Pragmatic representations of self- and others' action in the monkey putamen.

Social coordination in primates relies on parieto-frontal networks encoding self- and others' actions. These areas send convergent projections to the putamen, but its role in representing self- and others' actions remains unknown. We recorded neuronal activity from anatomically characterized putamen regions of two male macaques during a Mutual Action Task (MAT), where a monkey and a human took turns grasping a multi-affordance object based on sensory cues. Cortico-striatal synaptic input, indexed by local field potentials, mirrored known cortical dynamics during sensory instructions and moveme

Structural and functional abnormality of the putamen in children with developmental dyslexia.

There is currently debate with regards to the role of phonological deficit in Chinese reading difficulty, even though some researchers have suggested that the deficit of phonological processing is also a signature of developmental dyslexia in Chinese, as has been found in alphabetic languages. In this study, we examined the brain mechanisms of phonological deficit in Chinese children with developmental dyslexia (DD) during an auditory rhyming judgment task. First, we examined structural differences in Chinese dyslexia by comparing gray and white matter volume in Chinese children with DD, age-m

Accelerated aging of the putamen in patients with major depressive disorder.

BACKGROUND: Growing evidence indicates that major depressive disorder (MDD) is characterized by accelerated biological aging, including greater age-related changes in physiological functioning. The disorder is also associated with abnormal neural reward circuitry, particularly in the basal ganglia (BG). Here we assessed age-related changes in BG volume in both patients with MDD and healthy control participants. METHODS: We obtained whole-brain T1-weighted images from patients with MDD and healthy controls. We estimated grey matter volumes of the BG, including the nucleus accumbens, caudate, p

Disorganized somatotopy in the putamen of patients with focal hand dystonia.

OBJECTIVE: To explore the selectivity of neuronal somatotopic representation in the striatum of patients with unilateral task-specific dystonia of the right arm. MATERIALS AND METHODS: The authors used fMRI in 14 right-handed dystonic subjects to examine putaminal organization. Subjects performed flexion/extension of the right and left fingers and toes, and contraction of the lips. RESULTS: Compared to healthy volunteer subjects, dystonic subjects had altered somatotopic organization in the left putamen, contralateral to the affected hand. Disease severity correlated with underactivation and

Visualization of ischemic insult in caudate putamen with beta-CIT.

UNLABELLED: Dopamine (DA) has been considered to play an important role in the development of ischemic neuronal injury in the caudate putamen (CPu). The goal of this study was to examine the change in the dopamine transporter (DAT) after ischemic insult in CPu. METHODS: Male Mongolian gerbils (n = 10) were exposed to 10-min forebrain ischemia. Animals were decapitated 24 hr (n = 5) and 96 hr (n = 5) after ischemia. The change in the amount of DAT binding sites in CPu was evaluated by in vitro autoradiography with [125I]-beta-CIT (3 beta-(4-iodophenyl)tropan-2 beta-carboxylic acid methyl ester

Aesthetic appreciation correlates positively with putamen and inferior temporal gyrus.

In recent years, the effects of art on the human brain and cognitive function have been attracting attention. However, the relationship between the brain structure of healthy middle-aged people and artistic activity remained unclear. In this study, a cross-sectional analysis of 73 middle-aged men and women who considered themselves healthy revealed that gray matter volume (GMV) of the putamen and inferior temporal gyrus correlated with aesthetic appreciation measured by questionnaire. This is the first study to show that art may connect to more people's brain structures than previously thought

Altered putamen connectivity in patients with neurological post-COVID condition.

Although the exact aetiology of the post-COVID condition is still under investigation, there is increasing evidence for white matter pathology in patients with persistent cognitive and fatigue symptoms following an infection with SARS-CoV-2. Still, to date there are no studies that investigated the white matter connectome in patients with post-COVID condition. Based on previous findings, we analyzed the structural connectome of these patients, with a focus on the thalamus and basal ganglia. In this cross-sectional study, 43 patients (34 women, 9 men) and 41 (33 women, 8 men) healthy control pa

Widespread alterations in microRNA biogenesis in human Huntington's disease putamen.

Altered microRNA (miRNA) expression is a common feature of Huntington's disease (HD) and could participate in disease onset and progression. However, little is known about the underlying causes of miRNA disruption in HD. We and others have previously shown that mutant Huntingtin binds to Ago2, a central component of miRNA biogenesis, and disrupts mature miRNA levels. In this study, we sought to determine if miRNA maturation per se was compromised in HD. Towards this end, we characterized major miRNA biogenesis pathway components and miRNA maturation products (pri-miRNA, pre-miRNA, and mature)

Putamen dopamine synthesis, vesicular storage, and metabolism in Parkinson disease.

BACKGROUND: Severe putamen dopamine depletion in Parkinson disease (PD) has been attributed to nigrostriatal denervation; however, there are also functional abnormalities in extant terminals (the "sick-but-not-dead" phenomenon). Rates of intra-neuronal processes of synthesis, storage, and metabolism of dopamine complexly influence releasable dopamine stores but have not yet been systematically estimated. METHODS: Post-mortem empirical data were available about putamen tissue contents of 7 reactants, including the autotoxic dopamine metabolite 3,4-dihydroxyphenylacetaldehyde (DOPAL). We constr

Decreased [18F]spiperone binding in putamen in idiopathic focal dystonia.

In this study we have investigated the pathophysiology of two idiopathic focal dystonias: hand cramp with excessive cocontractions of agonist and antagonist hand or forearm muscles during specific tasks, such as writing, and facial dystonia manifested by involuntary eyelid spasms (blepharospasm) and lower facial and jaw spasms (oromandibular dystonia). We used positron emission tomography (PET) to measure the in vivo binding of the dopaminergic radioligand [18F]spiperone in putamen in 21 patients with these two focal dystonias and compared the findings with those from 13 normals. We measured r

Single cell studies of the primate putamen. I. Functional organization.

In order to clarify the functional organization of the putamen and the nature of sensory inputs to this structure we studied the relation of single cell activity to active movements and somatosensory stimulation in the awake primate. Neurons (N = 707) were categorized on the basis of their relation to active movements or responses to sensory stimulation of individual body parts. 38% of neurons studied were related to the arm, 9% to the leg, 11% to the mouth or face, and 3% to axial portions of the body. The remaining neurons exhibited non-specific activation which could not be confidently loca

Localization and connections of the tail of caudate and caudal putamen in mouse brain.

The neural circuits of the striatum (caudate and putamen) constitute a crucial component of the extrapyramidal motor system, and dysfunction in these circuits is correlated with significant neurological disorders including Parkinson's disease and Huntington's disease. Many previous studies in rodents revealed the neural connections of the rostral and intermediate parts of the striatum, but relatively fewer studies focused on the caudal striatum, which likely contains both the tail of caudate (CaT) and caudal putamen (PuC). In this study, we investigate the gene markers for the CaT and PuC and

Laminar Origin of Corticostriatal Projections to the Motor Putamen in the Macaque Brain.

In the macaque brain, projections from distant, interconnected cortical areas converge in specific zones of the striatum. For example, specific zones of the motor putamen are targets of projections from frontal motor, inferior parietal, and ventrolateral prefrontal hand-related areas and thus are integral part of the so-called "lateral grasping network." In the present study, we analyzed the laminar distribution of corticostriatal neurons projecting to different parts of the motor putamen. Retrograde neural tracers were injected in different parts of the putamen in 3 Macaca mulatta (one male)

A Case of Transient Global Amnesia with Small Left Putamen Infarction.

A 67-year-old right-handed woman experienced a sudden anterograde and retrograde short-term memory deficit that recovered on the next day. Magnetic resonance imaging (MRI) showed a spotty high-intensity lesion in the left putamen in the diffusion-weighted and fluid-attenuated inversion recovery images. Transient global amnesia due to lacunar infarction of the left putamen was diagnosed. Transient global amnesia is characterized by a sudden onset of anterograde amnesia that disappears within 24 hours. The cause of transient global amnesia is still uncertain. Some studies with MRI showed small l

Sustained increase of PKA activity in the postcommissural putamen of dyskinetic monkeys.

Levodopa-induced dyskinesias (LID) are a frequent complication of Parkinson's disease pharmacotherapy that causes significant disability and narrows the therapeutic window. Pharmacological management of LID is challenging partly because the precise molecular mechanisms are not completely understood. Here, our aim was to determine molecular changes that could unveil targetable mechanisms underlying this drug complication. We examined the expression and downstream activity of dopamine receptors (DR) in the striatum of 1-methyl-4-phenyl-1,2,3,6 tetrahydropiridine (MPTP)-lesioned monkeys with and

Geometric properties of caudate and putamen mark the progression of Huntington's disease.

Volumetric MRI of the caudate and putamen is a robust biomarker of striatal degeneration in Huntington's disease (HD). Striatal degeneration may be reflected beyond volume alone, in additional shape and intensity alterations. We evaluated five novel morphometric biomarkers of the caudate and putamen-curvature, flatness, eccentricity, elongation, and focal assessment of divergence estimation (FADE; T1-weighted signal shift)-using cross-sectional and longitudinal MRI data from healthy controls (N = 100), individuals with premanifest HD (preHD; N = 100), and individuals with early manifest HD (ea

Hyperuricaemia is associated with smaller volumes in the caudate nucleus head and putamen.

Hyperuricaemia is a risk factor for gout, kidney diseases, and cerebrovascular diseases. Uric acid (UA) is also known as an antioxidant and has been suggested to inhibit the progression of neurodegenerative diseases, such as Parkinson's disease. However, only a few studies have focused on the potential effects of UA on the basal ganglia. This study aimed to measure UA levels and basal ganglia volumes in community-dwelling adults and to evaluate the association thereof. Blood UA levels and brain MRI data were collected from individuals who underwent brain health checkups between January 2015 an

Dopaminergic dysfunction in the left putamen of patients with major depressive disorder.

INTRODUCTION: Dopaminergic transmission impairment has been identified as one of the main neurobiological correlates of both depression and clinical symptoms commonly associated with its spectrum such as anhedonia and psychomotor retardation. OBJECTIVES: We examined the relationship between dopaminergic deficit in the striatum, as measured by 123I-FP-CIT SPECT imaging, and specific psychopathological dimensions in patients with major depressive disorder. METHODS: To our knowledge this is the first study with a sample of >120 subjects. After check for inclusion and exclusion criteria, 121 (67

Imbalance between the caudate and putamen connectivity in obsessive-compulsive disorder.

BACKGROUND: Compulsive behaviors in obsessive-compulsive disorder (OCD) have been suggested to result from an imbalance in cortico-striatal connectivity. However, the nature of this impairment, the relative involvement of different striatal areas, their imbalance in genetically related but unimpaired individuals, and their relationship with cognitive dysfunction in OCD patients, remain unknown. METHODS: In the current study, striatal (i.e., caudate and putamen) whole-brain connectivity was computed in a sample of OCD patients (OCD, n = 62), unaffected first-degree relatives (UFDR, n = 53) and
