Harnessing membrane segregation of polysaccharide-degrading strains to mitigate fouling in membrane bioreactors: Insights from bacterial community ecology and metabolomics.

Polysaccharides are key biopolymers driving membrane fouling in membrane bioreactors (MBRs), and enhancing their in situ degradation represents a promising strategy for long-term fouling control. Here, we employed a "top-down" approach to isolate an efficient polysaccharide-degrading strain, Pseudonocardia sp. P-1, from activated sludge, and engineered a membrane-based segregator to retain the target strain while allowing metabolite exchange. When applied in an aerobic MBR (MBR-E, 1.66 kPa/day), the segregator markedly reduced fouling rates compared to the control MBR (MBR-C, 3.69 kPa/day, p <

[Purification, characterization and substrate selectivity analysis of transaminase from marine actinomycetes].

This study addresses the demand for green biosynthesis of chiral amine drugs by developing a novel R-selective amine transaminase (PamAT) derived from the marine actinomycete Pseudonocardia ammonioxydans. Following recombinant expression in Escherichia coli and purification via nickel-affinity chromatography, PamAT exhibited remarkable catalytic properties: strict stereoselectivity; a broad substrate spectrum; enhanced solvent tolerance (retaining>80% activity in 10% methanol or acetonitrile). Enzyme activity assays revealed that PamAT achieved the activity of (7.57±0.42) U/mg under mild condi

Changes in microbial composition during flue-cured tobacco aging and their effects on chemical composition: a review.

The aging is a crucial stage in tobacco processing, which contributes to the reduction of impurities and irritation, and the stabilization of the internal chemical composition of the leaves. However, it usually takes a long time (2-3 years) for the nature aging process of tobacco (20 °C-30 °C, relative humidity of 65-75%), which seriously affects the processing efficiency of tobacco. Microorganisms play an important role in the change of chemical composition and characteristic aromatic substances of tobacco. Acinetobacter, Sphingomonas Aspergillus, Bacilli, and Pseudonocardia is the main micro

A Synergistic Platform for Continuous Co-removal of 1,1,1-Trichloroethane, Trichloroethene, and 1,4-Dioxane via Catalytic Dechlorination Followed by Biodegradation.

Groundwater co-contaminated with 1,4-dioxane, 1,1,1-trichloroethane (TCA), and trichloroethene (TCE) is among the most urgent environmental concerns of the U.S. Department of Defense (DoD), U.S. Environmental Protection Agency (EPA), and industries related to chlorinated solvents. Inspired by the pressing need to remove all three contaminants at many sites, we tested a synergistic platform: catalytic reduction of 1,1,1-TCA and TCE to ethane in a H2-based membrane palladium-film reactor (H2-MPfR), followed by aerobic biodegradation of ethane and 1,4-dioxane in an O2-based membrane biofilm react

Diversity and Antimicrobial Activities of Actinobacteria Isolated from Mining Soils in Midelt Region, Morocco.

Multidrug-resistant bacteria have emerged as a serious global health threat that requires, more than ever before, an urgent need for novel and more effective drugs. In this regard, the present study sheds light on the diversity and antimicrobial potential of Actinobacteria isolates in mining ecosystems. We have indeed investigated the production of bioactive molecules by the Actinobacteria isolated from abandoned mining areas in Midelt, Morocco, where average contents of lead (Pb) and cadmium (Cd) are higher than normal world levels. One hundred and forty-five Actinobacteria isolates were isol

Metagenomic and microbiological analyses of historical manuscripts for bacterial community profiling and bacteria-related biodeterioration assessment.

Bacteria are important agents in the biodeterioration of cultural heritage objects, including historical manuscripts. Characterizing bacterial communities and generating robust microbiological data has therefore become crucial for conservation and restoration strategies. In this study, we investigated the bacterial communities associated with biodeterioration in six historical manuscripts using both culture-dependent and culture-independent (Illumina MiSeq) approaches. Culture-dependent methods yielded only 16 viable and culturable isolates, highlighting the limitations of traditional techniqu

Biodegradation Kinetics of 1,4-Dioxane in Chlorinated Solvent Mixtures.

This study investigated the impacts of individual chlorinated solvents and their mixtures on aerobic 1,4-dioxane biodegradation by Pseudonocardia dioxanivorans CB1190. The established association of these co-occurring compounds suggests important considerations for their respective biodegradation processes. Our kinetics and mechanistic studies demonstrated that individual solvents inhibited biodegradation of 1,4-dioxane in the following order: 1,1-dichloroethene (1,1-DCE) > cis-1,2-diochloroethene (cDCE) > trichloroethene (TCE) > 1,1,1-trichloroethane (TCA). The presence of 5 mg L(-1) 1,1-DCE

1,4-Dioxane-degrading consortia can be enriched from uncontaminated soils: prevalence of Mycobacterium and soluble di-iron monooxygenase genes.

Two bacterial consortia were enriched from uncontaminated soil by virtue of their ability to grow on 1,4-dioxane (dioxane) as a sole carbon and energy source. Their specific dioxane degradation rates at 30°C, pH = 7 (i.e. 5.7 to 7.1 g-dioxane per g-protein per day) were comparable to those of two dioxane-metabolizing archetypes: Pseudonocardia dioxanivoransCB1190 and Mycobacterium dioxanotrophicusPH-06. Based on 16S rRNA sequencing, Mycobacterium was the dominant genus. Acetylene inhibition tests suggest that dioxane degradation was mediated by monooxygenases. However, qPCR analyses targeting

Co-contaminant effects on 1,4-dioxane biodegradation in packed soil column flow-through systems.

Biodegradation of 1,4-dioxane was examined in packed quartz and soil column flow-through systems. The inhibitory effects of co-contaminants, specifically trichloroethene (TCE), 1,1-dichloroethene (1,1-DCE), and copper (Cu2+) ions, were investigated in the columns either with or without bioaugmentation with a 1,4-dioxane degrading bacterium Pseudonocardia dioxanivorans CB1190. Results indicate that CB1190 cells readily grew and colonized in the columns, leading to significant degradation of 1,4-dioxane under oxic conditions. Degradation of 1,4-dioxane was also observed in the native soil (witho

Polyene macrolide biosynthesis in streptomycetes and related bacteria: recent advances from genome sequencing and experimental studies.

The polyene macrolide group includes important antifungal drugs, to which resistance does not arise readily. Chemical and biological methods have been used in attempts to make polyene antibiotics with fewer toxic side effects. Genome sequencing of producer organisms is contributing to this endeavour, by providing access to new compounds and by enabling yield improvement for polyene analogues obtained by engineered biosynthesis. This recent work is also enhancing bioinformatic methods for deducing the structures of cryptic natural products from their biosynthetic enzymes. The stereostructure of

The lung microbiota in early rheumatoid arthritis and autoimmunity.

BACKGROUND: Airway abnormalities and lung tissue citrullination are found in both rheumatoid arthritis (RA) patients and individuals at-risk for disease development. This suggests the possibility that the lung could be a site of autoimmunity generation in RA, perhaps in response to microbiota changes. We therefore sought to test whether the RA lung microbiome contains distinct taxonomic features associated with local and/or systemic autoimmunity. METHODS: 16S rRNA gene high-throughput sequencing was utilized to compare the bacterial community composition of bronchoalveolar lavage fluid (BAL)

Deciphering environmental resistome and mobilome risks on the stone monument: A reservoir of antimicrobial resistance genes.

Antimicrobial resistance (AMR) in the environment has attracted increasing attention as an emerging global threat to public health. Stone is an essential ecosystem in nature and also an important material for human society, having architectural and aesthetic values. However, little is known about the AMR in stone ecosystems, particularly in the stone monument, where antimicrobials are often applied against biodeterioration. Here, we provide the first detailed metagenomic study of AMR genes across different types of biodeteriorated stone monuments, which revealed abundant and diverse AMR genes

Thioredoxin reductase from the malaria mosquito Anopheles gambiae.

Human pathogenic diseases received much attention recently due to their uncontrolled spread of antimicrobial resistance (AMR) which causes several threads every year. Effective alternate antimicrobials are urgently required to combat those disease causing infectious microbes. Halophilic actinobacteria revealed huge potentials and unexplored cultivable/non-cultivable actinobacterial species producing enormous antimicrobials have been proved in several genomics approaches. Root vertical spatial stress drives targeted enrichment of rhizospheric aerobic denitrifying microorganisms for enhanced nitrogen removal in constructed wetlands.

Aerobic denitrification (AD) can overcome the limitations of traditional denitrification in constructed wetlands (CWs) caused by heterogeneous dissolved oxygen distribution and carbon source scarcity, yet effectively enriching functional aerobic denitrifying microorganisms (ADMs) to enhance nitrogen removal efficiency remains a challenge. In this study, the microbial community structure and nitrogen removal pathway were regulated in small-scale CWs by root vertical spatial stress and different plants species. Reducing substrate depth from 60 cm to 10 cm significantly increased the average root

Characterization of multiple trichloroethene, cis-dichloroethene and 1,1-dichloroethene degrading propanotrophic communities.

Aerobic cometabolism offers a viable strategy for the remediation of chlorinated solvent plumes at oxic sites where anaerobic approaches are limited. Here, propane-enriched mixed cultures (derived from agricultural soils and an impacted site sediment) which previously degraded 1,4-dioxane, were evaluated for their capacity to also degrade trichloroethene (TCE), cis-1,2-dichloroethene (cDCE), and 1,1-dichloroethene (1,1-DCE) over successive transfers. Sustained biodegradation of TCE and cDCE was observed across multiple enrichments and cultures enriched on one compound generally degraded the ot

Shaping antibiotic resistance gene fate in soil-plant systems: Dual roles of biochar physicochemical traits mediated by pyrolysis conditions.

Antibiotic resistance genes (ARGs), emerging contaminants spreading via horizontal gene transfer, threaten ecosystems and human health. Biochar (BC) is a widely used agricultural soil amendment, yet its effects on ARG dissemination remain controversial, likely dependent on pyrolysis conditions. This study applied wheat straw BC prepared under three distinct pyrolysis conditions, including open-flame combustion (BC-ZJ), 500°C hypoxic pyrolysis, and 500°C anaerobic pyrolysis, to a Brassica rapa L.-soil system for exploring ARG transfer impacts and mechanisms. BC application increased plant stem/

Longitudinal study of mammary microbiota dynamics and mastitis prevention in Holstein cows by dry-off strategy.

INTRODUCTION: Conducting a dry-off period during the late lactation phase in dairy cows can reduce the incidence of clinical mastitis both during the dry period and after subsequent calving. The primary dry-off methods include the application of antibiotics alone or in combination with internal teat sealants. A thorough understanding of the mammary gland microbiota composition before and after dry-off is essential for developing scientifically sound dry-off protocols in practical dairy production. METHODS: Five Holstein cows approaching dry-off were selected for this study. The day of calving
