechanism in eukaryotes and it is exploited to protect them against viruses. The artificial micro RNA (amiRNA) mediated approach was employed to develop resistance against ZYMV. Four amiRNAs, amiZYMV_HC-115s and amiZYMV_HC-1162s (sense), amiZYMV_HC-182as and amiZYMV_HC-196as (antisense), were computationally designed and introduced into the AtMIR390a backbone. Guanosine tetraphosphate modulates salicylic acid signalling and the resistance of Arabidopsis thaliana to Turnip mosaic virus.

Chloroplasts can act as key players in the perception and acclimatization of plants to incoming environmental signals. A growing body of evidence indicates that chloroplasts play a critical role in plant immunity. Chloroplast function can be regulated by the nucleotides guanosine tetraphosphate and pentaphosphate [(p)ppGpp]. In plants, (p)ppGpp levels increase in response to abiotic stress and to plant hormones which are involved in abiotic and biotic stress signalling. In this study, we analysed the transcriptome of Arabidopsis plants that over-accumulate (p)ppGpp, and unexpectedly found a de

A benzo[b]thiophene-derived inhibitor of virus particle assembly via targeting capsid protein residue Arg157.

As a biological macromolecule, the coat protein (CP) of potato virus Y (PVY) mediates the virus' primary pathogenic behaviors. It has been gradually realized that certain residues on the CP are crucial for functions such as virus particle movement and assembly. However, there are few reports of potential drugs successfully targeting these key residues with unique mechanisms of action. Here, we disclose the first new phytovirucide that acts on the key site Arg157 (R157) on the PVY CP. In this investigation, we developed a series of benzo[b]thiophene-based compounds, strategically introducing su

Foliar Application of Nanoclay Promotes Potato (Solanum tuberosum L.) Growth and Induces Systemic Resistance against Potato Virus Y.

Potato virus Y (PVY) is one of the most harmful phytopathogens. It causes big problems for potatoes and other important crops around the world. Nanoclays have been extensively studied for various biomedical applications. However, reports on their interactions with phytopathogens, particularly viral infections, are still limited. In this study, the protective activity of Egyptian nanoclay (CE) and standard nanoclay (CS) against PVY was evaluated on potato (Solanum tuberosum L.) plants. Their physicochemical and morphological properties were examined with scanning electron microscopy (SEM), tran

Competition among co-infecting potyviral chimeras with adaptive, single-nucleotide differences in HCPro: Properties, and influence of environment factors on outcomes.

We previously showed that two natural, single-amino acid mutations in the HCPro of a potyviral chimera, separately increased virulence by enhancing either its silencing-suppression or proteolytic functions. To investigate how beneficial variants generated during infection compete with non-mutant forms, or among each other, we co-expressed them in pairs, in Nicotiana benthamiana. We assessed how variants co-exist and influence each other in the cell, and whether altered abiotic (elevated temperature) or biotic (a silencing-impaired plant) environmental factors, affected local and systemic infec

Development of axially chiral urazole scaffolds for antiplant virus applications against potato virus Y.

BACKGROUND: Potato virus Y (PVY) was first discovered by Smith in 1931 and is currently ranked as the fifth most significant plant virus. It can cause severe damage to plants from the family Solanaceae, which results in billions of dollars of economic loss worldwide every year. To discover new antiviral drugs, a class of multifunctional urazole derivatives bearing a stereogenic CN axis were synthesized with excellent optical purities for antiviral evaluations against PVY. RESULTS: The absolute configurations of the axially chiral compounds exhibited obvious distinctions in antiviral bioactiv

Turnip mosaic virus alters phosphorus metabolism and shoot-root allocation without resource competition.

Plant viruses affect production through symptom induction in host plants. These symptoms could partially arise from nutrient deprivation: The resource competition hypothesis posits that massive viral replication deprives hosts of essential nutrients, yet direct evidence for phosphorus (P) competition is lacking. Moreover, it is reported that biotic stresses can lead to alterations on P metabolism. Using a hydroponic system enabling separate analysis of shoots and roots in adult Arabidopsis thaliana plants, we investigated whether Turnip mosaic virus (TuMV) drawed significant P internal pools l

Synthetic Potato Systemin Enhances Solanum tuberosum Resistance to Potato Virus Y.

In recent years, short plant peptides have emerged as highly promising, innovative bioregulators of crop growth and resilience. Activating systemic defense using synthetic plant peptides represents a potent strategy to improve crop resistance against pathogenic threats. Potato virus Y (PVY) causes devastating economic losses in potato (Solanum tuberosum) crops. In this study, we investigated the protective activity of synthetic analogs of systemin, a classic defense peptide originally discovered within the Solanaceae family. We synthesized the putative potato octadecapeptide systemin (AVHSTPPS

Streptomyces fungicidicus-derived secondary metabolites as an antiviral agent to alleviate zucchini yellow mosaic virus in squash.

Zucchini yellow mosaic virus (ZYMV) poses a significant threat to squash crops, causing severe symptoms and substantial yield losses. This study investigates the potential of Streptomyces fungicidicus as a biocontrol agent for managing ZYMV in squash plants by inducing systemic resistance. Approximately 95% of field-collected squash samples were positive for ZYMV, exhibiting chlorotic mottling, vein banding, and leaf distortion. The virus was isolated, purified, and confirmed through RT-PCR (Accession no. PV131044) and transmission electron microscopy (TEM), which displayed flexuous, filamento
