A time-resolved metabolomics study of potato cultivar responses to Pseudomonas syringae.

INTRODUCTION: The bacterial plant pathogen Pseudomonas syringae poses a threat to various crops, including potato (Solanum tuberosum). Its pathogenicity and virulence are enabled by, among other factors, its type III secretion system, which delivers effectors into the host plant. While differences in cultivar responses offer a sustainable control strategy, the underlying temporal metabolic mechanisms in potatoes remain poorly characterised. METHODS: This study employed an untargeted LC-MS metabolomics to investigate dynamic metabolic reprogramming in contrasting potato cultivars, Sifra and Va…

Quantification of Mitochondrial Pyrroline-5-Carboxylate in Arabidopsis Under Pseudomonas Syringae Infection.

Pyrroline-5 carboxylic acid (P5C) is an intermediary metabolite formed during proline synthesis and catabolism. Proline catabolism in two steps occurs in mitochondria where proline dehydrogenase (ProDH) catabolizes proline into P5C, and finally, pyrroline-5 carboxylate dehydrogenase catabolizes P5C into glutamate. Estimating mitochondrial P5C is essential to know how the metabolism of P5C in mitochondria via proline catabolism genes is responsible for causing disease and defense reaction in the case of Pseudomonas syringae-Arabidopsis interaction. There is no protocol available in the literatu…

Immunomodulation by the Pseudomonas syringae HopZ type III effector family in Arabidopsis.

Pseudomonas syringae employs a type III secretion system to inject 20-30 different type III effector (T3SE) proteins into plant host cells. A major role of T3SEs is to suppress plant immune responses and promote bacterial infection. The YopJ/HopZ acetyltransferases are a superfamily of T3SEs found in both plant and animal pathogenic bacteria. In P. syringae, this superfamily includes the evolutionarily diverse HopZ1, HopZ2 and HopZ3 alleles. To investigate the roles of the HopZ family in immunomodulation, we generated dexamethasone-inducible T3SE transgenic lines of Arabidopsis for HopZ family…

The MicroRNA156a-SPL module regulates kiwifruit resistance to Pseudomonas syringae pv. actinidiae.

MicroRNAs (miRNAs) are key regulators of plant growth, development, and immunity. However, their role in kiwifruit and other perennial fruit trees remains poorly characterized. Here, we demonstrate that Ac-miR156a functions as a negative regulator of kiwifruit resistance to Pseudomonas syringae pv. actinidiae (Psa). In the resistant cultivar "Jinkui," Ac-miR156a expression was significantly suppressed upon Psa infection, whereas it was markedly induced in the susceptible cultivar "Hongyang." Overexpression of miR156a increased kiwifruit susceptibility to Psa, while silencing miR156a via short-…

Risk assessment and resistance mechanism of Pseudomonas syringae pv. actinidiae to streptothricins.

BACKGROUND: Streptothricins (STs) are microbial-derived compounds effective against Pseudomonas syringae pv. actinidiae (Psa), the causal agent of kiwifruit bacterial canker. However, the risk of resistance development and underlying mechanisms in Psa remain unclear. This study aims to establish baseline sensitivity of Psa to STs, characterize resistance-associated phenotypes, and identify molecular resistance mechanisms to inform sustainable STs use in agriculture. RESULTS: Sensitivity testing of 186 Psa isolates from Chinese kiwifruit-producing regions revealed no naturally resistant strain…

CATION-CHLORIDE CO-TRANSPORTER 1 (CCC1) Mediates Plant Resistance against Pseudomonas syringae.

Plasma membrane (PM) depolarization functions as an initial step in plant defense signaling pathways. However, only a few ion channels/transporters have been characterized in the context of plant immunity. Here, we show that the Arabidopsis (Arabidopsis thaliana) Na+:K+:2Cl- (NKCC) cotransporter CCC1 has a dual function in plant immunity. CCC1 functions independently of PM depolarization and negatively regulates pathogen-associated molecular pattern-triggered immunity. However, CCC1 positively regulates plant basal and effector-triggered resistance to Pseudomonas syringae pv. tomato (Pst) DC30…

Functional characterization of AcWRKY94 in response to Pseudomonas syringae pv. actinidiae in kiwifruit.

WRKY transcription factors are essential for coping with various biotic stresses. Pseudomonas syringae pv. actinidiae (Psa)-induced kiwifruit canker is a major problem restricting kiwifruit yield. Nevertheless, it's unclear how the kiwifruit WRKY genes respond to Psa. Through genome-wide identification, 112 WRKY members were found in 'Hongyang' genome in this work. Promoter analysis revealed that there were many cis-acting elements associated with stress responses in the AcWRKY gene's promoter region. According to transcriptomic analysis, 90 of the AcWRKY genes were differently expressed follo…

Colonization of Microplastics by Different Strains of Pseudomonas Syringae Increases Ice-Nucleation Activity.

Microplastics (MPs) are increasingly detected throughout the atmosphere, raising questions about their persistence and influence on cloud-relevant ice nucleation processes. Recent studies suggest that MPs may act as ice-nucleating particles (INPs), potentially enhanced by biological colonization. Here, we quantify the ice nucleation activity (INA) of polystyrene (PS) and polyethylene (PE) MPs before and after surface aging and microbial colonization. Strains of Pseudomonas syringae, spanning a range of INA, were cultured onto pristine and aged 100 μm PS and PE MPs. Uncolonized MPs (0.5 to 100 …

Evaluation of several strategies for controlling canker plant disease caused by Pseudomonas syringae.

Pseudomonas syringae is a gram-negative bacterium that causes a diversity of diseases in numerous plants. Strategies to inhibit P. syringae growth include protective procedures; however, controlling the disease is complicated due to its rapid spread. Several antimicrobial agents can prevent this disease, such as chemical compounds, biological agents, secondary metabolites, nanoparticles, bacteriophages, and antimicrobial peptides (AMPs). The most effective way to control the disease is through chemical control. Using copper compounds and antibiotics is a conventional practice to decrease canke…

Isolation, Characterization, and Pathogenicity of Two Pseudomonas syringae Pathovars from Populus trichocarpa Seeds.

Pseudomonas syringae is a ubiquitous plant pathogen, infecting both woody and herbaceous plants and resulting in devastating agricultural crop losses. Characterized by a remarkable specificity for plant hosts, P. syringae pathovars utilize a number of virulence factors including the type III secretion system and effector proteins to elicit disease in a particular host species. Here, two Pseudomonas syringae strains were isolated from diseased Populustrichocarpa seeds. The pathovars were capable of inhibiting poplar seed germination and were selective for the Populus genus. Sequencing of the ne…

Ethylene Response Factor ERF11 Activates BT4 Transcription to Regulate Immunity to Pseudomonas syringae.

Pseudomonas syringae, a major hemibiotrophic bacterial pathogen, causes many devastating plant diseases. However, the transcriptional regulation of plant defense responses to P. syringae remains largely unknown. Here, we found that gain-of-function of BTB AND TAZ DOMAIN PROTEIN 4 (BT4) enhanced the resistance of Arabidopsis (Arabidopsis thaliana) to Pst DC3000 (Pseudomonas syringae pv. tomato DC3000). Disruption of BT4 also weakened the salicylic acid (SA)-induced defense response to Pst DC3000 in bt4 mutants. Further investigation indicated that, under Pst infection, transcription of BT4 is m…

Thiamine Is Required for Virulence and Survival of Pseudomonas syringae pv. tomato DC3000 on Tomatoes.

Pseudomonas syringae pv. tomato DC3000 (PstDC3000) is an important plant pathogen that infects tomatoes and Arabidopsis. Thiamine and its derivative thiamine pyrophosphate (TPP) are cofactors that play an important role in the growth and survival of many bacterial microorganisms. However, the role of thiamine-related genes has not been determined in PstDC3000. Hence, to investigate the role of TPP in growth, resistance to stresses, and virulence of PstDC3000, double and quadruple mutants of thiamine biosynthesis-related genes (thiD/E, thiS/G, and thiD/E/S/G deletion mutants) as well as a singl…

Age-related resistance in Arabidopsis is a developmentally regulated defense response to Pseudomonas syringae.

Age-related resistance (ARR) has been observed in a number of plant species; however, little is known about the biochemical or molecular mechanisms involved in this response. Arabidopsis becomes more resistant, or less susceptible, to virulent Pseudomonas syringae (pv tomato or maculicola) as plants mature (in planta bacterial growth reduction of 10- to 100-fold). An ARR-like response also was observed in response to certain environmental conditions that accelerate Arabidopsis development. ARR occurs in the Arabidopsis mutants pad3-1, eds7-1, npr1-1, and etr1-4, suggesting that ARR is a distin…

The rpoS gene in Pseudomonas syringae is important in surviving exposure to the near-UV in sunlight.

The near-UV component of sunlight decreased culturability of the leaf epiphyte and plant pathogen Pseudomonas syringae. Exposure of the wild-type cells for 4 h to UV-A and UV-B in sunlight was ten fold more detrimental than exposure to sunlight with just UV-A. Sensitivity to UV-A especially increased in a mutant of P. syringae lacking the global regulatory sigma factor, RpoS. No RpoS-mutant cells were culturable after 4 h of exposure to near-UV sunlight. These findings suggest that both UV-A and UV-B wavelengths cause damage to the bacterial cell and that the RpoS protein regulates protective …

Characterization of two novel bacteriophages (PSV6 and PSV3) as biocontrol agents against Pseudomonas syringae.

Pseudomonas syringae is a prevalent plant-pathogenic bacterium. Conventional control strategies, such as fungicides, copper-based compounds, and antibiotics, often result in elevated environmental toxicity and promote the emergence of bacterial resistance. Consequently, bacteriophage-based biocontrol has garnered considerable attention as an eco-friendly alternative. In this study, two novel bacteriophages, PSV6 and PSV3, were isolated from wastewater using Pseudomonas syringae pv. syringae (Pss) as the host strain. Transmission electron microscopy and whole-genome phylogenetic analyses indica…
