Prenatal exposure to benzodiazepines and Z-drugs in humans and risk of adverse neurodevelopmental outcomes in offspring: A systematic review.

When used during pregnancy, benzodiazepines (BZDs) and related z-drugs could pass readily through the placenta and the foetal blood-brain barrier, where they can bind to γ-amino butyric acid (GABA) receptors in the developing foetal brain. Yet, data on long-term safety of prenatal BZD and z-drug use and its impact on offspring neurodevelopment are inconclusive. In this systematic review, we qualitatively synthetize the existing evidence on maternal exposure to various BZDs and z-drugs during pregnancy and offspring cognitive, emotional, behavioural, and motor skills developmental outcomes. Maternal Obesity and its Short- and Long-Term Maternal and Infantile Effects.

Obesity, in childhood or in adulthood, remains to be a global health problem. The worldwide prevalence of obesity has increased in the last few decades, and consequently, the women of our time suffer more gestational problems than women in the past. The prevalence of obesity is greater in older women than in younger ones and in women with low educational level than in their counterparts with a higher level of education. Maternal obesity during pregnancy may increase congenital malformations and neonatal morbidity and mortality. Maternal obesity is associated with a decreased intention to breas

The effects of prenatal exposure to methylxanthines.

This review discusses epidemiology and laboratory studies on the effects of prenatal methylxanthine administration on some systems developing organisms. They are mainly absorbed from coffee, tea and cocoa products such as cola beverages and chocolate bars. Prenatal methylxanthine exposure can induce several unfavourables changes in the developing organism, which are persistent even in later phases of life. Based on results obtained from animal studies, the effect on embryogenesis is not only poorly understood but also controversial. It is therefore important to study interspecies differences a

Nanoplastic toxicology following gestational and lactational exposure.

Nanoplatics (NPs), particularly polystyrene (PS)-NPs, can traverse the placental barrier upon maternal exposure, leading to bioaccumulation in both dam and offspring organs, and inducing widespread transplacental toxicity. The distribution and toxicity of NPs are influenced by a variety of factors, including NP properties (type, size, and charge), exposure parameters (dose, route, and timing), and biological variables (model and co-exposures). Due to their minute size, NPs pose significant threats to multiple systems in animal models. In rodent studies, reproductive and endocrine toxicity prim

Association between prenatal exposure to tetrachloroethylene and adverse birth outcomes: Systematic review and meta-analysis.

BACKGROUND: Tetrachloroethylene (PCE) is a ubiquitous chlorinated solvent with documented placental transfer. Despite widespread environmental and occupational exposure, no prior systematic review has synthesized evidence on prenatal PCE exposure and adverse birth outcomes. METHODS: We conducted a systematic review and meta-analysis of observational studies. PubMed, Web of Science, PsycINFO, EMBASE, and CINAHL were searched from inception to July 13, 2026. Eligible studies reported associations between prenatal PCE exposure (drinking water or inhalation) and adverse birth outcomes. Study qual

A metabolomic study on the gender-dependent effects of maternal exposure to fenvalerate on neurodevelopment in offspring mice.

BACKGROUND: The general population is widely exposed to fenvalerate. However, the effects of maternal exposure to fenvalerate on neurodevelopment in offspring and the underlying metabolic mechanism are largely unknown. METHODS: Pregnant mice were exposed to fenvalerate for 11 consecutive days. The forced swimming test (FST) was performed in 35 day-old offspring to investigate the effects of fenvalerate on neurobehavioral responses. Blood serum free T4 and free T3 concentrations were measured using commercial ELISA. Blood and thyroid samples were used for metabolomic analyses with UPLC Q-Exact

Maternal exposure to diluted diesel engine exhaust alters placental function and induces intergenerational effects in rabbits.

BACKGROUND: Airborne pollution is a rising concern in urban areas. Epidemiological studies in humans and animal experiments using rodent models indicate that gestational exposure to airborne pollution, in particular diesel engine exhaust (DE), reduces birth weight, but effects depend on exposure duration, gestational window and nanoparticle (NP) concentration. Our aim was to evaluate the effects of gestational exposure to diluted DE on feto-placental development in a rabbit model. Pregnant females were exposed to diluted (1 mg/m(3)), filtered DE (NP diameter ≈ 69 nm) or clean air (controls) fo

Low to Moderate Prenatal Alcohol Exposure and Neurodevelopmental Outcomes: A Narrative Review and Methodological Considerations.

PURPOSE: Although abstinence is recommended in pregnancy, many pregnancies are exposed to alcohol. Observational studies of the effects of low to moderate prenatal alcohol exposure (PAE) and neurodevelopmental outcomes have yielded inconsistent results, with some studies finding an increased risk of adverse neurobehavioral and cognitive outcomes, and other studies finding no changes or reduced risk of the same outcomes. The purpose of this narrative review is to summarize these inconsistencies and apply a methodological framework to discuss how different parameters contribute to the findings.

Fetal alcohol spectrum disorders-- implications for child neurology, part 1: prenatal exposure and dosimetry.

In the United States, approximately 80 000 women consume ethanol through all 3 trimesters of pregnancy each year. In this article, we review prevalence rates of prenatal alcohol exposure in the United States and discuss the mechanisms of prenatal alcohol exposure and placental-umbilical effects. Cigarette smoking and delayed prenatal care are often associated with prenatal alcohol exposure. In addition, increased risk for postnatal adversity is common, including maternal depression, foster care placement, and developmental delay. In part 2, we review prevalence rates and the diagnostic criteri

Annual Research Review: Exposure to environmental chemicals and psychosocial stress and the development of children's learning difficulties.

Although awareness of the role of environmental exposures in children's cognitive development is increasing, learning difficulties have not yet been a major focus of environmental health science. Learning difficulties disproportionately affect children living in economic disadvantage, yielding an 'achievement gap.' Studies examining the neurobiology of reading and math have mostly included economically advantaged youth, leaving a great deal unknown about the neural underpinnings of reading and math difficulties in youth living in disadvantaged contexts. Critically, due to environmental injusti

Exposure to Endocrine-Disrupting Chemicals and Congenital Heart Diseases: The Pooled Results Based on the Current Evidence.

The relationships between maternal exposure to endocrine-disrupting chemicals (EDCs) and congenital heart diseases (CHD) are not elucidated yet. The exposure levels of EDCs are generally estimated based on self-reported questionnaires or occupational exposure evaluations in the literature. Therefore, a study based on epidemiological data from human biospecimens is required to provide stronger evidence between maternal exposure to EDC and CHD. Embase, Pubmed, Scopus, and the Cochrane Library databases were searched for related research which provided risk estimates regarding the relationships b

Effects of Neonicotinoid Pesticide Exposure on Human Health: A Systematic Review.

BACKGROUND: Numerous studies have identified detectable levels of neonicotinoids (neonics) in the environment, adverse effects of neonics in many species, including mammals, and pathways through which human exposure to neonics could occur, yet little is known about the human health effects of neonic exposure. OBJECTIVE: In this systematic review, we sought to identify human population studies on the health effects of neonics. METHODS: Studies published in English between 2005 and 2015 were searched using PubMed, Scopus, and Web of Science databases. No restrictions were placed on the type of

Prenatal exposure to medication and risk of childhood cancer - a systematic review and meta-analysis.

BACKGROUND: The use of medication during pregnancy carries a potential health risk, including childhood cancer, for the unborn child. However, most drugs examined in previous observational studies have shown inconsistent results with the risk of childhood cancer, and these findings have not been consolidated in drug-specific meta-analyses. METHODS: We conducted a systematic search in the databases PubMed and Web of Science for studies on medication use during pregnancy and childhood cancer risk. Studies with exposure to diethylstilbestrol as a known teratogen were excluded. Meta-analyses were

Fetal Alcohol Spectrum Disorders.

Prenatal alcohol exposure (PAE) can cause a wide range of physical, neurobehavioral, and neurocognitive impairments that impact developmental trajectories throughout the lifespan. Clinically, individuals who have been exposed to alcohol prenatally and who show physical or neurobehavioral difficulties may be classified as having a condition included in fetal alcohol spectrum disorders (FASDs). The aim of this chapter is to summarize the current knowledge of FASD including diagnostic criteria, neurobehavioral outcomes, lifespan considerations, and interventions. Individuals with PAE exhibit chal

Epigenetic effects of endogenous and exogenous glucocorticosteroids during pregnancy on the offspring: a systematic-narrative review.

BACKROUND: Τhe epigenetic effects of glucocorticosteroids-also known as glucocorticoids-(GCs) on the human epigenome are under constant in-depth examination. During uncomplicated pregnancy, endogenous GCs are normally increased, this increase being increased in stressful maternal conditions, such GC excess potentially having a deleterious effect on the fetus. In addition, however, synthetic GCs have long been used during pregnancy, not only for lung maturation in pregnancies at risk for preterm birth but also therapeutically for a large number of maternal diseases. Although GCs can be administ

Intergenerational effect of an adverse intrauterine environment on perturbation of glucose metabolism.

Human epidemiological and animal studies have revealed the late consequences of malnutrition during gestation and early life on the health of the offspring. These studies have highlighted the inverse relationship between birth weight and the incidence of insulin resistance and type 2 diabetes later in life. The aim of this paper is to review the different means of achieving foetal malnutrition and its consequences even for a next generation, in animal models and to identify key area for further research. We address the impact of two models of maternal malnutrition (protein restriction and calo
