Cutibacterium Adaptation to Life on Humans Provides a Potential C acnes Infection Biomarker.

Propionibacteriaceae appear to have adapted to life on humans during the domestication of cattle. These microbial immigrants formed the genus Cutibacterium, and a descendent of those microbial trailblazers (C acnes) now dominates 25% of human skin. C acnes colonization of human skin requires the protein RoxP. Although all Cutibacteria encode this adaptation to life on humans, nothing like RoxP has been found in any other organism. In this study, we report an extensive assessment of 21 RoxP orthologs, which identified conserved molecular surfaces linked to heme-dependent oligomerization and low

Distinguishing bipolar depression, bipolar mania, and major depressive disorder by gut microbial characteristics.

BACKGROUND: Gut microbial disturbance has been widely confirmed in mood disorders. However, little is known about whether gut microbial characteristics can distinguish major depressive disorder (MDD), bipolar depression (BP-D), and bipolar mania (BP-M). METHODS: This was a prospective case-control study. The composition of gut microbiota was profiled using 16S ribosomal RNA (rRNA) gene sequencing of fecal samples and compared between healthy controls (HC; n = 46), MDD (n = 51), BP-D (n = 44), and patients with BP-M (n = 45). RESULTS: Gut microbial compositions were remarkably changed in the

Immune Activation and Glycolytic Responses to Cutibacterium acnes Cell Wall Polysaccharides.

Carbohydrates are key components of many microbial cell walls and play a versatile role in immune recognition. In this study, we analyzed the carbohydrate cell wall composition of Cutibacterium acnes strains associated with healthy skin (denoted as CH) and acne-prone skin (denoted as CA) to understand their influence on host immune responses in acne. We identified glucose, mannose, and galactose as the primary monosaccharides, with minor amounts of fucose, N-acetylgalactosamine, and N-acetylglucosamine. Linkage analysis revealed structural variations between CH and CA strains: CH strains showe

Growth in a biofilm sensitizes Cutibacterium acnes to nanosecond pulsed electric fields.

The Gram-positive anaerobic bacterium Cutibacterium acnes (C. acnes) is a commensal of the human skin, but also an opportunistic pathogen that contributes to the pathophysiology of the skin disease acne vulgaris. C. acnes can form biofilms; cells in biofilms are more resilient to antimicrobial stresses. Acne therapeutic options such as topical or systemic antimicrobial treatments often show incomplete responses. In this study we measured the efficacy of nanosecond pulsed electric fields (nsPEF), a new promising cell and tissue ablation technology, to inactivate C. acnes. Our results show that

Removal Mechanism of Cutibacterium acnes Biofilms: A Comparison between Synthetic Surfactants and Rhamnolipid Biosurfactants.

Biofilms are composed of autogenously secreted extracellular polymeric substances (EPS) and affect nearly all aspects of human activity. Biofilms with negative health and economic effects can be controlled through the integrated use of surfactants and mechanical force. However, the relationships between the surfactant molecular structures and biofilm removal pathways have not been sufficiently elucidated. Additionally, Cutibacterium acnes (C. acnes), a skin commensal bacterium with biofilm-forming capabilities that can exacerbate acne vulgaris, has been rarely investigated. In this study, we e

Study on Oleum cinnamomi Inhibiting Cutibacterium acnes and Its Covalent Inhibition Mechanism.

Oleum cinnamomi (OCM) is a volatile component of the Cinnamomum cassia Presl in the Lauraceae family, which displays broad-spectrum antibacterial properties. It has been found that OCM has a significant inhibitory effect against Cutibacterium acnes (C. acnes), but the precise target and molecular mechanism are still not fully understood. In this study, the antibacterial activity of OCM against C. acnes and its potential effect on cell membranes were elucidated. Metabolomics methods were used to reveal metabolic pathways, and proteomics was used to explore the targets of OCM inhibiting C. acnes

Multidrug-resistant Cutibacterium avidum isolated from patients with acne vulgaris and other infections.

OBJECTIVES: Cutibacterium avidum, a human skin commensal bacterium, rarely causes infections. It has recently been shown that Cutibacterium acnes, another member of the genus, acts as an opportunistic pathogen in surgical site infections. However, the antimicrobial susceptibility and pathogenicity of C. avidum remain unknown. METHODS: We investigated the epidemiological features and antimicrobial susceptibility of C. avidum isolated from patients with acne vulgaris and other infections. RESULTS: Cutibacterium avidum strains were isolated from patients with acne vulgaris (29 strains) and othe

Anti-biofilm activity of coated zinc oxide nanoparticles in a water-in-oil formulation against Cutibacterium acnes.

Coated ZnO-NPs demonstrated antibacterial and biofilm-disrupting activities against Cutibacterium acnes in a high-viscosity, skincare-mimicking water-in-oil formulation. They reduced viable biofilm-embedded cell count by more than 100-fold compared to the control formulation. The sebum-exposed in vitro biofilm model provides a practical platform for evaluating anti-biofilm skincare products.

BACKGROUND: Cutibacterium acnes (C. acnes) is a causative agent in the development of acne vulgaris, and this bacteria has been reported to show resistance against conventional antibiotics. One of the vital factors contributing to antibiotic resistance is the ability of C. acnes to form biofilms. Thus, the purpose of this review is to assess the efficacy of various recent developments and to identify acceptable methods for preventing infections associated with C. acnes biofilms. METHODOLOGY: A variety of criteria considered in the selection process, such as the site of infection, the mechanis

Reversal Potential of Multidrug Antibiotic Resistance in Cutibacterium acnes by Ethanol Extracts of Rhubarb.

Cutibacterium acnes (C. acnes) is a major contributor to acne inflammation and exhibits significant antibiotic resistance, but research focusing on reversing this resistance is limited. Rhubarb, a natural plant with known therapeutic effects, shows potential in combating antibiotic resistance, however, no studies have been explored before. The aim of this study was to investigate rhubarb's ability to reverse antibiotic resistance in C. acnes. Strains 11,827 and 6919 were cultured and passaged with 0.019 μg/mL ethanol extract, and antibiotic sensitivity was monitored from passage 0 to 12. The e

Extracellular traps released by antimicrobial TH17 cells contribute to host defense.

TH17 cell subpopulations have been defined that contribute to inflammation and homeostasis, yet the characteristics of TH17 cells that contribute to host defense against infection are not clear. To elucidate the antimicrobial machinery of the TH17 subset, we studied the response to Cutibacterium acnes, a skin commensal that is resistant to IL-26, the only known TH17-secreted protein with direct antimicrobial activity. We generated C. acnes-specific antimicrobial TH17 clones (AMTH17) with varying antimicrobial activity against C. acnes, which we correlated by RNA sequencing to the expression of

Photoinactivation of planktonic Cutibacterium acnes subsp. elongatum: A study on wavelength-dependent effects.

Bacterial inflammatory skin conditions, especially those caused by Cutibacterium acnes (C. acnes), are among the most common skin diseases globally. While C. acnes is a gram-positive bacterium found in healthy human skin, its overgrowth or reduced phylotype diversity have been associated with inflammatory conditions such as acne vulgaris. Traditional treatments often apply antibiotics. Alternatives, like photodynamic therapy (PDT) and intense pulsed light (IPL) therapy, apply visible light irradiation. These therapies utilize photosensitizers, such as coproporphyrin III (CP III), to generate r

Isolation and characterization of two novel oral bacteriophages with anti-biofilm activity against Cutibacterium acnes.

Bacteriophage therapy offers a promising solution to combat antibiotic-resistant infections, yet its potential against biofilm-associated pathogens in oral diseases remains underexplored. This study investigates the opportunistic bacterium Cutibacterium acnes, an overlooked contributor to dental implant and prosthetic joint infections. Biofilms formed by C. acnes are highly resilient and resistant to antibiotics, complicating treatment. Two novel lytic bacteriophages, Ristretto and Corretto, targeting C. acnes, were isolated from human saliva, with morphological analysis confirming their class

A Microtube Array Membrane (MTAM) Encapsulated Live Fermenting Staphylococcus epidermidis as a Skin Probiotic Patch against Cutibacterium acnes.

Antibiotics without selectivity for acne treatment may destroy the beneficial microbes in the human microbiome that helps to fight Cutibacterium acnes (C. acnes), a bacterium associated with inflammatory acne vulgaris. Probiotic treatment by direct application of live Staphylococcus epidermidis (S. epidermidis) onto the open acne lesions may run the risk of bloodstream infections. Here, we fabricated the polysulfone microtube array membranes (PSF MTAM) to encapsulate probiotic S. epidermidis. We demonstrate that the application of the encapsulation of S. epidermidis in PSF MTAM enhanced the gl

The Human Ocular Surface Microbiome and Its Associations with the Tear Proteome in Dry Eye Disease.

Although dry eye disease (DED) is one of the most common ocular surface diseases worldwide, its pathogenesis is incompletely understood, and treatment options are limited. There is growing evidence that complex interactions between the ocular surface microbiome (OSM) and tear fluid constituents, potentially leading to inflammatory processes, are associated with ocular surface diseases such as DED. In this study, we aimed to find unique compositional and functional features of the OSM associated with human and microbial tear proteins in patients with DED. Applying whole-metagenome shotgun seque

Lactobacilli are prominent in the initial stages of polymicrobial infection of dental pulp.

In earlier studies we used molecular methods to identify the major bacterial consortia associated with advanced dentin caries. These consortia are dominated by bacteria from the families Lactobacillaceae, Streptococcaceae, Veillonellaceae (formerly Acidaminococcaceae), Eubacteriaceae, and Lachnospiraceae from the phylum Firmicutes; Coriobacteriaceae, Bifidobacteriaceae, and Propionibacteriaceae from the phylum Actinobacteria; and Prevotellaceae from the phylum Bacteroidetes, as well as fusobacteria. The phases of infection of vital pulp tissue by dentin microorganisms remain obscure. In the pr

Metagenomic Next-Generation Sequencing of Nasopharyngeal Specimens Collected from Confirmed and Suspect COVID-19 Patients.

Metagenomic next-generation sequencing (mNGS) offers an agnostic approach for emerging pathogen detection directly from clinical specimens. In contrast to targeted methods, mNGS also provides valuable information on the composition of the microbiome and might uncover coinfections that may associate with disease progression and impact prognosis. To evaluate the use of mNGS for detecting severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) and/or other infecting pathogens, we applied direct Oxford Nanopore long-read third-generation metatranscriptomic and metagenomic sequencing. Structural and dynamic insights into acyl carrier protein in Cutibacterium acnes reveal mechanisms for fatty acid synthesis and transport.

Cutibacterium acnes thrives in anaerobic environments and plays a role in acne vulgaris and the emergence of antibiotic-resistant strains. To maintain membrane fluidity and protect against toxins under anaerobic conditions, C. acnes balances branched-chain fatty acids (BCFAs) and straight-chain fatty acids, produced by its fatty acid synthase. In this study, we investigated the molecular mechanisms of fatty acid synthesis in C. acnes by determining the first-solution structure and dynamics of the acyl carrier protein (CaACP) using NMR spectroscopy. Our analyses, integrating backbone dynamics a
