Functional bipartite invariance in mouse primary visual cortex receptive fields.

Sensory systems support generalization by representing features that persist under input variation; however, identifying the neuronal basis of these invariances remains difficult due to high-dimensional and nonlinear neural computations. Here we leverage the inception loop paradigm, iterating between large-scale recordings, predictive models and in silico experiments with in vivo verification, to characterize neuronal invariances in mouse primary visual cortex (V1). We synthesize varied exciting inputs (VEIs), dissimilar images that drive target neurons. These VEIs revealed a new bipartite inv

Circuit dynamics of binocular conflict in mouse primary visual cortex.

Binocular vision requires that the brain integrate input from both eyes to form a unified percept. Small interocular differences support depth perception (stereopsis), while larger disparities can cause diplopia or binocular rivalry. The neural mechanisms by which early visual circuits process concordant versus conflicting binocular signals remain incompletely understood. Here, we used visually evoked potentials (VEPs), unit recordings, and 2-photon calcium imaging in the binocular region of mouse primary visual cortex (bV1) to examine how distinct forms of binocular disparity engage local cir

NMDAR-Dependent Emergence of Behavioral Representation in Primary Visual Cortex.

Although neocortical sensory areas are generally thought to faithfully represent external stimuli, cortical networks exhibit considerable functional plasticity, allowing them to modify their output to reflect ongoing behavioral demands. We apply longitudinal 2-photon imaging of activity in the primary visual cortex (V1) of mice learning a conditioned eyeblink task to investigate the dynamic representations of task-relevant information. We find that, although all V1 neurons robustly and stably encode visual input, pyramidal cells and parvalbumin-expressing interneurons exhibit experience-depend

Plasticity, and Its Limits, in Adult Human Primary Visual Cortex.

There is an ongoing debate about whether adult human primary visual cortex (V1) is capable of large-scale cortical reorganization in response to bilateral retinal lesions. Animal models suggest that the visual neural circuitry maintains some plasticity through adulthood, and there are also a few human imaging studies in support this notion. However, the interpretation of these data has been brought into question, because there are factors besides cortical reorganization, such as the presence of sampling bias and/or the unmasking of task-dependent feedback signals from higher level visual areas

Altered contextual modulation of primary visual cortex responses in schizophrenia.

Schizophrenia is typically associated with higher-level cognitive symptoms, such as disorganized thoughts, delusions, and hallucinations. However, deficits in visual processing have been consistently reported with the illness. Here, we provide strong neurophysiological evidence for a marked perturbation at the earliest level of cortical visual processing in patients with paranoid schizophrenia. Using functional magnetic resonance imaging (fMRI) and adapting a well-established approach from electrophysiology, we found that orientation-specific contextual modulation of cortical responses in huma

Capillary Density and Neuronal Homeostasis in Human Primary Visual Cortex.

BACKGROUND: Hypoxia is a critical determinant of neuronal function and vulnerability in the brain. While neuronal density has been extensively characterized across cortical regions, the spatial relationship between neurons and the microvasculature represents an additional structural constraint on oxygen diffusion and metabolic support. In particular, the distance between neurons and nearby capillaries influences local oxygen availability required to sustain neuronal activity and maintain ionic homeostasis. METHODS: In this study, we quantified neuron-capillary spatial relationships across cor

Learned spatiotemporal sequence recognition and prediction in primary visual cortex.

Learning to recognize and predict temporal sequences is fundamental to sensory perception and is impaired in several neuropsychiatric disorders, but little is known about where and how this occurs in the brain. We discovered that repeated presentations of a visual sequence over a course of days resulted in evoked response potentiation in mouse V1 that was highly specific for stimulus order and timing. Notably, after V1 was trained to recognize a sequence, cortical activity regenerated the full sequence even when individual stimulus elements were omitted. Our results advance the understanding o

Distinct Response Selectivity Changes in the Primary Visual and Parietal Cortex during Visual Discrimination Learning.

When animals learn the behavioral relevance of sensory features, response selectivity in primary sensory areas increases for those features. However, the effect of learning on neuronal activity in higher-level areas associated with decision-making in the parietal cortex, compared with primary sensory cortex, is not yet known. We used two-photon calcium imaging to determine how learning modifies neural representations in the primary visual cortex (V1) and posterior parietal cortex (PPC) in a visual go/no-go orientation discrimination task in male and female mice. We found that behavior improvem

Stimulus-Selective Response Plasticity in Primary Visual Cortex: Progress and Puzzles.

Stimulus-selective response plasticity (SRP) is a robust and lasting modification of primary visual cortex (V1) that occurs in response to exposure to novel visual stimuli. It is readily observed as a pronounced increase in the magnitude of visual evoked potentials (VEPs) recorded in response to phase-reversing grating stimuli in neocortical layer 4. The expression of SRP at the individual neuron level is equally robust, but the qualities vary depending on the neuronal type and how activity is measured. This form of plasticity is highly selective for stimulus features such as stimulus orientat

Information transmission in the primary visual cortex of retinal degenerated rats.

To study the information transmission in the primary visual cortex (V1) of retinal degenerated (RD) models, wild type (WT) and RD rats were used in the experiments. The neural response in V1 was recorded extracellularly while the flicker with varied intensity levels was given as the visual stimulus. The mutual information (MI) and normalized mutual information (NMI) were determined for every isolated neuron, in order to quantify the amount and efficiency of information transmission in V1 of both control and experimental groups. The results showed that, on one hand, the RD group manifested rela

Complete pattern of ocular dominance columns in human primary visual cortex.

The occipital lobes were obtained after death from six adult subjects with monocular visual loss. Flat-mounts were processed for cytochrome oxidase (CO) to reveal metabolic activity in the primary (V1) and secondary (V2) visual cortices. Mean V1 surface area was 2643 mm2 (range, 1986-3477 mm2). Ocular dominance columns were present in all cases, having a mean width of 863 microm. There were 78-126 column pairs along the V1 perimeter. Human column patterns were highly variable, but in at least one person they resembled a scaled-up version of macaque columns. CO patches in the upper layers were

The influence of electrode types to the visually induced gamma oscillations in mouse primary visual cortex.

The local field potential (LFP) is an extracellular electrical signal associated with neural ensemble input and dendritic signaling. Previous studies have linked gamma band oscillations of the LFP in cortical circuits to sensory stimuli encoding, attention, memory, and perception. Inconsistent results regarding gamma tuning for visual features were reported, but it remains unclear whether these discrepancies are due to variations in electrode properties. Specifically, the surface area and impedance of the electrode are important characteristics in LFP recording. To comprehensively address thes

Crossmodal audio-visual interactions in the primary visual cortex of the visually deprived cat: a physiological and anatomical study.

Blind individuals often demonstrate enhanced non-visual perceptual abilities. Neuroimaging and transcranial magnetic stimulation experiments have suggested that computations carried out in the occipital cortex may underlie these enhanced somatosensory or auditory performances. Thus, cortical areas that are dedicated to the analysis of the visual scene may, in the blind, acquire the capacity to participate in other sensory processing. However, the neural substrate that underlies this transfer of function is not fully characterized. Here we studied the synaptic and anatomical basis of this pheno

Altered functional connectivity of the primary visual cortex in subjects with amblyopia.

Amblyopia, which usually occurs during early childhood and results in poor or blurred vision, is a disorder of the visual system that is characterized by a deficiency in an otherwise physically normal eye or by a deficiency that is out of proportion with the structural or functional abnormalities of the eye. Our previous study demonstrated alterations in the spontaneous activity patterns of some brain regions in individuals with anisometropic amblyopia compared to subjects with normal vision. To date, it remains unknown whether patients with amblyopia show characteristic alterations in the fun

Spatiotemporal dynamics of pattern formation in the primary visual cortex and hallucinations.

The existence of visual hallucinations with prominent temporal oscillations is well documented in conditions such as Charles Bonnett Syndrome. To explore these phenomena, a continuum model of cortical activity that includes additional physiological features of axonal propagation and synapto-dendritic time constants, is used to study the generation of hallucinations featuring both temporal and spatial oscillations. A detailed comparison of the physiological features of this model with those of two others used previously in the modeling of hallucinations is made, and differences, particularly re

Contrast coding in the primary visual cortex depends on temporal contexts.

Contrast response function in the primary visual cortex (V1) has long been described as following a sigmoid curve. However, this is mainly based on measuring neural responses to drifting contrast grating in a stable stimulation, a model that does not consider the effects of motion or length of stimulus presentation. During natural viewing, the visual system can obtain sufficient information for identifying the shapes defined by contrast from a single glance; acquiring greater knowledge of the neuronal response properties to contrast in such a short timescale is necessary to understand the unde

Specialized circuits from primary visual cortex to V2 and area MT.

Primary visual cortex recombines inputs from magnocellular (M) and parvocellular (P) streams to create functionally specialized outputs. Understanding these input-output relationships is complicated by the fact that layer 4B, which provides outputs to dorsal visual areas, contains multiple cell types. Using a modified rabies virus that expresses green fluorescent protein, we show that layer 4B neurons projecting to MT are a majority spiny stellate, whereas those projecting to V2 are overwhelmingly pyramidal. Regardless of cell type, MT-projecting neurons have larger cell bodies, more dendritic

Attentional gating in primary visual cortex: a physiological basis for dyslexia.

The visual magnocellular pathway is known to play a central part in visuospatial attention and in directing attention to specific parts of the visual world in serial search. It is proposed that, in the case of reading, this mechanism is trained to perform a sequential gating of visual information coming into the primary visual cortex to enable further orderly processing by the ventral stream. This scheme, taken together with the potential for plasticity between the different afferent channels in the case of a relative impairment of the magnocellular system, can provide some limited rationale f
