In North America, crown rust has emerged as a disease of Kentucky bluegrass (Poa pratensis) and may cause significant damage. After observing crown rust on Poa spp. in Minnesota and on three East Asian buckthorns (Rhamnus crenata, R. davurica, and R. japonica) in North Dakota, we sequenced one to four loci of nine samples for identification and to determine their placement within Puccinia series Coronata. Earlier studies had shown that the P. coronata var. avenae f. sp. avenae clade caused crown rust of Kentucky bluegrass in North America, but phylogenetic analysis identified our samples as a

Sterol 14α-Demethylase Cytochrome P450 (CYP51) protein involved in ergosterol biosynthesis pathways turn out to be a crucial target for the fungicidal compound. However, the recognition mechanism and dynamic behavior of CYP51 in wheat leaf rust pathogen, Puccinia triticina, is still obscure. Previously, a mutation at position 134 (Y134F) was reported in five European isolates of P. triticina, conversely, structural basis of this mutation remains unclear. To address this problem, three-dimensional structure of CYP51 protein from P. triticina was successfully built using homology modeling approa

Southern corn rust is a destructive maize disease caused by Puccinia polysora Underw that can lead to severe yield losses. However, genomic information and microsatellite markers are currently unavailable for this disease. In this study, we generated a total of 27,295,216 high-quality cDNA sequence reads using Illumina sequencing technology. These reads were assembled into 17,496 unigenes with an average length of 1015 bp. The functional annotation indicated that 8113 (46.37%), 1933 (11.04%) and 5516 (31.52%) unigenes showed significant similarity to known proteins in the NCBI Nr, Nt and Swiss

Yellow stripe-like (YSL) transporters are required for the transportation of metal-phytosiderophores and are structurally related to metal-nicotianamine complexes. Some studies also reported the involvement of YSL transporters in pathogen-induced defense. However, the molecular mechanisms of YSL genes involved in biotic stress responses are still not clear, especially in cereal crops. This study aimed to functionally characterize TaYS1A during the interaction of wheat and Puccinia striiformis f. sp. tritici (Pst), the causal agent of stripe rust disease. TaYS1A was localized in the cell membra

Wheat stem rust, caused by Puccinia graminis f. sp. tritici (Pgt), threatens food security owing to the pathogen's high virulence and evolutionary variability, as well as the risk of selecting fungicide-resistant forms. For the first time in Russia, we performed a comparative analysis of virulence and genetic diversity in fungicide-target genes (SdhA-SdhD, Cyp51) in 35 monopustular Pgt isolates collected from regions with contrasting fungicide pressure (Saratov, n = 12; Chelyabinsk, n = 12; Moscow, n = 11). Phenotyping on an extended set of nearly isogenic lines carrying different Sr genes rev

Glycerol-3-phosphate (G3P) is a proposed regulator of plant defense signaling in basal resistance and systemic acquired resistance (SAR). The GLY1-encoded glycerol-3-phosphate dehydrogenase (G3PDH) and GLI1-encoded glycerol kinase (GK) are two key enzymes involved in the G3P biosynthesis in plants. However, their physiological importance in wheat defense against pathogens remains unclear. In this study, quantification analysis revealed that G3P levels were significantly induced in wheat leaves challenged by the avirulent Puccinia striiformis f. sp. tritici (Pst) race CYR23. The transcriptional

SUMMARY Plant immunity against the majority of microbial pathogens is conveyed by a phenomenon termed non-host resistance (NHR). This multifactorial trait provides durable protection against a given pathogen species. We investigated the molecular basis of NHR in Arabidopsis against the wheat leaf rust pathogen, Puccinia triticina (Ptr). Urediospores germinated with high efficiency and grew randomly over the Arabidopsis leaf surface. However, only 12% of urediospores produced a germ tube that successfully located a stoma and just 0.2% of urediospores went on to produce a haustorium within a pen

In this research, the wheat cultivar 'Lovrin 10' and Puccinia triticina races 165 and 260 were used to constitute compatible and incompatible combinations to investigate the relationship between NO and H2O2 and between NO and calcium (Ca(2+)) signaling in the cell defense process by pharmacological means. The specific fluorescent probe DAF-FM DA was coupled with confocal laser scanning microscopy and used to label intracellular nitric oxide (NO) and monitoring the real-time NO dynamics during the processes of wheat defense response triggered by P. triticina infection. The results showed that a

Wheat stripe rust (yellow rust), a devastating disease caused by the phytopathogen Puccinia striiformis f. sp. tritici (Pst), poses a major threat to global wheat (Triticum aestivum L.) production. Under ongoing climate warming, the highly virulent Pst race CYR34 has become predominant in China and exhibits increased tolerance to elevated temperatures. Although temperature is known to influence Pst urediniospore germination, the molecular mechanisms underlying temperature sensitivity during this process remain poorly understood. In this study, we combined histological examinations with transcr

MYB transcription factors are a large family of proteins involved in the regulation of secondary metabolism and cell shape, the enhancement of disease resistance and the response to different stresses. In this study, the role of TaMYB4 in wheat against biotic and abiotic stresses was investigated. TaMYB4 was cloned from wheat cv. Suwan11 [the leaves were infected with Puccinia striiformis f.sp. tritici (Pst)]; the TaMYB4 protein is 243 amino acids in length. In addition, TaMYB4 exhibited high similarity with BdMYB4 from Brachypodium distachyon, which was also identified as a member of the R2R3

Isolates of the most important Puccinia species that have been reported on Chrysanthemum x morifolium were collected and the sequences of their ribosomal DNA internal transcribed spacers ITS1 and ITS2 were determined and used as phylogenetic markers. The focus of this study was on Puccinia horiana, due to its quarantine status and its impact in commercial chrysanthemum production. Three technical adjustments were needed to reliably obtain the nucleotide sequences starting from fresh or dried samples. The complete rDNA ITS nucleotide sequences of P. horiana, Puccinia chrysanthemi, and Puccinia

Wheat stripe rust, caused by the obligate biotroph Puccinia striiformis f. sp. tritici (Pst), remains one of the most destructive fungal diseases threatening global wheat production. Beyond traditional control through chemical application and resistant cultivar breeding, rapid advances in molecular biology have substantially reshaped our understanding of the intricate molecular dialogue between wheat and Pst. In recent years, research has increasingly focused on Pst pathogenicity factors, wheat immune perception, and the multilayered signaling networks that orchestrate defense responses. Parti

Stripe rust of wheat caused by Puccinia striiformis f. sp. tritici (Pst) remains a major constraint to wheat production in India due to the rapid evolution and frequent emergence of virulent pathotypes. Rapid and reliable discrimination of Pst pathotypes is essential for effective resistance deployment and surveillance. In the present study, transcriptome-wide simple sequence repeats (SSRs) and single nucleotide polymorphisms (SNPs) were exploited to develop and validate molecular markers for pathotype-specific detection of Pst pathotypes prevalent in North India (110S119, 238S119, 46S119, 110

Wheat yellow rust, caused by Puccinia striiformis f. sp. tritici (PST), is one of the most important wind-borne diseases in all wheat-growing regions, and its occurrence can lead to devastating yield losses in wheat. In China, the wheat fields in Gansu act as an inoculum reservoir during summer periods and provide PST spores for wheat in the fall. The wheat fields in Qinghai provide large amounts of oversummering inocula. The exchange and migration of spores between the two regions are essential to ensure the persistence of PST. To confirm this relationship, we studied the genetic diversity, s

The MYB transcription factor family is one of the largest gene families in plants and plays critical roles in how plants respond to biotic and abiotic stresses. However, the functions of MYB members in high-temperature seedling plant (HTSP) resistance in wheat to Puccinia striiformis f. sp. tritici (Pst) are not well understood. Previously, TaMYB73, an MYB transcription factor, was identified through an RNA-sequencing study of the wheat cultivar Xiaoyan 6 under infection by Pst. In this work, we characterized the molecular function of TaMYB73. The expression TaMYB73 was upregulated following P

Stripe rust, caused by Puccinia striiformis f. sp. tritici (Pst), is a devastating disease of wheat (Triticum aestivum) worldwide. Wheat high-temperature seedling plant (HTSP) resistance to Pst is non-race-specific and durable. WRKY transcription factors have been proven to play important roles in plant defence responses to attacks by several pathogens. However, there is no direct evidence as to whether WRKY transcription factors play a role in HTSP resistance to Pst. We isolated a WRKY gene, named TaWRKY70, from wheat cultivar Xiaoyan 6. The expression level of TaWRKY70 was increased signific

Wheat stripe rust, caused by Puccinia striiformis f. sp. tritici (Pst), remains a major threat to global wheat production, which necessitates a better understanding of the molecular mechanisms underlying resistance. WRKY transcription factors are pivotal regulators of plant immunity; however, their specific functions in the wheat-Pst pathosystem remain poorly characterized. In this study, we identified and functionally characterized a WRKY transcription factor gene, TaWRKY24, in wheat. Expression profiling revealed that TaWRKY24 was continuously induced upon Pst inoculation. Subcellular locali

Numerous studies have found that sucrose (Suc) metabolism plays a crucial role in the environmental stress response of many plant species. The majority of Suc metabolism-associated reports refer to acid invertases (Ac-Invs). However, alkaline/neutral Invs (A/N-Invs) have been poorly studied. In this study, a wheat A/N-Inv gene, Ta-A/N-Inv1, with three copies located on chromosomes 4A, 4B, and 4D, was cloned from a wheat-Puccinia striiformis f. sp. tritici (Pst) interaction cDNA library. Transcripts of the three Ta-A/N-Inv1 copies were up-regulated in wheat leaves that were infected by Pst or h

Stem rust, caused by the fungus Puccinia graminis f. sp. tritici (Pgt), is a harmful disease affecting grain crops. The traditional way to combat this and other infectious plant diseases is to use chemical pesticides. Biopesticides, as well as plant disease resistance inducers - in particular those based on chitosan, a derivative of chitin - are increasingly being considered as an effective and safe alternative. Recently, a globular form of chitosan, Novochizol, has been developed, which has a number of advantages and has shown its effectiveness in preliminary field and laboratory experiments.

Puccinia coronata f. sp. avenae (Pca) is an important fungal pathogen causing crown rust that impacts oat production worldwide. Genetic resistance for crop protection against Pca is often overcome by the rapid virulence evolution of the pathogen. This study investigated the factors shaping adaptive evolution of Pca using pathogen populations from distinct geographic regions within the United States and South Africa. Phenotypic and genome-wide sequencing data of these diverse Pca collections, including 217 isolates, uncovered phylogenetic relationships and established distinct genetic compositi
