Functional response of hippocampal CA1 pyramidal cells to neonatal hypoxic-ischemic brain damage.

Perinatal hypoxic-ischemic (H-I) is a major cause of brain injury in the newborn. The hippocampus is more sensitive to H-I injury than the other brain regions. It is believed that H-I brain damage causes a loss of neurons in the central nervous system. The patterns of neuronal death include apoptosis and necrosis. With regard to the responses of neurons, the neural functional changes should be earlier than the morphologic changes. The aim of the present study is to evaluate the electrophysiological characteristics and the synaptic transmission functions. Seven-day-old Sprague-Dawley rat pups w

Circuit-specific and neuronal subcellular-wide E-I balance in cortical pyramidal cells.

We used ChR2-assisted circuit mapping (CRACM) to examine neuronal/compartmental excitatory and inhibitory synaptic balance (E-I balance) in pyramidal cells (PCs) located in several brain regions (including both neocortices and paleocortices). Within the vS1, different inputs on the same neurons, or the same inputs formed on different targets, induced different E/I ratios. E/I ratios in PCs from different regions were largely different. Chemogenetic silencing of somatostatin (SOM)- or parvalbumin (PV)-containing interneurons (INs) while optogenetically activating long-range M1 inputs demonstrat

Synaptic properties of layer VI inverted pyramidal cells in the rodent somatosensory cortex.

The properties of specific cortical cell types enable greater understanding of how cortical microcircuits process and transmit sensory, motor, and cognitive information. Previous reports have characterized the intrinsic properties of the inverted pyramidal cell (IPC) where the most prominent dendrite is orientated towards the cortical white matter. Using whole cell patch clamp recordings from rat and mouse somatosensory cortex in conjunction with electric microstimulation of the white matter we characterized the synaptic inputs onto IPCs and the more common upright pyramidal cell (UPC) in the

Culture of pyramidal neural precursors, neural stem cells, and fibroblasts on various biomaterials.

A combinatory approach using biomaterials together with cells may improve the efficacy of cell therapy for treatment of various diseases/indications. In the current study, we cultured pyramidal neural precursors (PNPs), neural stem cells (NSCs), and fibroblasts on different materials that included fibrin, collagen, hyaluronic acid (HA), sciatic nerves, and matrigel, to search for the most suitable biomaterial for culture of each cell type. Collagen was fabricated in both an aligned collagen-poly (lactic-co-glycolic acid) (PLGA) composite and an alveolate form; fibrin and hyaluronic acid were m

Impact of dendritic size and dendritic topology on burst firing in pyramidal cells.

Neurons display a wide range of intrinsic firing patterns. A particularly relevant pattern for neuronal signaling and synaptic plasticity is burst firing, the generation of clusters of action potentials with short interspike intervals. Besides ion-channel composition, dendritic morphology appears to be an important factor modulating firing pattern. However, the underlying mechanisms are poorly understood, and the impact of morphology on burst firing remains insufficiently known. Dendritic morphology is not fixed but can undergo significant changes in many pathological conditions. Using computa

Hippocalcin gates the calcium activation of the slow afterhyperpolarization in hippocampal pyramidal cells.

In the brain, calcium influx following a train of action potentials activates potassium channels that mediate a slow afterhyperpolarization current (I(sAHP)). The key steps between calcium influx and potassium channel activation remain unknown. Here we report that the key intermediate between calcium and the sAHP channels is the diffusible calcium sensor hippocalcin. Brief depolarizations sufficient to activate the I(sAHP) in wild-type mice do not elicit the I(sAHP) in hippocalcin knockout mice. Introduction of hippocalcin in cultured hippocampal neurons leads to a pronounced I(sAHP), while ne

Coincident spiking activity induces long-term changes in inhibition of neocortical pyramidal cells.

In pyramidal cells, induction of long-term potentiation (LTP) and long-term depression (LTD) of excitatory synaptic transmission by coincidence of presynaptic and postsynaptic activity is considered relevant to learning processes in vivo. Here we show that temporally correlated spiking activity of a pyramidal cell and an inhibiting interneuron may cause LTD or LTP of unitary IPSPs. Polarity of change in synaptic efficacy depends on timing between Ca(2+) influx induced by a backpropagating train of action potentials (APs) in pyramidal cell dendrites (10 APs, 50 Hz) and subsequent activation of

Interaction between synaptic excitation and slow afterhyperpolarization current in rat hippocampal pyramidal cells.

1. Whole cell recordings from CA1 pyramidal cells were performed to investigate the interaction between excitatory postsynaptic potentials (EPSPs) or currents (EPSCs), and the slow Ca(2+)-dependent K(+) current, I(sAHP). Blockers of the slow afterhyperpolarization (sAHP) such as isoprenaline (ISO) or noradrenaline (NA) reduced the hyperpolarization that followed a short train of EPSPs, and slowed the decay of summated EPSPs or EPSCs. 2. ISO/NA action on synaptic responses was observed in the absence of action potentials, but was curtailed by Ca(2+) chelation (10 mM EGTA in the electrode) and w

Pyramidal cells of the frontal lobe: all the more spinous to think with.

The basal dendritic arbors of pyramidal cells in prefrontal areas 10, 11, and 12 of the macaque monkey were revealed by intracellular injection in fixed, flat-mounted, cortical slices. The size, number of branches, and spine density of the basal dendrites were quantified and compared with those of pyramidal cells in the occipital, parietal, and temporal lobes. These analyses revealed that cells in the frontal lobe were significantly more spinous than those in the other lobes, having as many as 16 times more spines than cells in the primary visual area (V1), four times more those in area 7a, an

Inhibition of voltage-gated calcium channels by fluoxetine in rat hippocampal pyramidal cells.

Fluoxetine, an antidepressant which is used world-wide, is a prominent member of the class of selective serotonin re-uptake inhibitors. Recently, inhibition of voltage-gated Na(+) and K(+) channels by fluoxetine has also been reported. We examined the effect of fluoxetine on voltage-gated calcium channels using the patch-clamp technique in the whole-cell configuration. In hippocampal pyramidal cells, fluoxetine inhibited the low-voltage-activated (T-type) calcium current with an IC(50) of 6.8 microM. Fluoxetine decreased the high-voltage-activated (HVA) calcium current with an IC(50) between 1

Action potential repolarization and a fast after-hyperpolarization in rat hippocampal pyramidal cells.

1. The repolarization of the action potential, and a fast after-hyperpolarization (a.h.p.) were studied in CA1 pyramidal cells (n = 76) in rat hippocampal slices (28-37 degrees C). Single spikes were elicited by brief (1-3 ms) current pulses, at membrane potentials close to rest (-60 to -70 mV). 2. Each action potential was followed by four after-potentials: (a) the fast a.h.p., lasting 2-5 ms; (b) an after-depolarization; (c) a medium a.h.p., (50-100 ms); and (d) a slow a.h.p. (1-2 s). Both the fast a.h.p. and the slow a.h.p. (but not the medium a.h.p.) were inhibited by Ca2+-free medium or C

Differential roles of NaV1.2 and NaV1.6 in neocortical pyramidal cell excitability.

Mature neocortical pyramidal cells functionally express two sodium channel (NaV) isoforms: NaV1.2 and NaV1.6. These isoforms are differentially localized to pyramidal cell compartments, and as such are thought to contribute to different aspects of neuronal excitability. But determining their precise roles in pyramidal cell excitability has been hampered by a lack of tools that allow for selective, acute block of each isoform individually. Here, we leveraged aryl sulfonamide-based molecule (ASC) inhibitors of NaV channels that exhibit state-dependent block of both NaV1.2 and NaV1.6, along with

Morphological alterations of the pyramidal and stellate cells of the visual cortex in schizophrenia.

Schizophrenia is a severe brain disorder characterized by certain types of delusion, hallucination and thought disorder. Studies have revealed impaired synaptic plasticity and reduced gamma-aminobutyric acid levels of the visual cortex in patients with schizophrenia. While previous work established a critical role for interneurons and cortical connectivity in the generation of hallucinations, the present study set out to examine the morphology of pyramidal cells and interneurons from layers 3 and 4 in the primary visual cortex from schizophrenic brains and to identify any dendritic and spinal

Layer-specific modulation of pyramidal cell excitability by electroconvulsive shock.

Dysregulation of cortical excitability crucially involves in behavioral and cognitive deficits of neurodegenerative and neuropsychiatric diseases. Electroconvulsive shock (ECS) changes neuronal excitability and has been used in the therapy of major depressive disorder and mood disorders. However, the action and the targets of the ECS in the cortical circuits are still poorly understood. Here we show that the ECS differently changes intrinsic properties of pyramidal cells (PCs) among superficial and deep layers. In layer 2/3 PCs, the ECS induced membrane hyperpolarization and the reduction of i

Synaptic density on the axonal tree of a pyramidal cell in the cortex of the mouse.

The aim of this study was to investigate the density and distribution of synapses on the axonal tree of a pyramidal cell in the mouse cortex. The method used was a new variation of Golgi-electron microscopy involving the modification of Colonnier, the photochemical method of Blackstad and the use of phosphotungstic acid. A segment of axon collateral of 87 microns carried 17 synapses, the proximal 27 microns being free of synapses. The main axon was postsynaptic down to a length of 30 microns. On the following 163 microns there were six presynaptic elements. On the proximal parts of four other

Effects of ketamine and propofol on muscarinic plateau potentials in rat neocortical pyramidal cells.

Propofol and ketamine are widely used general anaesthetics, but have different effects on consciousness: propofol gives a deeply unconscious state, with little or no dream reports, whereas vivid dreams are often reported after ketamine anaesthesia. Ketamine is an N-methyl-D-aspartate (NMDA) receptor antagonist, while propofol is a γ-aminobutyric-acid (GABAA) receptor positive allosteric modulator, but these mechanisms do not fully explain how these drugs alter consciousness. Most previous in vitro studies of cellular mechanisms of anaesthetics have used brain slices or neurons in a nearly "com

Altered dendritic morphology of MEC II pyramidal and stellate cells in Rett syndrome mice.

INTRODUCTION: Mutations in the methyl-CpG-binding protein-2 gene (MECP2), which cause Rett syndrome (RTT), disrupt neuronal activity; however, the impact of the MECP2 loss-of-function on the cytoarchitecture of medial entorhinal cortex layer II (MECII) neurons-crucial for spatial memory and learning-remains poorly understood. METHODS: In this study, we utilized Golgi staining and neuron tracing in the Mecp2+/- mouse model of RTT to investigate the pyramidal and stellate cell alterations in MECII. RESULTS AND DISCUSSION: Our findings revealed that pyramidal cells displayed a significant reduc

Dendritic development of hippocampal CA1 pyramidal cells in a neonatal hypoxia-ischemia injury model.

It is believed that neonatal hypoxia-ischemia (HI) brain injury causes neuron loss and brain functional defects. However, the effect of HI brain injury on dendritic development of the remaining pyramidal cells of the hippocampus and the reaction of contralateral hippocampal neurons require further studies. The Morris water maze and Golgi-Cox staining were used to evaluate the learning and memory and dendritic morphology of pyramidal cells. The results of Golgi-Cox staining showed CA1 pyramidal neurons of HI injury models with fewer bifurcations and shorter dendrite length than the naive contro

Neocortical pyramidal cells with axon-carrying basal dendrites: a prominent feature regulated by activity.

Axon-carrying dendrites, AcDs, have been reported in hippocampus to be privileged dendrites which can directly feed synaptic inputs into an axonal output bypassing the soma and escaping somatic inhibition. In non-primate neocortex, 15-20% of the pyramidal cells have basal AcDs and comparable proportions occurred in organotypic cortex cultures. How does an axon happen to emerge from a dendrite? Very immature migrating pyramidal neurons have no basal dendrites but already have an axon. Sprouts were seen to emerge from the very proximal portion of axons suggesting that an AcD cell starts out as a
