Zinc protoporphyrin (ZPP) is produced instead of heme as soon as iron support to erythropoiesis becomes insufficient. In iron deficiency the intra-erythrocytic ZPP concentration is increased. The aim of this study was to investigate whether ZPP is influenced by increased iron levels in hereditary hemochromatosis (HE) and is useful in the clarification of hyperferritinemia. Twenty HE patients and 160 patients with hyperferritinemic caused by anemia of chronic disorders, liver diseases, transfusional iron overload and hematologic or solid malignancies were enrolled. ZPP was measured using the Av

The solid-substrate room-temperature phosphorescence (SS-RTP) of two commercially available metalloporphyrin compounds, zinc(II) protoporphyrin (ZnPP) and tin(IV) protoporphyrin (SnPP) has been studied. Strong and stable RTP signals of the two metalloporphyrins in neutral to weakly basic solutions can be simply induced on filter paper without addition of external heavy atom perturbers. Their emission bands appeared at 723 nm for ZnPP and 718 nm for SnPP at an excitation wavelength of 417 nm. Compared with SnPP, ZnPP is a better RTP probe for DNA because its RTP enhancement effect is much highe

Protoporphyrins are organic compounds with cyclic structure that are synthesised by a wide variety of organisms. In humans, these compounds are detected in blood and urine, with significantly higher levels in blood. Their potential as biomarkers of anemia and other diseases is currently being investigated, as their levels change according to the biochemical processes associated with the disease. The most widely used biomarker of anemia is serum ferritin, but it is unreliable in patients with inflammatory bowel disease (IBD) because its levels can be altered by acute inflammation and/or infecti

Superficial siderosis of the human central nervous system is caused by small continuous or recurrent subarachnoid hemorrhages that lead to the destructive deposition of hemosiderin. The excessive tissue iron derives from heme that is oxidized in a rate-limiting step by the enzyme heme oxygenase (HO), and especially the inducible form, HO-1. We postulated that competitive inhibition of HO by tin-protoporphyrin IX (SnPP) could prevent experimental superficial siderosis. Since synthetic metalloporphyrins do not cross the blood-brain barrier, SnPP was delivered directly into the cisterna magna. Ra

To investigate the mechanisms responsible for the distinct cutaneous manifestations of erythropoietic protoporphyria and porphyria cutanea tarda, the effects of protoporphyrin (PP) and uroporphyrin (URO), the predominant porphyrins in the respective disease, on mast cells were examined. Release of preformed and generated mediators was assessed by the release of radioactivity from cells labeled with [3H]serotonin and [14C]arachidonic acid, respectively. Clinically relevant doses of PP (25-500 ng/ml) and 396-407 nm irradiation (3-16 X 10(2)J/m2) induced maximal net release of preformed mediators

The effect of zinc (II) protoporphyrin IX (ZnPP) administration (40 mumol/kg, i.v.) on neonatal heme catabolism and the associated bilirubin production was investigated in rhesus (Macaca mulatta) neonates. Carbon monoxide excretion rates (VECO), tissue heme oxygenase activity, and plasma bilirubin concentrations were measured during a 26-hour postnatal period. In ZnPP-treated neonates (n = 4), VECO values were significantly (p = 0.002) diminished by 24% within 24 h. When compared to controls (n = 3), tissue heme oxygenase activity in ZnPP-treated neonates was greatly reduced in both liver (94%

Suppression of hyperbilirubinemia was studied in jaundiced adult and neonatal Gunn rats following treatment with tin-protoporphyrin. The effects of tin-protoporphyrin treatment on heme oxygenase, NADPH cytochrome c reductase, and biliverdin reductase activities were studied in adult jaundiced Gunn rat renal, hepatic and splenic tissues. Hepatic heme oxygenase activity was studied in nonjaundiced and jaundiced Gunn neonates. Significant decreases in plasma bilirubin levels were observed for both adult and neonatal rats treated with 50 or 100 mumol tin-protoporphyrin/kg body weight. Tin-protopor

Rhesus monkey infants were injected subcutaneously at birth with 12 to 100 mumol of tin-protoporphyrin IX, a competitive inhibitor of microsomal heme oxygenase. The elevated unconjugated serum bilirubin levels of the neonates receiving this metalloporphyrin rapidly declined to near adult levels by 24-30 h. Control neonates which received an injection of saline exhibited normal physiologic hyperbilirubinemias of from 3-6 mg/dl by 12-24 h as expected. These studies establish the effectiveness of tin-protoporphyrin IX in depressing bilirubin production and preventing physiologic hyperbilirubinemi

To further define the pathogenesis of protoporphyric liver disease, perfused rat livers received varying doses of protoporphyrin, and aberrations of hepatic ultrastructure and function were correlated. Results indicated that the relative canalicular volume was equally increased in all protoporphyrin-perfused livers; however, bile flow was only minimally diminished at the smallest protoporphyrin dose employed. Protoporphyrin injured microvilli at the sinusoidal pole and reduced the surface density of the endoplasmic reticulum in a dose-related fashion. No crystalline or amorphous material was d

The protoporphyrinemia of iron deficiency is well recognized. Clinically, information on the protoporphyrin/heme molar ratio in whole blood offers certain advantages over protoporphyrin measurement alone. A procedure for determining this ratio is reported. Protoporphyrin is extracted, solubilized, and measured fluorometrically. Heme (as hemin chloride) is precipitated with the blood proteins, the precipitate is dissolved in an alkaline/pyridine solvent, and the resulting bispyridine ferriprotoporphyrin is measured spectrophotometrically. The molar ratio of these two metabolites correlates well

Deficiency of ferrochelatase (FECH) in erythropoietic protoporphyria (EPP) leads to accumulation of its substrate, protoporphyrin IX (PPIX), the final intermediate in the heme biosynthesis pathway. PPIX is produced primarily in the bone marrow and subsequently delivered to the liver, where it can cause cholestatic liver injury and, in severe cases, liver failure. A key unresolved question is how circulating PPIX is transported to the liver to initiate hepatic damage. Given the structural similarity between PPIX and heme, we investigated whether the heme carrier hemopexin (HPX) mediates this pr

Photodynamic therapy (PDT) is a non-invasive treatment involving a photosensitizer (PS), light source, and tissue oxygen. Protoporphyrin IX (PpIX) is commonly used as a PS due to its tumor-targeting properties and phototoxicity. However, the physicochemical properties of PpIX foster self-aggregation, which is a challenge for its incorporation into pharmaceutical formulations. This study aimed to evaluate the solubility of PpIX in distinct solvent systems to support the development of novel pharmaceutical formulations. The shake-flask method was employed, using purified water, 50% ethanol (EtOH

OBJECTIVE: Iron deficiency anemia is known to impair cognitive and psychomotor development. The zinc protoporphyrin/heme (ZPP/H) ratio is a simple, accurate, and sensitive laboratory screening test that detects early iron depletion before the onset of anemia. The objective of this work was to evaluate this test in a primary pediatric practice setting.
METHODS: The iron status of a cohort of 361 children was screened during routine examinations at a community pediatric practice. Whole blood hemoglobin concentration, hematocrit ratio, serum transferrin saturation, ferritin concentration, and th

The most common screening tests for lead poisoning are reviewed. Stippled red cells are inconstant and appear too late. Blood lead concentrations are difficult to measure accurately and often are uncorrelated with body lead stores. Excess of delta aminolevulinic acid and coproporphyrin in urine does not appear early enough. A marked increase of erythrocyte protoporphyrin level practically clinches the diagnosis, later confirmed by increase of lead in urine after an EDTA lead-mobilization test. Measurements of erythrocyte protoporphyrin levels are easy to perform, more sensitive, more precociou

The substantial increases of hepatic, splenic, and renal heme oxygenase levels that occur shortly after birth in neonatal rats were prevented by a single administration of tin-protoporphyrin (10 micromoles per kilogram of body weight). With this treatment serum bilirubin levels declined within 24 hours to near-normal adult levels and remained low throughout the postnatal period. Zinc-protoporphyrin at doses up to 50-fold greater than the effective dose of tin-protoporphyrin did not prevent the immediate increases in tissue heme oxygenase activities and in serum bilirubin levels that occur post

Inhibition of superoxide dismutase by diethyldithiocarbamate or cyanide increases the rate of red blood cells lysis after irradiation in the presence of protoporphyrin IX. Catalase activity, which is decreased during the photohemolytic process, appears to be not essential for the lytic event. No relationship between catalase activity and hemolysis rate was found. Superoxide dismutase appears to prevent only in part catalase inactivation by singlet oxygen.

A detailed study of a Jewish erythrohepatic protoporphyria (EHP)-affected family with a sibship consisting of 2 brothers and a sister, all of whom manifested protodetermined hepatopathy which varied from relatively mild hepatic involvement in the sister to a fulminant fatal illness in the eldest brother. The nature and course of his illness as well as the autopsy findings are described in detail. This sibship is also unique in that the 2 brothers and biochemical evidence of a severe degree of G6PD deficiency, while the sister was shown to be a carrier.
