New magnetic nanocomposite Fe3O4@Saponin/Cu(II) as an effective recyclable catalyst for the synthesis of aminoalkylnaphthols via Betti reaction.

In this research, a new magnetic nanocomposite Fe3O4@Saponin/Cu(II) based on quillaja saponin was prepared and the catalyst structure was characterized thoroughly using FT-IR, EDS, TGA, XRD, VSM, HR-TEM, SEM, ICP, BET analyzes. The catalyst prepared in the three-component synthesis of several Betti bases, 1-(α-aminoalkyl)naphthols, under environmentally friendly conditions was used. The advantage of this reaction is the high efficiency of the products and the short reaction time. Furthermore, Fe3O4@Saponin/Cu(II) nano-catalyst is recoverable magnetically and is reusable for other processes wit

Designing Adjuvant Formulations to Promote Immunogenicity and Protective Efficacy of Leptospira Immunoglobulin-Like Protein A Subunit Vaccine.

The leptospirosis burden on humans, especially in high-risk occupational groups and livestock, leads to public health and economic problems. Leptospirosis subunit vaccines have been under development and require further improvement to provide complete protection. Adjuvants can be used to enhance the amplitude, quality, and durability of immune responses. Previously, we demonstrated that LMQ adjuvant (neutral liposomes containing monophosphoryl lipid A (MPL) and Quillaja saponaria derived QS21 saponin) promoted protective efficacy of LigAc vaccine against Leptospira challenge. To promote immuno

Evaluation of the Adjuvant Activity of Herniaria glabra Saponin-Based Nanoadjuvants.

The increasing use of subunit and recombinant vaccines has created a demand for safe and effective adjuvants capable of enhancing immune responses. Here, we investigated a saponin-rich fraction isolated from Herniaria glabra as a source of novel nanoparticulate vaccine adjuvants. The saponins spontaneously self-assembled with lipid components to form stable nanocomplexes with favorable physicochemical characteristics. In a murine immunization model using viral glycoprotein antigens, these nanocomplexes significantly enhanced antigen-specific immune responses, increasing antibody titers by 8- t

The influence of steroidal and triterpenoid saponins on monolayer models of the outer leaflets of human erythrocytes, E. coli and S. cerevisiae cell membranes.

The present paper discusses the use of monolayers of lipid mixtures mimicking the composition of biological membranes of bacteria, erythrocyte and yeast in the context of the anti-bacterial, hemolytic and anti-fungal activity of saponins. Saponins are plant-produced glycosidic biosurfactants with either steroidal or triterpenoidal aglycone. In the present study we used digitonin as a representative steroidal saponin (extracted from Digitalis purpurea) and a mixture of triterpenoid saponins from Quillaja saponaria Molina. The effect of saponins was studied first on monolayers consisting of sing

Possible Molecular Mechanisms by Which an Essential Oil Blend from Star Anise, Rosemary, Thyme, and Oregano and Saponins Increase the Performance and Ileal Protein Digestibility of Growing Broilers.

Phytogenic feed additives represent a potential alternative to antibiotics with attributed health and growth-promoting effects. Chickens supplemented with an essential oil blend, a Quillaja saponin blend, or a combination of both phytogenic preparations showed a comprehensively and significantly improved apparent ileal digestibility of crude protein and amino acids compared to control birds. Accordingly, holistic transcriptomic analyses of jejunum and liver samples indicated alterations of macromolecule transporters and processing pathways likely culminating in an increased uptake and metaboli

Susceptibility of Salmonella enterica Isolates from Tomato Farm Environments to Fatty Acids Naturally Found on Tomato Fruit.

Salmonella enterica subsp. enterica can colonize tomato fruit as it interacts with fruit surface compounds. The exometabolome of tomato fruit contains a mixture of compounds, including fatty acids, which could affect Salmonella fitness. Fatty acids detected in fruit exudates were investigated for Salmonella inhibition. Pelargonic, lauric, myristic, palmitic, margaric, stearic, and oleic acids were suspended in water dissolved in dimethyl sulfoxide (DMSO) or emulsified in water and quillaja saponin to assess how bioavailability impacted Salmonella growth. The minimum inhibitory concentrations o

Food-Grade Saponin Extract as an Emulsifier and Immunostimulant in Emulsion-Based Subunit Vaccine for Pigs.

Subunit vaccines consisting of highly purified antigens require the presence of adjuvants to create effective and long-lasting protective immunity. Advances on adjuvant research include designing combination adjuvants which incorporate two or more adjuvants to enhance vaccine efficacy. Previously, an oil-in-water emulsion adjuvant (OW-14) composed of mineral oil and an inexpensive gum Arabic emulsifier has been reported demonstrating enhanced and robust immune responses when used as an adjuvant in swine subunit vaccines. This study presents a modified version of OW-14 prepared with food-grade

Lowering of cholesterol bioaccessibility and serum concentrations by saponins: in vitro and in vivo studies.

Using an in vitro digestion model, we studied the effect of six saponin extracts on the bioaccessibility of cholesterol and saturated fatty acids (SFAs). In the absence of saponins, around 78% of the available cholesterol was solubilized in the simulated intestinal fluids. The addition of two extracts, Quillaja Dry (QD) and Sapindin (SAP), was found to decrease cholesterol bioaccessibility to 19% and 44%, respectively. For both extracts, the main mechanism of this effect is the displacement of cholesterol molecules from the bile salt micelles, leading to formation of cholesterol precipitates t

The Matrix-M™ adjuvant: A critical component of vaccines for the 21st century.

Matrix-M™ adjuvant is a key component of several novel vaccine candidates. The Matrix-M adjuvant consists of two distinct fractions of saponins purified from the Quillaja saponaria Molina tree, combined with cholesterol and phospholipids to form 40-nm open cage-like nanoparticles, achieving potent adjuvanticity with a favorable safety profile. Matrix-M induces early activation of innate immune cells at the injection site and in the draining lymph nodes. This translates into improved magnitude and quality of the antibody response to the antigen, broadened epitope recognition, and the induction

Assessment of the monoterpene, glycidic and triterpene-moieties' contributions to the adjuvant function of the CP05 saponin of Calliandra pulcherrima Benth during vaccination against experimental visceral leishmaniasis.

The CP05 saponin from Calliandra pulcherrima Benth, shows remarkable similarities to the QS21 saponin of Quillaja saponaria Molina. Both shared a monoterpene hydrophobic moiety, a glycidic chain attached to the triterpene C28, and three sugars attached to C3. Different from QS21, the CP05 does not show the aldehyde group in triterpene C4 involved in TH1 response. Balb/c mice were immunized either intact saponin (CP05), the monoterpene-deprived (BS), the C28 carbohydrate-deprived (HS) or the sapogenin fraction, in formulation with the FML antigen of Leishmania donovani and challenged with 2 x 1

Evaluation of adjuvant activity of Astragaloside VII and its combination with different immunostimulating agents in Newcastle Disease vaccine.

Astragaloside VII (AST-VII), a major cycloartane saponin isolated from Turkish Astragalus species, turned out to be one of the most active metabolites demonstrating Th1/Th2 balanced immune response. As Quillaja saponins are extensively used in adjuvant systems, this study made an attempt to improve AST-VII based adjuvant systems by using different immunostimulatory/delivery agents (monophosphoryllipid A (MPL), Astragalus polysaccharide (APS) and squalene) and to induce cellular and humoral immune response against a viral vaccine. For this purpose, Newcastle Disease vaccine (NDV) was chosen as

Enhancement of paracellular transport of heparin disaccharide across Caco-2 cell monolayers.

The enhancement of paracellular transport of heparin disaccharide using several absorption enhancers across Caco-2 cell monolayers was tested. The cytotoxicity of these enhancers was also examined. The enhancing effects by Quillaja saponin, dipotassium glycyrrhizinate, 18beta-glycyrrhetinic acid, sodium caprate and taurine were determined by changes in transepithelial electrical resistance (TEER) and the amount of heparin disaccharide transported across Caco-2 cell monolayers. Among the absorption enhancers, 18beta-glycyrrhetinic acid and taurine decreased TEER and increased the permeability o

Zein Monolayers: Characterization and Interaction with (Bio)surfactants.

Zein is the main protein of corn seeds, which is often employed in food packaging and as a model of keratin. In this study, zein monolayers were deposited from nonconventional solvents: aqueous ethanol and acetic acid, on pure water that was later exchanged for 1% (bio)surfactant solutions: SDS, CTAB, Triton X-100, and the saponin-rich plant extracts of soapwort (Saponaria officinalis L.) and cowherb (Saponaria vaccaria [P. Mill.] Rauschert), as well as Quillaja bark saponins (QBS). The monolayers on pure water could be reversibly compressed up to ∼47 mN/m. On the basis of neutron reflectivity

A designed self-microemulsion delivery system for dihydromyricetin and its dietary intervention effect on high-fat-diet fed mice.

The present study aims to design a self-microemulsion delivery system (d-α-tocopheryl polyethylene glycol 1000 succinate - quillaja saponin) to enhance the absorptivity of dihydromyricetin (DMY-S), and to investigate its dietary intervention effect on high-fat-diet (HFD) fed mice. We find DMY-S can inhibit the increase of body weight and fat mass, preventing non-alcoholic fatty liver disease. Compared to the model group, the abundance of mice intestinal flora is mainly changed in certain bacterial genera of Firmicutes and Bacteroides, including norank_f_Muribaculaceae and Blautia. The result o

QS-21: Structural insights, immunological mechanisms, and prospects for rational design of safer vaccine adjuvants.

QS-21, a refined adjuvant based on saponins, is isolated from the bark of the soap-bark tree (Quillaja saponaria). It has garnered significant interest in the creation of next-generation vaccine adjuvants due to its robust ability to elicit Th1-type and Th2-type immunological responses. Despite its successful incorporation into licensed vaccines such as Shingrix® (herpes zoster) and Mosquirix® (malaria), the widespread application of QS-21 remains constrained by several limitations, including limited natural availability, chemical instability, dose-dependent toxicity, and an incompletely eluci

Induction of delayed-type hypersensitivity and interferon-gamma to Candida albicans and anti-hen-egg white lysozyme antibody as phenotypic markers of enhanced bovine immune response.

Delayed-type hypersensitivity (DTH) is a protective localized cell-mediated immune response (CMIR), primarily against intracellular pathogens. DTH is widely used in research to assess immune responsiveness and has been a valuable diagnostic test in commercial settings. In pigs and other species both antibody (AMIR) and CMIR have been considered as reliable phenotypic markers of selection programs for disease resistance. Therefore in cattle, it was also considered important to find antigen/adjuvant combinations capable of inducing AMIR and CMIR without interfering with diagnostic tests. The obj
