Copper-Mediated Homocoupling of N-propargylcytisine-Synthesis and Spectral Characterization of Novel Cytisine-Based Diyne Dimer.

Cytisine, a naturally occurring alkaloid and partial agonist of nicotinic acetylcholine receptors (nAChRs), has long been used as a smoking cessation aid and serves as the pharmacophore for varenicline. Recent research has expanded its therapeutic scope to neurodegenerative and neurological disorders, motivating the development of new cytisine derivatives. Among these, N-propargylcytisine combines the biological activity of the parent compound with the synthetic versatility of the terminal alkyne group. Herein, we report the synthesis and characterization of N-propargylcytisine, and its symmet

A Pyridine Dearomatization Approach for the Gram Scale Synthesis of (±)-Sparteine.

Both enantiomers of sparteine have suffered from pricing and supply chain variability, which has inspired efforts toward efficient chemical synthesis. Here, we build upon our reported synthesis of the matrine-type lupin alkaloids in order to synthesize (±)-sparteine. Specifically, selective quenching of the cyclization between glutaryl chloride and pyridine with methanol provides a functionalized quinolizidine core that was elaborated to (±)-sparteine in six additional steps on gram scale. This synthesis provides a scalable route to sparteine from inexpensive commodity chemicals utilizing a de

Promising alkaloids and flavonoids compounds as anti-hepatitis c virus agents: a review.

BACKGROUND: Virus infections are presently seen as a major public health problem. Hepatitis C Virus (HCV) is recognized as a "silent killer" because the acute infection has no symptoms, and it develops as a chronic infection that causes hepatocellular carcinoma and liver damage. The World Health Organization (WHO) predicts that between 130-170 million people are estimated to have chronic Hepatitis C. Plants have various phytochemical compounds such as alkaloids and flavonoids that have prominent antiviral effects especially anti-HCV. The current HCV treatment still has limitations related to s

Transcript profiling of a bitter variety of narrow-leafed lupin to discover alkaloid biosynthetic genes.

Lupins (Lupinus spp.) are nitrogen-fixing legumes that accumulate toxic alkaloids in their protein-rich beans. These anti-nutritional compounds belong to the family of quinolizidine alkaloids (QAs), which are of interest to the pharmaceutical and chemical industries. To unleash the potential of lupins as protein crops and as sources of QAs, a thorough understanding of the QA pathway is needed. However, only the first enzyme in the pathway, lysine decarboxylase (LDC), is known. Here, we report the transcriptome of a high-QA variety of narrow-leafed lupin (L. angustifolius), obtained using eight…

Measurements of cadmium levels in relation to tobacco dependence and as a function of cytisine administration.

Cigarette smoking delivers a number of heavy metals, including cadmium (Cd), into the body. Bioaccumulation may result in an increase in pathological consequences over time. The assessment of changes in serum Cd concentrations during the treatment of cigarette dependence with cytisine was performed for the first time. Parameters assessing smoking habits, strength of addiction, and effectiveness of therapy were analyzed. Cd was determined before, during, and after the end of treatment. The serum Cd levels were significantly higher in the smokers than in the nonsmokers. Significant differences i

Electron impact mass spectral study of halogenated N-acetyl- and N-propionylcytisines.

(-)-Cytisine and its derivatives, characterised by high affinity to neuronal nicotinic acetylcholine receptors with specificity for the α4β2 subtype, have been shown to be important probes in central nervous system (CNS) research. Electron impact mass spectral (EI-MS) fragmentations of halogenated derivatives of N-acetylcytisine and N-propionylcytisine have been investigated. Detailed fragmentation pathways have been identified for all significant ions including a few characteristic fragment ions. The principal mass spectral fragmentation routes of iodine and bromine compounds have been determ

[Medicinal plants--concentrators of chromium. The role of chromium in alkaloid metabolism].

Mass screening of medicinal plants of the flora of Russia (196 species) was performed for chromium content. A total of 124 species-chromium concentrators were found, in which the chromium content markedly exceeded the mean values, this excess being 4- to 6-fold in 54 species, 7- to 3-fold in 62 species, and 37- to 114-fold in seven species. The greatest capacity of chromium accumulation was shown for the sand immortelle, foxglove, Alexandrian laurel, Greek valerian, marsh cudweed, adenostilis, and lobelia. These species are considered as potential sources of chromium for correction of its defi

Chiral high-performance liquid chromatographic separations of vinca alkaloid analogues on alpha 1-acid glycoprotein and human serum albumin columns.

Separations of the stereoisomers of a series of tetracyclic and pentacyclic vinca alkaloid analogues having two or three chiral centres were performed on Chiral-AGP and Chiral-HSA high-performance liquid chromatographic columns. Phosphate buffers with pH 5-7 containing 5-35% acetonitrile or 2-propanol were used as mobile phases. The results were in accordance with previous binding data obtained with native AGP and on an HSA-Sepharose column. Whereas on Chiral-AGP the retention of the trans isomers having 1(R),12b(S)-indolo[2,3-a]quinolizidine or the corresponding 3(S),16(R)-eburnane absolute c

Sophoridine induces parthanatos in mouse renal tubular epithelial cells via mitochondrial dysfunction and apoptosis-inducing factor (AIF) translocation.

Sophoridine, a quinolizidine alkaloid from Sophora flavescens, shows therapeutic promise but its nephrotoxicity is poorly understood. This study investigated sophoridine-induced renal toxicity and its mechanisms in vivo (C57BL/6 mice, 45 mg/kg daily i.p. for 30 days) and in vitro (TCMK-1 renal tubular epithelial cells, 0-1.5 mg/mL). Subacute exposure caused severe tubular damage, elevated serum urea nitrogen and creatinine, and widespread downregulation of renal metabolic pathways. Mechanistically, sophoridine induced reactive oxygen species accumulation, ATP depletion, and mitochondrial membr

The therapeutic potential of cytisine and its derivatives in disorders of the central nervous system.

Cytisine is a quinolizidine alkaloid that is widely used in smoking cessation, exerting its neurological and other biological actions primarily through binding to the nicotinic acetylcholine receptor (nAChR). Cytisine binds to the nAChR (herein represented by α4β2) and exhibits distinct receptor recognition characteristics that provide a basis for the structural optimization of its derivatives in terms of affinity and selectivity. An increasing number of studies indicate that cytisine and its derivatives exhibit a variety of neuroprotective and functional benefits in disorders of the central n

Direct formylation of 2-pyridone core of 3-N-methylcytisine via Duff reaction; synthesis of 9-enyl, 9-ynyl and 9-imino derivatives.

The first direct synthesis of 3-N-methyl-9-formylcytisine via electrophylic formylation is described. It is established, that Vilsmeier-Haack and Gatterman variants of this reaction are unsuccessful in the case with 3-substituted (-)-cytisine derivatives, but Duff procedure (with hexamethylenetetramine in trifluoroacetic acid) gives a possibility to obtain the target pseudo aromatic aldehyde with the 69% yield. Convenient precursors for [4 + 2]- or [3 + 2]-cycloaddition reactions are obtained by means of condensation of synthesized 3-N-methyl-9-formylcytisine with acetone, nitromethane and pho
