Post-translational formylglycine modification of bacterial sulfatases by the radical S-adenosylmethionine protein AtsB.

C(alpha)-Formylglycine (FGly) is the catalytic residue of sulfatases. FGly is generated by post-translational modification of a cysteine (prokaryotes and eukaryotes) or serine (prokaryotes) located in a conserved (C/S)XPXR motif. AtsB of Klebsiella pneumoniae is directly involved in FGly generation from serine. AtsB is predicted to belong to the newly discovered radical S-adenosylmethionine (SAM) superfamily. By in vivo and in vitro studies we show that SAM is the critical co-factor for formation of a functional AtsB.SAM.sulfatase complex and for FGly formation by AtsB. The SAM-binding site of

Post-translational modifications of seminal proteins and their importance in male fertility potential.

Introduction: The seminal proteome has been shown to directly influence the male fertile potential. Post-translational modifications (PTMs) are significant changes that play a role in the biological regulation of proteins. Sperm cells are transcriptionally and translationally inactive and these modifications are essential to control protein function.Areas covered: Here we reviewed seven PTMs which importance for male reproductive function investigated in the past decade, namely S-nitrosylation and tyrosine nitration (both occurring by the action of NO), glycosylation, ubiquitination, acetylati

Beyond citrullination: other post-translational protein modifications in rheumatoid arthritis.

The presence of autoantibodies is one of the hallmarks of rheumatoid arthritis (RA). In the past few decades, rheumatoid factors (autoantibodies that recognize the Fc-tail of immunoglobulins) as well as anti-citrullinated protein antibodies (ACPAs) have been studied intensively. ACPAs recognize post-translationally modified proteins in which the amino acid arginine has been converted into a citrulline. More recently, other autoantibody systems recognizing post-translationally modified proteins have also gained attention, including autoantibodies recognizing fragmented immunoglobulin (anti-hing

Overview of xeroderma pigmentosum proteins architecture, mutations and post-translational modifications.

The xeroderma pigmentosum complementation group proteins (XPs), which include XPA through XPG, play a critical role in coordinating and promoting global genome and transcription-coupled nucleotide excision repair (GG-NER and TC-NER, respectively) pathways in eukaryotic cells. GG-NER and TC-NER are both required for the repair of bulky DNA lesions, such as those induced by UV radiation. Mutations in genes that encode XPs lead to the clinical condition xeroderma pigmentosum (XP). Although the roles of XPs in the GG-NER/TC-NER subpathways have been extensively studied, complete knowledge of their

Carbamate post-translational modifications: bridging CO2 biology and protein regulation.

Carbon dioxide (CO2) is a bioactive gas essential for several physiologically important biological processes, including metabolism, acid-base homoeostasis, and chemosensing. CO2 can directly modulate these processes by stimulating or inhibiting specific cell signalling pathways, including the nuclear factor-κB, calcium, and Wingless (Wnt) pathways. One mechanism by which CO2 interacts with proteins to modulate cell signalling is through the reversible and nonenzymatic modification of α- N-terminal and lysine ε-amino groups to form the carbamate post-translational modification (PTM). Advances in the interaction between gut microbiota and post-translational modifications of proteins.

Gut microbiota intricately regulate host cells through diverse mechanisms, with numerous pathways involving protein post-translational modifications (PTMs). This review comprehensively summarizes the impacts of the gut microbiota on various PTMs in host cells. It elaborates on how these modifications contribute to the development of host diseases, delving into mediating pathways, including changes in microbial metabolites, key enzymes, and the microenvironment. Conversely, it also explores how PTMs influence gut microbiota abundance. To overcome current research limitations, such as narrow per

Emerging Insights into Protein Post-Translational Modifications in Chlamydia-Host Interactions.

Chlamydia species are responsible for significant diseases in both humans and animals, with their infection processes involving complex interactions with host cells. Protein post-translational modifications (PTMs) have emerged as a critical focus in understanding the complex interplay between Chlamydia and its host. PTMs, including phosphorylation, glycosylation, ubiquitination, acetylation, and methylation, play pivotal roles in regulating key processes during Chlamydia infection, such as bacterial invasion, intracellular survival, immune evasion, and manipulation of host signaling pathways.

Differential Effects of Post-translational Modifications on the Membrane Interaction of Huntingtin Protein.

Huntington's disease is a neurodegenerative disorder caused by an expanded polyglutamine stretch near the N-terminus of the huntingtin (HTT) protein, rendering the protein more prone to aggregate. The first 17 residues in HTT (Nt17) interact with lipid membranes and harbor multiple post-translational modifications (PTMs) that can modulate HTT conformation and aggregation. In this study, we used a combination of biophysical studies and molecular simulations to investigate the effect of PTMs on the helicity of Nt17 in the presence of various lipid membranes. We demonstrate that anionic lipids su

Human T-lymphotropic virus proteins and post-translational modification pathways.

Cell life from the cell cycle to the signaling transduction and response to stimuli is finely tuned by protein post-translational modifications (PTMs). PTMs alter the conformation, the stability, the localization, and hence the pattern of interactions of the targeted protein. Cell pathways involve the activation of enzymes, like kinases, ligases and transferases, that, once activated, act on many proteins simultaneously, altering the state of the cell and triggering the processes they are involved in. Viruses enter a balanced system and hijack the cell, exploiting the potential of PTMs either

The effect of replication protein A inhibition and post-translational modification on ATR kinase signaling.

The ATR kinase responds to elevated levels of single-stranded DNA (ssDNA) to activate the G2/M checkpoint, regulate origin utilization, preserve fork stability, and allow DNA repair to ensure genome integrity. The intrinsic replication stress in cancer cells makes this pathway an attractive therapeutic target. The ssDNA that drives ATR signaling is sensed by the ssDNA-binding protein replication protein A (RPA), which acts as a platform for ATRIP recruitment and subsequent ATR activation by TopBP1. We have developed chemical RPA inhibitors (RPAi) that block RPA-ssDNA interactions (RPA-DBi) and

Altered post-translational modification of redox factor 1 protein in human uterine smooth muscle tumors.

Uterine leiomyomas are the most common benign smooth muscle tumors in the myometrium. The expression of redox factor 1 (Ref-1), a DNA repair enzyme and redox-modifying factor, was studied in the myometrium and uterine smooth muscle tumors to investigate the relevance of Ref-1 in the growth regulation of the tumors. Two forms of Ref-1 protein were detected, using three antibodies against different epitopes of Ref-1. The abundance of the large form of Ref-1 was increased in leiomyoma extracts relative to myometrial tissue extracts, and the large form was dominant in cell lines derived from leiom

Non-oxidative modification of lens crystallins by kynurenine: a novel post-translational protein modification with possible relevance to ageing and cataract.

In humans, the crystallin proteins of the ocular lens become yellow-coloured and fluorescent with ageing. With the development of senile nuclear cataract, the crystallins become brown and additional fluorophores are formed. The mechanism underlying crystallin colouration is not known but may involve interaction with kynurenine-derived UV filter compounds. We have recently identified a sulphur-linked glutathionyl-3-hydroxykynurenine glucoside adduct in the lens and speculated that kynurenine may also form adducts with GSH and possibly with nucleophilic amino acids of the crystallins (e.g. Cys).

Regulation of HDAC6 Catalytic Activity in Cancer: The Role of Post-Translational Modifications and Protein-Protein Interactions.

Histone deacetylase 6 (HDAC6) is a large multidomain protein that deacetylates lysine residues on cytoplasmic proteins, influencing numerous cellular processes. Both the catalytic and noncatalytic functions of HDAC6 have been implicated in cancer development and progression. Over a decade of research on catalytic domain inhibitors has shown that these drugs are well tolerated, exhibit anticancer activity, and can alleviate chemotherapy-induced peripheral neuropathies. However, their effectiveness in treating solid tumours remains uncertain. HDAC6 activity is regulated by protein-protein intera

Pathogenic Protein Post-Translational Modifications in Alzheimer's Disease: Mechanisms and Therapeutic Strategies.

BACKGROUND: Alzheimer's Disease (AD) pathology involves much more than just Amyloid- Beta (Aβ) and tau (Tau) deposition. A broad network of Post-Translational Modifications (PTMs) drives pathogenic protein conformations that accelerate neuroinflammation, synaptic dysfunction, aggregation, and trans-neuronal spread. Characterising PTM-dependent biochemical signatures allows earlier diagnosis and supports the development of molecularly targeted therapies. METHODS: A comprehensive review was performed with clinical-trial registries, high-confidence proteomic repositories, and major bibliographic

Bacterial protein post-translational modifications: a dynamic regulatory layer of antibiotic resistance.

Antibiotic resistance has become a major threat to global public health. It is commonly explained by target mutations, acquisition of resistance genes, drug inactivation, enhanced efflux, and reduced permeability. However, even in an unchanged genetic background, bacterial susceptibility to antibiotics can shift rapidly with nutritional, metabolic, and stress states, indicating that the functional output of resistance determinants is also subject to post-translational regulation. Recent quantitative analyses of bacterial post-translational modifications (PTMs) have shown that PTMs can directly

Myelin Basic Protein Post-Translational Modifications Orchestrate Astrocyte Regulatory Networks.

Multiple sclerosis (MS) pathogenesis involves not only immune-mediated myelin injury but also glial responses. We examined how three charge isomers of myelin basic protein (MBP)-native (C1), phosphorylated (C4), and citrullinated (C8)-modulate rat astrocytes. Cytokines were quantified and grouped (pro/anti-inflammatory, chemotactic, neurotrophic, angiogenic, tissue remodeling), and regulatory markers assessed. C1 strongly upregulated the lipid-sensing receptor LXR, and reduced global DNA methylation; C4 moderately enhanced LXR; C8 failed to activate LXR or alter methylation. Functionally, C1 a

Post-Translational Modifications of Proteins: Novel Insights in the Autoimmune Response in Rheumatoid Arthritis.

Post-translational modifications (PTM) are chemical changes mostly catalyzed by enzymes that recognize specific target sequences in specific proteins. These modifications play a key role in regulating the folding of proteins, their targeting to specific subcellular compartments, their interaction with ligands or other proteins, and eventually their immunogenic properties. Citrullination is the best characterized PTM in the field of rheumatology, with antibodies anticyclic citrullinated peptides being the gold standard for the diagnosis of rheumatoid arthritis (RA). Quantitative proteomic characterization of redox-dependent post-translational modifications on protein cysteines.

Protein thiols play a crucial role in redox signaling, in the regulation of enzymatic activity and protein function, and in maintaining redox homeostasis in living systems. The unique chemical reactivity of the thiol group makes protein cysteines susceptible to reactions with reactive oxygen and nitrogen species that form various reversible and irreversible post-translational modifications (PTMs). The reversible PTMs in particular are major components of redox signaling and are involved in the regulation of various cellular processes under physiological and pathological conditions. Post-translational Modifications and Protein Quality Control in Motor Neuron and Polyglutamine Diseases.

Neurodegenerative diseases, including motor neuron and polyglutamine (polyQ) diseases, are a broad class of neurological disorders. These diseases are characterized by neuronal dysfunction and death, and by the accumulation of toxic aggregation-prone proteins in the forms of inclusions and micro-aggregates. Protein quality control is a cellular mechanism to reduce the burden of accumulation of misfolded proteins, a function that results from the coordinated actions of chaperones and degradation systems, such as the ubiquitin-proteasome system (UPS) and autophagy-lysosomal degradation system. The landscape of protein post-translational modifications in the pathogenesis of acute respiratory distress syndrome.

Acute respiratory distress syndrome (ARDS) remains one of the most severe forms of acute lung injury, characterized by diffuse alveolar damage and refractory hypoxemia secondary to non-cardiogenic pulmonary edema. After many years of research and incremental improvements in ventilatory and supportive strategies, mortality rates are still disappointingly high and no pharmacological agent has convincingly demonstrated a mortality benefit in large trials. Elucidating the rapid molecular processes driving the acute phase of lung injury, particularly post-translational modifications (PTMs), may be
